2,4-dichloro-5-fluorophenyl, 4-fluoro-3-(Phenoxy)phenyl, 4-fluorophen-yl groups are known pharmacophores and can be used in the synthesis of new biologically active molecules. Therefore, 4-amino-6-arylmethyl-3-mercapto-1,2,4-triazin5(4H)-ones 1 are condensed with 3-phenoxy-4-fluoro-benzoic acid, 2,4-dichloro-5-fluorobenzoic acid, and 4-fluorobenzoic acid 2 to give 7-substituted-3-arylmethyl-4H-1,3,4-thiadiazolo[2,3-c]-1,2,4-triazin-4-ones 3 . Phosphorous oxychloride was used as cyclizing agent. All the newly synthesized compounds 3a-l was screened for their antibacterial activities. Most of them showed promising activity in the range of 10 w g/mL concentrations.  相似文献

Summary. A series of dichlorofluorophenyl containing triazolothiadiazoles were obtained by cyclocondensation of triazole with substituted benzoic, aryloxyacetic, and anilinoacetic acids using POCl3 as cyclizing agent. The structures of the newly synthesized compounds were characterized and confirmed by IR, 1H NMR, mass spectra, and elemental analysis. Selected compounds were screened for their antitubercular activity against Mycobacterium tuberculosis. The compound bearing 2,4-dichloro-5-fluorophenyl moiety at position 3, and 6 of the triazolothiadiazole showed excellent activity in vitro primary screening. Compounds with 3-chloro-4-fluorophenyl and 4-fluorophenoxymethyl moieties at position 6 of the triazolothiadiazole showed very good analgesic activity. Triazolothiadiazole with 4-chlorophenyl, 4-fluoro-3-phenoxyphenyl, and 2,4-dichloro-5-fluorophenyl moieties showed excellent antimicrobial activity against the tested strains at 6.25 μg cm−3 concentrations.  相似文献

以4-氨基-5-苯氧甲基-3-巯基-1,2,4-三唑与取代苯氧乙酸为原料,三氯氧磷为溶剂,回流反应, 合成了3,6-二苯氧甲基-1,2,4-三唑并[3,4-b]-1,3,4-噻二唑衍生物。通过IR、1H NMR和元素分析确定了各化合物分子结构,并对其进行了室内毒力测试,结果表明此类化合物具有较好的杀菌、抑菌活性。  相似文献

To discover new 1,2,4-triazole derivatives which may possess significant biological activities, we synthesized a series of novel 6-aryl-3-(D-galactopentitol-1-yl)-7H-1,2,4-triazolo[3,4-b][1,3,4]thiadiazines and 4-(arylmethylidene)amino-5-(D-galactopentitol-1-yl)-3-mercapto-4H-1,2,4-triazoles from 4-amino-3-(D-galactopentitol-1-yl)-5-mercapto-1,2,4-triazole. All the title compounds were characterized by elemental analysis, IR, 1H- and 13C-NMR. Plant growth-regulating activity tests showed that these compounds have remarkable effects on the growth of radish and wheat.  相似文献

The structures of the newly synthesized compounds were characterized and confirmed by IR, 1H NMR, mass spectra, and elemental analysis. Compounds bearing the 4-methylthiophenyl, 3,4-methylenedioxyphenyl, and 2,3,5-trichlorophenyl moiety showed excellent antiinflammatory activity. The antimicrobial screening studies revealed that compounds with 4-anisyl, 4-methylthiophenyl, 3,4-methylenedioxyphenyl, and 2,3,5-trichlorophenyl at position 5 of the arylidenetriazolothiazolidinone moiety showed excellent activity against all tested strains at 6.25 μg cm−3 concentrations.  相似文献

A series of 6-aryl-3-(3-hydroxypropyl)-7H-1,2,4-triazolo[3,4-b][1,3,4]-thiadiazines were synthesized by the reaction of 4-amino-3-(3-hydroxypropyl)-5-mercapto-1,2,4-triazole (1) with substituted omega-haloacetophenones. Their structures were confirmed by elemental analysis, IR, 1H-NMR, and 13C-NMR. The reaction of thiocarbohydrazide with carboxylic acids at the melting temperature allows an improved preparation of the S-substituted 4-amino-3-mercapto-1,2,4-triazole heterocycles. The interaction of ethoxycarbonyl isothiocyanate with semicarbazides or thiosemicarbazides produces 1-ethoxycarbonyl-2-thiobiureas or bithioureas, respectively. The former are cyclisable to 2-ethoxycarbonamido-5-hydroxy-l,3,4-thiadiazole in acid, or to 2(H)-carbamoyl-3-hydroxy-5-alkylthio-1,2,4-triazoles, and thence to 3-hydroxy-5-mercapto-1,2,4-triazole in alkaline media. The ring-closure of the 1-ethoxycarbonylbithioureas proceeds similarly affording, under the influence of acids, 5-amino (or mercapto)-2-ethoxycarbonamido-1,3,4-thiadiazoles. The action of alkali produces compounds derived from 3-hydroxy-5-mercapto-1,2,4-triazole; the isolation, in a selected example, of the 2(H)-phenylthiocarbamoyl-derivative, elucidates the course of this reaction. Hydrazinolysis of 1-ethoxycarbonylbithiourea yields 4-amino-3-hydrazino-5-mercapto-1,2,4-triazole.  相似文献

以1,2,4-三唑为核心结构的杂环分子,因其优良而广泛的生物活性,已成为药物筛选中重要的研究对象。 本文为了研究其构效关系,针对1,2,4-三唑结构中3个位点分别进行不同的结构修饰,并将其与哌嗪和均三嗪分别偶联,首次合成了两类共28个双三唑多杂环目标分子[TM1TM3(af)]和[TM4TM5(ae)]。 为了比较三唑中4-氨基对生物活性的影响,还与化合物6-吗啉-2,4-(5-硫醚基-4-氨基-3-正戊基1,2,4-三唑)-1,3,5-三嗪(C)和6-吡咯-2,4-(5-巯基-4-氨基-3-正戊基-1,2,4-三唑)-1,3,5-三嗪(D)进行了比较。 通过傅里叶变换红外光谱仪(IR)、核磁共振谱(1H NMR)和高分辨率质谱仪(HRMS)等技术手段对28个目标分子进行了结构表征,研究了其对细胞分裂周期25磷酸酯酶B(Cdc25B)的抑制活性。 结果表明,21个目标分子表现出优良的抑制活性,9个目标分子的抑制活性优于阳性参照物,有望成为抗肿瘤药物先导物。  相似文献

The reaction of benzoyl isothiocyanates and methoxycarbonyl isothiocyanate with 4-amino-4,5-dihydro-3-(methylthio)-1,2,4-triazin-5-ones in acetonitrile gave several substituted 4H-1,3,4-thiadiazolo[2,3-c]-1,2,4-triazin-4-ones VIa-h instead of the expected thioureas. In addition, benzoyl isothiocyanate reacted with 4-amino-3-(methylthio)-5-(trifluoromethyl)-4H-1,2,4-triazole to give the benzoyl thiourea IX and with 4-amino-3-mercapto-5-(trifluoromethyl)-4H-1,2,4-triazole to give the bicyclic compound N-[3-(trifluoromethyl)-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazol-6-yl]benzamide IX .  相似文献

7-羟基黄酮与过量1,2-二溴乙烷反应得到7-溴乙氧基黄酮,将其分别与3-取代-4-氨基-1,2,4-三唑-5-硫酮肉桂醛席夫碱、3-取代-4-苯基-5-巯基-1,2,4-三唑、3-(α-萘亚甲基)-5-巯基-1,2,4-三唑及3-巯基-5-氨基-1,2,4-三唑肉桂醛席夫碱反应,得到4类共16个7-三唑硫乙氧基黄酮类衍生物.采用红外光谱(IR)、核磁共振氢谱(1H NMR)、质谱(MS)及元素分析(EA)等方法对化合物的结构进行了确证.测定了目标化合物清除超氧自由基(O-·2)、羟自由基(·OH)和2,2-二苯基-1-苦味酰基自由基(DPPH·)的活性及总还原能力,并测定了其抗菌活性.结果表明,多数化合物在0.5 mg/m L浓度时具有抗DPPH·活性,其中7-(5-苯亚甲基-4-苯基烯丙亚胺基-1,2,4-三唑-3-硫乙氧基)黄酮(1i)活性较强;多数化合物表现了较好的抑菌活性,其中7-(5-苯亚甲基-4-苯基-1,2,4-三唑-3-硫乙氧基)黄酮(2c)对大肠杆菌、金黄色葡萄球菌和黑曲霉均具有较强的抑制作用.  相似文献

微波辐射条件下, 首先由2-苯并呋喃甲酰肼依次与二硫化碳和水合肼反应合成3-(2-苯并呋喃基)-4-氨基-5-巯基- 1,2,4-三唑, 进一步在微波辐射条件下由4-氨基-5-巯基-1,2,4-三唑分别与芳甲酸/芳氧基乙酸、α-溴代苯乙酮及芳醛反应以较高产率制得了相应的1,2,4-三唑并[3,4-b]-1,3,4-噻二唑、1,2,4-三唑并[3,4-b]-1,3,4-噻二嗪及4-芳亚甲基亚胺基-5-巯基-1,2,4-三唑. 产物结构经IR, 1H NMR, MS及元素分析进行了表征.  相似文献

<正>A novel series of 3,6-disubstituted 1,2,4-triazolo[3,4-b]-1,3,4-thiadiazoles were synthesized by the condensation of 4-amino-5- [2-(4-chlorophenoxymethylbenzimidazole)-1-methylene]-3-mercapto-1,2,4-triazole with various(un)substituted aromatic acids in the presence of phosphorous oxychloride.These compounds were investigated for their inhibitory activity to E.coli methionine aminopeptidase(EcMetAP1).Some of the tested compounds showed significant inhibitory activity.  相似文献
