A novel amphiphilic diblock copolymer, consisting of dendronized polymethacrylate‐b‐poly(ethylene oxide), was synthesized via atom transfer radical polymerization; from it, micellelike aggregates of various morphologies, prepared under near‐equilibrium conditions, were studied with transmission electron microscopy and scanning electron microscopy. The effects of various factors on the aggregate morphologies of the amphiphilic copolymer, such as the water content, the copolymer concentration, and the type of common solvent, were investigated systematically. The unique architecture of the block copolymer led to morphological variety and peculiarities such as dendritic and shuttle‐shaped aggregates, which could be attributed to the effective packing of the bulky side chains, that is, another driving force for the aggregates. © 2005 Wiley Periodicals, Inc. A series of novel amphiphilic brush‐dendritic‐linear poly[poly(ethylene glycol) methyl ether methacrylate]‐b‐polyamidoamine‐b‐poly(ε‐caprolactone) copolymers (PPEGMEMA‐b‐Dmb‐PCL) (m = 1, 2, and 3: the generation number of dendron) were synthesized by the combination techniques of click chemistry, atom transfer radical polymerization (ATRP), and ring‐opening polymerization (ROP). The brush‐dendritic copolymers bearing hydrophilic brush PPEGMEMA and hydrophobic dendron polyamidoamine protected by the tert‐butoxycarbonyl (Boc) groups [Dm‐(Boc) (m = 1, 2, and 3)] were for the first time prepared by ATRP of poly(ethylene glycol) methyl ether methacrylate monomer (PEGMEMA) initiated with the dendron initiator, which was prepared from 2′‐azidoethyl‐2‐bromoisobutyrate (AEBIB) and Dm‐(Boc) terminated with a clickable alkyne by click chemistry. Then, the brush‐dendritic copolymers with primary amine groups (PPEGMEMA‐b‐Dm) were obtained from the removal of the protected Boc groups of the brush‐dendritic copolymers in the presence of trifluoroacetic acid. The brush‐dendritic‐linear PPEGMEMA‐b‐Dmb‐PCL copolymers were synthesized from ROP of ε‐caprolactone monomer using PPEGMEMA‐b‐Dm as the macroinitiators and stannous octoate as catalyst in toluene at 130 °C. To the best of our knowledge, this is the first report that integrates hydrophilic brush polymer PPEGMEMA with hydrophobic polyamidoamine (PAMAM) dendron and PCL to form amphiphilic brush‐dendritic‐linear copolymers. The amphiphilic brush‐dendritic‐linear copolymers can self‐assemble into spherical micellar structures in aqueous solution. © 2012 Wiley Periodicals, Inc. In this work, a series of PLGA‐PEG diblock copolymers were synthesized by ring‐opening polymerization of L‐lactide and glycolide using mPEG as macroinitiator and stannous octoate as catalyst. Spherical micelles were obtained from the various copolymers by using co‐solvent evaporation method. The biocompatibility of micelles was evaluated with the aim of assessing their potential in the development of drug delivery systems. Various aspects of biocompatibility were considered, including MTT assay, agar diffusion test, release of cytokines, hemolytic test, dynamic clotting time, protein adsorption in vitro, and zebrafish embryonic compatibility in vivo. The combined results revealed that the micelles present good cytocompatibility and hemocompatibility in vitro. Moreover, the cumulative effects of micelles throughout embryos developing stages have no toxicity in vivo. It is thus concluded that micelles prepared from PLGA‐PEG copolymers present good biocompatibility as potential drug carrier.  相似文献

The poly(ethylene glycol)/poly(2‐(N,N‐dimethylamino)ethyl methacrylate) (PEG/PDMAEMA) double hydrophilic block copolymers were synthesized by atom transfer radical polymerization using mPEG‐Br or Br‐PEG‐Br as macroinitiators. The narrow molecular weight distribution of PEG/PDMAEMA block copolymers was identified by gel permeation chromatography results. The thermosensitivity of PEG/PDMAEMA block copolymers in aqueous solution was revealed to depend significantly on pH, ionic strength, chain structure, and concentration of the block copolymers. By optimizing these factors, the cloud point temperature of PEG/PDMAEMA block copolymers can be limited within body temperature range (30–37 °C), which suggests that PEG/PDMAEMA block copolymers could be a good candidate for drug delivery systems. © 2010 Wiley Periodicals, Inc. Poly(ethylene oxide) (PEO) star polymer with a microgel core was prepared by atom transfer radical poylmerization (ATRP) of divinyl benzene (DVB) with mono‐2‐bromoisobutyryl PEO ester as a macroinitiator. Several factors, such as the feed ratio of DVB to the initiator, type of catalysts, and purity of DVB, play important roles during star formation. The crosslinked poly(divinyl benzene) (PDVB) core was further obtained by the hydrolysis of PEO star to remove PEO arms. Size exclusion chromatography (SEC) traces revealed the bare core has a broad molecular weight distribution. PEO–polystyrene (PS) heteroarm star polymer was synthesized through grafting PS from the core of PEO star by another ATRP of styrene (St) because of the presence of initiating groups in the core inherited from PEO star. Characterizations by SEC, 1H NMR, and DSC revealed the successful preparation of the target star copolymers. Scanning electron microscopy images suggested that PEO–PS heteroarm star can form spherical micelles in water/tetrahydrofuran mixture solvents, which further demonstrated the amphiphilic nature of the star polymer. © 2004 Wiley Periodicals, Inc. Amphiphilic diblock copolymers of polystyrene (PS) and poly(N‐vinylpyrrolidone) (PNVP) were prepared by a combination of ATRP and MADIX. Well‐defined PS with bromine end group was synthesized by ATRP in bulk at 110 °C using (1‐bromoethyl) benzene as an initiator. The Br‐ end group was then converted to xanthate as verified by 1H NMR spectroscopy, elemental analysis, and UV‐spectroscopy. PS‐b‐PNVP copolymers were produced by MADIX of NVP in bulk at 60 °C using PS‐xanthate as a macro‐chain transfer agent and the kinetics of polymerization were investigated. The structures of PS‐b‐PNVP were characterized using GPC and 1H NMR. Amphiphilic PS‐b‐PNVP could form spherical micelles with PS cores and PNVP shells in aqueous solution as confirmed by 1H NMR and laser light scattering (LLS). The values of critical micelle concentration of PS‐b‐PNVP and the average aggregation number of PS‐b‐PNVP in the micelles were measured using pyrene as a probe and static LLS, respectively. The aggregation number increases concomitantly with temperature (10–50 °C), but the hydrodynamic radius of the micelles remains almost constant over the same temperature range, which may indicate shell dehydration at a higher temperature. © 2008 Wiley Periodicals, Inc. A series of peptides of general primary structure (VSSLESK)n (n = 2, 3, 4, 5 and 6) were designed and synthesized by fluorenylmethyloxycarbonyl solid-phase synthesis using a convergent approach. Peptides containing 21, 28, 35 and 42 residues were modified with α-methoxy poly(ethylene glycol) (mPEG; mol. wt. 2000) by reaction of mPEG–succinimidyl carbonate with the α-amino group of the resin-attached protected peptides. The conformation and thermal stability of the peptides and of their AB block copolymers (A is the mPEG block, B the (VSSLESK)n block) in aqueous medium were investigated by circular dichroism, size-exclusion chromatography and by analytical ultracentrifugation. The helicity of peptides increased with increasing length in a cooperative manner. The peptides and mPEG–peptides with 35 and 42 amino acid residues (block copolymers) adopted a two-stranded α-helical coiled-coil conformation in aqueous solution. The presence of the polymer chain in the diblock hybrid copolymers had no disturbing effect with respect to the stability of the α-helical peptide part in these constructs. Moreover, the thermal stability of mPEG-modified 42-peptide was substantially higher than that of the native 42-peptide. Analytical ultracentrifugation data revealed that in phosphate-buffered saline solution (25–200 μM ) the block copolymer mPEG-block-(VSSLESK)6 ( PEG42 ) associated into stable intermolecular coiled-coil dimers.

The synthesis of poly(methyl acrylate)-block-poly(gamma-benzyl-L-glutamate) (PMA-b-PBLG) diblock copolymers, using atom-transfer radical polymerization (ATRP) of methyl acrylate and living polymerization of gamma-benzyl-L-glutamate-N-carboxyanhydride (Glu-NCA) is described. Amido-amidate nickelacycle end groups were incorporated onto amino-terminated poly(methyl acrylates), and the resulting complexes were successfully used as macroinitiators for the growth of polypeptide segments. This method allows the controlled preparation of polypeptide-block-poly(methyl acrylate) diblock architectures with control over polypeptide chain length and without the formation of homopolypeptide contaminants.  相似文献

A series of side‐chain liquid‐crystalline (LC) homopolymers of poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] with different degrees of polymerization were synthesized by atom transfer radical polymerization (ATRP), which were prepared with a wide range of number‐average molecular weights from 5.1 × 103 to 20.6 × 103 with narrow polydispersities of around 1.17. Thermal investigation showed that the homopolymers exhibit two mesophases, a smectic phase, and a nematic phase, and the phase‐transition temperatures of the homopolymers increase clearly with increasing molecular weights. A series of novel LC coil triblock copolymers with narrow polydispersities was synthesized by ATRP, and their thermotropic phase behavior was investigated with differential scanning calorimetry and polarized optical microscopy. The LC coil triblocks were designed to have an LC conformation of poly[6‐(4‐methoxy‐4′‐oxy‐azobenzene) hexyl methacrylate] with a wide range of molecular weights from 3.5 × 103 to 1.7 × 104 and the coil conformation of poly(ethylene glycol) (PEG) (number‐average molecular weight: 6000 or 12,000) segment. Their characterization was investigated with 1H NMR, Fourier transform infrared spectra, and gel permeation chromatography. Triblock copolymers exhibited a crystalline phase, a smectic phase, and a nematic phase. The phase‐transition temperatures from the smectic to nematic phase and from the nematic to isotropic phase increased, and the crystallization of PEG depressed with increasing molecular weight of the LC block. © 2003 Wiley Periodicals, Inc. Novel AB2‐type amphiphilic block copolymers of poly(ethylene glycol) and poly(N‐isopropylacrylamide), PEG‐b‐(PNIPAM)2, were successfully synthesized through single‐electron transfer living radical polymerization (SET‐LRP). A difunctional macroinitiator was prepared by esterification of 2,2‐dichloroacetyl chloride with poly(ethylene glycol) monomethyl ether (PEG). The copolymers were obtained via the SET‐LRP of N‐isopropylacrylamide (NIPAM) with CuCl/tris(2‐(dimethylamino)ethyl)amine (Me6TREN) as catalytic system and DMF/H2O (v/v = 3:1) mixture as solvent. The resulting copolymers were characterized by gel permeation chromatography and 1H NMR. These block copolymers show controllable molecular weights and narrow molecular weight distributions (PDI < 1.15). Their phase transition temperatures and the corresponding enthalpy changes in aqueous solution were measured by differential scanning calorimetry. As a result, the phase transition temperature of PEG44b‐(PNIPAM55)2 is similar to that in the case of PEG44b‐PNIPAM110; however, the corresponding enthalpy change is much lower, indicating the significant influence of the macromolecular architecture on the phase transition. This is the first study into the effect of macromolecular architecture on the phase transition using AB2‐type amphiphilic block copolymer composed of PEG and PNIPAM. © 2009 Wiley Periodicals, Inc. Ring-opening polymerization of D,L-lactide was carried out in the presence of poly(ethylene glycol), using Zn powder as catalyst. The hydroxyl-capped PLA-PEG-PLA triblock copolymers were coupled with adipoyl chloride at different molar ratios under mild conditions. N-Dimethylaminopyridine (DMAP) was used as catalyst of the coupling reaction. The resulting PLA/PEG multiblock copolymers were characterized by various analytical techniques such as IR, 1H NMR, SEC, and DSC. Sol-gel transition properties of the multiblock copolymers were investigated by mechanical rheology. The data showed that the sol-gel transition temperature and the transition modulus increased with increasing molecular weight and the solution concentration of the multiblock copolymers. Using 2‐chloropropionamide derivative of poly(propyleneimine) dendrimer DAB‐dendr‐(NH2)32 (DAB‐32‐Cl) as the macroinitiator, atom transfer radical polymerization of styrene was successfully carried out in DMF medium. The monodisperse poly(propyleneimine)–polystyrene (dendrimer–PSt) particles with diameters smaller than 100 nm could be prepared. The morphology, size, and size distribution of the dendrimer–PSt particles were characterized by transmission electron microscopy (TEM) and photon correlation spectroscopy (PCS). The effects of reaction temperature, the ratio of St/macroinitiator, and reaction time on the size, and size distribution of the dendrimer–PSt nanoparticles were investigated. In a selective solvent (DMF/H2O), polymers can self‐assemble into different aggregate configurations such as regular microsphere and wire‐like thread. © 2008 Wiley Periodicals, Inc. Biodegradable star‐shaped poly(ethylene glycol)‐block‐poly(lactide) copolymers were synthesized by ring‐opening polymerization of lactide, using star poly(ethylene glycol) as an initiator and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature. Two series of three‐ and four‐armed PEG‐PLA copolymers were synthesized and characterized by gel permeation chromatography (GPC) as well as 1H and 13C NMR spectroscopy. The polymerization under the used conditions is very fast, yielding copolymers of controlled molecular weight and tailored molecular architecture. The chemical structure of the copolymers investigated by 1H and 13C NMR indicates the formation of block copolymers. The monomodal profile of molecular weight distribution by GPC provided further evidence of controlled and defined star‐shaped copolymers as well as the absence of cyclic oligomeric species. The effects of copolymer composition and lactide stereochemistry on the physical properties were investigated by GPC and differential scanning calorimetry. For the same PLA chain length, the materials obtained in the case of linear copolymers are more viscous, whereas in the case of star copolymer, solid materials are obtained with reduction in their Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. The copolymer of polystyrene‐block‐poly(ethylene oxide)‐block‐poly (tert‐butyl acrylate) (PS‐b‐PEO‐b‐PtBA) was prepared, the synthesis process involved ring‐opening polymerization (ROP), nitroxide‐mediated polymerization (NMP), and atom transfer radical polymerization (ATRP), and 4‐hydroxyl‐2,2,6,6‐tetramethylpiperidinyl‐1‐oxy (HTEMPO) was used as parent compound. The PEO precursors with α‐hydroxyl‐ω‐2,2,6,6‐tetramethylpiperidinyl‐1‐oxy end groups(TEMPO‐PEO‐OH) were first obtained by ROP of EO using HTEMPO and diphenylmethylpotassium (DPMK) as the coinitiator. The TEMPO at one end of PEO chain mediated the polymerization of St using benzoyl peroxide as initiator. The resultant PS‐b‐PEO‐OH reacted further with 2‐bromoisobutyryl bromide and then initiated the polymerization of tBA in the presence of CuBr and PMDETA by ATRP. The ternary block copolymers PS‐b‐PEO‐b‐PtBA and intermediates were characterized by gel permeation chromatography, Fourier transform infrared, and nuclear magnetic resonance spectroscopy in detail. Differential scanning calorimetry measurements confirmed that the PS‐b‐PEO‐b‐PtBA with PEO as middle block can weaken the interaction between PS and PtBA blocks, the glass transition temperature (Tg) for two blocks were approximate to their corresponding homopolymers comparing with the PEO‐b‐PS‐b‐PtBA with PEO as the first block. © 2008 Wiley Periodicals, Inc. Model diblock copolymers of poly(1,4‐butadiene) (PB) and poly(dimethylsiloxane) (PDMS), PB‐b‐PDMS, were synthesized by the sequential anionic polymerization (high vacuum techniques) of butadiene and hexamethylciclotrisiloxane (D3) in the presence of sec‐BuLi. By homogeneous hydrogenation of PB‐b‐PDMS, the corresponding poly(ethylene) and poly(dimethylsiloxane) block copolymers, PE‐b‐PDMS, were obtained. The synthesized block copolymers were characterized by nuclear magnetic resonance (1H and 13C NMR), size‐exclusion chromatography (SEC), Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), transmission electron microscopy (TEM), and rheology. SEC combined with 1H NMR analysis indicates that the polydispersity index of the samples (Mw/Mn) is low, and that the chemical composition of the copolymers varies from low to medium PDMS content. According to DSC and TGA experiments, the thermal stability of these block copolymers depends on the PDMS content, whereas TEM analysis reveals ordered arrangements of the microphases. The morphologies observed vary from spherical and cylindrical to lamellar domains. This ordered state (even at high temperatures) was further confirmed by small‐amplitude oscillatory shear flow tests. © 2006 Wiley Periodicals, Inc. Poly(ethylene glycol) (PEG) is often used to biocompatibilize surfaces of implantable biomedical devices. Here, block copolymers consisting of PEG and l ‐cysteine‐containing poly(amino acid)s (PAA's) were synthesized as polymeric multianchor systems for the covalent attachment to gold surfaces or surfaces decorated with gold nanoparticles. Amino‐terminated PEG was used as macroinitiator in the ring‐opening polymerization, (ROP), of respective amino acid N‐carboxyanhydrides (NCA's) of l ‐cysteine (l ‐Cys), l ‐glutamate (l ‐Glu), and l ‐lysine (l ‐Lys). The resulting block copolymers formed either diblock copolymers, PEG‐b‐p(l ‐Gluxcol ‐Cysy) or triblock copolymers, PEG‐b‐p(l ‐Glu)xb‐p(l ‐Cys)y. The monomer feed ratio matches the actual copolymer composition, which, together with high yields and a low polydispersity, indicates that the NCA ROP follows a living mechanism. The l ‐Cys repeat units act as anchors to the gold surface or the gold nanoparticles and the l ‐Glu repeat units act as spacers for the reactive l ‐Cys units. Surface analysis by atomic force microscopy revealed that all block copolymers formed homogenous and pin‐hole free surface coatings and the phase separation of mutually immiscible PEG and PAA blocks was observed. A different concept for the biocompatibilization of surfaces was followed when thiol‐terminated p(l ‐Lys) homopolymer was first grafted to the surface and then covalently decorated with HOOC‐CH2‐PEG‐b‐p(Bz‐l ‐Glu) polymeric micelles. © 2013 Wiley Periodicals, Inc. A new series of segmented copolymers were synthesized from poly(ethylene terephthalate) (PET) oligomers and poly(ethylene glycol) (PEG) by a two‐step solution polymerization reaction. PET oligomers were obtained by glycolysis depolymerization. Structural features were defined by infrared and nuclear magnetic resonance (NMR) spectroscopy. The copolymer composition was calculated via 1H NMR spectroscopy. The content of soft PEG segments was higher than that of hard PET segments. A single glass‐transition temperature was detected for all the synthesized segmented copolymers. This observation was found to be independent of the initial PET‐to‐PEG molar ratio.
