We have studied the dynamics of binding of 19F labelled haptens by mouse plasmacytoma antibody MOPC-315, a protein which shows specificity for nitrophenyl haptens. The off-rates for the dissociation of hapten from MOPC-315 proteins (7S, Fab′ and Fv) were determined by the application of several methods including a technique which, in certain cases, allows the direct determination of the rate of exchange of nuclei between magnetically non-equivalent sites without requiring prior knowledge of intrinsic line widths. Used in conjunction with independently determined data on line widths, chemical shifts, and binding affinities, these studies show that the rates for hapten association to, or dissociation from, the intact 7S antibody, the Fab′ fragment, and the Fv fragment of MOPC-315 are essentially the same. They also indicate the presence, probably in the Fv region, of a low affinity (K~ 103 M?1) site(s) for hapten binding. The mobility of the hapten combining site decreases as the size of the protein increases. These rate data, which were determined at relatively high protein concentrations (up to 40 mg ml?1), agree with on-rates determined at much lower protein concentrations (≤1 mg ml?1); we therefore conclude that protein aggregation, if it does occur, does not significantly affect binding in these systems.  相似文献

The design and synthesis of functional chemical derivatives of small organic molecules is usually a key step for the intricate production of a variety of bioconjugates. In this respect, the derivatization site at which the spacer arm is introduced in immunizing conjugates constitutes a highly critical parameter for the generation of high-affinity and selective antibodies. However, due to the usual complexity of the required synthetic procedures, the appropriate comparison of alternative tethering positions has often been neglected. In the present study, meticulous strategies were followed to prepare synthetic derivatives of pyraclostrobin with the same linkers located at diverse rationally-chosen sites. Activity appraisal of antibodies and bioconjugates was carried out by bidimensional competitive direct and indirect immunoassays, and a superior performance of two of the three synthesized haptens was found. Finally, a detailed analysis of the conformations of the target molecule and the synthesized haptens in aqueous solution was done using computer assisted molecular modeling techniques. This study suggested that the lower titers and affinities of one set of antibodies are most probably due to conformational effects of the spacer arm in the immunizing bioconjugate.  相似文献

Five haptens with different spacer-arm attachment sites on the structure of the organophosphorus insecticide fenthion were designed and synthesized. All of the haptens were conjugated with ovalbumin (OVA) for the coating antigen, and three haptens containing all or most of the structure of fenthion were conjugated with bovine serum albumin (BSA) for the immunogen. Six polyclonal antisera were raised against the three BSA conjugates, and 30 antibody/coating conjugate combinations were selected for studies of assay sensitivity and specificity for fenthion. The study revealed the best combination with high sensitivity (I50 of 0.08 ng mL−1) and high assay specificity, which indicated that when structural difference between the analyte and an immunizing hapten is less than that between a coating hapten and the immunizing hapten, a high sensitive enzyme-linked immunosorbent assay (ELISA) in the heterologous system may stand a good chance to be developed. The immunity results showed that heterology in the hapten spacer-arm attachment site of the immunogen could achieve a remarkable improvement in the quantity, sensitivity, and/or specificity of antibody, and that the moiety of an analyte, which is the same as the moiety near/on the immunizing spacer-arm hapten attachment site, contributes greatly to the interaction of antibody and hapten.  相似文献

In our continuing effort to develop effective anti-heroin vaccines as potential medications for the treatment of opioid use disorder, herein we present the design and synthesis of the haptens: 1-AmidoMorHap (1), 1-AmidoMorHap epimer (2), 1 Amido-DihydroMorHap (3), and 1 Amido-DihydroMorHap epimer (4). This is the first report of hydrolytically stable haptenic surrogates of heroin with the attachment site at the C1 position in the 4,5-epoxymorophinan nucleus. We prepared respective tetanus toxoid (TT)–hapten conjugates as heroin vaccine immunogens and evaluated their efficacy in vivo. We showed that all TT–hapten conjugates induced high antibody endpoint titers against the targets but only haptens 2 and 3 can induce protective effects against heroin in vivo. The epimeric analogues of these haptens, 1 and 4, failed to protect mice from the effects of heroin. We also showed that the in vivo efficacy is consistent with the results of the in vitro drug sequestration assay. Attachment of the linker at the C1 position induced antibodies with weak binding to the target drugs. Only TT-2 and TT-3 yielded antibodies that bound heroin and 6-acetyl morphine. None of the TT–hapten conjugates induced antibodies that cross-reacted with morphine, methadone, naloxone, or naltrexone, and only TT-3 interacted weakly with buprenorphine, and that subtle structural difference, especially at the C6 position, can vastly alter the specificity of the induced antibodies. This study is an important contribution in the field of vaccine development against small-molecule targets, providing proof that the chirality at C6 in these epoxymorphinans is a vital key to their effectiveness.  相似文献

A strategy for pinpointing favorable noncovalent interactions between transition states and active sites of biological catalysts is described. This strategy combines high-level quantum mechanical calculations of transition state geometries with an automated docking procedure using AutoDock. By applying this methodology to antibody-catalyzed hydrolyses of aryl esters (by the 48G7, CNJ206, and 17E8 families of antibodies), varying levels of catalysis are explained in terms of specific hydrogen bonding interactions between combining site residues and transition states. Although these families of antibodies were produced in separate experiments by different researchers using related but different haptens, the mechanism of transition state stabilization appears to be highly conserved. Despite being elicited in response to anionic phosphonate haptens, the best catalysts often utilize hydrogen bond acceptors to stabilize transition states. A mutant of antibody CNJ206, designed based on this observation and predicted to be a better catalyst, is proposed. In the case of antibody 48G7, affinity maturation is shown to produce a catalyst that is highly selective for one of two enantiomeric transition states from a nonselective germline precursor.  相似文献

The solar spectra at selected sites over hemispherical, conical and pinnacle plant canopy models has been evaluated with a dosimetric technique. The irradiance at the sites varies by up to a factor of 0.31 compared to the irradiance on a horizontal plane. The biologically effective (UVBE) exposures evaluated with the dosimetric technique at sites over the plant canopy are up to 19% of that on a horizontal plane. Compared to a spectroradiometer, the technique provides a more practicable method of measuring the UVBE exposures at multiple sites over a plant canopy. Usage of a dosimeter at one site to provide the exposures at that site for different sun angles introduces an error of more than 50%. Knowledge of the spectra allowed the UV and UVBE exposures to be calculated at each site along with the exposures to the entire canopies. These were dependent on the sun angle and the canopy shape. For plant damage, the UVBE was a maximum of about 1.4 mJ cm 2 min?1. Compared to the hemispherical canopy, the UVBE exposure for generalized plant damage was 45% less for the pinnacle canopy and 23% less for the conical canopy. The canopy exposures could not be determined from measurements of the ambient exposure.  相似文献

A simple synthetic method for haptens of organophosphorus (OP) pesticides with a spacer arm (aminocarboxylic acid) attached at the pesticide thiophosphate group was developed. While the previous synthetic approach for this type of haptens requires seven steps, the present method involves only two steps. Using this method, four haptens of the OP insecticide parathion-methyl were synthesized. Rabbits were immunized with either one of the two haptens coupled to bovine serum albumin for production of polyclonal antibodies. Using the serum with the highest specificity, an antigen-coated ELISA was developed, which showed an IC50 of 6.4 ng/ml with a detection limit of 0.2 ng/ml. An antibody-coated ELISA using an enzyme tracer was also developed, which showed an IC50 of 3.5 ng/ml with a detection limit of 0.3 ng/ml. The antibodies showed negligible cross-reactivity with other OP pesticides tested except with the insecticides parathion and paraoxon only in the antigen-coated ELISA.  相似文献

Glutathione peroxidase (GPX) is an important antioxidant enzyme, which plays an important role in scavenging reactive oxygen species. To obtain humanized GPX catalytic antibodies, the phage displayed human antibody library on the surface of the filamentous bacteriophage was used to select novel antibodies by repetitive screening. Phage antibodies B8, H6 and C1 with the GSH-binding site were obtained from the library by enzyme-linked immunosorbent assay (ELISA) analysis with four rounds of selection against three haptens, S-2,4-dinitrophenyl t-butyl ester [GSH-S-DNP-Bu (B)], S-2,4-dinitrophenyl t-hexyl ester [GSH-S-DNP-He (H)] and S-2,4-dinitrophenyl cycle-hexyl ester [GSH-S-DNP-cHe (C)], and characterized using surface plasmon resonance (SPR) biosensor. The gold layer was modified by dithiodiglycolic acid (DDA) and three haptens were easily attached to DDA by self-assembling to form a biosensor membrane. The membrane bounds specifically corresponding antibodies. The kinetic process of the reaction between phage antibodies and their haptens was studied by SPR biosensor. In order to improve selectivity, chemical modification was used to incorporate directly catalytic group selenocysteine (Sec) into selected phage clone B8, H6 and C1 to form Se-B8, Se-H6 and Se-C1, respectively. The GPX activities of Se-B8, Se-H6 and Se-C1 were found to be 3000, 2000 and 700 units/μmol, respectively. Compared with conventional ELISA analysis, the proposed method based on SPR biosensor is much more rapid and simpler.  相似文献

ABSTRACT

A planar array evanescent immunosensor, equipped with a charge-coupled device (CCD) as a detector was used to simultaneously detect pesticides, paraoxon, carbofuran and tetrachloroethylene. Wells approximately 2.5 mm in diameter were formed on glass slides using a photoactivated optical adhesive. Coating haptens were covalently attached to the bottoms of the circular wells to form a sensing surface. The detection of the hapten was completed through competitive immuno-reaction between the coasting haptens, and haptens in solution, with the corresponding antibody in solution. The identification and amount of antibody bound at each location on the slide were determined by quantitative image analysis. Concentrations as low as 0.5ng/mL of paraoxon, 0.Ing/mL of carbofuran, and 0.2ng/mL of tetrachloroethlene could be measured.  相似文献

应用5-参数Morse势方法模拟了O-Ni表面相互作用势,考察了氧原子在镍三个低指数表面的吸附特性.同时构造了羟基与Ni(100)、Ni(110)和Ni(111)表面相互作用的推广LEPS势,获得了羟基在表面的吸附位、吸附几何、结合能及本征振动等数据.理论结果表明,羟基垂直吸附于镍表面的高对称位是稳定的,垂直吸附于Ni(100)表面4-重洞位的吸附能为96.98 kJ•mol-1,垂直吸附于Ni(111)表面3-重洞位的吸附能为96.00 kJ•mol-1,在Ni(110)表面存在两种吸附态:垂直吸附于长桥位的吸附能为99.38 kJ•mol-1,倾斜14°吸附于赝势三重位吸附能为96.98 kJ•mol-1.理论结果与实验结果符合得较好.  相似文献

A good strategy was brought forward for designing efficient haptens and complete antigens for 3-amino-2-oxazolidinone (AOZ). Haptens designed newly were achieved facilely in good yield by using LiCI-N(Et)_3 as new catalysis system,the structure of which was elucidated by spectroscopy analysis,such as NMR and MS.Novel antigens for AOZ were prepared successfully by convenient active ester method.The ratios of haptens 3 and 4 to carrier proteins were proven respectively as 41:1(5a),39:1(6a),11:1(5b) and 9:1(6b) by trinitrobenzene sulfonic acid(TNBS) method.The results of indirect competitive ELISA (ic-ELISA) of antiserums indicated that the haptens with a short unsaturated side chain can evoke specific immune response effectively.  相似文献

A direct competitive ELISA using an enzyme tracer was also developed, which showed an IC50 of 5.6 ng/mL with a detection limit of 0.4 ng/mL. The antibodies in both assays showed negligible cross-reactivity with other OP pesticides except with the insecticide parathion-methyl only in the direct ELISA. Recoveries of fenitrothion from fortified lettuce and rice samples ranged 84-116 and 100-121%, respectively.  相似文献

Crystals of Zn2+/Mn2+ yeast enolase with the inhibitor PhAH (phosphonoacetohydroxamate) were grown under conditions with a slight preference for binding of Zn2+ at the higher affinity site, site I. The structure of the Zn2+/Mn2+-PhAH complex was solved at a resolution of 1.54 A, and the two catalytic metal binding sites, I and II, show only subtle displacement compared to that of the corresponding complex with the native Mg2+ ions. Low-temperature echo-detected high-field (W-band, 95 GHz) EPR (electron paramagnetic resonance) and 1H ENDOR (electron-nuclear double resonance) were carried out on a single crystal, and rotation patterns were acquired in two perpendicular planes. Analysis of the rotation patterns resolved a total of six Mn2+ sites, four symmetry-related sites of one type and two out of the four of the other type. The observation of two chemically inequivalent Mn2+ sites shows that Mn2+ ions populate both sites I and II and the zero-field splitting (ZFS) tensors of the Mn2+ in the two sites were determined. The Mn2+ site with the larger D value was assigned to site I based on the 1H ENDOR spectra, which identified the relevant water ligands. This assignment is consistent with the seemingly larger deviation of site I from octahedral symmetry, compared to that of site II. The ENDOR results gave the coordinates of the protons of two water ligands, and adding them to the crystal structure revealed their involvement in a network of H bonds stabilizing the binding of the metal ions and PhAH. Although specific hyperfine interactions with the inhibitor were not determined, the spectroscopic properties of the Mn2+ in the two sites were consistent with the crystal structure. Density functional theory (DFT) calculations carried out on a cluster representing the catalytic site, with Mn2+ in site I and Zn2+ in site II, and vice versa, gave overestimated D values on the order of the experimental ones, although the larger D value was found for Mn2+ in site II rather than in site I. This discrepancy was attributed to the high sensitivity of the ZFS parameters to the Mn-O bond lengths and orientations, such that small, but significant, differences between the optimized and crystal structures alter the ZFS considerably, well above the difference between the two sites.  相似文献

在25,60和100°C下分别测定了甲醇及二甲醚在SAPO-34分子筛上的吸附等温线,同时用微量热法测定了微分吸附热与覆盖率的关系曲线(量热线),提出了吸附数据需要利用双吸附位Langmuir方程拟合,并获取了相应的吸附参数.对比测得的吸附等温线与量热线发现,在一定压力下,当甲醇及二甲醚在SAPO-34上达到一定吸附量后,随着吸附质分压增加,量热线快速下降,而吸附等温线显示出吸附量仍然继续增加.由此推断,在SAPO-34分子筛上存在两种吸附位——常规吸附位及弱吸附位,其中弱吸附位在高分压下继续吸附.如缺乏量热数据提供的常规吸附位饱和吸附量数据,对吸附等温线进行单吸附位拟合获取吸附参数极易导致错误结果,尤其是当吸附质分压较高时.建议采用双吸附位Langmuir方程,参照量热线提供的常规吸附位的饱和吸附量,通过拟合可以获得两种吸附位的吸附参数.  相似文献

The rapid expansion of structural information for protein-ligand binding sites is potentially an important source of information in structure-based drug design and in understanding ligand cross reactivity and toxicity. We have developed a large database of ligand binding sites extracted automatically from the Protein Data Bank. This has been combined with a method for calculating binding site similarity based on geometric hashing to create a relational database for the retrieval of site similarity and binding site superposition. It contains an all-against-all comparison of binding sites and holds known protein-ligand binding sites, which are made accessible to data mining. Here we demonstrate its utility in two structure-based applications: in determining site similarity and in aiding the derivation of a receptor-based pharmacophore model. A competitive chemiluminescence immunoassay which can be used for point-of-care testing and blood screening of metoprolol is reported. Four haptens for metoprolol were synthesized. An octanedioic acid-modified hapten was conjugated with bovine serum albumin to serve as the immunogen and the haptens were conjugated with ovalbumin for the coating antigen. Polyclonal antibodies for metoprolol were produced and the detection conditions were optimized. A competitive chemiluminescence immunoassay was established based on the produced antibody with potential for bedside therapeutic monitoring of metoprolol. The limit of detection in phosphate-buffered saline was 2?ng?mL?1. Satisfactory recovery values from 89.3 to 107.6% in plasma were achieved. The results provided by the reported method were consistent with values obtained by high-performance liquid chromatography for pharmacokinetic studies. The immunoassay has potential to be developed as a test kit offering a simple and cost-effective approach for on-site monitoring of metoprolol.  相似文献

The adsorption energetics of NO and CO on Pt(111) are studied using an ab initio embedding theory. The Pt(111) surface is modeled as a three-layer, 28-atom cluster with the Pt atoms fixed at bulk lattice sites. Molecular NO is adsorbed at high symmetry sites on Pt(111), with the fcc threefold site energetically more favorable than the hcp threefold and bridge sites. The calculated adsorption energy at the fcc threefold site is 1.90 eV, with an N-surface distance of 1.23 Å. The NO molecular axis is perpendicular to the Pt(111) surface. Tilting the O atom away from the surface normal destablizes adsorbed NO at all adsorption sites considered. On-top Pt adsorption has been ruled out. The Pt(111) potential surface is very flat for CO adsorption, and the diffusion barriers from hcp to fcc sites are 0.03 eV and less than 0.06 eV across the bridge and the atop sites, respectively. Calculated adsorption energies are 1.67, 1.54, 1.51, and 1.60 eV at the fcc threefold, hcp threefold, bridge, and atop sites, respectively. Calculated C-surface distances are 1.24 Å at the fcc threefold site and 1.83 Å at the atop site. It is concluded that NO and CO adsorption energetics and geometries are different on Pt(111).  相似文献

Summary Instrumental neutron activation analysis (INAA) has been employed to investigate the relationship between three ancient occupation sites — Samaru-west, Pantaki and Tsauni in Kaduna State, Nigeria, based on the analysis of cultural deposits at the sites. Samaru-west was found to be related with Pantaki site but not so with the Tsauni site. Elemental profiles for the pottery excavated from each site have been established.  相似文献
