2,4-Dihydro-5-methyl-2-phenyl-4-(9H-xanthen-9-yl)-3H-pyrazol-3-one ( 3 ) was prepared by the condensation of phenylhydrazine and ethyl α-acetyl-9H-xanthene-9-acetate ( 2 ), or 9H-xanthen-9-ol ( 1 ) and 2,4-dihydro-5-methyl-2-phenyl-3H-pyrazol-3-one ( 4 ). 5-Amino-2,4-dihydro-2-phenyl-4-(9H-xanthen-9-yl)-3H-pyrazol-3-one ( 6 ) was obtained by the condensation of 1 and 5-amino-2,4-dihydro-2-phenyl-3H-pyrazol-3-one ( 5 ). 相似文献

2-(9H-Xanthen-9-ylmethyl)-1H-benzimidazole ( 2a ) was prepared by condensing 9H-xanthene-9-acetic acid ( 1a ) with 1,2-benzenediamine. Similarly, 2-(9H-thioxanthen-9-ylmethyl)-1H-benzimidazole ( 2b ) and its S,S-dioxide ( 2d ) were obtained. Compound 2d was also prepared by oxidizing 2b with hydrogen peroxide in acetic acid. Heating of 9H-thioxanthene-9-acetic acid 10-oxide ( 1c ) with 1,2-benzenediamine gave 9-methylene-9H-thioxanthene ( 3 ). 2-(9H-Thioxanthen-9-ylmethyl)-1H-benzimidazole S-oxide ( 2c ) was obtained by oxidizing 2b with m-chloroperbenzoic acid in acetone. 相似文献

A series of new 1,4-pentadien-3-one derivatives containing 1,2,4-triazole moiety were synthesized. The structures of the synthesized compounds were charactered via 1H NMR, 13C NMR and HRMS. Antibacterial bioassays indicated that some of compounds showed potential antibacterial activities against Ralstonia solanacearum (Rs), Xanthomonas oryzae pv. Oryzae (Xoo) and Xanthomonas axonopodis pv. Citri (Xac). Compounds F8 and F17 showed good in vitro antibacterial activities against Rs, with the EC50 values of 18.6 and 18.6 μg/mL, respectively, which were better than commercial agent bismerthiazol (55.2 μg/mL). Furthermore, compounds F12 and F15 showed good in vitro antibacterial activities against Xoo, with the EC50 values of 10.9 and 17.5 μg/mL, which were better than commercial agent bismerthiazol (69.3 μg/mL). Moreover, compounds F2, F9, F16 and F17 showed good in vitro antibacterial activities against Xac, with the EC50 values of 6.6, 5.4, 7.5 and 7.8 μg/mL, respectively, which were better than commercial agent bismerthiazol (54.9 μg/mL). The effect of compound F9 on Xac bacterial cell membrane rupture was observed by scanning electron microscopy (SEM). In addition, antiviral bioassays indicated that some of compounds showed excellent protection activities against tobacco mosaic virus (TMV). Compounds F5 and F15 showed good protecting activity against TMV, with the EC50 values of 108.3 and 105.4 μg/mL, respectively, which were better than commercial agent ningnanmycin (214.7 μg/mL). Microscale thermophoresis (MST) also showed that the binding of compound F2 to TMV coat protein (TMV-CP) yielded a Kd value of 1.260 ± 0.654 μmol/L, which was very close to ningnanmycin (1.058 ± 0.286 μmol/L). Similarly, the molecular docking studies for F2 and F5 with TMV-CP (PDB code: 1EI7, ID: 4QGH) indicated that compounds F2 and F5 had partially interacted with TMV-CP. 相似文献

Eighteen novel 3-substituted-indolin-2-ones containing chloropyrroles were synthesized and their biological activities were evaluated. The presence of a chlorine atom on the pyrrole ring was crucial to reduce cardiotoxicity. The presence of a 2-(ethyl-amino)ethylcarbamoyl group as a substituent at the C-4' position of the pyrrole enhanced the antitumor activities notably. IC50 values as low as 0.32, 0.67, 1.19 and 1.22 μM were achieved against non-small cell lung cancer (A549), oral epithelial (KB), melanoma (K111) and large cell lung cancer cell lines (NCI-H460), respectively. 相似文献

As a part of a research on anti-inflammatory analgesic compounds 3-methyl substituted pyrazolotria-zolopyrimidin-4-one and pyrazolothiazolopyrimidin-4-one derivatives were prepared by previously reported procedures. A series of novel pyrazole oxime derivatives containing oxazole ring were designed and synthesized. The title compounds were structurally confirmed by 1H NMR, 13C NMR spectra and elemental analyses. Preliminary bioassay results showed that some of the title compounds displayed promising fungicidal activity besides insecticidal and acaricidal activity. Particularly, compound 8c exhibited potent fungicidal activity against cucumber Pseudoperonospora cubensis beyond good insecticidal activity against Aphis craccivora and Nilaparvata lugens. 相似文献

To discover new chemotypes of nematicides with proper toxicological profiles, a series of novel 1,2,3- benzotriazin-4-one derivatives were synthesized and further bioevaluated. The bioassay results showed that most of the synthesized compounds were endowed with moderate to good control efficacy against Meloidogyne incognita at 10.0 mg/L in vivo. Among them, compounds 6k and 6p displayed 100% inhibitory activities at this concentration, which implied that they could be used as lead compounds for promising nematicides. 相似文献

A simple, efficient, and general method has been developed for the synthesis of various 3-alkylquinazolin-4-one derivatives from quinazolin-4-one treated with alkyl bromides under phase transfer catalysis condition. The structures of the compounds were characterized by elemental analysis, IR, (1)H-NMR and (13)C-NMR spectra. trans-Perhydro-1,4-benzoxazepin-3-ones 2a-c were synthesized and transformed to condensed-skeleton perhydro-trans-1,4-benzoxazepines 3a,b , the thiones 4a,b , the urea derivatives 5a,b , and N-acylated compounds 6a-e . Compounds 6b,d were ring-opened by hydrochloric acid in ethanol to yield trans-2-(1-carbethoxyethoxy)-1-acylaminomethylcyclohexane derivatives 7b,d . The 1H- and 13C-nmr investigation and X-ray analysis of 5b and 6c,d proved that the expected N-acylated derivatives were formed and that both rings of the trans anellated compounds have a chair conformation. 相似文献

A novel derivative of 2',4'-bridged nucleic acid, named hydroxamate-bridged nucleic acid (HxNA), containing a six-membered perhydro-1,2-oxazin-3-one ring, was designed and synthesized. The introduction of a carbonyl function along with an N-O linkage in the six-membered bridged structure is the unique structural feature of the novel 2',4'-bridged nucleic acid analogue. The design was carried out to restrict the flexibility of the sugar moiety through the trigonal planarity of carbonyl function, which would improve the properties of the modification. The synthesized monomer was incorporated into oligonucleotides, and their properties were examined. The HxNA-modified oligonucleotides exhibited selectively high affinity toward complementary ssRNA. Furthermore, the nuclease resistance of the HxNA-modified oligonucleotide was found to be higher than that of the corresponding natural and 2',4'-BNA/LNA-modified oligonucleotides. Interestingly, exposure of HxNA modified oligonucleotide to 3'-exonuclease resulted in gradual opening of the bridge, which stopped further digestion. Moreover, ring-opening of only one modification at the 3'-end of the oligonucleotides was observed, even if two or three HxNA modifications were present in the sequence. The results demonstrate the strong potential of the HxNA modification as a switch for the generation of highly nuclease-resistant RNA selective oligonucleotide in situ, which could have potential applications in antisense technology. 相似文献

Reactions of α-ketols with diethyl malonate at the molar ratio 2:1 in the presence of K2CO3 were investigated. The heterospiranes obtained contained a dihydrofuran and a γ-lactone rings. The optimum conditions for the synthesis of heterospiranes were found by varying the ratio of the initial compounds: α-ketoalcohol (or 2-ethoxycarbonyl-, 2-methoxycarbonyl-, 2-cyano-3,4,4-trimethyl-2-buten-4-olides), diethyl malonate, K2CO3, by varying the environment, and also the temperature and the time of the reaction. 相似文献

As a part of systematic investigation of synthesis and biologically active compounds of thiazolidine (TZD) derivatives containing pyrazole ring system, several new pyrazole–TZD derivatives 8a , 8b , 8c , 8d and 9a , 9b , 9c , 9d have been synthesized. Compounds 8a , 8b , 8c , 8d were prepared from N‐substituted TZDs 6a , 6b , 6c , 6d and 1H‐pyrazole‐4‐carboxaldehyde 7 by Knoevenagel‐type reaction. Treatment of 8a , 8b , 8c , 8d with sodium hydride at room temperature caused dimerization reaction to afford the corresponding spirocompounds 9a , 9b , 9c , 9d . All the synthesized compounds were characterized by spectroscopic analysis. In vitro, the synthesized compounds 8a , 8b , 8c , 8d and 9a , 9b , 9c , 9d were tested for their growth inhibitory activity in A549 lung cancer, B16F10 murine melanoma, and HeLa human uterine carcinoma cells and for their differentiation of 3T3‐L1 preadipocytes to adipocytes. The results showed that compound 8c possessed growth inhibitory effect of B16F10 cells (IC50 = 27 μM) and compounds 9c , 9d had induction effect on the differentiation of 3T3‐L1 preadipocytes. J.
