Synthesis of benzo[4,5]phenaleno[1,9-bc]thiophene and benzo[4,5]phenaleno[9,1–6c]thiophene

Two pentacyclic thiophenes, benzo[4,5]phenaleno[1,9–6c]thiophene ( 1 ) and benzo[4,5]phenaleno[9,1-bc]-thiophene (2) were synthesized via the corresponding 3-methylphenanthro[2,1-b]thiophene (7) and 1-methylanthra[2,1–6]thiphene ( 19 ).     

Synthesis of anthra[b]thiophenes and benzo[b]naphtho[d]thiophenes

All isomers of the parent anthra[b]thiophenes and benzo[b]naphtho[d]thiophenes, namely anthra[2,3-b]thio-phene, anthra[2,1-b]thiophene, anthra[1,2-b]thiophene, benzo[b]naphtho[2,3-d]thiophene, benzo[b]naphtho[2,1-d]thiophene and benzo[b]naphtho[1,2-d]thiophene were synthesized using a new procedure.     

Synthesis of benzo[b]phenanthro[d]thiophenes

The synthesis of benzo[b]phenanthro[1,2-d]thiphene ( 1 ), benzo[b]phenanthro[4,3-d]thiophene ( 2 ), benzo-[b]phenanthro[2,1-d]thiophene ( 3 ) and benzo[b]phenanthro[3,4-d]thiophene ( 4 ) from appropriately substituted olefines by photochemical cyclodehydrogenation is described. The photolysis of olefin 9 gave a mixture of 4 and anthra[1,2-b]benzo[d]thiophene ( 5 ).     

Synthesis of phenaleno[1,9-bc]thiophene

The cyclization of 2-acetonylthionaphthalene, prepared from 2-mercaptonaphthalene (1) and chloro-acetone in the presence of sodium hydroxide, with polyphosphoric acid gave 1-methylnaphtho[2,1-b]thiophene (4) in 64% overall yield from 1. By bromination with N-bromosuccinimide, 4 was converted in 40% yield into l-bromomethylnaphtho[2,1-b]thiophene (8) . Treatment of 8 with potassium cyanide in a phase-transfer medium gave l-cyanomethylnaphtho[2,1-b]thiophene (10) in good yield. Compound 10 was reduced to the corresponding aldehyde 11 and then cyclized with polyphosphoric acid to phenaleno[l,9-bc]thiophene (12) in 24% overall yield from 10.     

The synthesis of benzo[b]phenanthro[d]thiophenes and anthra[b]benzo[d]thiophenes

The synthesis of benzo[b]phenanthro[2, 3-d]thiophene ( 5 ), benzo[b]phenanthro[4, 3-d]thiophene ( 6 ), benzo-[b]phenanthro[2, 1-d]thiophene ( 9 ), benzo[b]phenanthro[3, 2-d]thiophene ( 14a ), anthra[1, 2-b]benzo[d]thiophene ( 24 ), anthra[2, 3-b]benzo[d]thiophene ( 29 ) and anthra[2, 1-b]benzo[d]thiophene ( 30 ) is described as well as the preparation of 13-methylbenzo[b]phenanthro[3, 2-d]thiophene ( 14b ).     

Benzannelated analogs of phenanthro[1,2-b]- and [2,1-b]thiophene: Synthesis and structural characterization by two-dimensional NMR and X-ray techniques

Syntheses of benzo[3,4]phenanthro[1,2-b]thiophene, benzo[3,4]phenanthro[2,1-b]thiophene and their 1-methyl analogs are reported as potential constituents of solvent refined coal liquids and for mutagenicity testing. The attempted synthesis of the 13-methyl analogs which gave the 11-methyl isomers is also described. Total assignments of the ¹H- and ¹³C-nmr spectra based on long range optimized heteronuclear protoncarbon two-dimensional chemical shift correlation are reported. Carbon assignments obtained for benzo[3,4]-phenanthro[1,2-b]thiophene using this approach were confirmed with a 125 MHz ¹³C–¹³C INADEQUATE spectrum. X-Ray crystal structures were determined for benzo[3,4]phenanthro[1,2-b]thiophene and 1-methyl-benzo[3,4]phenanthro[2,1-b]thiophene. Both molecules were helically distorted from planarity. Close intramolecular contacts between the bay region H1–H13 and ClMe-H13 of 2.03 and 2.28 Å, respectively, were responsible for the distortions. There were no close intermolecular contacts of <3.5Å. both molecules refined to an R value of <0.05.     

Benzo[b] thiophene derivatives. XIV. Derivatives of naphtho[1,8-c] thiophene

In the course of our efforts to synthesize the sulfur isostere of lysergic acid, we have prepared a number of derivatives of the little known naphtho[1,8-bc]thiophene ring system. Two previously reported compounds, 2-bromo-3,4-dihydro-5-oxo-5H-naphtho[1,8-bc]thiophene and 2-bromo-5-hydroxy-3,4-dihydro-5H-naphtho[1,8-bc] thiophene have been obtained pure for the first time, and the syntheses and spectral data of eighteen new compounds are presented. The new compounds include alcohols, ketones, and halogen derivatives of partially reduced naphtho-[1,8-bc] thiophene, as well as the sulfones of several of these compounds.

The preparation of the benzo[4,5]cyclohepta[1,2-b]pyrazine and benzo[4,5]cyclohepta[1,2-b]quinoxaline systems

Benzo[4,5]cyclohepta[1,2-b]quinoxaIine 2 , benzo[4,5]cyclohepta[1,2-b]pyrazine 3a and benzo[4,5]cyclohepta[1,2-b]quinoxaline 4 were prepared from 4,5-benzotropolone and 1,2-phenylenediamine, ethylenediamine and 1,2-diaminocyclohexane, respectively. Compound 3a was methylated to 3b .     

K-region oxides and imines of benzo[b]phenanthro[2,3-d]thiophene and benzo[b]phenanthro[3,2-d]thiophene

The syntheses of the K-oxides and K-imine derivatives of benzo[b]phenanthro[2,3-d]thiophene and benzo-[b]phenanthro[3,2-d]thiophene are described. The parent hydrocarbons 1 and 2 were oxidized with osmium tetroxide and sodium metaperiodate, and the dialdehydes 12 and 18 so formed, cyclized to the corresponding epoxides 1a,12b-dihydrobenz[b]oxireno[9,10]phenanthro[2,3-d]thiophene ( 7 ) and 1a,12b-dihydrobenz-[b]oxireno[9,10]phenanthro[3,2-d]thiophene ( 13 ). Reaction of the oxiranes with sodium azide gave mixtures of azido-alcohols that, in turn, were transformed to the thiaarene imines 1a,12b-dihydro-1H-benz[b]azirino-[9,10]phenanthro[2,3-d]thiophene ( 8 ) and 1a,12b-dihydro-1H-benz[b]azirino[9,10]phenanthro[3,2-d]thiophene ( 14 ), respectively, with the aid of tri-n-butylphosphine.     

Synthesis of phenanthro[b]thiophenes

All isomers of the parent phenanthro[b]thiophenes, namely, phenanthro[1,2-b]thiophene, phenanthro-[2,1-b]thiophene, phenanthro[2,3-b]thiophene, phenanthro[3,4-b]thiophene, phenanthro[3,4-b]thiophene, phenanthro[4,3-b]thiophene and phenanthro[9,10-b]thiophene have been synthesized.     

Synthesis of pyreno[b]thiophenes

The synthesis of all the isomers of pyreno[b]thiophene namely, pyreno[1,2-b]thiophene, pyreno[2,1-b]thiophene and pyreno[4,5-b]thiophene is described. Their spectral characteristics are also discussed.     

The synthesis of novel polycyclic heterocyclic ring systems via photocyclization. 8 . [1]Benzothieno[2,3-c]naphtho[1,2-h]quinoline and [1]benzothieno[2,3-c]naphtho[1,2-h][1,2,4]triazolo[4,3-a]quinoline

The previously unknown polycyclic heterocyclic ring systems, namely, [1]benzothieno[2,3-c]naphtho[1,2-h]-quinoline and [1]benzothieno[2,3-c]naphtho[1,2-h][1,2,4]triazolo[4,3-a]quinoline were synthesized via photocyclization of 3-chloro-N-(1′-phenanthryl)benzo[b]thiophene-2-carboxamide.     

Synthesis of fluorantheno[1,2-b]thiophene and fluorantheno[3,2-b]thiophene,isosteres of benzofluoranthenes

The total syntheses of fluorantheno[1,2-b]thiophene and fluorantheno[3,2-b]thiophene are reported. The common cyclobutanone intermediate which was obtained by [2 + 2] addition of fluorenylidene ketene with 2,3-dihydrothiophene underwent regioselectively α or β ring opening to lead to linear or angular PAC skeleton molecules under desired conditions. 2,3-Dichloro-5,6-dicyanobenzoquinone was applied for aromatization to achieve the formation of PAC's. These two isosteres exhibit very similar uv/visible spectra to benzo[a] and [b]fluoranthenes respectively. Spectroscopic data used for their structural assignments is also discussed.

Intramolecular cyclization reactions of 3-phenyl-benzo[b]thiophene-2-carbonyl chloride and of 2-phenyl-benzo[b]-thiophene-3-carbonyl chloride gave 6-oxo-6H-benz[b]indeno-[1,2-d]thiophene and 10-oxo-10H-benz[b]indeno[2,1-d]thiophene, respectively.

Synthesis of Benzo[c]thiophene Analogs Containing Benzimidazole,Benzothiazole, and Oxazole

Iodine-mediated cyclization of benzo[c]thiophene aldehyde with 1,2-diphenylamine/2-aminophenylthiol led to the formation of benzimidazole/benzothiazole-incorporated benzo[c]thiophenes. Similarly, reaction of benzo[c]thiophene aldehyde with p-toluenesulfonylmethyl isocyanide (TOSMIC) reagent in the presence of K₂CO₃ as a base furnished oxazole-containing benzo[c]thiophene analogs.     

Quino[1,2-c]quinazolines. I. Synthesis of quino[1,2-c]quinazolinium derivatives and the related indazolo[2,3-a]quinoline derivatives as analogs of the antitumor benzo[c]phenanthridine alkaloids

9,10-Dimethoxy-1,2,3,4,12,13-hexahydro-1-oxoquino[1,2-c]quinazolinium perchlorate, 1,2,3,4,13,24-hexahydro-1-oxo[1,3]dioxolo[4,5-g]quino[1,2-c]quinazolinium perchlorate, 6-methyl-2,3,9,10-tetramethoxyquino-[1,2-c]quinazolinium perchlorate and 2,3-dimethoxy-13-methyl[1,3]dioxolo[4′,5′:6,7]quino[1,2-c]quinazolinium perchlorate were synthesized as analogs of the potent antitumor benzo[c]phenanthridine alkaloids nitidine and fagaronine. The related 2,3,8,9-tetramethoxyindazolo[2,3-a]quinoline and 2,3-dimethoxy[1,3]dioxolo-[4,5-g]indazolo[2,3-a]quinoline were also synthesized. Further, the novel formation of 6,7-dimethoxy-2-(2-ethylamino-4,5-dimethoxyphenyl)quinoline via reductive alkylation with Raney nickel in refluxing ethanol is also reported.

Photochemical synthesis of heteroatom-containing polycyclic aromatic compounds

Anthra[2,1-b]furan, anthra[2,1-b]benzo[d]furan, anthra[2,1-b]thiophene, anthra[1,2-b]thiophene, anthra[2,1-b]benzo[d]thiophene, anthra[2,1-b]pyrrole and naphtho[2,3-c]carbazole derivatives were synthesized in fairly good yields by a one-pot photocycloaddition reaction of 2,3-disubstituted 1,4-naphthoquinone with 1,1-diarylethylene. This is the first reported synthesis of these aromatic compounds.
