通过静态试验比较了不同树脂对速溶茶副产品中茶多酚的吸附和解析特性,筛选出对茶多酚有较好的吸附率和解析率的NKA-9型树脂。动态试验确定了最佳的吸附和解析参数:上样流速为0.8mL/min、上样液温度为60℃、pH为5,洗脱流速为0.9mL/min、乙醇浓度为80%、洗脱剂用量为3BV。经过NKA-9纯化后速溶茶副产品中茶多酚含量达到58.7%,得率为14%。  相似文献

不同树脂对瓜果腐霉代谢产物中除草活性成分的吸附分析   总被引:1,自引:0,他引:1  
利用HPD400、HPD500、HPD600、HPD700、HPD850、ADS-17、D101、DM130大孔吸附树脂对瓜果腐霉培养滤液中除草活性成分的吸附情况进行了研究,通过对树脂吸附后的流出液和洗脱液的浓缩物进行HPLC检测分析,确定了其除草活性成分。实验发现,在所选择的8种树脂中以HPD500、HPD850、HPD600、ADS-17对除草活性成分的吸附能力最强,其中以HPD500和HPD600树脂的10%的乙醇洗脱液中除草活性成分含量最高。综合结果表明,以HPD500树脂作为吸附瓜果腐霉培养滤液中除草活性成分的适宜树脂。研究发现,HPD500树脂对除草活性成分吸附和解吸附的最佳条件是:吸附最适温度为20℃,树脂吸附饱和度为33.75mL/g,较适宜吸附流速为2BV/h,较适宜脱附流速为1BV/h,洗脱剂浓度为10%乙醇,洗脱剂用量为2.5BV。  相似文献

采用大孔吸附树脂分离纯化拳参黄酮,筛选其与VC协同抗氧化最佳活性部位。以静态吸附率和解吸率为指标,从中筛选出富集拳参黄酮的最佳树脂,研究其吸附热力学特性,并以黄酮液p H值、浓度等性能确定其动态吸附、洗脱条件。结果表明,NKA-II大孔吸附树脂对拳参黄酮的吸附可用Freundlich方程进行描述,此过程为优惠吸附过程。NKA-II大孔吸附树脂纯化拳参黄酮的最佳条件为:以p H值为2,0.4296mg/m L黄酮液上样,使用40%、60%、80%、95%乙醇溶液洗脱。通过·DPPH清除实验和IC50mix值、相互作用指数等对各复配物抗氧化性进行评价,其中95%乙醇洗脱部位与VC以1:2复配比组成的复配组,协同抗氧化效果最佳。实验结果说明,拳参黄酮是一种很好的天然抗氧化剂。  相似文献

采用温浸法设计四因素三水平正交试验,对黄芪多糖最佳提取工艺进行了优化,结果表明:四因素对黄芪多糖提取的影响顺序为提取温度>提取次数>料液比>提取时间,提取最佳工艺为:料液比1∶6,提取时间90 min,提取温度100℃时提取3次;采用乙醇沉淀法设计三因素三水平正交实验对其最佳分离工艺进行研究,研究发现:三因素三水平对黄芪多糖分离影响顺序为乙醇浓度>乙醇加入量>沉淀时间,分离的最佳工艺为乙醇浓度为90%,加入量5倍体积,沉淀时间4 h;选用AB-8大孔吸附树脂和聚酰胺为吸附剂,不同浓度乙醇为洗脱剂对黄芪多糖最佳纯化工艺进行了探索,确定了最佳纯化工艺为:AB-8大孔吸附树脂吸附,30%乙醇洗脱.这些条件的确定为黄芪的大规模开发和应用奠定了基础.  相似文献

茶叶提取液经大孔吸附树脂二级阶段洗脱层析分离茶多酚咖啡碱.研究了α酸作咖啡碱洗脱剂的可行性及洗脱效果,解决了酸性溶液中茶多酚及咖啡碱的分析方法问题.确定XDA-1树脂为吸附剂,α酸及85%乙醇二级阶段洗脱层析分离茶多酚咖啡碱的优惠工艺.研究了XAD-7树脂再生α酸回收咖啡碱的工艺条件,形成了全树脂法生产茶多酚咖啡碱的新工艺.所得茶多酚产品纯度可达93.83%,咖啡碱含量为2.78%,收率可达13%.本工艺过程简单,避免使用有毒溶剂,安全无污染.  相似文献

比较了NKA-9、D311、S-8、HPD600、NKA-2、A型、D140和聚酰胺等8种树脂对杜仲中降血压活性成分京尼平甙酸的吸附及脱附性能,从中筛选出吸附率及脱附率均较高的A型树脂进行试验.确定了最佳工艺条件:杜仲皮粉末用50%乙醇水溶液提取后,95%乙醇沉淀,上清液回收乙醇后,用蒸馏水稀释,调节pH=6~9后用A型树脂吸附,15%乙醇溶液洗脱,流速为1.5ml/min,洗脱液浓缩后冷冻干燥得产品.  相似文献

本文采用大孔树脂分离黄酒中的活性多肽,对5种大孔吸附树脂进行了筛选,并对筛选出的DA201-C树脂的吸附解吸条件进行了探究。结果表明:DA201-C具有最佳的吸附率和解吸率,并且在25℃能很好的符合langmuir等温吸附式和Freundlich方程。在上样浓度为3.2mg/mL,洗脱剂为70%乙醇,洗脱流速为1BV/h时,能达到最佳的分离效果,多肽的含量达到66.7%,纯化倍数为9.8。  相似文献

伊枯草菌素A是一种具有代表性的微生物源两性环脂肽类化合物。采用DA201大孔吸附树脂从枯草芽孢杆菌发酵液中分离提取伊枯草菌素A,考察了不同吸附、解吸条件对其动态吸附性能的影响。结果表明,DA201型树脂对发酵液中伊枯草菌素A的饱和吸附容量可达到3.48mg/g;动态吸附中最佳吸附流速为1BV/h,其吸附率可以达到95.56%;采用95%乙醇作为洗脱剂,以1BV/h的流速洗脱,其洗脱率可以达到92.41%;DA201型树脂是一种从发酵液中分离和提取伊枯草菌素A的较理想的吸附剂。  相似文献

筛选6种不同性能的吸附树脂,对比了这些树脂对大豆异黄酮的吸附规律.结果表明,以大豆胚为原料,用乙醇提取,经选择性吸附树脂ADS-22的吸附,再用75%的乙醇洗脱,可得到含量达74.37%大豆异黄酮提取物,表明ADS-22树脂对大豆异黄酮有良好的吸附选择性.  相似文献

采用微波和超声波联合提取技术提取川佛手中的黄酮.通过对大孔吸附树脂种类和洗脱液浓度条件优化,获得了最佳实验参数.在此基础上,选择D101树脂,通过石油醚、水和无水乙醇梯度洗脱对川佛手黄酮粗提物进行了分离纯化,得到川佛手黄酮纯化物纯度为92.65%.将川佛手黄酮粗产物、纯化产物、Vc和芦丁进行了抗氧化比较研究,川佛手黄酮纯化产物对DPPH·和·OH的清除率分别可达到82.22%和69.46%,结果表明川佛手黄酮纯化产物具有较好的清除自由基能力和较强的抗氧化活性.  相似文献

Blackberry polyphenols possess various health-promoting properties. Since they are very sensitive to environmental conditions such as the presence of light, oxygen and high temperatures, the application of such compounds is restricted. Fibers are recognized as efficient carriers of polyphenols and are often used in polyphenols encapsulation. In the present study, the ability of apple fiber to adsorb blackberry juice polyphenols was examined. Freeze-dried apple fiber/blackberry juice complexes were prepared with different amounts of fibers (1%, 2%, 4%, 6%, 8% and 10%) and a constant amount of blackberry juice. Polyphenol profile, antioxidant activity, inhibition of the α-amylase, color parameters, as well as the IR spectra, of the obtained complexes were assessed. The results showed a negative effect of higher amounts of fiber (more than 2%) on the adsorption of polyphenols and the antioxidant activity of complexes. With the proper formulation, apple fibers can serve as polyphenol carriers, and thus the application as novel food additives can be considered.  相似文献

Spiny burs of Castanea mollissima Blume (Chinese chestnut) are usually discarded as industrial waste during post-harvesting processing. The objective of this study was to establish an extraction and isolation procedure for tannins from chestnut burs, and to assess their potential antioxidant activity. Aqueous ethanol solution was used as extraction solvent, and HPD 100 macroporous resin column was applied for isolation. The influence of solvent concentration in the extraction and elution process on extraction yield, tannins and polyphenols content, as well as antioxidant potential, including DPPH and ABTS radical scavenging ability, reducing power ability and cellular antioxidant ability were assessed. In both the extraction and isolation process, 50% aqueous ethanol led to superior total tannins and polyphenols content as well as significantly higher antioxidant activity. In addition, the antioxidant activity and the total tannins content in extracts and fractions had a positive linear correlation, and the predominant components responsible for antioxidant activities were characterized as hydrolysable tannins. To the best of our knowledge, this is the first report on the enrichment of tannins from burs of C. mollissim using macroporous resin chromatography, and to assess the cellular antioxidant activity of them.  相似文献

本文研究了大孔吸附树脂吸附金莲花黄色素的性能.研究结果表明:采用静态吸附法,从X-5、AB-8、D-296R、NKA、S-8 五种吸附树脂中筛选出一种吸附、解吸性能都较为理想的大孔吸附树脂X-5,并对其进行了静态吸附动力学的研究,得出的静态吸附动力学曲线反映了树脂的吸附量随时间的变化关系,其吸附动力学方程符合Langmiur提出的吸附速率方程.采用动态吸附法,得到动态吸附透过曲线和动态解吸曲线,其中采用80%乙醇溶液作为洗脱剂,其动态解吸率为88.79%.  相似文献

White-rot fungus Inonotus obliquus grown in submerged culture produces antioxidative phenolic compounds. In this study, addition of lignocellulosic materials into the liquid culture increased the production and antioxidant activity of extra- and intra-cellular phenolic compounds (EPC and IPC, respectively). The production of EPC and IPC was significantly enhanced by wheat straw (by 151.2 and 45.3 %), sugarcane bagasse (by 106.9 and 26.1 %), and rice straw (by 67.6 and 38.9 %). Both of the EPC and IPC extracts from the three substrates showed a higher hydroxyl and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity than those from the control medium. The highly active polyphenols such as tea catechins of epicatechin-3-gallate (ECG) and epigallocatechin-3-gallate (EGCG), and phelligridin G in the EPC extracts increased by 113.1, 75.0, and 86.3 % in the sugarcane bagasse medium. Davallialactone and inoscavin B in the EPC extracts were generated in large amounts in the lignocellulose media but not found in the control medium. The IPC extract from the wheat straw medium had the highest production of EGCG and ECG (17.6 and 18.1 mg/l). The different enhancement among the materials was attributed to the content and degradation rate of cellulose, hemicellulose, and lignin. The different antioxidant activity of the EPC and IPC extracts was related to their phenolic compositions.  相似文献

A simple and rapid HPLC method for determination of chlorogenic acid (5-O-caffeoylquinic acid) in mate tea extracts was developed and validated. The chromatography used isocratic elution with a mobile phase of aqueous 1.5% acetic acid-methanol (85:15, v/v). The flow rate was 0.8 mL/min and detection by UV at 325 nm. The method showed good selectivity, accuracy, repeatability and robustness, with detection limit of 0.26 mg/L and recovery of 97.76%. The developed method was applied for the determination of chlorogenic acid in mate tea extracts obtained by ethanol extraction and liquid carbon dioxide extraction with ethanol as co-solvent. Different ethanol concentrations were used (40, 50 and 60%, v/v) and liquid CO? extraction was performed at different pressures (50 and 100 bar) and constant temperature (27 ± 1 °C). Significant influence of extraction methods, conditions and solvent polarity on chlorogenic acid content, antioxidant activity and total phenolic and flavonoid content of mate tea extracts was established. The most efficient extraction solvent was liquid CO? with aqueous ethanol (40%) as co-solvent using an extraction pressure of 100 bar.  相似文献
