Recombinant scfv antibodies specific for CYP1A1 and CYP1B1 P450 enzymes were combined with targeted imaging mass spectrometry to simultaneously detect the P450 enzymes present in archived, paraffin-embedded, human breast cancer tissue sections. By using CYP1A1 and CYP1B1 specific scfv, each coupled to a unique reporter molecule (i.e., a mass tag) it was possible to simultaneously detect multiple antigens within a single tissue sample with high sensitivity and specificity using mass spectrometry. The capability of imaging multiple antigens at the same time is a significant advance that overcomes technical barriers encountered when using present day approaches to develop assays that can simultaneously detect more than a single antigen in the same tissue sample.  相似文献

High‐throughput particle counting by a differential resistive pulse sensing method in a microfluidic chip is presented in this paper. A sensitive differential microfluidic sensor with multiple detecting channels and one common reference channel was devised. To test the particle counting performance of this chip, an experimental system which consists of the microfluidic chip, electric resistors, an amplification circuit, a LabView based data acquisition device was developed. The influence of the common reference channel on the S/N of particle detection was investigated. The relationship between the hydraulic pressure drop applied across the detecting channel and the counting throughput was experimentally obtained. The experimental results show that the reference channel designed in this work can improve the S/N by ten times, thus enabling sensitive high‐throughput particle counting. Because of the greatly improved S/N, the sensing gate with a size of 25 × 50 × 10 μm (W × L × H) in our chips can detect and count particles larger than 1.5 μm in diameter. The counting throughput increases with the increase in the flowing velocity of the sample solution. An average throughput of 7140/min under a flow rate of 10 μL/min was achieved. Comparing with other methods, the structure of the chip and particle detecting mechanism reported in this paper is simple and sensitive, and does not have the crosstalking problem. Counting throughput can be adjusted simply by changing the number of the detecting channels.  相似文献

An integrated printed circuit board (PCB) based array sensing chip was developed to simultaneously detect lactate and glucose in mouse serum. The novelty of the chip relies on a concept demonstration of inexpensive high-throughput electronic biochip, a chip design for high signal to noise ratio and high sensitivity by construction of positively charged chitosan/redox polymer Polyvinylimidazole-Os (PVI-Os)/carbon nanotube (CNT) composite sensing platform, in which the positively charged chitosan/PVI-Os is mediator and electrostatically immobilizes the negatively charged enzyme, while CNTs function as an essential cross-linker to network PVI-Os and chitosan due to its negative charged nature. Additional electrodes on the chip with the same sensing layer but without enzymes were prepared to correct the interferences for high specificity. Low detection limits of 0.6 μM and 5 μM were achieved for lactate and glucose, respectively. This work could be extended to inexpensive array sensing chips with high sensitivity, good specificity and high reproducibility for various sensor applications.  相似文献

We demonstrate conical pores etched in tracked glass chips for fabricating patterns at the micrometer scale. Highly fluorescent patterns based on photopolymerization of diacetylene films were formed by irradiating UV light through conical pores called "photo-pens". The properties of photopens were investigated through experiments, finite-difference-time-domain (FDTD) simulations and numerical calculations based on Fresnel equations. We show that the pattern dimensions are easily controlled by adjusting the exposure time. Thus, patterns with a range of dimensions can be fabricated without any need of changes in the pore diameter. Parallel patterning was also demonstrated by simultaneously exposing the films to photons through multiple pores in the chip. Our method provides an inexpensive, versatile, and efficient way for patterning without the use of sophisticated masks.  相似文献

Performing bioassay formats based on enzyme and antibody recognition reactions with a single detection chip remains an unmet challenge owing to the different requirements of such bioassays. Herein, we describe a dual‐marker biosensor chip, integrating enzyme and antibody‐based assays for simultaneous electrochemical measurements of insulin (I) and glucose (G). Simultaneous G/I sensing has been realized by addressing key fabrication and operational challenges associated with the different assay requirements and surface chemistry. The I immunosensor relies on a peroxidase‐labeled sandwich immunoassay, while G is monitored through reaction with glucose oxidase. The dual diabetes biomarker chip offers selective and reproducible detection of picomolar I and millimolar G concentrations in a single microliter sample droplet within less than 30 min, including direct measurements in whole blood and saliva samples. The resulting integrated enzymatic‐immunoassay biosensor chip opens a new realm in point‐of‐care multiplexed biomarker detection.  相似文献

A critical requirement for the successful recognition of multiple analytes is the acquisition of abundant sensing information. However, for this to be achieved requires massive chemical sensors or multiplex materials, which complicates the multianalysis. Thus, there is a need to develop a strategy for the facile and efficient recognition of multiple analytes. Herein, we explore the angle‐dependent structural colors of photonic crystals to provide abundant optical information, thereby generating a rainbow‐color chip to realize the convenient recognition of multiple analytes. By simply using a multiangle analysis method, the monophotonic crystal matrix chip can differentially enhance fluorescence signals over broad spectral ranges, thereby resulting in abundant sensing information for highly efficient multiple analysis. Twelve saccharides with similar structures, as well as saccharides in different concentrations and mixtures, were successfully discriminated.  相似文献

Autoimmune diseases are characterized by the presence of autoantibodies in serum of affected patients. The heterogeneity of autoimmune relevant antigens creates a variety of different antibodies, which requires a simultaneous detection mode. For this reason, we developed a tool for parallelized, label-free, optical detection that accomplishes the characterization of multiple antigen–antibody interactions within a single measurement on a timescale of minutes. Using 11-aminoundecyltrimethoxysilane, we were able to immobilize proteinogenic antigens as well as an amino-functionalized cardiolipin on a glass surface. Assay conditions were optimized for serum measurements with a single spot antigen chip on a single spot 1-λ detection system. Minimized background signal allows a differentiation between patients and healthy controls with a good sensitivity and specificity. Applying polarized imaging reflectometric interference spectroscopy, we evaluated samples from three APS patients and three control subjects for this proof-of-principle and already obtained good results for β2-glycoprotein I and cardiolipin.  相似文献

Cyclodextrins (CDs) have been widely used in host-guest molecular recognition because of their chiral and hydrophobic cavities. For example, β-cyclodextrin (βCD) lodged as a molecular adaptor in protein pores such as α-hemolysin (αHL) is used for stochastic sensing. Here, we have tuned the cavity and overall size of βCD by replacing a single oxygen atom in its ring skeleton by a disulfide unit in two different configurations to both expand our ability to detect analytes and understand the interactions of βCD with protein pores. The three-dimensional structures of the two stereoisomeric CDs have been determined by the combined application of NMR spectroscopy and molecular simulation and show distorted conformations as compared to natural βCD. The interactions of these synthetic βCD analogues with mutant αHL protein pores and guest molecules were studied by single-channel electrical recording. The dissociation rate constants for both disulfide CDs from the mutant pores show ~1000-fold increase as compared to those of unaltered βCD, but are ~10-fold lower than the dissociation rate constants for βCD from wild-type αHL. Both of the skeleton-modified CDs show altered selectivity toward guest molecules. Our approach expands the breadth in sensitivity and diversity of sensing with protein pores and suggests structural parameters useful for CD design, particularly in the creation of asymmetric cavities.  相似文献

In this work, we explored a novel fabrication method to construct Au and Ag electrodes on chip, utilizing the different solubility of gold and silver in different etching solutions. KI-I2 etching solution and 50% HNO3 were chosen to dissolve the metal layers alternatively. Planar electrodes with gold and silver could be simultaneously and accurately patterned on chip using photolithographic technique. The as-prepared electrode could be directly served as integrated three-electrode system for electrochemical measurement. Based on it, a sensing strategy has been carried out using home-made electrochemical sensing (ECS) chip, which depended on the competition of double strand DNA and Hg(II)-mediated T–T base pairs (T-Hg(II)-T). Actually, a mercury specific oligonucleotide (MSO) was immobilized onto the thus-fabricated gold working electrode and employed as the sensing element. Chronocoulometry (CC) was chosen to monitor the differences of surface charge volume and quantify the concentrations of Hg(II) ions with a low detection limit down to 1 nM. Therefore, a facile method to fabricate Au and Ag electrodes has been demonstrated to simplify the production of ECS chip. The ECS chip was finally used for constructing an effective sensing platform for sensitive Hg(II) determination, which held promising potential for designing ECS chip in lab-on-a-chip device or point-of-care diagnosis.  相似文献

A robust, portable chip containing a single protein nanopore would be a significant development in the practical application of stochastic sensing technology. Here, we describe a chip in which a single alpha-hemolysin (alphaHL) pore in a planar phospholipid bilayer is sandwiched between two layers of agarose gel. These encapsulated nanopore chips remain functional after storage for weeks. The detection of the second messenger inositol 1,4,5-trisphosphate (IP3) was demonstrated with a chip containing a genetically engineered alphaHL pore as the sensor element.  相似文献

This paper reports a lab-on-a-chip device that counts the number of bacteria flowing through a microchannel. The bacteria number counting is realized by a microfluidic differential Resistive Pulse Sensor (RPS). By using a single microfluidic channel with two detecting arm channels placed at the two ends of the sensing section, the microfluidic differential RPS can achieve a high signal-to-noise ratio. This method is applied to detect and count bacteria in aqueous solution. The detected RPS signals amplitude for Pseudomonas aeruginosa ranges from 0.05 V to 0.17 V and the signal-to-noise ratio is 5-17. The number rate of the bacteria flowing through the sensing gate per minute is a linear function of the sample concentration. Using this experimentally obtained correlation curve, the concentration of bacteria in the sample solution can be evaluated within several minutes by measuring the number rate of the bacteria flowing through the sensing gate of this microfluidic differential RPS chip. The method described in this paper is simple and automatic, and have wide applications in determining the bacteria and cell concentrations for microbiological and other biological applications.  相似文献

Silicon photonic microring resonators have established their potential for label-free and low-cost biosensing applications. However, the long-term performance of this optical sensing platform requires robust surface modification and biofunctionalization. Herein, we demonstrate a conjugation strategy based on an organophosphonate surface coating and vinyl sulfone linker to biofunctionalize silicon resonators for biomolecular sensing. To validate this method, a series of glycans, including carbohydrates and glycoconjugates, were immobilized on divinyl sulfone (DVS)/organophosphonate-modified microrings and used to characterize carbohydrate-protein and norovirus particle interactions. This biofunctional platform was able to orthogonally detect multiple specific carbohydrate-protein interactions simultaneously. Additionally, the platform was capable of reproducible binding after multiple regenerations by high-salt, high-pH, or low-pH solutions and after 1 month storage in ambient conditions. This remarkable stability and durability of the organophosphonate immobilization strategy will facilitate the application of silicon microring resonators in various sensing conditions, prolong their lifetime, and minimize the cost for storage and delivery; these characteristics are requisite for developing biosensors for point-of-care and distributed diagnostics and other biomedical applications. In addition, the platform demonstrated its ability to characterize carbohydrate-mediated host-virus interactions, providing a facile method for discovering new antiviral agents to prevent infectious disease.  相似文献

In this article we present a novel droplet microfluidic chip enabling amplification-free detection of single pathogenic cells. The device streamlines multiple functionalities to carry out sample digitization, cell lysis, probe-target hybridization for subsequent fluorescent detection. A peptide nucleic acid fluorescence resonance energy transfer probe (PNA beacon) is used to detect 16S rRNA present in pathogenic cells. Initially the sensitivity and quantification abilities of the platform are tested using a synthetic target mimicking the actual expression level of 16S rRNA in single cells. The capability of the device to perform "sample-to-answer" pathogen detection of single cells is demonstrated using E. coli as a model pathogen.  相似文献

Fluorescence cross-correlation spectroscopy (FCCS) is a highly sensitive fluorescence technique with distinct advantages in many bioanalytical applications involving interaction and binding of multiple components. Due to the use of multiple beams, bulk optical FCCS setups require delicate and complex alignment procedures. We demonstrate the first implementation of dual-color FCCS on a planar, integrated optofluidic chip based on liquid-core waveguides that can guide liquid and light simultaneously. In this configuration, the excitation beams are delivered in predefined locations and automatically aligned within the excitation waveguides. We implement two canonical applications of FCCS in the optofluidic lab-on-chip environment: particle colocalization and binding/dissociation dynamics. Colocalization is demonstrated in the detection and discrimination of single-color and double-color fluorescently labeled nanobeads. FCCS in combination with fluorescence resonance energy transfer (FRET) is used to detect the denaturation process of double-stranded DNA at nanomolar concentration.  相似文献

A bioarray chip was developed for the rapid and sensitive detection of the antibody (Ab) in nanoliter volumes of biological fluids. The chip was used to detect the anti-hemagglutinin (anti-HA) Ab from mouse ascites fluids. On the bioarray chip, lanes of antigens (i.e., a 13-amino acid peptide, HA, that is bound by anti-HA Ab) were printed in one dimension on the chip surface. The antigens probed the protein samples (i.e., mouse anti-HA) obtained from biological fluids flowing in channels arranged in the perpendicular dimension. To determine if the detection sensitivity was increased by using different fluorescent labeling methods, two types of labeling reagents were compared, namely AlexaFluor 647-labeled goat anti-mouse Ab (AxIgG), and biotin-labeled anti-mouse Ab (BioIgG) followed by cyanine 5-tagged streptavidin (SA-Cy5). In addition, the assay procedure was shortened by omitting the reduction and blocking steps, while maintaining the sensitivity. In conclusion, a bioarray chip was adopted to detect as low as 10?pM mouse monoclonal antibody (anti-HA) in 500 nL of biological fluids (i.e., corresponding to 5 attomoles of antibody). Our results demonstrate that on-chip detection using this bioarray format is more sensitive and less time-consuming than traditional analyses.  相似文献

Techniques that can effectively separate protein–peptide complexes from free peptides have shown great value in major histocompatibility complex (MHC)–peptide binding studies. However, most of the available techniques are limited to measuring the binding of a single peptide to an MHC molecule. As antigen presentation in vivo involves both endogenous ligands and exogenous antigens, the deconvolution of multiple binding events necessitates the implementation of a more powerful technique. Here we show that capillary electrophoresis coupled to fluorescence detection (CE–FL) can resolve multiple MHC–peptide binding events owing to its superior resolution and the ability to simultaneously monitor multiple emission channels. We utilized CE–FL to investigate competition and displacement of endogenous peptides by an immunogenic gluten peptide for binding to HLA-DQ2. Remarkably, this immunogenic peptide could displace CLIP peptides from the DQ2 binding site at neutral but not acidic pH. This unusual ability of the gluten peptide supports a direct loading mechanism of antigen presentation in extracellular environment, a property that could explain the antigenicity of dietary gluten in celiac disease.  相似文献

Analysis of genetic and functional variability in populations of living cells requires experimental techniques capable of monitoring cellular processes such as cell signaling of many single cells in parallel while offering the possibility to sort interesting cell phenotypes for further investigations. Although flow cytometry is able to sequentially probe and sort thousands of cells per second, dynamic processes cannot be experimentally accessed on single cells due to the sub-second sampling time. Cellular dynamics can be measured by image cytometry of surface-immobilized cells, however, cell sorting is complicated under these conditions due to cell attachment. We here developed a cytometric tool based on refractive multiple optical tweezers combined with microfluidics and optical microscopy. We demonstrate contact-free immobilization of more than 200 yeast cells into a high-density array of optical traps in a microfluidic chip. The cell array could be moved to specific locations of the chip enabling us to expose in a controlled manner the cells to reagents and to analyze the responses of individual cells in a highly parallel format using fluorescence microscopy. We further established a method to sort single cells within the microfluidic device using an additional steerable optical trap. Ratiometric fluorescence imaging of intracellular pH of trapped yeast cells allowed us on the one hand to measure the effect of the trapping laser on the cells' viability and on the other hand to probe the dynamic response of the cells upon glucose sensing.  相似文献

Rapid identification of infectious disease pathogens such as foot-and-mouth disease virus (FMDV) during new outbreaks of disease is of fundamental importance in disease control. SpectroSensTM optical microchip sensors demonstrating rapid, label-free detection of FMDV are presented; these contain multiple high-precision planar Bragg gratings and function as low-cost, robust refractive-index sensors. Sensor selectivity to FMDV is imparted by functionalising the top-surface of specific sensing channels with anti-FMDV monoclonal antibodies (mAbs). Selective binding of cognate antigens within the test sample to surface-immobilised FMDV mAbs results in localised changes in refractive index within specific sensing channels; these antibody-antigen interactions manifest as increases in wavelength of light reflected from the multi-channel sensor chip (light is coupled into and out of the chip via optical fibres). Selective identification of FMDV within minutes of sample introduction has been demonstrated by referenced measurement of changes in sensor reflected wavelength from anti-FMDV channels against sensor controls; simplified ‘snap-shot’ assay data are displayed in the form of a simple yes/no readout using a robust, hand-portable device, with further semi-quantitative information available to the ‘super-user’. The characteristics of the SpectroSensTM multiplexed detection platform highlight its potential for in-field detection of foot-and-mouth disease and prospective expansion into diagnoses of other infectious veterinary diseases.  相似文献

The simultaneous detection of multiple analytes is an important consideration for the advancement of biosensor technology. Currently, few sensor systems possess the capability to accurately and precisely detect multiple antigens. This work presents a simple approach for the functionalization of sensor surfaces suitable for multichannel detection. This approach utilizes self-assembled monolayer (SAM) chemistry to create a nonfouling, functional sensor platform based on biotinylated single-stranded DNA immobilized via a streptavidin bridge to a mixed SAM of biotinylated alkanethiol and oligo(ethylene glycol). Nonspecific binding is minimized with the nonfouling background of the sensor surface. A usable protein chip is generated by applying protein-DNA conjugates which are directed to specific sites on the sensor chip surface by utilizing the specificity of DNA hybridization. The described platform is demonstrated in a custom-built surface plasmon resonance biosensor. The detection capabilities of a sensor using this protein array have been characterized using human chorionic gonadotropin (hCG). The platform shows a higher sensitivity in detection of hCG than that observed using biotinylated antibodies. Results also show excellent specificity in protein immobilization to the proper locations in the array. The vast number of possible DNA sequences combine with the selectivity of base-pairing makes this platform an excellent candidate for a sensor capable of multichannel protein detection.  相似文献
