A method for the identification of fatty acid methyl esters (FAME) based on the analysis of shifts in equivalent chain lengths (ECL) is described. The method is based on two-dimensional retention data achieved on one capillary column. Various temperature and pressure programs are applied on the same cyanopropyl column and the shifts in the ECL values are analysed by multivariate methods. The chain length, number of double bonds, and the double bond positions can be determined with high accuracy. The same procedure is suitable for determination of the number of trans and cis double bonds in trans fatty acids, and for detection of artefacts in fatty acid methyl ester chromatograms.  相似文献

A method has been developed for separation of linolenic acid and its seven isomers by HPLC on a silver-ion-loaded column. The standard 18:3 isomers, isolated from a heated linseed oil or prepared by isomerization of linolenic acid, were converted into phenacyl esters and detected by UV at 238 nm. The use of low temperature (10 °C) combined with a gradient of dichloromethane and methanol enabled separation of all the cis/trans isomers. The peaks were identified by comparison of ECL values with those of a standard mixture, by chromatographing collected HPLC fractions on a polar GC column. HPLC quantification was compared with GC analysis. There was satisfactory agreement between the tow methods. This method could be used for seperation, collection and quantification of 18:3 fatty acids with trans double bonds in different oils and foods.  相似文献

The equivalent chain length (ECL) values of the methyl esters of 83 defined fatty acids (FAs) have been determined by gas chromatog-raphy (GC) on three fused silica DB-WAX and three DB-1 columns. ECL values of further 46 FAs were calculated by different methods. Conditions of chromatography, methods of ECL values calculation and differences between ECL values on individual columns and between trans- and cis-isomers of corresponding FAs are also discussed.  相似文献

In order to get information on the radiolytic changes in 1,2-polybutadiene (1,2-PB) the sol and gel fractions, the conversion of double bonds, the structure and concentration of radicals, the formation of dienes and the formation of gaseous products were measured. In addition, the dose rate dependence and temperature dependence for the conversion of double bonds were determined. G values for double bond conversion depend on molecular weight and range from 20 to 200. G values for crosslinking are about 10. A mechanism for the double bond conversion is proposed which involves initiation by a transformation of the primary radical ion in the vinyl group into a carbonium ion and a radical. This is supported by ESR measurement. Reaction of the carbonium ion with a vinyl group in the same chain gives rise to cyclization, whereas reaction with a vinyl group in a neighboring chain results in crosslinking. A comparison of the G values for conversion of double bonds with the G values for crosslinking shows that the formation of cyclic rings exceeds the formation of crosslinks by a factor of about 10. The corresponding values in 1,4-cis- and 1,4-trans-polybutadiene are much smaller [G(cl) ? 2; G(db) ? 7]. The pendent vinyl groups in 1,2-polybutadiene therefore are more reactive than the vinylidene groups in 1,4-polybutadienes.  相似文献

基于点击化学和重氮盐法的双共价键固定化方法,制备了一种高灵敏、可重复使用的电化学发光(ECL)适体传感器. 该方法以可卡因为分析物,以可卡因适体为分子识别物质,以钌联吡啶衍生物为ECL信号物质. 采用电化学方法在玻碳电极表面重氮化叠氮苯胺,通过点击反应连接炔基功能化的钌联吡啶衍生物标记可卡因适体,获得适体传感器. 该传感器在共反应剂存在下,产生弱的电化学发光信号,可卡因存在下,电化学发光信号增加. 基于此,建立了“信号增强”型检测可卡因的电化学发光分析新方法. 电化学发光信号与可卡因浓度在0.1 nmol·L-1 ~ 100 nmol·L-1范围内呈良好的线性关系,检出限为60 pmol·L-1. 该传感器具有良好的稳定性,可重复多次使用. 该双共价键法在构建ECL传感器方面具有很好的应用前景.  相似文献

Magnetically induced current densities have been extensively utilized to investigate aromaticity. However, an applied external magnetic field induces a current in all chemical bonds. The present work investigates the values and patterns of magnetically induced current densities in open hydrocarbon chains containing two or more CC double bonds, considering the three possibilities for the mutual positions of the double bonds (conjugated, isolated, and consecutive) and comparing the corresponding current patterns. The patterns for chains containing three conjugated double bonds are also compared with the patterns in the aromatic benzene ring. The study highlights different current patterns for the three categories of double-bonds arrangements. The differences can be interpreted in terms of presence or absence of electron delocalization (present only in the case of conjugated double bonds) and, for consecutive double bonds, also in terms of the mutual perpendicularity of the π orbitals of adjacent double bonds.  相似文献

Calculations involving molecular orbitals have appeared in the chemical literature over the past few years, but all have used smalltrans-polyenes. We now report extended Huckel molecular orbital calculations ontrans-polyenes of up to 18 double bonds (36 carbons and 38 hydrogen atoms). Information obtained from these calculations include total energies, bond gaps, and net charges on each atom. Also found is that the band gap approaches 1.41 eV as the number of double bonds approaches infinity. This value is quite close to reported experimental values. Data are also presented for polyenes assuming equal C-C bond lengths.  相似文献

In this paper we report a transparent bipolar electrode based microfluidic chip-electrochemiluminescence (ECL) system for sensitive detection of folate receptors (FR) on cell membranes. This integrated system consists of a poly(dimethylsiloxane) (PDMS) layer containing a microchannel and a glass bottom sheet with indium tin oxide (ITO) strips as bipolar detectors. The ITO strips are fabricated using a PDMS micromold with carbon ink as a protective layer in place of traditional photoresist. The configuration of the bipolar electrode has great influence on the ECL intensity of Ru(bpy)(3)(2+)/tripropylamine(TPA) system. Further studies show that folic acid (FA) can strongly inhibit the ECL of the Ru(bpy)(3)(2+)/TPA system. Based on specific recognition between FA and FR on cell membrane, this microfluidic chip-ECL system is successfully applied for detecting the level of FR on human cervical tumor (HL-60) cells and MEF cells. It is found that the ECL intensity increases with the number of HL-60 cells in the range of 21 to 3.28 × 10(4) cells/mL. The average level of FR on HL-60 cells is calculated to be 8.05 ± 0.75 × 10(-18) mol/cell. While for MEF cells, it shows a much slower ECL increment than HL-60 cells due to the much lower FR level on MEF cells (5.30 ± 0.61 × 10(-19) mol/cell). Moreover, exocytosis of FA after FR mediated endocytosis was observed according to the change of the ECL signal with the incubation time of HL-60 cells in the FA- Ru(bpy)(3)(2+)/TPA system.  相似文献

The hydrogen bonding interactions between cysteine (Cys) and formaldehyde (FA) were studied with density functional theory regarding their geometries, energies, vibrational frequencies, and topological features of the electron density. The quantum theory of atoms in molecules and natural bond orbital analyses were employed to elucidate the interaction characteristics in the Cys‐FA complexes. The intramolecular hydrogen bonds (H‐bonds) formed between the hydroxyl and the N atom of cysteine moiety in some Cys‐FA complexes were strengthened because of the cooperativity. Most of intermolecular H‐bonds involve the O atom of cysteine/FA moiety as proton acceptors, while the strongest H‐bond involves the O atom of FA moiety as proton acceptor, which indicates that FA would rather accept proton than providing one. The H‐bonds formed between the CH group of FA and the S atom of cysteine in some complexes are so weak that no hydrogen bonding interactions exist among them. In most of complexes, the orbital interaction of H‐bond is predominant during the formation of complex. The electron density (ρb) and its Laplace (?2ρb) at the bond critical point significantly correlate with the H‐bond parameter δR, while a linearly relationship between the second‐perturbation energy E(2) and ρb has been found as well. © 2011 Wiley Periodicals, Inc. Crystals of poly-trans-1,4-butadiene of uniform size have been grown from three solvents (n-heptane, methyl isobutyl ketone, and toluene) by using a minimum dissolution temeprature technique. The percentage of double bonds available for reaction in the crystals was determined by epoxidation in suspension; crystal thicknesses were measured by electron microscopy. These values were used to calculate the number of monomer units per fold. The number of available double bonds was found to increase with decreasing molecular weight, evidence for the presence of non-reentrant chains (cilia) at the crystal surfaces. The nature of the chain fold in poly-trans-1,4-butadiene crystals is discussed.  相似文献

A novel electrogenerated chemiluminescence (ECL) aptamer-based biosensing method for the determination of thrombin was developed on basis of a structure-switching ECL-dequenching mechanism. A thiolated ss-DNA capture probe, composing of a ss-DNA sequence to adopt two distinct structures-a DNA double strand with a complementary DNA sequence tagged with a ECL signal producer tris(2,2-bipyridyl)ruthenium derivative and a DNA duplex with a thrombin-binding aptamer tagged with a ECL-quencher ferrocene (FcDNA), was self-assembled onto surface of a gold electrode. In the presence of thrombin, the aptamer sequence prefers to form the aptamer-thrombin complex and the switch of the binding partners occurs in conjunction with the generation of a strong ECL signal owing to the dissociation of FcDNA. The integrated ECL intensity versus the concentration of thrombin was linear in the range from 2.0 × 10−10 M to 2.0 × 10−7 M. The detection limit was 6 × 10−11 M. The relative standard derivation for 2.0 × 10−9 M was 5.7% (n = 7). This work demonstrates that the sensitivity and specificity of ECL aptamer-based method for proteins can be greatly improved using quenching ECL signal producer by a chemical quencher such as ferrocene.  相似文献

Hot electron induced cathodic electrochemiluminescence (ECL) was observed at screen printed carbon electrodes (SPCEs) during pulse polarization. The thin insulating film resulted from the printing inks was found to be suitable for generating hot electrons, which can further be converted to hydrated electrons and induce the subsequent luminescence. Compared with disposable Al/Al2O3 electrode, SPCEs show more stable and reproducible ECL in a wider pH range without background emission. A sensitive ECL method for determination of quercetin is proposed. The detection limit is 8.0×10?10 mol L?1(S/N=3), which is two magnitudes lower than that of common ECL method.  相似文献

LC with atmospheric pressure chemical ionization (ACPI) MS with RP and chiral phase was used for separation of triacylglycerols (TAGs) from yeasts of the genera Candida, Kluyveromyces, Rhodotorula, Saccharomyces, Torulospora, Trichosporon, and Yarrowia. Chiral LC–APCI‐MS is based on using two columns in series packed with a 3,5‐dimethylphenyl carbamate modified β‐cyclodextrin chiral phase. All regioisomers and enantiomers of TAGs containing one to five double bonds were separated. Molecular species of TAGs, i.e. regioisomers and enantiomers, were identified and quantified by MS/MS. Among the 94 identified TAGs, the most abundant were triolein, oleopalmitoleoolein, and dipalmitoleoolein. In strains producing palmitoleic acid in amounts >25% of total fatty acids (FAs), this acid, or unsaturated FA is bound in sn‐1. In strains containing palmitoleic acid at 10–25% total FAs this acid is mainly bound in sn‐3, saturated FA being bound in sn‐1. Strains containing <10% palmitoleic acid form preferentially symmetrical TAGs.  相似文献

Phenolic compounds such as vanillic and p-coumaric acids are pollutants of major concern in the agro-industrial processing, thereby their effective detection in the industrial environment is essential to reduce exposure. Herein, we present the quenching effect of these compounds on the electrochemiluminescence (ECL) of the Ru(bpy)32+/TPrA (TPrA=tri-n-propylamine) system at a disposable screen-printed carbon electrode. Transient ECL profiles are obtained from multiple video frames following 1.2 V application by a smartphone-based ECL sensor. A wide range of detection was achieved using the sensor with limit of detection of 0.26 μM and 0.68 μM for vanillic and p-coumaric acids, respectively. The estimated quenching constants determined that the quenching efficiency of vanillic acid is at least two-fold that of p-coumaric acid under the current detection conditions. The present ECL quenching approach provided an effective method to detect phenolic compounds using a low-cost, portable smartphone-based ECL sensor.  相似文献

Algae from cold water (Canada) and warm water (China) were analyzed for their total lipid content, and for their fatty acid (FA) composition and content. The major findings are that FA from Canadian algae are generally richer in polyunsaturated FA (PUFA), with a higher n-3/n-6 FA ratio, and a higher degree of total unsaturation. The 18 C, 4 double bonds FA (18 : 4 stearidonic acid, morotic acid as synonym) was detected in greater amounts in cold water samples. The high levels of total PUFA, and especially of n-3 FA in Canadian algae, suggests their possible utilizations for nutritional purposes.  相似文献

It is shown thatab initio open-shell SCF calculations yield acceptable results for rotation barriers about double bonds in contrast to more conventional closed-shell SCF calculations. Using basis sets of double zeta+polarization quality the SCF values for the rotation barriers of ethylene and allene are obtained to be 48 and 50 kcal/mole, respectively. An IEPA estimate of the influence of electron correlation leads to values of 64 and 52 kcal/mole, respectively, which are in reasonable agreement with the experimental values.  相似文献

Anodic electrochemiluminescence (ECL) of 3‐mercaptopropionic acid (MPA)‐ capped CdTe/CdS core‐shell quantum dots (QDs) with tripropylamine (TPrA) as the co‐reactant were studied in aqueous (Tris buffer) solution for the first time. The results suggest that the oxidation of TPrA at a glassy carbon electrode (GCE) surface participated in the ECL of QDs, and the onset potential and the intensity of ECL of CdTe/CdS QDs were affected seriously by TPrA, as the co‐reactant, in Tris buffer solution. The onset potential of ECL in this new system was about +0.5 V (vs. Ag/AgCl) and the ECL intensity greatly enhanced when TPrA was present. Various influencing factors, such as the electrolyte, pH, QDs concentration, potential range and scan rates on the ECL were studied. Based on the selective quenching by Cu2+ to the light emission from CdTe/CdS QDs/TPrA system, a highly sensitive and selective method for the determination of Cu2+ was developed. At the optimal conditions, the relative ECL intensity, I0/I, was proportional to the concentration of Cu2+ from 14 nM to 0.21 μM with the detection limit of 6.1 nM based on the signal‐to‐noise ratio of 3. The possible ECL mechanism of QDs and the quenching mechanism of ECL were proposed.  相似文献

A highly selective and sensitive detection method based on tris(2,2′-bipyridyl)ruthenium(II) [Ru(bpy)32+] electrogenerated chemiluminescence (ECL) has been developed for the quantitative determination of β-blockers in both pharmaceutical preparations and human urine samples. The ECL emission is based on the reaction between electro-oxidized Ru(bpy)33+ and the secondary amino groups on the β-blockers. The ECL intensities for the β-blockers were strongly dependent on the pH at which the ECL detections were conducted, with the maximum intensities being obtained at pH 9.0. Under the optimal condition, the detection limit for atenolol and metoprolol were almost 0.5 μM (50 pmol) and 0.08 μM (8 pmol), respectively, with S/N of 3 and a linear working range that extends four orders of magnitude with relative standard deviations below 5% for 10 replicate injected samples. The concentrations of atenolol and metoprolol were determined in pharmaceutical preparations using flow injection analysis with Ru(bpy)32+ ECL detection based on standard addition method. The determined values by the present method showed acceptable agreement with the stated values by manufacturers. The determination of the five β-blockers in human urine samples was performed using HPLC-Ru(bpy)32+ ECL detection. The resulting chromatogram was much simpler than that obtained with HPLC-UV absorbance detection.  相似文献

An ultrasensitive electrochemiluminescence (ECL) immunosensor for the detection of tetrodotoxin (TTX) is proposed, which are composed of the branched poly‐(ethylenimine) (BPEI) functionalized graphene (BGNs)/Fe3O4‐Au magnetic capture probes and luminol‐capped gold nanocomposites (luminol‐AuNPs) as the signal tag. Herein, a typical sandwich immunecomplex was constructed on the glassy carbon electrode. The BGNs/Fe3O4‐Au hybrids could efficiently conjugate primary antibody via the Au−S chemical bonds or Au−N chemical bonds and rapidly separate under external magnetic field. The introduction of BPEI to GO could enhance the luminol‐ECL intensity. Meanwhile, the multifunctional nanocomposites have been proved with good water‐solubility, excellent electron transfer, outstanding stability, etc. The luminescent luminol‐AuNPs, a high efficient electrochemiluminescence marker, can be assembled on the second antibody, which can produce the ECL signal to achieve the determination of TTX. This proposed ECL immunosensor with a linear range from 0.01–100 ng/mL can be applied in the detection of TTX in real samples with satisfactory results.  相似文献
