The title compounds 2-(8-quinoltnoxy)-4H-imidazolin-4-ones (4) were synthesized by means of base catalytic reactions of 8-hydroxyquinoline with each of carbodiimides 2, which were obtained via aza-Wittig reactions of iminophosphoranes 1 with the corresponding aromatic isocyanates.  相似文献

The reactions of diorganotin dichloride [Ph_2SnCl_2, (PhCH_2)_2-SnCl_2 or (n-Bu)_2SnCl_2] with potassium salt of 2,5-dimercapto-4-phenyl-1, 3, 4-thiodiazole gave complexes R_2Sn (S_3N_2C_8H_5)_2(4: R=Ph; 5: R=PhCH_2 and 6: R=n-Bu), respectively.Characterizations were carried out for all complexes by IR, ~1HNMR spectra and X-ray crystallography analysis. Including theSn…N interaction, the three complexes all have six-coordinateddistorted octahedral geometry. The crystal of 2, 4, 6-tristyryl-s-triazine (TSTA) has been prepared and its crystal structure been determined to be in the polar non-centrosymmetric space group Cmc21. The molecular structure of TSTA is characterized by the slightly curved planar configuration and the octupolar C3v molecular symmetry. As exoected. TSTA crvstal shows a quite novel nonlinear optical (NLO) property with its powder second ha~onic generating(SHG) intensity of 1.8 times as that of urea. It also shows excellent transparency (with the peak position of 322 nm in absorption spectrum) and good thermal stability (with the melting point of 225-229℃).  相似文献

A synthetic cationic surfactant, 5,5-ditetradecyl-2-(2-trimethyl-ammonioethyl)-l,3-dioxane bromide (DTDB), was used to construct a supported bilayer lipid membrane (s-BLM) coatedon an underlying glassy carbon electrode (GCE). Electrochemical impedance spectroscopy (EIS), small-angle X-ray diffraction (SAXD) and cyclic voltammetry were used to characterize the s-BLM. Both EIS and SAXD data indicated that the synthetic lipid exists as a well-oriented bilayer in the membrane.The voltammetric study showed that the lipid membrane can open ion channels in the presence of ClO4^- stimulant with Ru(bpy)3^2 as marker ions and give distinct channel currents.The channels can be dosed and open up again many times by removing or introducing ClO4^- anions.  相似文献

A series of 3-pyridinyl-6-aryl-1, 2, 4-triazolo[3, 4-b][1, 3, 4]thiadiazoles(PATT) were prepared, the structures were confirmed by 1R and ^1H NMR spectra. The results of cyclic voltammetry measurements imply that all these compounds have a higher electron affinity (EA) than 2-(4-biphenyl)-5-(4-tert-butyl phenyl)-1, 3. 4-oxadiazole (PBD) which implies that PATT could be acting as better electron acceptors than widely used electron transporting material PBD.  相似文献

HgBa2Can-1CunO2n+2+( (n=1, 2, 3, 4) 1 are tetragonal with space group space P4/mmm. For n=1,2,3, nearly single-phase crystals were obtained, while for n=4, the sample was primarily a mixture of the n=3 and 4 phases. These materials also possessed the highest Tc values yet observed for any superconductors. 3, 3-Dimethyl-1-(1,2,4-triazol)-2-butanone was treated with aqueous formaldehyde to give an additional product, and subsequent elimination by acetic anhydride yielded 4,4-dimethyl-2-(1,2,4-triazol)-1-penten-3-one. Further addition with substituted amines provideda series of (1,2,4-triazol)-4,4-dimethyl-3-pentanone, which were then reduced by KBH4 to obtaina series of (1,2,4-triazol)-4,4-dimethyl-3-pentanol. Their structures were confirmed by ^1HNMR and elemental analysis. The results of bioassay showed that the title products possess good fungicidal activities.  相似文献

Variable benzo[b]furan derivatives having (E)- and (Z)-2-alkylcarbamoyl-1-methylvinyl groups at the 2-, 4- and 5-positions and a carboxylpropoxy or (1-phenyl)ethoxy group at the 7-position were prepared to find novel and selective leukotriene B4(LTB4) receptor antagonists. (E)-2-(2-diethylcarbamoyl-1-methylvinyl)-7-(1-phenylethoxy)benzo[b]furan (4v) showed selective inhibition to the human BLT2 receptor (hBLT2). On the other hand, (E)-2-acetyl-4-(2-diethylcarbamoyl-1-methylvinyl)-7-(1-phenylethoxy)benzo[b]furan (7v) inhibited both human BLT(1) receptor (hBLT1) and hBLT2. The (E)-2-(2-diethylcarbamoyl-1-methylvinyl) group lay on approximately the same plane as the benzo[b]furan ring, whereas the (E)-4-(2-diethylcarbamoyl-1-methylvinyl) group had the torsion angle (45.7 degree) from the benzo[b]furan ring plane. However, the (Z)-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans were inactive. The inhibitory activity depended on the conformation of the 2-diethylcarbamoyl-1-methylvinyl group.  相似文献

(E)-2-Acetyl-4-(2-diethylcarbamoyl-1-methylvinyl)-7-(1-phenylethoxy)benzo[b]furan (4b) with a characteristic conformation and (E)-2-(2-morpholinocarbo-1-methylvinyl)-7-ethoxycarbopropoxybenzo[b]furan ((E)-3b) were prepared and evaluated for their leukotriene B4(LTB4) antagonistic activity. Compound 4b showed potent antagonistic activity against human BLT1 and BLT2 receptors. Compound (E)-3b displayed selective BLT2 receptor antagonistic activity. A series of 3-(4-chlorophenyl)-2-(2-aminothiazol-4-yl)benzo[b]furan derivatives 6-10 were prepared and their leukotriene B(4) inhibitory activity was evaluated. We found that several compounds showed strong inhibition of calcium mobilization in CHO cells overexpressing human BLT(1) and BLT(2) receptors. Among them, 3-(4-chlorophenyl)-2-[5-formyl-2-[(dimethylamino)methyleneamino]thiazol-4-yl]-5-methoxybenzo[b]furan 9b showed the most potent and selective inhibition for the human BLT(2) receptor, and its IC(50) value was smaller than that of the selected positive control compound, ZK-158252.  相似文献

A series of 8'-substituted N-(endo-8-azabicyclo[3.2.1]oct-3-yl)-1-isopropyl-2-oxo-1,2-dihydro-3-quinolinecarboxamides were synthesized. The 5-HT4 receptor agonistic activity was evaluated using the isolated guinea pig ileum preparation. Of the compounds synthesized, N-(endo-8-(3-hydroxypropyl)-8-azabicyclo[3.2.1]oct-3-yl)-1-isopropyl-2-oxo-1,2-dihydro-3-quinolinecarboxamide (15a, TS-951) exhibited the most potent serotonin 5-HT4 receptor agonistic activity. This compound had a high affinity for the serotonin 5-HT4 receptor although it had no affinities for other broad spectrum receptors. Furthermore, it remarkably enhanced gastrointestinal motility in conscious fed dogs without unfavorable effects that non-selective serotonin 5-HT4 receptor agonist has. TS-951 may be useful in improving gastrointestinal dysfunction.  相似文献

Several 2-alkylcarbamoyl-1-alkylvinylbenzo[b]furans were designed to find a selective leukotriene B4 (LTB4) receptor antagonist. 2-(2-Alkylcarbamoyl-1-alkylvinyl)benzo[b]furans having a substituent group at the 3-position, 4-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans having a substituent group at the 3-position, and 7-(2-alkylcarbamoyl-1-methylvinyl)benzo[b]furans and 3-(2-alkylcarbamoyl-1-alkylvinyl)benzo[b]furans were prepared and evaluated for LTB4 receptor (BLT1 and BLT2) inhibitory activities. (E)-3-Amino-4-[2-[2-(3,4-dimethoxyphenyl)ethylcarbamoyl]-1-methylvinyl]benzo[b]furan ((E)-17c) showed potent and selective inhibitory activity for BLT2. On the other hand, (E)-7-(2-diethylcarbamoyl-1-methylvinyl)benzo[b]furan ((E)-27a) showed potent inhibitory activity for both BLT1 and BLT2.  相似文献

以间氯苯胺与乙氧基亚甲基丙二酸二乙酯(EMME)为原料, 经缩合、 高温环合、 水解、 氯代和亲核取代等反应设计并合成了16个3位为(4'-吗啉)-羰基或乙氧羰基的4-苯氨基喹啉化合物(Ⅰ1~Ⅰ10, Ⅱ1~Ⅱ6). 目标化合物结构经MS及1H NMR确证. 以MTT法, 采用表皮生长因子受体(EGFRs)高表达的人癌细胞(HeLa, HepG2, BGC-823)对目标化合物进行活性测试, 结果表明, 该类化合物对HeLa, HepG2和BGC-823细胞增殖具有一定程度的抑制作用. 其中化合物Ⅱ4~Ⅱ6 对 HepG2 细胞抑制作用较强, 化合物Ⅱ1和Ⅱ5对BGC-823细胞抑制作用较强. 初步探讨了化合物结构与生物活性的关系.  相似文献

The stereochemical diversity-oriented conformational restriction strategy can be an efficient method for developing specific ligands for drug target proteins. To develop potent histamine H(3) and/or H(4) receptor ligands, a series of conformationally restricted analogs of histamine with a chiral trans- or cis-4-amino-2,3-methano-1-(1H-imidazol-4-yl)butane structure was designed based on this strategy. These stereochemically diverse compounds were synthesized from previously developed versatile chiral cyclopropane units. Among these analogs, a trans-cyclopropane-type compound, (2S,3R)-4-(4-chlorobenzylamino)-2,3-methano-1-(1H-imidazol-4-yl)butane (5b), has remarkable antagonistic activity to both the H(3) (K(i) = 4.4 nM) and H(4) (K(i) = 5.5 nM) receptors, and a cis-cyclopropane-type compound, (2R,3R)-4-amino-2,3-methano-1-(1H-imidazol-4-yl)butane (6a), is a potent and selective H(3) receptor partial agonist (K(i) = 5.4 nM). Although (2S,3R)-4-amino-2,3-methano-1-(1H-imidazol-4-yl)butane (5a) does not have a hydrophobic group which the usual H(3) receptor antagonists have, it was found to be a potent H(3) receptor antagonist (K(i) = 20.1 nM). Thus, a variety of compounds with different pharmacological properties depending on the cyclopropane backbones and also on the side-chain functional groups were identified. In addition to the previously used 1,2-methanobutane backbone, the 2,3-methanobutane backbone also worked effectively as a cyclopropane-based conformational restriction structure. Therefore, the combination of these two cyclopropane backbones increases the stereochemical and three-dimensional diversity of compounds in this strategy, which can provide a variety of useful compounds with different pharmacological properties.  相似文献

Two heteroditopic receptors based on a calix[4]arene crown ether containing amidoanthraquinone pendants in cone and 1,3-alternate conformations (1 and 2, respectively) were synthesized. Photophysical properties of 1 and 2 were studied by UV-vis and fluorescence spectrophotometry in dried CH(3)CN. Both 1 and 2 showed the highest sensitivity towards F(-) through the appearance of a new charge transfer band at 500 nm and the enhancement of the emission spectra at λ(em) = 542 nm and 528 nm respectively. Interestingly, in the presence of K(+), the fluorescence intensity of 1 at 542 nm increased around 2 fold compared to that in the absence of K(+) upon addition of F(-), while this phenomenon was not observed in the case of receptor 2. Cyclic voltammograms of receptors 1 and 2 showed two consecutive one-electron reversible waves in 40% v/v CH(3)CN in CH(2)Cl(2), corresponding to two single-electron reductions to give mono- and dianions species at E(1/2)I = -1.21 V and E(1/2)II = -1.66 V as well as E(1/2)I = -1.25 V and E(1/2)II = -1.71 V, respectively. H(2)PO(4)(-) gave remarkable potential shifts (ca. 200 mV) of the second reduction waves (E(1/2)II) of both free 1 and 2. In the presence of K(+), only receptor 1 gave remarkable potential shifts in its redox wave II upon adding F(-) and AcO(-). Therefore, receptors 1 and 2 exhibited dual sensing modes by fluorescence spectrophotometry and cyclic voltammetry. The topology of ligands also played an important role in cooperative binding properties of heteroditopic receptor 1 possessing a closer distance between a cation and an anion binding. On the other hand, the two ion binding sites of receptor 2 were separated by a longer distance and did not support the cooperative binding. This resulted in the abstraction of K(+) from receptor 2 upon addition of anions.  相似文献

A McMurry coupling reaction and selective crystallization were used to develop a simple and efficient two-step synthesis of (Z)-4-hydroxytamoxifen (2a). This compound is an active metabolite of tamoxifen, a selective estrogen receptor (ER) modulator widely used to treat breast cancer. A series of cyclic imides bearing a omega-(4-aryl and 4-heteroaryl-1-piperazinyl)alkyl moieties was synthesized and tested in vivo for anxiolytic activity. The in vitro binding affinities of these compounds were also examined for 5-HT1A receptor sites. Structure-activity relationships within these series are discussed. One of these compounds, (1R*,2S*,-3R*,4S*)-N-[4-[4-(2-pyrimidinyl)-1-piperazinyl]butyl]-2,3- bicyclo[2.2.1]heptanedicarboximide (1: tandospirone), was found to be equipotent with buspirone in its anxiolytic activity and more anxio-selective than buspirone and diazepam. Tandospirone (1) is currently undergoing clinical evaluation as a selective anxiolytic agent.  相似文献
