We observed a strong, more than 16-fold, enhancement of Texas Red-labeled BSA fluorescence emission when deposited on silver colloid coated surfaces (SCCS). The same labeled protein deposited on silver island films (SIFs) showed an approximate 8-fold fluorescence enhancement. The lifetimes of Texas Red-BSA fluorescence are significantly shorter on silvered surfaces than on uncoated quartz substrate indicating a strong change in radiative decay rate of the dyes. We also observed a 36-fold increased brightness of overlabeled fluorescein-HSA deposited on silver colloid coated surface. Stronger enhancement observed for overlabeled Fl-HSA protein indicates that presence of silver particles partially decreased self-quenching. Our results indicate that surfaces coated with silver colloids are valuable substrates for metal-enhanced fluorescence.  相似文献

We have studied enhancement of the fluorescence of fluorescein isothiocyanate (FITC), bound to albumin and near an annealed silver island film, as a function of the distance between the protein molecules and the metal. As the intermediate spacer layer between the albumin and the silver substrate, we used multilayer films based on polyelectrolytes. The maximum nine-fold enhancement coefficient for the fluorescence of FITC corresponds to a thickness of the intermediate layer of ≈4 nm, or three layers of the polyelectrolyte. We report the first findings of Metal-Enhanced Fluorescence (MEF) from modified plastic substrates. In the past several years our laboratories have reported the favorable effects of fluorophores in close proximity to silver nanoparticles. These effects include, enhanced fluorescence intensities, (increased detectability), and reduced lifetimes, (enhanced fluorophore photostability). All of these reports have featured silver nanostructures and fluorophores which have been immobilized onto clean glass or quartz surfaces. In this report we show how plastic surfaces can be modified to obtain surface functionality, which in turn allows for silver deposition and therefore metal-enhanced fluorescence of fluorophores positioned above the silver using a protein spacer. Our findings show that plastic substrates are ideal surfaces for metal-enhanced phenomena, producing similar enhancements as compared to clean glass surfaces. Subsequently, we speculate that plastic substrates for MEF will find common place, as compared to the more expensive and less versatile traditional silica based supports.  相似文献

采用表面自组装技术,在玻璃表面构筑银纳米粒子的二维组装结构。银纳米粒子组装结构的表面等离子共振光谱中偶极子表面等离子体共振对组装结构更为敏感而表现出较大位移。组装银纳米粒子可极大增强罗丹明B的荧光。荧光的表面增强效应主要来自银纳米粒子对荧光分子所处区域的局部电磁场增强,银纳米粒子的表面分子修饰对其表面增强效应有较大的影响。  相似文献

The dependence of surface-enhanced fluorescence of the semiconducting polymer poly (3-hexylthiophene) on the diameter of silver nanoparticles was studied. Particle suspensions with a high degree of monodispersity, ranging in diameter from 15 to 153 nm, were synthesized. Polymer films were spin-cast onto substrates containing immobilized silver particles. Fluorescence enhancement factors ranged from 0.9 to 4.9 and generally improved with increasing particle size. Normalization of the fluorescence enhancement to the number of interrogated particles showed an 800-fold increase in enhancement between the smallest and largest particles.  相似文献

本文提出一种大尺度的金属-电介质复合微纳结构(银-硅结构),用于提高荧光生物检测的灵敏度及解决荧光物质距离结构远场范围时荧光增强的近场局限。这种大尺度的金属-电介质复合微纳结构与之前的金属-电介质复合微纳结构不同,其通过光的散射和干涉实现了荧光物质距离结构远场范围时的荧光增强。在本文中,通过采用时域有限差分法,主要从荧光激发和荧光发射两个过程研究银-硅结构。结果表明,在激发过程中,银-硅结构的荧光强度高于玻璃结构且位于银-硅结构两柱之间的狭缝中的电场分布比金属结构(银结构)更均匀,因此在银-硅结构中可以实现荧光增强,而且分子运动行为的检测更准确。在发射过程中,当荧光纳米粒子距离结构远场范围内时,与玻璃相比,银-硅结构可以实现更好的荧光增强效果。利用银-硅结构实现荧光增强的机理是光的散射和干涉,荧光被银膜向上散射,同时,结构两侧的银/硅柱也散射一部分荧光,荧光相互干涉传播至远场实现荧光增强。此外,银-硅结构易于制备和集成。因此,其可以很好地应用于生物传感领域。  相似文献

In this short paper, we reported the enhanced fluorescence from a single fluorophore bound to a 50nm silver nanoparticle. We found that on average the Cy5 molecules bound to metal nanoparticles are approximately 15-fold brighter than that of free dyes, and that single molecule lifetimes are shorter as compared to free fluorophores. The increased emission rate is primarily the result of local plasmon enhancement. These results demonstrate that the use of fluorophore-metal interactions can increase the brightness and photostability of fluorophores for single molecule detection.  相似文献

We have explored the opportunities for enhanced ratiometric pH sensing using the well-known carboxy seminaphthofluorescein (SNAFL-2) and silver island films (SiFs). Our results show that the metallic surfaces can provide up to a 40-fold increase in probe fluorescence intensity as compared to nonmetallic surfaces with the same probe coverage. However, while the S/N is significantly better for pH sensing, the emission wavelength ratiometric values are similar to that obtained in solution, due to the fact that the emission of both the acidic and basic forms of the probe are enhanced to similar extents. To the best of our knowledge this is the first report of enhanced ratiometric fluorescence sensing on metallic surfaces.  相似文献

本文研究了 Rh B分子的 539.0 nm和 572 .5nm两荧光峰在表面增强活性银胶颗粒表面上的增强和淬灭效应 :在同一分子体系中同时观察到 530 .0 nm荧光峰的增强和 572 .5nm荧光峰的淬灭 ,并对其进行了理论计算。结果表明这两个荧光峰的增强或淬灭主要取决于局域电磁场增强和分子到金属表面无辐射能量转移衰减过程的竞争效应。当满足吸收共振增强和辐射共振增强时 ,荧光被增强 ;反之 ,荧光被淬灭。计算表明 ,荧光增强因子最高为 1  相似文献

采用微乳液法合成了不同粒径的纳米银粒子,考察了环己烷和甲苯作为油相对制备纳米银的影响.对纳米银粒子的尺寸与结构进行了表征,观察到近球形多晶粒子,并有孪晶结构存在,对晶体结构的分析表明银粒子存在不同程度的点阵畸变,晶面间距增大.不同粒径的纳米银粒子氯仿体系可呈现荧光光谱,而纳米银甲苯体系则无荧光发射.结合紫外—可见吸收光谱和电子自旋共振谱对该体系的荧光发射机理进行了分析 关键词: 微乳液 纳米银粒子 纳米晶结构 荧光  相似文献

In recent years our laboratories have reported the favorable effects for fluorophores placed in close proximity to surface immobilized silver nanostructures. These include; greater quantum yields, reduced lifetimes (increased photostability) and directional emission. However, while these findings are likely to find multifarious applications for surface assays based on enhanced fluorescence detection, a solution based enhanced sensing platform has yet to be realized. In this short, note, we show how SiO2-coated silver colloids, indeed provide for a solution based enhanced fluorescence sensing platform with a 3-5 fold enhancement typically observed.  相似文献

在以聚赖氨酸为表面耦联层分子的玻片基底制备了银纳米粒子阵列。SEM表征结果表明,银粒子以亚单层的形式排列在基底表面。比较银溶胶和纳米粒子阵列的紫外可见光谱可见聚赖氨酸耦联层对银纳米粒子的粒径具有一定的选择性,甲基紫精在银纳米粒子阵列上的表面增强FT拉曼光谱表明在近红外区拉曼散射的表面增强主要来自于化学增强效应。  相似文献

银纳米离子的SERS技术和SEF技术的信号检测灵敏度非常高,可以用在微流控芯片的定量分析中。为了提高微流控芯片光学检测技术的检测精度,提出一种在微通道中添加银纳米粒子来增强SYBR GreenⅠ拉曼和荧光信号的方法,并对该方法的原理和增强效果进行了研究。首先,利用准分子激光器在PMMA基板上直写刻蚀出宽200 μm、深68 μm的微通道,接着将制备的银前体溶液加入微通道,通过加热制备出表面增强拉曼(SERS)和表面增强荧光(SEF)基板,接下来对添加银纳米粒子前后的拉曼和荧光信号分别进行对比,进一步研究了微通道中不同浓度银纳米粒子对SYBR GREEN I的拉曼和荧光信号增强效果。添加银纳米粒子后,表面增强拉曼(SERS)实验的增强因子为3.5×103,添加银纳米粒子的样品的荧光信号强度与不含银纳米粒子样品的荧光信号强度相比,约增加了1倍。结果表明,在微通道中检测SYBR Green I时通过增加银纳米粒子显著地增强了拉曼和荧光信号,这种方法可以用在以SYBR GreenⅠ做染料的微流控芯片检测技术中。  相似文献

文章报道了吸附在银纳米颗粒表面的四苯基卟啉(TPP)的Q带荧光寿命的变化.实验发现将银纳米颗粒的表面等离激元共振吸收峰调至与TPP的Q吸收带共振时,会使TPP的Q带荧光寿命得到延长.这是由于银纳米颗粒对表面的光电场有较大的表面等离激元共振增强效应,而处于激发态的TPP分子有较高的极性,这种增强的光电场会对有较高极性的TPP分子有稳定作用,所以延长了其Q带寿命.这对于以卟啉为光敏剂的光动力治疗有重要的意义.  相似文献

本文将三种金属纳米颗粒(球形银纳米颗粒、三角形银纳米薄片和三角形金纳米薄片)分别置于Eu(TTFA)3掺杂的Su8薄膜上研究了它们对铕配合物薄膜发光的影响.结果显示,金属纳米颗粒的局域等离子体共振(LSPR)谱对薄膜发光有很好的调控作用.对于LSPR谱分别与铕配合物激发谱和辐射谱重叠较好的球形和三角形两种银颗粒,相比于未放置颗粒时薄膜的发光,放置有颗粒的发光有明显的增强,其中球形银颗粒的样品的发光增强效果更为明显.而对于LSPR谱与铕配合物激发谱和辐射谱都不重叠的三角形片状金颗粒,该颗粒对样品的发光则起到了猝灭作用.  相似文献

利用银胶和银镜两种SERS活性表面,对吸附分子的荧光增强或淬灭机制进行了实验和理论研究.结果表明,荧光增强或淬灭主要取决于局域电磁场增强和分子到金属表面无辐射能量转移过程的竞争效应.当满足吸收共振增强和辐射共振增强时,荧光被增强,反之荧光被淬灭.荧光增强最大一般只为10—102倍,远小于喇曼截面的增强. 关键词:  相似文献

Substantial increases in fluorescence emission from fluorophore-protein–coated fractal-like silver structures have been observed. We review two methods for silver fractal structure preparation, which have been employed and studied. The first, a roughened silver electrode, typically yielded a 100-fold increase in fluorophore emission, and the second, silver fractal-like structures grown on glass between two silver electrodes, produced a 500-fold increase. In addition, significant increases in probe photostability were observed for probes coated on the silver fractal like structures. These results further serve to compliment our recent work on the effects of nobel metal particles with fluorophores, a relatively new phenomenon in fluorescence we have termed both metal-enhanced fluorescence [1] and radiative decay engineering [2,3]. These results are explained by the metallic surfaces modifying the radiative decay rate () of the fluorescent labels. We believe that this new silver-surface preparation, which results in ultrabright and photostable fluorophores, offers a new generic technology platform for increased fluorescence signal levels, with widespread potential applications to the analytical sciences, imaging, and medical diagnostics.  相似文献

We report surface-enhanced Raman scattering (SERS) from 4-mercaptopyridine adsorbed on nanotextured silver surfaces as the coverage of silver is varied. The degree of surface enhancement is strongly dependent on silver coverage and correlated to the extinction of the surface at the Raman excitation wavelength, that extinction being determined by multiparticle surface plasmon resonances. The coverage dependence of the Raman intensity is consistent with signals being dominated by molecules at junctions inside nanoparticle aggregates where electromagnetic energy is localized into “hot spots” by interactions of the incident and scattered fields with the surface plasmons. The Raman intensity drops precipitously near the conductivity percolation threshold because these hot spots are destroyed when conducting paths allow plasmons to propagate. Our approach to substrate preparation provides clean surfaces with average enhancements ≥107, an order of magnitude larger than typical for SERS. PACS 78.67.-n; 78.68.+m; 33.20.Fb  相似文献

We study the fluorescence enhancement of dye molecules adsorbed on regular two-dimensional arrays of designed silver nanoparticles. The silver particles show two orthogonal optical resonances at different wavelengths because of their elongated shape. The short-wavelength resonance was designed to fit to the absorption maximum of the fluorophore. When the excitation light drives the short-wavelength resonance, the measured fluorescence intensity is strongly enhanced compared to that for the orthogonal particle orientation. This shows directly a strong electromagnetic coupling between the nanoparticles and the fluorophore. Additionally enhanced photochemical bleaching is observed due to the interaction of fluorophores with the particles. Using a rate model describing the fluorescence enhancement and the bleaching enhancement, an average value for the particle-induced increase in the radiative fluorescence rate is obtained, together with a lower limit for the averaged particle-induced field intensity enhancement factor. 采用物理吸附法,在机械抛光纯银表面引入对巯基苯胺(p-Amiothiophenol,PATP)分子充当隔离层,运用激光光谱学方法研究隔离层对位于银表面附近的罗丹明6G(Rh6G)分子的荧光增强效应影响。实验结果表明,未经PATP分子修饰的机械抛光金属衬底对Rh6G分子表现为猝灭效应,而经过PATP分子修饰后的银表面对Rh6G分子的荧光发射具有增强效应。根据局域表面等离子共振及辐射能量转移模型对实验观测所得结果进行了分析研究,结果表明,PATP有机分子隔离层的引入有效地减小了荧光分子与金属衬底之间的无辐射能量速率,提高了荧光辐射强度。  相似文献
