高效液相色谱法分析元宝枫叶中儿茶素类物质   总被引:7,自引:2,他引:5  
本文建立了元宝枫树叶中儿茶素种类及其含量的高效液相色谱(HPLC)测定方法。采用反相C18色谱柱,以甲醇/水(含0.5%乙酸)=25/75(V/V)为流动相,对没食子儿茶素(GC)、表没食子儿茶素(EGC)、儿茶素(C)、表没食子儿茶素没食子酸酯(EGCG)、表儿茶素(EC)和没食子儿茶素没食子酸酯(GCG)进行定性、定量分析;以甲醇/水(含0.5%乙酸)=35/65(V/V)为流动相,对表儿茶素没食子酸酯(ECG)和儿茶素没食子酸酯(CG)进行定性分析,柱温均为35℃,检测波长为278 nm,流速为1.0mL/min。结果表明:元宝枫叶中有EGC、EC和GCG,其它五种则无。EGC平均含量为0.0389 mg/g,方法精密度(RSD)为0.42%(n=6);EC平均含量为0.0289 mg/g,方法RSD为1.5%(n=6);GCG平均含量为0.284 mg/g,方法RSD为0.32%(n=6)。该方法简便、准确、分离效果好,为元宝枫叶开发成茶叶、饮料以及医疗保健品提供重要依据。  相似文献

用紫外-可见(UV-Vis)吸收光谱和1H核磁共振(NMR)谱研究了茶多酚类衍生物表没食子儿茶素没食子酸酯(EGCG)与Zn(Ⅱ)离子的相互作用,并用密度泛函理论(DFT)计算了EGCG与Zn(Ⅱ)离子络合前后的空间结构及其紫外和核磁共振谱.实验与理论研究结果表明:EGCG主要构象是其芳香B环以e键(平伏键)及芳香D环以a键(直立键)形式共同与C环链接.EGCG通过其芳香D环上酚羟基与Zn(Ⅱ)离子相互作用,生成稳定的Zn(Ⅱ)与EGCG摩尔比为1:1的Zn(Ⅱ)-EGCG四面体络合物.  相似文献

应用高效液相色谱法测定了茶叶,特别是绿茶中的8种多酚,包括儿荼素(C)、表没食子儿茶素(EGC)、没食子儿荼素(GC)、表儿茶素(EC)、表没食子儿茶素没食子酸酯(EGCG)、没食子儿荼素没食子酸酯(GCG)、表儿茶素没食子酸酯(ECG)及儿茶素没食子酸酯(CG).在C18>反相柱(250 min×4.6 mm,5 μm)上进行分离,用不同体积比混合的流动相A及流动相B的混合液作梯度淋洗,流速为1.0 mL·min-1,柱温为40℃,在280 nm波长处进行紫外检测.上述8组分在0.004~4.0 g·L-范围内呈线性关系,方法的检出限在0.6~2.6 mg·L-1范围内,平均回收率为83%.  相似文献

 采用毛细管区带电泳法测定了 10种中国产葡萄籽中的 4个主要儿茶素类化合物 :(+)儿茶素、(- )表儿茶素、(± )表没食子儿茶素、(± )表儿茶素没食子酸酯的含量。在 0 0 2mol/L硼砂和 0 0 0 5mol/L磷酸盐的混合缓冲体系 (pH 10 0 )的背景缓冲液中 ,4个化合物在 10min内取得了令人满意的分离。迁移时间的重现性(RSD)小于 2 % ,峰面积的重现性 (RSD)小于 5 %。在质量浓度为 0 0 0 5g/L~ 0 5 g/L时 ,线性相关系数大于0 995。检测限为 3mg/L~ 10mg/L。该方法简单、快速、准确 ,可作为葡萄籽分析和药用开发过程中分析儿茶素类化合物的有效方法推广使用。  相似文献

建立了从茶叶鲜叶中分离纯化7种儿茶素类化合物(没食子儿茶素(GC)、表没食子儿茶素(EGC)、儿茶素(C)、表没食子儿茶素没食子酸酯(EGCG)、表儿茶素(EC)、表没食子儿茶素3-O-(3-O-甲基)没食子酸酯(EGCG3"Me)和表儿茶素没食子酸酯(ECG))的半制备色谱法。铁观音鲜叶经甲醇超声浸提、浓缩、氯仿萃取后,向水相中加入碱式醋酸铅沉淀,得到茶多酚粗品。分别以甲醇-水、乙腈-水作为流动相,采用半制备色谱法纯化7种儿茶素类化合物,纯度均达到90%。此外,利用同样的方法分离纯化另外两种茶叶鲜叶中的7种儿茶素类化合物,得到相似的结果。该方法以溶剂提取、离子沉淀结合半制备色谱,适于简单、高效地同时分离制备多种儿茶素类化合物。  相似文献

采用高效液相色谱-质谱联用技术及高效液相色谱法对生熟普洱茶中的主要成分进行定性和定量分析。鉴定出普洱茶水溶液中8种主要成分,分别为没食子酸(GA)、没食子酸儿茶素(GC)、表没食子酸儿茶素(EGC)、儿茶素(C)、咖啡因(CAF)、表儿茶素(EC)、表没食子酸儿茶素没食子酸酯(EGCG)和表儿茶素没食子酸酯(ECG)。以这8种成分的含量为指标,对普洱生茶和熟茶各20批进行主成分分析、聚类分析和判别分析,能准确地区分普洱生茶与熟茶。  相似文献

儿茶素(catechins)分为酯型儿茶素(EGCG和ECG)和游离型儿茶素(EC和EGC)。酯型儿茶素的抗氧化活性比游离型儿茶素的强[1]。儿茶素类化合物的基本结构均为α 连(或邻)苯酚基苯并吡喃。由于儿茶素分子具有α 连(或邻)苯酚基结构,使它们具有强抗氧化的特性[2]。深入研究儿茶素类抗氧化的构效关系,对天然抗氧化剂的筛选具有重要意义。1 方法  选用EGCG、ECG、EGC、EC作为模型分子。这四种分子中A环的5 、7 位均带酚羟基,不同的是在C环的2 位带邻苯酚基或连苯三酚基,3 位上带羟基或没食子酸结构。分子的原子编号及结构式见图1,…  相似文献

采用反相高效液相色谱法同时测定茶叶中L (+) 表儿茶素(L EC),L (-) 表儿茶素没食子酸酯(L ECG),L (-) 表没食子儿茶素没食子酸酯(L EGCG),L (-) 表没食子儿茶素(L EGC),(+) 没食子儿茶素没食子酸酯(GCG),DL 儿茶素(DL C)六种儿茶素组分,以乙醇∶乙酸∶水=14.5∶1.0∶84.5(体积比)为流动相(1.0ml·min-1),C18柱,UV检测器(278nm)。六种儿茶素组分的相对标准偏差分别为0.67%,0.45%,0.24%,0.56%,3.11%,4 41%,回收率分别为95.2%,98.2%,101.3%,97.0%,98.3%,97.5%。该法采用乙醇、乙酸等无毒溶剂作流动相,回收率高,用于实际样品测定,结果满意。  相似文献

采用HPLC-ESI-Q-TOF和MALD I-TOF质谱技术分析儿茶素组成与结构,发现儿茶素中的表没食子儿茶素没食子酸酯(EGCG)具有多聚体特性。有机化学法合成EGCG-Ge4 化合物,并经HPLC进一步分离纯化。红外光谱和质谱技术分析并证实EGCG不仅能形成多聚体,而且还能络合金属锗(包括其他金属离子),成为EGCG-Ge4 和EGCG-Ge4 多聚体。EGCG-Ge4 抑制肺癌细胞生长速率明显高于EGCG,其提高幅度约为30%~40%左右。  相似文献

The electron ionization (EI) mass spectral fragmentation of derivatized 4,5- and 5,6-epoxysterols was investigated. Interesting fragmentation processes involving a transannular cleavage of the epoxide ring after transfer of the trimethylsilyl group are significant in the case of 4,5-epoxysterol trimethylsilyl ethers (affording abundant fragment ions at m/z 403 and 404). Different pathways, which have been substantiated by deuterium labelling, are proposed in order to explain the formation of these ions. In contrast, this transfer is not significant in the case of 5,6-epoxysterol trimethylsilyl ethers. The EI mass spectra of these latter compounds appear to be very complex and to differ slightly according to the stereochemistry of the epoxy group. Acetate and trifluoroacetate derivatives of 4,5-epoxysterols display interesting EI mass spectra dominated by a fragment ion at m/z 332 resulting from cleavage of the steroid ring A.  相似文献

The fragment ion formation characteristics of the radical anions generated from hexahydro-1,3,5-trinitrotriazine (RDX) and its three nitroso metabolites were studied using GC/MS with negative chemical ionization (NCI) to understand the fragmentation mechanisms responsible for the formation of the most abundant ions observed in their NCI mass spectra. Ab initio and density functional theory calculations were used to calculate relative free energies for different fragment ion structures suggested by the m/z values of the most abundant ions observed in the NCI mass spectra. The NCI mass spectra of the four nitramines are dominated by ions formed by the cleavage of nitrogen-nitrogen and carbon-nitrogen bonds in the atrazine ring. The most abundant anions in the NCI mass spectra of these nitramines have the general formulas C(2)H(4)N(3)O (m/z 86) and C(2)H(4)N(3)O(2) (m/z 102). The analyses of isotope-labeled standards indicate that these two ions are formed by neutral losses that include two exocylic nitrogens and one atrazine ring nitrogen. Our calculations and observations of the nitramine mass spectra suggest that the m/z 86 and m/z 102 ions are formed from either the (M--NO)(-) or (M--NO(2))(-) fragment anions by a single fragmentation reaction producing neutral losses of CH(2)N(2)O or CH(2)N(2)O(2) rather than a set of sequential reactions involving neutral losses of HNO(2) or HNO and HCN.  相似文献

The fragmentation of bisphenol A diglycidyl ether (BADGE), bisphenol F diglycidyl ether (BFDGE) and their derivatives was studied by electrospray ionization tandem mass spectrometry. Multiple-stage mass spectrometry and accurate mass measurements were combined to establish the fragmentation pathways. BADGEs and BFDGEs tend to form ammonium adducts under electrospray conditions which fragmented easily. The fragmentation of [M+NH(4)](+) for BADGEs started with the cleavage of the phenyl-alkyl bond, which was followed by the α-cleavage of the ether group to generate the characteristic product ions at m/z 135, [C(9)H(11)O](+), and m/z 107, [C(7)H(7)O](+). The fragmentation of the BFDGE isomer mixtures was studied by on-line reversed-phase liquid chromatography coupled to multiple-stage mass spectrometry (LC/MS(n)). Information obtained from product ion spectra for each BFDGE isomer and its comparison with the fragmentation pathway of BADGE allowed each isomer and the chromatographic elution order to be identified.  相似文献

High-resolution electrospray ionization multistage tandem mass spectrometry (MS(1-7)) in positive ion mode was used to determine the accurate masses and the fragmentation pathways of two furofurans, sesamin and gmelinol. The protonated molecules [M+H]+ were absent in the conventional mass spectra of both compounds, but two characteristic ions, [M+H-H(2)O]+ and [M+H-H(2)]+, were always observed. Successive losses of CH(2)O and CO are the common characteristic fragmentations. Based on the exact masses acquired from 21 different tandem mass spectra, two or three fragmentation pathways for each compound are proposed. The consecutive losses of two H(2)O molecules and one H(2) molecule readily take place from the furan rings for both sesamin and gmelinol, resulting in the absence of the protonated molecules in the single-stage experiments. HCHO loss is observed at least three times in the tandem mass spectra, mainly from methylenedioxy groups (OCH(2)O) for sesamin but only from tetrahydrofuran rings for gmelinol. Moreover, CO loss is found at least three times in the tandem mass spectra of both sesamin and gmelinol from the 3,4-methylenedioxyphenyl (ArOCH(2)O) moieties for sesamin and from both the dimethoxyphenyl and the tetrahydrofuran ring moieties for gmelinol. In addition, the disubstituted benzyl cation ArCH(2)+ at m/z 135 for sesamin and at m/z 151 for gmelinol was found in the MS(3) spectra of both sesamin and gmelinol, which is very helpful in the identification of the compositions of 3,4-disubstituted groups on the benzene rings of the furofurans.  相似文献

High-resolution electrospray ionization multistage tandem mass spectrometry (MS(1-9)) was used to determine the accurate masses and the fragmentation pathways of protonated podophyllotoxin (1) and its corresponding 4'-demethyl-4beta-substituted derivatives (2-4). The protonated molecules, [M + H](+), of all the four compounds were observed in the conventional single-stage mass spectra. Two fragmentation pathways, that appear to be characteristic of the four compounds, are proposed on the basis of their multistage tandem mass spectrometric data. The characteristic elimination, from the precursor protonated ions, of the neutral groups 4-R(1)H, 1-ArH, CO, CH(2)O and C(4)H(4)O(2), in which R is located on C-4, is the common elimination, and the product ions at m/z 267, 239, 229, 181, 173, 153, 143 and 115 are the common diagnostic masses. The elimination of the R(1) group substituent located on the C-4 position of compounds 1-4 has a significant influence on the fragmentation pathway obtained in the conventional single-stage mass spectra. A large R(1) group would be unfavorable for this elimination, unless the collision energy is raised. Apart from the common fragmentations obtained for the protonated molecules 1-4, significant additional product ions were detected in the various multistage tandem mass spectrometric analyses, particularly in the case of the product ions derived initially from the phenolic hydroxyl group of 2-4, which are different from those of 1. Based on these additional formed product ions, several additional fragmentation pathways for 1 or 2-4 are also presented.  相似文献

A mass spectrometric study was carried out on two nonylphenoxycarboxylic acids, NP1EC and NP2EC (where 1 and 2 indicate the number of ethoxylate units attached to the nonylphenoxy moiety), that are persistent metabolites of widely used nonionic surfactant nonylphenol ethoxylates. In a gas chromatographic/mass spectrometric (GC/MS) study of the methyl esters of NP1EC and NP2EC, two series of fragment ions were observed in electron ionization (EI) mass spectra; m/z (179 + 14n, n = 0-7) and m/z (105 + 14n, n = 0-4) for NP1ECMe and m/z (223 + 14n, n = 0-7) and m/z (107 + 14n, n = 0-5) for NP2ECMe. Similarity indices were used to compare quantitatively the mass spectra of isomers. The mass spectra of two isomers were found to be similar whereas those of the remaining isomers were readily distinguishable from each other. The abundant fragment ions of the two NPECMes were investigated further by GC/MS/MS; product ions resulting from cleavage in the alkyl moiety, cleavage in the ECMe moiety and cleavage in both moieties were detected. Possible structures of the nonyl groups in the two esters were inferred. GC/chemical ionization (CI) mass spectra of the NPECMes with isobutane as reagent gas showed characteristic hydride ion-abstracted fragment ions shifted by 1 Da from those in the corresponding EI mass spectra. The sensitivity of a selected ion monitoring quantitation method for the NPECMes is enhanced under CI conditions compared with that under EI conditions. With electrospray ionization MS/MS, [M - H](-) ions of NP1EC (m/z 277) and NP2EC (m/z 321) were observed and, upon collision-induced dissociation of [M - H](-) of each of the two acids, fragment ions of m/z 219 corresponding to deprotonated nonylphenol, were observed in each case. Based on this observation, a rapid, simple and reliable selected product ion quantitation method is proposed for NP1EC and NP2EC.  相似文献

Three cyclitol derivatives were isolated from the marine sponge Sarcotragus sp. by reversed-phase high-performance liquid chromatography and analyzed by fast-atom bombardment mass spectrometry (FAB-MS). Their structural elucidation was carried out with FAB tandem mass spectrometry (FAB-MS/MS). FAB-MS spectra produced a significant abundance of the sodium adducts [M+Na]+ and [M+2Na-H]+ from a mixture of m-NBA and NaI. In addition, trifluoroacetylation of the cyclitol derivatives was used for confirmation of the presence of the cyclitol ring. High abundance [M-5H+5CF3CO+Na]+ ions were observed in the FAB-MS spectra of the trifluoroacetyl-cyclitol derivatives. Collision-induced dissociation (CID) of the [M+Na]+ ions produced diverse product ions via a series of dissociative processes. Charge-remote fragmentation (CRF) patterns of [M+Na]+ ions were very useful for the identification of product ions which are characteristic for the cyclitol ring and long hydrocarbon chains substituted at the glycerol backbone. Moreover, the CID-MS/MS spectra of the [M+Na]+ ions yielded characteristic product ions at m/z 53, 83, 113, 155 and 171 for the cyclitol moiety, and at m/z 213, 229 and 245 for the glycerol backbone attached to the cyclitol ring.  相似文献

The mass spectra of the controlled substance 3,4-MDMA and its regioisomer 2,3-MDMA are characterized by an imine fragment base peak at m/z 58 and additional fragments at m/z 135/136 for the methylenedioxybenzyl cation and radical cation, respectively. Three positional ring methoxy isomers of N-methyl-2-(methoxyphenyl)-3-butanamine (MPBA) have an isobaric relationship to 2,3- and 3,4-MDMA. All five compounds have the same molecular weight and produce similar EI mass spectra. This lack of mass spectral specificity for the isomers in addition to the possibility of chromatographic co-elution could result in misidentification. The lack of reference materials for the potential imposter molecules constitutes a significant analytical challenge. Perfluoroacylation of the amine group reduced the nitrogen basicity and provided individual fragmentation pathways for discrimination among these compounds based on unique fragment ions and the relative abundance of common ions. Studies using gas chromatography with infrared detection provided additional structure-IR spectra relationships. The underivatized amines and the perfluoroacylated derivatives (PFPA and HFBA) were resolved by capillary gas chromatography on a 100% dimethylpolysiloxane stationary phase. The perfluoroacylated derivatives showed better resolution on a cyclodextrin modified stationary phase.  相似文献

Low-energy collision-induced electrospray ionization tandem mass spectrometry ESI-CID-MS/MS (in the positive ion mode) was used for the structural characterization of a series of five representative epioplythiodioxopipreazines: dethiotetra(methylthio)chemotin, chaetocochins A, B and C, and chemotin isolated from the fungus Chaetomium cochliodes. The fragmentation pathways were elucidated by ESI-IT-MS(n). The elemental compositions of most of the product ions were confirmed by low-energy ESI-CID-QTOF-MS/MS analyses. The loss of the S(2) molecule seems always to be the first when the S--S bond is present. The loss of 77 Da corresponding to the loss of the [CH(3)SCH(2)O]' radical was diagnostic for chaetocochins A and B, in which the two piperazines rings are linked by an acetal group. It was found that a McLafferty rearrangement plays a significant role in the skeleton fragmentation of theses series of studied complex multicyclic piperazine compounds. This MacLafferty rearrangement affords the product ions at m/z 416 and 400, containing the two piperazine rings belonging to the epipolythiodioxopipreazines. In addition, the pentacyclic rearrangement involving the loss of the SMe(.) radical seems to occur in the presence of the unfused ring. Finally the product ions at m/z 635 and 591 seem to be the characteristic ions for chaetocochin A.  相似文献
