Twelve novel 5,6-disubstituted-3-phenyl-1,2,3-triazolo[4,5-d]pyrimidine-7-ones 5a–5l were designed and successfully synthesized via tandem aza-Wittig and annulation reactions of the corresponding iminophosphorances 2, aromatic isocyanate, and substituted thiophenols in satisfactory yields. The results from the preliminary bioassay indicated that some of these compounds possess inhibitory activities against the root and stalk of Brassica napus(rape) as well as Echinochloa crusgalli (barnyard grass) at the dosages of 100 mg/L and 10 mg/L, respectively.  相似文献

Twelve novel pyrazolo [4,3-d] pyrimidin-7-ones were synthesized and their structures were confirmed by IR, ^1H NMR and MS. Their in vitro bronchodilatory activities were tested in guinea-pigs. The pharmacological results show that compound 11c has more potent activity than aminophylline.  相似文献

The reaction of acyl derivatives of ethyl 3-aminothieno[2,3-b]pyridine-2-carboxylate with hydrazine hydrate gives a series of tricyclic 3-amino-3,4-dihydropyrido[3′,2′:4,5]thieno[3,2-d]pyrimidin-4-ones containing aliphatic, aromatic, or heteroaromatic fragments at C(2). The title compound 3-benzyl-5-cyclohexylamino-6-phenyI-3,6-dihydro-1,2,3-triazolo[4,5-d]pyrimidin-7-one (C23H24N6O, Mr = 400.48) has been synthesized by means of tandem aza-Wittig reaction, and its crystal structure was determined by single-crystal X-ray diffraction. The crystal belongs to monoclinic, space group P21/n with a = 6.5550(6), b - 10.3711(10), c = 31.265(3)(A), β = 93.827(2)° ,V = 2120.8(4) (A)3, Z = 4, Dc = 1.254 g/cm3, S = 1.025, μ(MoKa) = 0.081 mm-1, F(000) = 848, R = 0.0576 and wR = 0.1368. X-ray analysis reveals that all ring atoms in the triazolopyrimidinone moiety are almost coplanar and the cyclohexyl ring adopts a chair conformation.  相似文献

New methods for annelation of a pyridine ring to a pyrimidine ring were suggested. Substituted 8H-pyrido[2,3-d]pyrimidin-5-ones (8a-f) were synthesized by the interaction of 2,6-disubstituted 4-amino-5-acetylpyrimidines (1–4) with formamide or acetamide acetals followed by cyclization under the action of sodium methoxide in methanol. 2,4-Disubstituted 7-phenyl-8H-pyrido[2,3-d]pyrimidin-5-ones (11a-c) were prepared by the reaction of 2,6-disubstituted 5-acetyl-4-benzoylaminopyrimidines (10a-c) with MeONa in boiling BuOH.Translated fromIzvestiya Akademii Nauk. The effects of the nature of the nucleophile and the structure of 6-nitrotriazolo[1,5-a]pyrimidinones on the direction of the ring opening were investigated. The triazole ring is opened under the action of strong bases, such as alkoxides or alkalis, to form 2-cyanoaminopyrimidinones and then 2-aminopyrimidinones. The results of the reactions with N-nucleophiles depend on the accessibility of position 5 of the heterocycle. Thus, sterically hindered secondary amines react with 5-methyltriazolopyrimidinones to give 2-guanidinopyrimidinones and then 2-aminopyrimidinones. In the absence of a substituent at position 5, the azine ring is opened to form 4-alkyl-3-amino-1,2,4-triazoles and 3-amino-2-nitroacrylamides. Under the action of primary amines, only the pyrimidine fragment is cleaved.  相似文献

Abstract  A new route to the synthesis of 2-arylthieno[2,3-d]pyrimidin-4(3H)-ones has been developed through heterocyclization of 2-amino-4,5-dimethylthiophene-3-carboxamide with aromatic aldehydes in boiling glacial acetic acid followed by air oxidation. The unoxidized intermediates, 2-aryl-2,3-dihydrothieno[2,3-d]pyrimidin-4(1H)-ones, are isolated when the reactions are carried out either at room temperature or under a nitrogen atmosphere. Summary A convenient and simple synthesis of 5-mercapto-3-phenyl-2-thioxo-2,3-dihydrothiazolo-[4,5-d]pyrimidin-7(6H-one(2) and its 5,7-dichloro (3), 5,7-diazido (4), 5,7-diamino (5), 5,7-dimerapto (6), 5,7-dimethylthio (7), and 6-methyl-5-methylthio (8) derivatives is described. Summary Some 3-substituted 2-thioxothiazolo[4,5-d]pyrimidin-7(6H)-ones (2a,b;4) have been synthesized and converted to their 7-chloro (3,5), 7-diethanolamino (6a,b), 7-bis(2-chloroethyl)amino (7), 7-azido (8), 7-amino (9), 7-hydrazino (16), 7-mercapto (11a,b), and 7-methylthio (12) derivatives. Eighteen novel 5-alkylamino-6-aryl-3-phenyl-2-thioxo-2,3-dihydrothiazolo[4,5-d]pyrimidin-7(6H)-ones 5a-r were designed and easily synthesized via a tandem aza-Wittig reaction. Treatment of iminophosphorane 2 with aromatic isocyanate gave carbodiimide 3, which reacted with fluoro-substituted alkylamines to provide the title compounds in 65-87% isolated yields using sodium ethoxide as catalyst. All compounds 5 were confirmed by IR, 1H NMR, MS and elemental analysis, and compound 5p was further analyzed by single-crystal X-ray diffraction. The preliminary bioassay indicated that some compounds showed significant inhibition activities against the root growth of rape and barnyard grass at a dosage of 100 mg/L.  相似文献
