A simple and rapid method is described for the quantitative extraction and determination of Cd and Pb in mussel tissue (Mytilus edulis). The method is based on the quantitative ultrasound-assisted extraction (i.e. sample mass at mg level) of the two metals using diluted nitric acid as extractant. The extraction procedure is carried out in autosampler cups of the graphite furnace (typically, less than 20 mg). A two-level full factorial design (24) was applied to optimize the variables influencing the ultrasound extraction process. These variables were: extraction time, ultrasound amplitude, nitric acid concentration and particle size. Optimization results showed that acid concentration and particle size were the more significant variables. Determination of Cd and Pb in extracts obtained after ultrasound treatment was carried out by Electrothermal Atomic Absorption Spectrometry. The method was validated by statistically comparing the metal contents found with the certified ones corresponding to the BCR 278 mussel tissue. No significant differences were observed for P = 0.05. LODs for Cd and Pb in mussel tissue were 0.019 and 0.37 μg g–1. RSDs values (corresponding to between-batch precision for n = 5) were 2.2 and 6.7% for Cd and Pb, respectively. The method was applied to measure the contents of Cd and Pb in mussels used as pollution bioindicators from the Galician coast (Ria de Vigo, Spain).  相似文献

In this work it was evaluated the determination of Cd and Pb in mussels by tungsten coil electrothermal atomic absorption spectrometry (TCA-AAS). A critical and comprehensive study of the effects caused by Pd, Mg, ascorbic acid, and binary mixtures of these compounds on the atomization of Cd and Pb in acid digested solutions of mussels was performed. Palladium and mixtures containing it were useful to increase sensitivity and thermal stability of Cd and Pb. Additionally, the coil lifetime was increased and the background signals were decreased in these modifiers. All these favorable effects were analytically exploited to determine Cd and Pb in samples of mussels. The proposed methodology was validated using two certified reference materials (oyster tissue and mussel). No statistical difference was observed between determined and certified values at a 95% confidence level. Cadmium and Pb in Chilean bivalve mussels were determined by TCA and by graphite furnace-AAS. Again, the results showed statistical agreement.  相似文献

A dynamic ultrasound-assisted extraction step is proposed for the quantitative extraction of Cd and Pb from plant leaves prior to determination by electrothermal atomic absorption spectrometry (ETAAS). Beech leaves (a certified reference material—CRM 100—where the target analytes were not certified) were used for optimizing the extraction step by a multivariate approach. The samples (0.25 g) were subjected to dynamic ultrasound-assisted extraction with dilute nitric acid as extractant. The method was validated with a certified reference material BCR-062 (olive leaves) where both Cd and Pb are certified. The good agreement between the certified values and those found using the proposed method demonstrates its usefulness. The repeatability was 2.0 and 0.9% and the within-laboratory reproducibility was 7.1 and 2.8% for Cd and Pb, respectively. The precision of the method, together with its efficiency, rapidity, low cost and environmental acceptability makes it a good alternative for the determination of trace metals from plant material.  相似文献

The applicability of a DETATA sorbent to the preconcentration of lead and cadmium followed by the determination of these elements in urine using atomic absorption spectrometry with electrothermal atomization was demonstrated. After preconcentration by a factor of 10, the limits of detection were 0.01 and 0.2 μg/L for cadmium and lead, respectively. The accuracy of the results was supported by the analysis of Seronorm Trace Elements Urine (Batch no. 101021).  相似文献

The concentration of lead and cadmium in different kinds of milk samples (powdered, infant formula, market, buffalo, condensed and human) were determined using electrothermal atomic absorption spectrophotometric technique. Among all the varieties of milk analysed, condensed milk was found to contain much higher amount of lead. Human milk as expected was found to have lowest concentration of these elements. The results were compared with the reported values of other countries. Daily intake of these toxic elements by adults and babies up to the age of six months through the consumption of various types of milk was estimated and compared with the tolerance levels.  相似文献

A method has been developed for the direct simultaneous determination of Cd and Pb in white and red wine by electrothermal atomic absorption spectrometry (ET-AAS) using a transversely heated graphite tube atomizer (THGA) with longitudinal Zeeman-effect background correction. The thermal behavior of both analytes during pyrolysis and atomization stages were investigated in 0.028 mol l−1 HNO3 and in 1+1 v/v diluted wine using mixtures of Pd(NO3)2+Mg(NO3)2 and NH4H2PO4+Mg(NO3)2 as chemical modifiers. With 5 μg Pd+3 μg Mg as the modifiers and a two-step pyrolysis (10 s at 400°C and 10 s at 600°C), the formation of carbonaceous residues inside the atomizer was avoided. For 20 μl of sample (wine+0.056 mol l−1 HNO3, 1+1, v/v) dispensed into the graphite tube, analytical curves in the 0.10–1.0 μg l−1 Cd and 5.0–50 μg l−1 Pb ranges were established. The characteristic mass was approximately 0.6 pg for Cd and 33 pg for Pb, and the lifetime of the tube was approximately 400 firings. The limits of detection (LOD) based on integrated absorbance (0.03 μg l−1 for Cd, 0.8 μg l−1 for Pb) exceeded the requirements of Brazilian Food Regulations (decree #55871 from Health Department), which establish the maximum permissible level for Cd at 200 μg l−1 and for Pb at 500 μg l−1. The relative standard deviations (n=12) were typically <8% for Cd and <6% for Pb. The recoveries of Cd and Pb added to wine samples varied from 88 to 107% and 93 to 103%, respectively. The accuracy of the direct determination of Cd and Pb was checked for 10 table wines by comparing the results with those obtained for digested wine using single-element ET-AAS, which were in agreement at the 95% confidence level.  相似文献

Electrodeposition on the graphite electrode under conditions of controlled current in a flow-through mode, followed by electrothermal atomic absorption spectrometry, is proposed for the determination of cadmium. After electrolysis in a microcell of 2.6 μl volume, deposited metal was dissolved in 40 μl 0.2 mol l−1 HNO3 and the whole volume was direct injected into the atomizer. Using this on-line arrangement and electrodeposition from 1.75 ml of sample solution detection limit of 25 ng l−1 Cd was attained. A method was developed for the quantitative determination of total vanadium concentration in mussels via electrothermal atomic absorption spectrometry (ETAAS). After the microwave digestion of the samples, a program using temperatures of 1600 °C and 2600 °C for ashing and atomization respectively, without any matrix modifiers, allowed us to obtain results that were satisfactory since they agreed closely with certified reference material values. The detection limit was 0.03 mg kg–1 (dry weight), indicating that the method is suitable for the analysis of mussel samples. This determination was compared with matrix modifiers that have been reported previously. The method was applied to various cultivated and wild mussels from the Galician coast, yielding levels below 1 mg kg–1 (wet weight).  相似文献

This paper proposes a method for the determination of lead in aluminum and magnesium antacids employing electrothermal atomic absorption spectrometry (ET AAS). The pyrolysis and atomization temperatures established during the optimization step were 700 and 2200 °C, respectively, using phosphate as the chemical modifier. Under these conditions, a characteristic mass of 25 pg, and limits of detection and quantification of 0.40 and 1.35 μg L−1, respectively were obtained. Some experiments demonstrated that the calibration can be performed employing the external calibration technique using aqueous standards. The precision expressed as relative standard deviation (RSD %) was 4.03% for an antacid sample with lead concentrations of 284.5 μg L−1. The proposed method was applied for the determination of lead in five antacid samples acquired in Salvador City, Brazil. The lead content was varied from 87 to 943 μg g−1. The samples were also analyzed after complete dissolution by inductively coupled plasma mass spectrometry (ICP-MS). No statistical difference was observed between the results obtained by both of the procedures performed.  相似文献

A procedure for the determination of cadmium, chromium, and lead in marine sediment slurries by electrothermal atomic absorption spectrometry is proposed. Slurry was prepared by mixing 10 mg of ground sample with particle size smaller than 50 μm completed to the weight of 1.0 g with a 3% nitric acid and 10% hydrogen peroxide solution. The slurry was maintained homogeneous with an aquarium air pump. For cadmium, the best results were obtained using iridium permanent with optimum pyrolysis and atomization temperatures of 400 and 1300 °C, respectively, a characteristic mass, mo (1% absorption), of 2.3 pg (recommended 1 pg). Without modifier use, zirconium, ruthenium, and rhodium mo were 3.4, 4.1, 4.6, and 4.8 pg, respectively. For chromium, the most sensitive condition was obtained with zirconium permanent with optimum pyrolysis and atomization temperatures of 1500 and 2500 °C, mo of 6.6 pg (recommended 5.5 pg); and without modifier use, rhodium, iridium, and ruthenium mo were 5.3, 8.8, 8.8, and 8.9 pg, respectively. For lead, the best modifier was also zirconium, mo of 8.3 pg for the optimum pyrolysis and atomization temperatures of 600 and 1400 °C, respectively, (recommended mo of 9.0 pg). For iridium, ruthenium, without modifier, and rhodium, mo were 14.7, 15.5, 16.5, and 16.5 pg, respectively. For all the modifiers selected in each case, the peaks were symmetrical with r2 higher than 0.99. Being analyzed (n = 10), two marine sediment reference materials (PACS-2 and MESS-2 from NRCC), the determined values, μg l−1, and certified values in brackets, were 2.17 ± 0.05 (2.11 ± 0.15) and 0.25 ± 0.03 (0.24 ± 0.01) for cadmium in PACS-2 and MESS-2, respectively. For chromium in PACS-2 and MESS-2 the values were 94.7 ± 5.6 (90.7 ± 4.6) and 102.3 ± 10.7 (106 ± 8), respectively. Finally, for lead in PACS-2 and MESS-2, the results obtained were 184 ± 7 (183 ± 8) and of 25.2 ± 0.40 (21.9 ± 1.2), respectively. For cadmium and lead in both samples and chromium in PACS-2, calibration was accomplished with aqueous calibration curves. For chromium in MESS-2, only with the standard addition technique results were in agreement with the certified ones. The limits of detection (k = 3, n = 10) obtained with the diluents were 0.1, 3.4, and 3.6 μg l−1 for cadmium, chromium, and lead, respectively.  相似文献

A comparison is made between a platform-equipped Perkin-Elmer HGA-500 and a recently designed two-step constant-temperature graphite furnace with respect to susceptibility to interference effects as well as attainable detection limits. It is shown that larger quantities of biological samples can be used with the constant-temperature furnace without suppressing the analyte signal, resulting in lower detection limits. Materials studied include NBS Olyster Tissue and Bovine Liver, and human lung tissue and human blood.  相似文献

A method for the determination of lead in blood using a tungsten coil atomizer is described. A 100 μl volume of the whole blood sample is transferred to a sampler cup containing 100 μl of water plus 300 μl of 0.25% v/v Triton X-100. After lysis of blood cells, 500 μl of 10% w/v trichloroacetic acid is added for protein precipitation and 10 μl of the supernatant solution is automatically delivered into the tungsten coil. The furnace heating program is implemented in 41 s. It is shown by the paired t-test that there is no significant difference at the 5% probability level between results obtained by the proposed method and by using a transversely heated graphite atomizer with a longitudinal Zeeman background correction. Accuracy is also assessed by employing reference materials. The proposed tungsten coil procedure presents a characteristic mass of 15 pg Pb and a detection limit of 1.9 μg Pb dl−1.  相似文献

Procedures for the electrothermal atomic absorption spectrometric determination of lead and cadmium in urine, serum and blood are developed. For serum and blood, the samples are diluted by incorporating 0.015% (w/v) Triton X-100 and 0.1% (w/v) ammonium dihydrogenphosphate to the solutions, which are then introduced directly into the furnace. A solution containing 15% (w/v) hydrogen peroxide and 0.65% (w/v) nitric acid is also introduced into the atomizer by means of a separate injection. Zeeman-based correction is recommended. Both conventional and fast-heating programs are discussed. Calibration is carried out using the standard additions method. The reliability of the procedures is checked by analyzing three certified reference materials and by recovery studies.  相似文献

Summary The electrothermal atomization of lead has been investigated for accurate determination of lead in water samples. Thiourea served to lower the atomization temperature of lead and to eliminate the interferences from chloride matrix. The addition of thiourea also allowed the accurate determination of lead irrespective of its chemical form. The absolute sensitivity (1% absorption) was 1.1 × 10–12g of lead. The method permits the direct rapid determination of lead in water samples including sea water.

Zusammenfassung Zur Verringerung der Atomisie-rungstemperatur und zur Beseitigung von Störungen durch die Chloridmatrix wird ein Zusatz von Thioharnstoff empfohlen. Dieser Zusatz gestattet außerdem eine genaue Bestimmung unabhängig von der chemischen Form, in der das Blei vorliegt. Die absolute Empfindlichkeit (1% Absorption) beträgt 1,1 · 10–12 g Pb. Das Verfahren wurde zur direkten Bleibestimmung in Wässern, einschl. Meereswasser, angewendet.

We have devised a new method for bis(tributyltin)oxide (TBTO) determination in marine sediments and mussels. This technique involves an n-hexane/methylene chloride mixture extraction and extract purification with a sodium hydroxide wash in order to eliminate interfering compounds. TBTO is then extracted again by nitric acid and converted into an inorganic tin species; the analysis has been effected using Zeeman graphite furnace-atomic absorption spectrophotometry. The method detection limit for the matrices examined is 0.004 μg TBTO g?1 (wet weight) and is sufficient for the analysis in real samples. The percentage recovery of TBTO from sediments and mussels samples is higher than 85% and 95% respectively. This method has been applied to TBTO level determination in sediments and mussels (Mytilus galloprovincialis) sampled in the harbour area in Taranto, where mussel culture activities are much developed; the TBTO levels obtained in sediments and mussels were in the range 15-47 ng g?1 (wet weight) and 11-30 ng g?1 (wet weight) respectively. Such values are comparable with those found in other harbour areas in the Mediterranean Sea.  相似文献
