Monooxo Mo(V) complexes of a N2O heteroscorpionate ligand designated (L10O) are found to exist as isolable cis and trans isomers. We have been able to trap the kinetically labile cis isomer and follow its isomerization to the thermodynamically more stable trans form. We have also followed the kinetics of isomerization between the cis and trans isomers of the corresponding dioxo Mo(VI) and W(VI) species. Here the trans is the labile isomer that spontaneously converts to the thermodynamically more stable cis. It is observed that at 60 degrees C in DMSO the Mo(VI) complex isomerizes approximately 6.5 times faster than the Mo(V) and nearly 5 times faster than the corresponding W(VI) analogs. The temperature dependence to the kinetics of the Mo(V) and Mo(VI) isomerizations give activation parameters that are similar for both oxidation states and consistent with those previously observed in [(L1O)MoOCl2] suggesting a similar twist mechanism is operating in all cases. Thus there are oxidation state, metal ion and donor atom dependent differences in isomeric stability that could have significant implications for understanding the mechanisms of both enzymatic and nonenzymatic oxo atom transfer reactions catalyzed by complexes of Mo, W and Re.  相似文献

We have used the RHF/6-31G* method to optimize the geometric parameters of three isomers of the (ClP–CCl2)2 molecule. We have established that the trans isomer is the most stable, and that it is energetically more favorable than the two cis isomers by 7.8 and 14.2 kJ/mole respectively.  相似文献

Ab initio SCF and Cl calculations have shown that the most important factor in determining the relative energies of conformational isomers in free radicals is the movement of electron density into the singly occupied orbital. This is most favorable when an electron pair is situated trans to the radical site (trans correlation effect). In the HOCO radical we find that the trans isomer is more stable than the cis by 3.3 kcal/mol while SCF calculations yield virtually no energy difference between the two isomers.  相似文献

Silver-ion high-performance liquid chromatography (Ag+-HPLC) has been shown to be effective in the resolution of most of the isomers of conjugated octadecadienoic acids (18:2), also known as conjugated linoleic acid (CLA). The CLA isomers identified in natural fats from ruminants are a mixture of numerous positional and geometric isomers from 7,9- to 12,14-18:2. Ag+-HPLC separates both geometric (trans,trans < cis/trans < cis,cis) and positional CLA isomers using the mobile phase hexane/acetonitrile (99.9:0.1). The elution volumes for the CLA isomers were not only affected by the concentration of acetonitrile (in the prepared mobile phase) but also with successive runs during the day using a prepared mobile phase batch, due to the partial solubility of acetonitrile in hexane. However, this drift does not affect the relative resolution of the CLA isomers. The addition of diethyl ether to the mobile phase partly stabilizes the solvent mixture. In order to facilitate the interpretation of Ag-+HPLC chromatograms, the relative retention volumes (RRV) were calculated for each CLA isomer. Toluene was added to all the test portions and served as an estimator of dead volume, whereas the elution of the ubiquitous 9c,11t-CLA isomer was chosen as unity (1.00). Expressing the elution of all the CLA isomers as their RRV greatly helped to standardize each CLA isomer, resulting in relatively small coefficients of variation (% CV) for the trans,trans (<1.5%) and cis/trans (<0.5%) CLA isomers. The identification of the CLA isomers was further facilitated by synthesis of authentic CLA isomers. All the geometric CLA fatty acid methyl esters (FAME) from positions 6,8- to 13,15-CLA were commercially available or synthesized by a combination of partial hydrazine reduction of known polyunsaturated fatty acids followed by alkali isomerization, isolation of products, and further iodine-catalyzed geometric isomerization. Based on expressing the elution volume as RRV and the availability of the synthetic CLA isomers, a unique reversal of the elution order of the c/t CLA isomers was found. It is also proposed that the retention times of CLA isomers by gas chromatography (GC) should be expressed as their relative retention times (RRT) relative to methyl gamma-linoleneate. The availability of CLA reference materials and the application of RRV and RRT to Ag+-HPLC and GC separations, respectively, will greatly improve in the identifications of CLA isomers.  相似文献

Chemical investigation of the marine red alga (Rhodophyta) Ceratodictyon spongiosum containing the symbiotic sponge Sigmadocia symbiotica collected from Biaro Island, Indonesia, yielded two isomers of a new and bioactive thiazole-containing cyclic heptapeptide, cis,cis-ceratospongamide (1) and trans, trans-ceratospongamide (2). Isolation of these peptides was assisted by bioassay-guided fractionation using a brine shrimp toxicity assay (Artemia salina). The structures of the ceratospongamides, which each consist of two L-phenylalanine residues, one (L-isoleucine)-L-methyloxazoline residue, one L-proline residue, and one (L-proline)thiazole residue, were established through extensive NMR spectroscopy, including (1)H-(13)C HMQC-TOCSY, and (1)H-(15)N HMBC experiments, as well as chemical degradation and chiral analysis. cis,cis- and trans,trans-ceratospongamide are stable conformational isomers of the two proline amide bonds. Molecular modeling of these two ceratospongamide isomers showed the trans, trans isomer to be quite planar, whereas the cis,cis isomer has a more puckered overall conformation. trans,trans-Ceratospongamide exhibits potent inhibition of sPLA(2) expression in a cell-based model for antiinflammation (ED(50) 32 nM), whereas the cis,cis isomer is inactive. trans,trans-Ceratospongamide was also shown to inhibit the expression of a human-sPLA(2) promoter-based reporter by 90%.  相似文献

We have previously demonstrated that the complex [(L1O)MoOCl(2)], where L1OH = (2-hydroxy-3-tert-butyl-5-methylphenyl)bis(3,5-dimethylpyrazolyl)methane, exists as both cis and trans isomers (Kail, B.; Nemykin, V. N.; Davie, S. R.; Carrano, C. J.; Hammes, B. S.; Basu, P. Inorg. Chem. 2002, 41, 1281-1291). Here, the cis isomer is defined as the geometry with the heteroatom in the equatorial position, and the trans isomer is designated as the geometry with the heteroatom positioned trans to the terminal oxo group. The trans isomer represents the thermodynamically more stable geometry as indicated by its spontaneous formation from the cis isomer. In this report, we show that for complexes of [(LO)MoOCl(2)], where LOH is the sterically less restrictive (2-hydroxyphenyl)bis(3,5-dimethylpyrazolyl)methane, only the trans isomer could be isolated, while in the corresponding thiolate containing ligand (2-dimethylethanethiol)bis(3,5-dimethylpyrazolyl)methane (L3SH) only the cis isomer could be observed. In addition, we have isolated and structurally characterized the complex [(L1O)MoO(OPh)(Cl)], a rare example of a species possessing both cis and trans phenolates. Using DFT calculations, we have investigated the origins of the differences in stability between the cis and trans isomers in these complexes and suggest that they are related to the trans influence of the oxo-group. Crystal data for [(LO)MoOCl(2)] (1) include that it crystallizes in the triclinic space group P(-)1 with cell dimensions a = 8.9607 (12) A, b = 10.596 (4) A, c = 13.2998 (13) A, alpha = 98.03 (2) degrees, beta = 103.21 (2) degrees, gamma = 110.05(2) degrees, and Z = 2. [(L1O)MoO(OPh)Cl].2CH(2)Cl(2) (2.2CH(2)Cl(2)) crystallizes in the triclinic space group P(-)1 with cell dimensions a = 12.2740 (5) A, b = 13.0403 (5) A, c = 13.6141 (6) A, alpha = 65.799 (2) degrees, beta = 64.487 (2) degrees, gamma = 65.750 (2) degrees, and Z = 2. [(L3S)Mo(O)Cl(2)] (3) crystallizes in the orthorhombic space group Pna2(1), with cell dimensions a = 13.2213 (13) A, b = 8.817 (2) A, c = 15.649 (4) A, and Z = 4. The implications of these results on the function of mononuclear molybdoenzymes such as sulfite oxidase, and the DMSO reductase, are discussed.  相似文献

Both dioxo Mo(VI) and mono-oxo Mo(V) complexes of a sterically restrictive N2O heteroscorpionate ligand are found to exist as cis and trans isomers. The thermodynamically stable isomer differs for the two oxidation states, but in each case, we have isolated the kinetically labile isomer and followed its isomerization to the thermodynamically stable form. The Mo(VI) complex is more stable in the cis geometry and isomerizes more than 6 times faster than the Mo(V) complex, which prefers the trans geometry. In OAT reactions with PPh3, the trans isomer of the dioxo-Mo(VI) reacts approximately 20 times faster than the cis isomer. Thus, there are both oxidation state and donor atom dependent differences in isomeric stability and reactivity that could have significant functional implications for molybdoenzymes such as DMSO reductase.  相似文献

Abstract

Lewisite is generally a mixture of several components with the trans isomer of lewisite being the predominant compound. A geminal isomer has not been previously reported as one of the components of the mixture. In the lewisite samples we examined, the geminal isomer, dichloro(l-chlorovinyl)arsine, comprised 2.7 per cent of the total material compared to 95.2 and less than 1 per cent, respectively, for the trans and cis isomers. The remaining fraction was not identified. The geminal isomer of lewisite has been characterized along with the trans and cis isomers using several spectroscopic techniques. Proton NMR of the geminal isomer produced a coupling constant consistent with vinylic protons in a geminal configuration. Mass spectrometry and infrared spectroscopy characterizations were based on an ethanedithiol derivative of the lewisite isomers with gas chromatography used to first separate the derivatized isomers. The electron ionization massspectra of the trans and cis derivatives were very similar, but significant differences were observed in the mass spectrum of the geminal form. Infrared absorption spectra were obtained for the trans and geminal derivatives with significant differences observed between the two, but the method was not sensitive enough to detect the cis isomer.

Antioxidant activity of hydroxystilbene derivatives in homogeneous solution   总被引:3,自引:0,他引:3  
The antioxidant activity of the cis and trans isomers of several analogues of resveratrol and pterostilbene has been investigated, especially with regard to the effect of the stereochemistry about the olefinic double bond. The antioxidant power of these compounds was estimated by measuring the rate constants for their reactions with peroxyl radicals and, with two of them, the bond dissociation enthalpy (BDE) of the phenolic O-H bond which is cleaved in the inhibition reaction. The present data show that in homogeneous solution the various hydroxystilbenes investigated behave as mild antioxidants with the notable exceptions of the trans isomer of 4 and 6, whose activities are only slightly lower than that of alpha-tocopherol (vitamin E). The rate constants of the inhibition reaction show that the antioxidant activity of the cis-hydroxystilbene is in all the examined cases worse, by a factor ranging between 2 and 6, than that of the corresponding trans isomers. This lower reactivity depends on enthalpy factors as it can be inferred by the experimental values of the O-H bond dissociation enthalpy in the two geometric isomers of 3',5'-di-tert-butyl-4'-hydroxy-3,5-dimethoxystilbene showing that the strength of the O-H bond in the cis isomer is larger by 1.8 kcal/mol. DFT calculations provide a rationalization of this result, indicating that, although the cis geometry implies a destabilization with respect to the trans species of both phenoxyl radical and parent hydroxystilbene, the destabilization of the radical is larger because the folding of the structure strongly reduces the delocalization of the unpaired electron on the styryl group. A comparison of these results with previously reported data on the proapoptotic activity of these stilbenoids suggests that these two properties are not correlated.  相似文献

Dehydrodidemnin B (DDB or aplidine), a potent antitumoral natural product currently in phase II clinical trials, exists as an approximately 1:1 mixture of two slowly interconverting conformations. These are sufficiently long-lived so as to allow their resolution by HPLC. NMR spectroscopy shows that this phenomenon is a consequence of restricted rotation about the Pyr-Pro(8) terminal amide bond of the molecule's side chain. The same technique also indicates that the overall three-dimensional structures of both the cis and trans isomers of DDB are similar despite the conformational change. Molecular dynamics simulations with different implicit and explicit solvent models show that the ensembles of three-dimensional structures produced are indeed similar for both the cis and trans isomers. These studies also show that hydrogen bonding patterns in both isomers are alike and that each one is stabilized by a hydrogen bond between the pyruvyl unit at the terminus of the molecule's side chain and the Thr(6) residue situated at the junction betwen the macrocycle and the molecule's side chain. Nevertheless, each conformational isomer forms this hydrogen bond using a different pyruvyl carbonyl group: CO(2) in the case of the cis isomer and CO(1) in the case of the trans isomer.  相似文献

The geometries and their harmonic frequencies of the difluoroethylene isomers were gradient optimized using ab initio methods at the different levels. High-level methods give very good structural parameters in comparison to experiment. The single energy of stable structure and the frontier orbital energy gap were calculated using MP4SDTQ/6-311++G(2df,2pd) method, and it is found that the single energy of stable structure of cis isomer is lower than that of trans isomer. This is consistent with the experimental result. Mulliken population analyses were also performed to interpret the internal nature of cis preference of difluoroethylene isomers.  相似文献

Ab initio calculations at the MP4(SDTQ)/6-311G//MP2/6-31G level were performed to study the structures and stabilities of the dimer of ethyl cation, (C(2)H(+)(5))(2), and related C(4)H(10)(2+) isomers. Two doubly hydrogen bridged diborane type trans 1 and cis 2 isomers were located as minima. The trans isomer was found to be more favorable than cis isomer by only 0.6 kcal/mol. Several other minima for C(4)H(10)(2+) were also located. However, the global energy minimum corresponds to C-H (C(4) position) protonated 2-butyl cation 10. Structure 10 was computed to be substantially more stable than 1 by 31.7 kcal/mol. The structure 10 was found to be lower in energy than 2-butyl cation 13 by 34.4 kcal/mol.  相似文献

The kinetics of Z-(cis)/E-(trans) isomerization of enalapril was investigated by reversed phase high-performance liquid chromatography (RP-HPLC) using a monolith ODS column under a series of different temperature and pH conditions. At a neutral pH 7, the rate (k(obs)) of Z-(cis)/E-(trans) isomerization of enalapril at 4 degrees C (9.4 x 10(-3)min(-1)) is much lower than at 23 degrees C (1.8 x 10(-1)min(-1)), while the fractional concentration of Z-(cis) isomer is always higher than that of E-(trans) isomer in the pH range 2-7. The fractional concentration of the E-(trans) isomer becomes a maximum (about 40%) in the pH range 3-6, where enalapril exists as a zwitterion. The hydrophobicity (logP(O/W)) of both isomers was estimated by high-speed counter-current chromatography (HSCCC). Normal phase HSCCC separation using a tert-butyl methyl ether-acetonitrile-20mM potassium phosphate buffer (pH 5) two-phase solvent system (2:2:3, v/v/v) at 4 degrees C was effective in partially separating the isomers, and the partition coefficient (K) of each isomer was directly calculated from the retention volume (V(R)). The logP(O/W) values of Z-(cis) and E-(trans) isomers were -0.46 and -0.65, respectively.  相似文献

[formula: see text] [1.1](3,3')-Azobenzenophane, in which two azobenzenes are cyclically connected by -CH2- chains at the meta positions, has been synthesized. The crystal structures of all isomers have been revealed. This is the first report on the crystal structure of the cis isomer of macrocyclic azobenzenes. The trans,trans isomer was slightly distorted, the trans,cis isomer highly deformed, and the cis,cis isomer unstrained. The thermal stability of cis isomers in solutions are deducible from the crystal structures.  相似文献

The radical addition reactions of organobromine compounds, XBr (X = CH2COOMe, PhCH2, CHBr2 and CCl3) with cyclohexene afforded mixtures of cis/trans isomer pairs of 1-X-2-Br-cyclohexanes. In addition to benzyl benzoyloxy derivatives are formed also, when benzoyl peroxide is used as an initiator. Owing to the great difficulties in separating these cis/trans isomer pairs, they are identified directly in their mixtures by NMR spectroscopy. In addition to one-dimensional (ID) 1H, proton decoupled 13C and DEPT-135, also two-dimensional (2D) 13C-13C INADEQUATE as well as 1H-13C HMQC experiments have been used in assigning the signals of each compound in their mixtures. The identification of each isomer was based on comparison of experimental 3J(H,H) coupling constants with theoretical ones based on the well-known Karplus type relationship. The more stable conformation for each isomer was estimated using the semiempirical AM1 molecular orbital method. The calculations support the isomer pair elucidations.  相似文献

The 1 : 1 reactions of [ClP(mu-NtBu)]2 with the difunctional aromatic amines 1,2-1-YH-2-NH2-C6H4 in the presence of Et3N give the dimeric phosph(III)azane macrocycles [{P(mu-NtBu)2(1-Y-2-HN-C6H4)]2, predominantly as the cis isomer in the case of Y=O (1.cis) and as the trans isomer for Y=S (2.trans). Model M.O. calculations suggest that the selection of the cis and trans isomers is not thermodynamically controlled. The alternative isomers 1.trans and 2.cis are generated exclusively by the deprotonation of the model intermediates [(1-Y-2-NH2-C6H4)P(mu-NtBu)]2[Y=O (3), S (4)] with nBuLi followed by cyclisation with [ClP(mu-NtBu)]2. The solid-state structures of 1.cis/trans(50 : 50), 2.cis, 3 and 4 are reported.  相似文献

The molecular structure of the trans isomer of metal-free phthalocyanine (H2Pc) is determined using the gas electron diffraction (GED) method and high-level quantum chemical calculations. B3LYP calculations employing the basis sets 6-31G**, 6-311++G**, and cc-pVTZ give two tautomeric isomers for the inner H atoms, a trans isomer having D2h symmetry and a cis isomer having C2v symmetry. The trans isomer is calculated to be 41.6 (B3LYP/6-311++G**, zero-point corrected) and 37.3 kJ/mol (B3LYP/cc-pVTZ, not zero-point corrected) more stable than the cis isomer. However, Hartree-Fock (HF) calculations using different basis sets predict that cis is preferred and that trans does not exist as a stable form of the molecule. The equilibrium composition in the gas phase at 471 degrees C (the temperature of the GED experiment) calculated at the B3LYP/6-311++G** level is 99.8% trans and 0.2% cis. This is in very good agreement with the GED data, which indicate that the mole fraction of the cis isomer is close to zero. The transition states for two mechanisms of the NH tautomerization have been characterized. A concerted mechanism where the two H atoms move simultaneously yields a transition state of D2h symmetry and an energy barrier of 95.8 kJ/mol. A two-step mechanism where a trans isomer is converted to a cis isomer, which is converted into another trans isomer, proceeds via two transition states of C(s) symmetry and an energy barrier of 64.2 kJ/mol according to the B3LYP/6-311++G** calculation. The molecular geometry determined from GED is in very good agreement with the geometry obtained from the quantum chemical calculations. Vibrational frequencies, IR, and Raman intensities have been calculated using B3LYP/6-311++G**. These calculations indicate that the molecule is rather flexible with six vibrational frequencies in the range of 20-84 cm(-1) for the trans isomer. The cis isomer might be detected by infrared matrix spectroscopy since the N-H stretching frequencies are very different for the two isomers.  相似文献

Thermal degradation of polybutadiene (PBD) in anaerobic atmosphere at 250 °C had been studied by carbon-13 nuclear magnetic resonance spectroscopy (13C NMR) before complete crosslinking. In this investigation four types of low molecular weight PBD with different 1,2-vinyl isomer content had been chosen, then pure and mixed samples of PBD were heated in different time periods. 13C NMR spectra showed that two kinds of crosslinking mechanisms occur that both of them produce methyl groups. The first mechanism is a reaction between 1,2-vinyl isomers of two PBD chains, and the second one occurs between 1,2-vinyl isomer of one chain via methylene carbon of cis or trans isomer in another chain. Also 13C NMR results showed that the presence of 1,2-vinyl isomer in the PBD structure is necessary and without it none of the mentioned reactions will occur. Furthermore isomers sequence is another important parameter which affects crosslinking. Results show that cis or trans isomer which is not adjacent to 1,2-vinyl isomer does not take part in crosslinking reaction. Moreover such cis or trans isomer can take part in second mechanism of crosslinking that 1,2-vinyl isomer was attached from head to cis or trans isomer, thus in this arrangement of isomers second mechanism of crosslinking will become dominant rather than first mechanism of crosslinking.  相似文献

The boron and aluminium dimers [Me2E(micro-py)]2 [E=B (1); Al (2)] are formed as mixtures of two isomers in which the group 13 centres are coordinated by the bridging 2-py ligands in a cis or trans manner, however, in contrast to previous studies, we find that simply heating the mixtures of these isomers of and gives the more thermodynamically stable (synthetically useful) trans isomers exclusively (the trans isomer being the only product in the case of the gallium analogue ).  相似文献
