In this work, the development of two solid-phase extraction procedures (off-line and on-line formats) for the identification and quantification of several (fluoro)quinolones in hospital sewage water by HPLC-UV is described. Both procedures are based on the use of C18 and anion exchange (SAX) sorbents for the preconcentration and clean-up steps, respectively, and all variables influencing both steps were optimised. In the off-line format, after its pH was adjusted to 2.5, sample was preconcentrated on a C18 cartridge and eluted with 4 mL of methanol/ammonia (94/6). The methanolic extract must be diluted up to 10 mL with water to allow quantitative retention of the analytes on the SAX cartridge. In the on-line format, the addition of 2.5% of NH4Cl to the sewage water sample (pH = 2.5) was necessary to increase the breakthrough volumes of the analytes in the C18 precolumn. Quantitative transfer of the (fluoro)quinolones from the C18 precolumn to the SAX precolumn was accomplished by pumping 2 mL of a mixture methanol/water (40/60, pH = 9.2) at 2 mL min(-1). Elution of the analytes from the SAX precolumn by means of the chromatographic mobile phase required the inclusion of an additional isocratic step at the beginning of the gradient program. Both off-line and on-line solid phase extraction procedures coupled to HPLC-UV were applied to the analysis of a sewage water sample collected in the sewer system at the output of the St Dimphna Hospital (Geel, Belgium). The fluoroquinolone ciprofloxacin was found in this sample and quantified at 5.8 +/- 0.4 microg L(-1) (off-line method) and 5.6 +/- 0.5 microg L(-1) (on-line method). The analysis of spiked samples containing the seven (fluoro)quinolones studied provided quantitative recoveries in all cases with low RSD values (from 6 to 12%), and all the analytes could be identified by means of their UV spectra with match factors varying from 950 to 985 depending on the (fluoro)quinolone.  相似文献

Benzotriazole ultra‐violet stabilisers are compounds widely used in personal care products, which can reach the environment after passing through wastewater treatment plants. In this work, we develop a novel method to evaluate the presence of seven compounds in marine sediments and sewage sludges using microwave‐assisted extraction followed by a clean‐up step based in on‐line solid phase extraction coupled to ultra‐high‐performance liquid chromatography with MS/MS detection. This method allows for fast and efficient extraction from the solid matrix, subsequent automatic on‐line purification and preconcentration, and analysis. For the optimised method, LOD were from 53.3 to 146 ng/kg and LOQ were in the range of 176–486 ng/kg. The method was validated for different environmental solid samples with satisfactory recoveries and relative standard deviations, between 46.1 and 83.9 and 7.8 and 15.5% (sludges) and 50.1 and 87.1% and 8.83 and 16.3% (sediments), respectively. Finally, the studied analytes were quantified in concentrations between 0.18 and 24.0 ng/g in real samples of marine sediments and sewage sludges from Gran Canaria Island (Spain).  相似文献

This study describes an SPE coupled with large volume injection (LVI) analytical method for the analysis of organochlorine pesticides, BHC (alpha, beta, delta), aldrin, endosulfan (alpha, beta), endrin, dieldrin, and DDT, from aqueous samples. Determination was carried out by GC with MS. The LODs of organochlorine pesticides were determined at 10 ng/L concentration levels, and the results show that SPE-LVI-GC/MS has the potential to accurately determine organochlorine pesticides in water, as it avoids analyte classes in the various steps of a typical extraction procedure.  相似文献

A method based on microwave‐assisted extraction (MAE) has been developed for the determination of paclitaxel and five related taxoids, namely 10‐deacetylbaccatin III (10‐DAB III), cephalomannine, 10‐deacetylpaclitaxel (10‐DAT), 7‐xyl‐10‐ deacetylpaclitaxel (7‐xyl‐10‐DAT), and 7‐epi‐10‐deacetylpaclitaxel (7‐epi‐10‐DAT) in Taxus species in this study. The influential parameters of the MAE procedure were optimized, and the optimal conditions were as follows: extraction solvent 80% ethanol solution, solid/liquid ratio 1:10 (g/mL), temperature 50°C, and three extraction cycles, each cycle 10 min. The method validation for LC‐MS/MS analysis was performed. The LOD and LOQ were 3.16–9.20 and 12.20–30.45 ng/mL, respectively. Repeatability and reproducibility for the six taxiods with RSD ranged from 2.78 to 3.85% and from 5.26 to 6.60%. The recoveries of the method for the six taxoids were 92.6–105.6%. The developed MAE‐LC‐MS/MS method was also successfully applied to determine the contents of six taxoids in different Taxus species.  相似文献

Dieldrin is a moth-proofing agent that was banned by the Stockholm Convention in 2001. The amount of dieldrin in wool products was measured by a microwave-assisted extraction (MAE) method. The optimal conditions were as follows: extraction solvent, acetone/n-hexane (1:1 v/v); extraction temperature, 110 degrees C; extraction time, 10 min; solvent volume, 25 mL. When six samples were used, dieldrin contents determined by GC with the proposed MAE agreed closely with those by the Japanese official method using GC with solvent extraction and cleanup by column chromatography. The proposed MAE has two merits. First, the pretreatment of the MAE needs only 4 h for 11 samples, while that using the Japanese official method needs 2 days for six samples. Second, the volume of organic solvents used for the proposed method was only about one-tenth of that used in the Japanese official method. Our proposed method seems to be easy and useful for daily (routine) tests. Dieldrin contents of 28 used wool products, which were obtained from local clothing shops and ordinary homes, were determined by GC with the proposed MAE, and six products contained dieldrin (0.310-175 ppm). The dry cleaning of the woolen yarn containing 175 ppm dieldrin did not remove a significant amount of dieldrin. Therefore, it seems likely that dieldrin is still distributed slightly but widely throughout the world.  相似文献

Veltol® (2-methyl 3-hydroxy 4-pyrone) and Veltol-Plus® (2-ethyl 3-hydroxy 4-pyrone) are patented flavor ingredients in food products. Only Veltol® can occur naturally, but both Veltol® and Veltol-Plus® are often added to food products. In order to monitor the use of these compounds in food products, lower detection limits were needed. The Solid Phase Microextraction (SPME) technique for beverages and SPME coupled with Microwave Assisted Extraction (MAE) for solid food samples were studied. The influence of the pH, salt content, SPME adsorption time, GC inlet conditions, and the conditioning of the SPME fiber were investigated. Different food products were tested including coffee, beverages, chewing gums and potato chips. The coupled MAE and SPME shows good results for solid food samples. The reproducibility of the technique was less than 13%RSD and the detection limit was 10 ppb for Veltol® and 2 ppb for Veltol-Plus® using the SIM mode in GC/MS. The technique also shows good selectivity for the target compounds investigated in different food samples.  相似文献

A rapid, sensitive and selective liquid chromatography-tandem spectrometry method was developed and validated for determination of paeoniflorin in rat plasma using geniposide as the internal standard. The samples were pretreated with solid-phase extraction using Extract-Clean cartridges. Separation of paeoniflorin and IS was achieved on a reversed-phase C18 column (50x4.6 mm i.d.) with a mobile phase made up of acetonitrile and 0.05% formic acid (25:75, v/v) at a flow rate of 0.5 mL/min. Detection was carried out on a triple quadrupole tandem mass spectrometer by multiple-reaction monitoring and an electrospray ionization source was employed as the ionization source. The lower limit of quantification obtained was 4 ng/mL (n=6) using 200 microL plasma with an accuracy of -3.67% (relative error) and a precision of 4.13% (relative standard deviation). A good linearity was found in the range of 4-1000 ng/mL. The intra- and inter-day relative standard deviations in the measurement of quality control samples 10, 150 and 800 ng/mL ranged from 3.73 to 4.94% and from 4.31 to 6.56%, respectively. The accuracy was from -3.93 to -1.11% in terms of relative error. The analyte and IS were stable in the battery of stability studies. This method was successfully applied to a pharmacokinetic study of paeoniflorin after a single oral administration of 53.36 mg/kg paeoniflorin to rats.  相似文献

建立了基质分散固相萃取-液相色谱-串联质谱(dSPE-LC-MS/MS)定量检测4种动物源性食品基质中硝基咪唑类药物及其代谢物的方法。样品(2.0 g)用乙酸乙酯提取后浓缩,经正己烷脱脂、50 mg乙二胺-N-丙基硅烷(PSA)吸附剂吸附净化后,过0.22 μm亲水聚四氟乙烯(PTFE)滤膜。采用C18柱分离,在电喷雾电离(ESI)源和选择反应监测(SRM)模式下检测,基质匹配内标法定量。在0.5~20.0 μg/L范围内,硝基咪唑类药物及其代谢物呈现良好的线性关系,相关系数(r2)>0.99;方法的检出限为0.1~0.5 μg/kg;在1.0、3.0和10.0 μg/kg的加标水平下,硝基咪唑类药物及其代谢物的回收率为84.2%~120.8%,相对标准偏差为2.0%~16.2%(n=6)。该法准确、快速,成本低,易操作,能够满足动物源性食品中硝基咪唑类药物及其代谢物残留的监测要求。  相似文献

A sensitive and efficient analytical method for triclosan (TCS) determination in water, which involves enrichment with bamboo‐activated charcoal and detection with HPLC‐ESI‐MS, was developed. The influence of several operational parameters, including the eluant and its volume, the flow rate, the volume andacidity of the sample, and the amount of bamboo‐activated charcoal, were investigated and optimized. Under the optimum conditions, linearity of the method was observed in the range of 0.02–20 μg/L, with correlation coefficients (r2) >0.9990. The limit of detection was 0.002 μg/L based on the ratio of chromatographic signal to baseline noise (S/N = 3). The spiked recoveries of TCS in real water samples were achieved in the range of 97.6–112.5%. The proposed method was applied to analyze TCS in real aqueous samples. All the surface water samples collected in Xiaoqing River had detectable levels of TCS with concentrations of 42–197 ng/L.  相似文献

A solid‐phase extraction (SPE) method was developed to extract 14 pesticides simultaneously from environment samples using cigarette filter as the sorbent before gas chromatography‐mass spectrometry (GC‐MS) analysis. Parameters influencing the extraction efficiency, such as the sample loading flow rate, eluent and elution volume, were optimized. The optimum sample loading rate was 3 mL/min, and the retained compounds were eluted with 6 mL of eluent at 1 mL/min under vacuum. Good linearity was obtained for all the 14 pesticides (r2>0.99) from 0.1 to 20 μg/L for water and from 2 to 400 μg/kg for soil samples. The detection limits (signal‐to‐noise=3) of the proposed method ranged from 0.01 to 0.20 μg/L for water samples and from 0.42 to 6.95 μg/kg for soil samples. The developed method was successfully applied for determination of the analytes in real environmental samples, and the mean recoveries ranged from 76.4 to 103.7% for water samples and from 79.9 to 105.3% for soil samples with the precisions (relative standard deviation) between 2.0 and 13.6%.  相似文献

A simple and specific analytical method was developed and tested for the determination of pharmaceuticals in mollusc samples. A combination of microwave-assisted micellar extraction (MAME) and solid-phase extraction (SPE) using a non-ionic surfactant, polyoxyethylene 10 lauryl ether, was examined to extract and determine simultaneously a group of pharmaceuticals such as carbamazepine, clorfibric acid, ketoprofen, naproxen, bezafibrate and ibuprofen by liquid chromatography using UV-diode array detector. The MAME extraction performance was evaluated by studying various parameters such as the volume and concentration of surfactant and microwave conditions. Finally, an OASIS HLB cartridge was used as an optimum SPE sorbent to clean up the extracts and preconcentrate the selected analytes. The proposed method showed satisfactory linearity and reproducibility (between 3 and 15%), as well as detection limits ranging from 30 to 220 ng/g. Finally, the method was successfully applied to the determination of the target pharmaceuticals in various kinds of mollusc samples. This study has demonstrated that microwave-assisted micellar extraction with solid-phase extraction may be used as a viable alternative to conventional methods for the extraction of pharmaceuticals in this type of matrices.  相似文献

A multiresidue method for the determination of more than 80 pesticides in water has been developed and validated. The proposed method is based on SPE followed by GC coupled to MS/MS. Different variables affecting SPE procedure, such as cartridges, sample volume and solvents were studied, and mass spectrometric conditions were optimised in order to increase selectivity and sensitivity. Calibration curves were linear over the range of 0.03-0.5 microg/L. Recoveries were in the range of 70-110% and repeatability was below 20% for the lowest calibration point. LODs ranged from 0.001 to 0.025 microg/L and LOQs from 0.003 to 0.076 microg/L. Finally, the method was successfully applied to the analysis of water samples from southeast of Spain.  相似文献

A rapid and solvent‐free procedure for the determination of 4‐tert‐octylphenol and 4‐nonylphenol isomers in aqueous samples is described. The method involves in‐situ acetylation and microwave‐assisted headspace solid‐phase microextraction prior to their determination using gas chromatography–ion trap mass spectrometry operated in the selected ion storage mode. The dual experimental protocols to evaluate the effects of various derivatization and extraction parameters were investigated and the conditions optimized. Under optimized conditions, 300 μL of acetic anhydride mixed with 1 g of potassium hydrogencarbonate and 2 g of sodium chloride in a 20 mL aqueous sample were efficiently extracted by a 65 μm polydimethylsiloxane‐divinylbenzene fiber that was located in the headspace when the system was microwave irradiated at 80 W for 5 min. The limits of quantitation were 5 and 50 ng/L for 4‐tert‐octylphenol and 4‐nonylphenol isomers, respectively. The precision for these analytes, as indicated by relative standard deviations, were less than 8% for both intra‐ and inter‐day analysis. Accuracy, expressed as the mean extraction recovery, was between 74 to 88%. A standard addition method was used to quantitate 4‐tert‐octylphenol and 4‐nonylphenol isomers, and the concentrations ranged from 120 to 930 ng/L in various environmental water samples.  相似文献

In the work, a rapid, simple and high-throughput sample preparation method was developed for the determination of sulfonamide (SA) antibiotic residues in chicken breast muscle. The extraction and clean-up were online combined and up to 20 samples can be treated simultaneously in 6 min. The SAs were first extracted with acetonitrile under the action of microwave energy, and then the extract was directly introduced into the SPE column for on-line clean-up and concentration. Subsequently, the SAs eluted from the SPE column were determined by liquid chromatography-tandem mass spectrometry. The precisions of extraction results of 20 samples were in the range of 4.9-7.4%. The limits of detection and quantification obtained were in the range of 2.4-3.6 ng/g and 8.6-11.3 ng/g for SAs, respectively. The recoveries of SAs obtained by analyzing chicken muscles at three fortified levels (10, 50 and 500 ng/g) were in the range of 82.6-93.2%. The results of the validation process prove that the proposed method is suitable for treating numbers of complex samples simultaneously in a short time.  相似文献

Investigations were performed along the Danube river at Budapest (Hungary) by collecting water and sediment samples simultaneously for 1 year in order to clarify the possible hazard of selected acidic pharmaceuticals (ibuprofen, naproxen, ketoprofen, and diclofenac) on the water supply used for the production of drinking water by bank filtration. In the case of water samples, the sample preparation procedure included solid phase extraction (SPE), meanwhile, in the case of sediment samples, microwave-assisted extraction (MAE) followed by dispersive matrix extraction (DME) for pre-cleaning as well as SPE for enrichment. The quantification was carried out using gas chromatography–mass spectrometry (GC–MS). The calculated recoveries were 97–99% (± 7%) for the water and 95–103% (± 12%) for the sediment samples. In the river water, ketoprofen concentration was always below the limit of quantification (LOQ) level; ibuprofen, naproxen and diclofenac could be quantified in the range of 8–50, 2–30, 7–90 ng/L. In sediments, only naproxen and diclofenac were found in the range of 2–20 and 5–38 ng/g, respectively. According to the obtained results, the concentration ratios of the two phases linearly depended on the total organic carbon content (TOC) of the sediments at each sampling date. The linear regressions were 0.925 and 0.946 for naproxen and diclofenac, respectively.  相似文献

A simple and reliable ultrasound‐assisted solid‐phase dispersion extraction coupled with ion chromatography was developed for the determination of aminophenols and phenol. The highly viscous hair colorant was dispersed in solvents using anhydrous sodium sulfite having dual functions of dispersant and antioxidant. The use of anhydrous sodium sulfite did not change the sample volume because it could completely dissolve in solution after matrix dispersion. The extraction and cleanup were combined in one single step for simplifying operation. The extraction process could be rapidly accomplished within 9 min with high sample throughput under the synergistic effects of vibration, ultrasound, and heating. Satisfactory linearity was observed with correlation coefficients higher than 0.9992, and the limits of detection varied from 0.02 to 0.09 mg/L. The applicability of the proposed method was demonstrated by measuring the concentrations of aminophenols and phenol in 32 different commercial hair color products. The recoveries ranged from 86.4–101.2% with the relative standard deviations in the range of 0.52–4.3%. The method offers an attractive alternative for the analysis of trace phenols in complex matrices.  相似文献

Tea polyphenols are well known for their beneficial health effects that involve their anti-carcinogenic, anti-mutagenic, anti-pathogenic and anti-oxidative properties. The main polyphenols of green tea are favan-3-ols (catechins) and their corresponding gallate compounds, which constitute about one-third of the dry weight of tea leaves. Their main ingredients are (+)-catechin (C), (−)-epicatechin (EC), (−)-gallocatechin (GC), (−)-epigallocatechin (EGC), (−)-catechin gallate (CG), (−)-epicatechin gallate (ECG), (−)-gallocatechin gallate (GCG) and (−)-epigallocatechin gallate (EGCG). Each has slightly different biological properties. We have developed a method to simultaneously analyze all these compounds in plasma and urine. The samples were first incubated with β-d-glucuronidase and sulfatase to release the catechin residues from their corresponding conjugates for subsequent extraction by selective solid phase column, Waters Oasis HLB extraction cartridges. The extracted molecules were resolved by reversed phase HPLC and monitored by coulometric chemical detection on a CoulArray detector. All eight catechin compounds were analyzed in a single chromatogram within 25 min. For plasma and urine analyses, good linearity (>0.9950) was validated in the range 10-2000 and 10-5000 ng/ml, respectively. The coefficients of variance (CV) were less than 5%. Absolute recovery was greater than 85% and detection limit was 5 ng/ml. The chromatogram exhibited minimal interference as a result of the highly selective solid phase extraction and CoulArray detection.  相似文献

根据新试剂对若丹宁偶氮苯甲酸(RABA)与钯的显色反应及C18固相萃取小柱对显色络合物的固相萃取,建立了一种测定痕量钯的新方法,在pH为2.0~4.0的HCl-邻苯二甲酸氢钾(HCl-KHP)缓冲介质中,在CTMAB存在下,钯与RABA发生反应形成1:1的稳定络合物,该络合物可用C18固相萃取小柱富集,小柱上富集的络合物用乙醇洗脱后用光度法测定,在富集后的测定液中,络合物最大吸收波长为500 nm,摩尔吸光系数ε=1.36×105 L·mol-1·cm-1,Pd2 量在0.1~1.0 μg/mL内符合比尔定律,方法用于催化剂中钯的测定.  相似文献
