The native fluorescence intensity of desipramine was enhanced in the presence of β-cyclodextrin in aqueous solution. The inclusion complex formation between these compounds was studied by spectrofluorimetry. A stable complex with a 2: 1 stoichiometry of β-cyclodextrin to desipramine was formed (logβ2 = 9.29 ± 0.01). In the presence of an optimum concentration of β-cyclodextrin, the fluorescence intensity was linearly proportional to desipramine concentration in the range of 0.1–100 μg/mL (7.2 × 10?7?1.0 × 10?4 M) with a limit of detection of 7 × 10?8 M. The method was successfully applied to the detection of desipramine in its tablets.  相似文献

Aqueous solutions of -cyclodextrin (-CD) or 2,6-di-o-methyl--cyclodextrin (DM--CD) and dodecylethyldimethylammonium bromide (D12EDMAB) have been studied from speed of sound (u) data at 298.15 K, using a pulse-echo-overlap technique. The molecular encapsulation process of the surfactant monomer into the cyclodextrin cavity and its effect in the micellization process of the surfactant have been analyzed from theu measurements: I) as a function of [D12EDMAB] in the presence of several initial cyclodextrin concentrations (-CD or.DM--CD); II) as a function of [cyclodextrin] (-CD or DM--CD), for an initial micellar solution of D12EDMAB and; III) as a function of the [cyclodextrin]/[surfactant] stoichiometric concentrations. Both inclusion complexes formed (-CDD12EDMAB) and (DM--CDD12EDMAB) have stoichiometries of 11, and their association constantK have been determined using a model proposed in this work, based on the additivity of the different contributions of the involved species to the speed of sound. The apparent critical micellar concentration, cmc*, of D12EDMAB is found to increase linearly upon the addition of cyclodextrin (-CD or DM--CD). The free surfactant concentration in the micellar region, [D12EDMAB]f, decreases in the presence of -CD and slightly increases in the presence of DM--CD. The influence of the parcial methylation of the -cyclodextrin (-CDDM--CD) and of the polar head of the surfactant (D12TAB D12EDMAB) on the complextion and micellar parameters are also discussed.Supplementary material available: Tables of speed of sound (14 pages) are available from the authors.  相似文献

The penetration of ampicillin molecule into the β-cyclodextrin cavity in aqueous solution is studied by 1 H NMR, 2D COSY and ROESY spectroscopy. Obtained data suggest that the phenyl moiety of ampicillin is included inside the hydrophobic cavity of β-cyclodextrin molecule in the solution, and the mode and depth of this inclusion is confirmed on the basis of 2D ROESY spectral data.  相似文献

It is established that the rate of peroxidase-catalyzed oxidation of flavonoid quercetin is increased by 20% in the presence of macrocyclic complexing agent β-cyclodextrin. The comparison of the kinetic parameters of the indicated reaction in the presence and in the absence of β-cyclodextrin shows that its introduction does not significantly influence the specificity of the enzyme with respect to the reducing substrate (characterized as k cat/K M ratio), while the increase in the reaction rate does not depend on the duration of the incubation of quercetin with β-cyclodextrin. It is assumed that the increase of the reaction rate is associated with nonspecific interaction between β-cyclodextrin and quercetin oxidation product.  相似文献

The inclusion complexes of 2-naphthyloxyacetic acid (NOA) and 1-naphthylacetic acid (NAA) with -cyclodextrin have been investigated in aqueous solution. It has been demonstrated that the naphthalene derivatives form 1:1 complexes when included in the cyclodextrin. A possible structure is proposed, having an axial inclusion of the naphthalene derivatives. In the case of the -CD: NOA complex, the naphthyl moiety is included in the cyclodextrin and the acetic acid group protrudes from the cavity, while NAA is only partially included because of the steric effect of the group in position 1. Association constants of 560±100 M–1 and 100±50 M–1 have been calculated for the -CD: NOA and -CD: NAA complexes, making use of the increment in the fluorescene emission produced in the inclusion process.  相似文献

Nanocomposite of β-cyclodextrin and graphene sheet (β-CD/GS) was successfully prepared, which exhibited high stability in aqueous solution. When used in electrochemical detection of dopamine, the β-CD/GS modified carbon electrode showed low detection limit, broad linear range, along with good ability to suppress the background current from large excess ascorbic acid. The electrochemical reaction of dopamine on the β-CD/GS showed a mass diffusion-controlled process, which was different from the adsorption-controlled process on the unmodified graphene sheet.  相似文献

SAPO-34 molecular sieves were synthesized by the addition of β-cyclodextrin(β-CD) as crystal growth inhibitor using metakaolin as silicon and aluminum sources. Properties of the obtained samples were characterized by XRD,SEM,N2adsorption–desorption,FTIR,XRF,EDX,NH293-TPD andSi MAS NMR. When β-CD was added,crystal size of the SAPO-34 crystals decreased. Variation of Si content from the crystal center to surface decreased while total Si content hardly changed.29 Si MAS NMR results showed that β-CD contributed to better Si dispersion and decreased the size of Si(4Si) patches. Moreover,the MTO(methanol-to-olefin) process was conducted to investigate the influence of β-CD on catalytic performance. The synthesized sample with molar ratio of β-CD/Al2O3 equaling 0.055 remained active for 610 min while the sample synthesized without β-CD for only 280 min,which indicates that the lifetime of catalyst synthesized with β-CD is greatly prolonged.  相似文献

We simulated the docking of α-lipoic acid (α-LA) in β-cyclodextrin (β-CD) using two models. We considered in this study complexes formed by 1:1 host–guest stoichiometry in vacuo and in aqueous phase, using PM6, DFT and ONIOM2 hybrid calculations. The results obtained with PM6 method clearly indicate that the complexes formed are energetically favored with or without solvent, model 2 (α-LA entering the cavity of β-CD from its wide side by COOH group) is found more favored than model 1 (α-LA entering into the cavity of β-CD from its wide side by cyclic group), the preference being greater in the case of ONIOM2 calculations. In addition, NBO analysis gives that mutual interactions between the donor and acceptor orbitals of α-lipoic acid and β-CD plays an important role to the stabilization of such a complex. Finally, 1H nuclear magnetic resonance (NMR) chemical shifts of free and complexed α-LA were calculated by the Gauge-Including Atomic Orbital (GIAO) method and compared with available experimental data. The results of GIAO calculations were analyzed and discussed.  相似文献

An efficient and facile method for the synthesis of N-substituted pyrroles in moderate to good yields by the Paal–Knorr reaction of γ-diketones with amines in the presence of β-cyclodextrin in aqueous media has been developed.Moreover,this process tolerated diamines(e.g.,para-,meta- or orthophenylenediamine)to construct bis-pyrrole or mono-pyrrole derivates.β-Cyclodextrin can be recovered easily after the reactions and reused without evident loss in activity.  相似文献

Acetaminophen is a well-known drug commonly used to provide pain relief, but it can also lead to acute liver failure at high concentrations. Therefore, there is considerable interest in monitoring its concentrations. Sensitive and selective acetaminophen electrochemical sensors were designed by cycling a glassy carbon electrode (GCE) to high potentials in the presence of β-CD in a phosphate electrolyte, or by simply activating the GCE electrode in the phosphate solution. Using cyclic voltammetry, adsorption-like voltammograms were recorded. The acetaminophen oxidation product, N-acetyl benzoquinone imine, was protected from hydrolysis, and this was attributed to the adsorption of acetaminophen at the modified GCE. The rate constants for the oxidation of acetaminophen were estimated as 4.3 × 10–3 cm2 s–1 and 3.4 × 10–3 cm2 s–1 for the β-CD-modified and -activated electrodes, respectively. Using differential pulse voltammetry, the limit of detection was calculated as 9.7 × 10–8 M with a linear concentration range extending from 0.1 to 80 μM. Furthermore, good selectivity was achieved in the presence of caffeine, ascorbic acid and aspirin, enabling the determination of acetaminophen in a commercial tablet. Similar electrochemical data were obtained for both the β-CD-modified and activated GCE surfaces, suggesting that the enhanced detection of acetaminophen is connected mainly to the activation and oxidation of the GCE. Using SEM, EDX and FTIR, no evidence was obtained to indicate that the β-CD was electropolymerised at the GCE.

Crystal structure of β-cyclodextrin (β-CD) complexes with sorbic acid, usually as food preservative, has been determined by single-crystal X-ray diffraction at 113 K. The space group of β-cyclodextrin-sorbic acid complex is P1 with unit cell dimensions of a = 15.284(3) Å, b = 15.402(3) Å, c = 17.981(4) Å, α = 99.67(3)°, β = 112.83(3)°, γ = 102.48(3)° and Z = 1. The result indicates that the β-CD molecules form head-to-head dimers which pack in the intermediate mode. Each dimer contains two guest molecules whose methyl groups are located at the dimer interface while the carboxyl groups protrude from the β-CD primary faces. Water molecules (25.5) are distributed outside the cyclodextrin cavity over 31 sites. Furthermore, nuclear magnetic resonance spectroscopy (1H NMR) has been employed to investigate the inclusion behavior between the host β-CD and guest sorbic acid in aqueous solution. The results obtained enabled us to structurally characterize the β-CD inclusion complex with sorbic acid.  相似文献

The inclusion of α-lipoic acid (LA) in β-cyclodextrin (β-CD) by increasing the aqueous solubility and photostability can enhance its medicinal use in the oral administration. Different preparation methods were employed to obtain an α-lipoic acid-β-cyclodextrin (LA-β-CD) inclusion complex and to determine the physical–chemical characteristics and the interactions present in this compound. The formation of the solid inclusion compound was confirmed by X-ray powder diffraction, differential scanning calorimetry (DSC) and infrared spectroscopy (FTIR). FTIR and DSC data confirm the new obtained compound. The crystalline structure of this compound belongs to the monoclinic system with four molecules in the unit cell. 1H NMR spectroscopic method was employed to study the inclusion process in aqueous solution. Job plots derived from the 1H NMR spectral data demonstrated an 1:1 stoichiometry of the inclusion complex in liquid state. 2D NMR data suggest the orientation of LA with the carboxyl group near to narrower rim of the β-CD.  相似文献

The ligand substitution in Eu(AA)3 · Phen-CDCl3-TFA systems, where AA is acetylacetone, Phen is 1,10-phenanthroline, and TFA is trifluoroacetic acid was studied at various molar ratios (m) of competing ligands (m = TFA/AA) using NMR (1H, 19F) and luminescence spectroscopy. The formation of mixed Eu(AA)2(TFA) · Phen and Eu(AA)(TFA)2 · Phen complexes in the resultant solutions was attested. The luminescence spectra were studied. The competitive capacity of TFA was ascertained to be higher than that of AA. Depending on the m value, the substitution of acido ligands was shown to occur successively according equations Eu(AA)3 · Phen + (TFA) n = Eu(AA)3 − n (TFA) n · Phen + (AA) n (n = 1, 2, 3).  相似文献

UV spectroscopy was used to study the protolytic properties and determine the ionization constants of p-aminobenzoic acid in the presence of β-cyclodextrin. Formation of supramolecular structures of 1: 1 composition was established. Stability constants of the β-cyclodextrin-p-aminobenzoic acid inclusion complex were calculated, as were the thermodynamic parameters (ΔG°, ΔH°, and ΔS°) of its formation. The complexing process between β-cyclodextrin and p-aminobenzoic acid was found to occur spontaneously in the temperature range under investigation while being accompanied by energy liberation and leading to a reduction in the system’s entropy.  相似文献

The complexation process between racemic flurbiprofen and β-cyclodextrin in solution was investigated by 1D and 2D proton NMR spectroscopy. In the presence of β-cyclodextrin, the aromatic protons of flurbiprofen were the most affected, suggesting a strong involvement of the phenyl groups in the inclusion mechanism. The stoichiometry of the complex was determined by the method of continuous variation, using the chemical induced shifts of both host and guest protons. The association constant, Ka of the obtained complex was calculated and found to be 2483.8 M?1. On the other hand, signals belonging to the protons associated with the carboxyl group are split in the presence of β-cyclodextrin indicating enantiomeric differentiation. Rotating frame NOE spectroscopy, (ROESY), was used to ascertain the solution geometry of the host–guest complex. The result suggested that the flurbiprofen molecule fully penetrates the β-cyclodextrin cavity with the carboxyl group protruding from the primary hydroxyl side and the phenyl group close to the secondary rim.  相似文献

The interactions of brilliant cresyl violet (BCV) with herring sperm DNA in γ-cyclodextrin (γ-CD) supramolecular system were studied by UV-vis absorption spectroscopy and cyclic voltammetry (CV). Both UV-vis absorption and CV data show that the interaction of BCV with DNA depends on the concentration ratio of BCV to DNA (R), the initial concentration of BCV and γ-CD. The binding constants of BCV monomer, (BCV)2 dimer and (BCV)2-γ-CD inclusion complex with DNA are 1.64 × 105, 2.56 × 104 and 2.32 × 103 M−1, respectively. It was observed that γ-CD can affect the interactive mode of BCV with DNA. If R is larger than 0.5, the (BCV)2-γ-CD inclusion complex will retain intact and bind to DNA via the electrostatic attraction forces. By contrast, when R is smaller than 0.5, the inclusion complex will be partially dissociated and the free BCV monomer is intercalated into the double-helix structure of DNA attributing to the more favorable microenvironment of DNA for the BCV monomer. Our work postulates the importance of the initial concentration of dye and host molecule on the interaction of dye with DNA in living bodies.  相似文献

Fluconazole (FLZ) is a synthetic, bistriazole antifungal agent, effective in treating superficial and systemic infections caused by Candida species. Major challenges in formulating this drug for clinical applications include solubility enhancement and improving stability in biological systems. Cyclodextrins (CDs) are chiral, truncated cone shaped macrocyles, and can easily encapsulate fluconazole inside their hydrophobic cavity. NMR spectroscopy has been recognized as an important tool for the interaction study of cyclodextrin and pharmaceutical compounds in solution state.  相似文献

Studies on the effect of-cyclodextrin (BCD) and its derivatives on the selectivity in hydroxymethylation of guaiacol by formaldehyde was carried out. Fairly high selectivity with respect to isovanillyl alcohol formation was achieved. Significantly, the selectivity-enhancing effects of 2,6-di-O-methyl-BCD was much larger, giving rise to 22% more of isovanillyl alcohol formation than BCD and its polymer. UV, fluorescence,1H-NMR spectroscopic and potentiometric studies were also carried out to determine the orientation of guaiacol inside the BCD cavity.  相似文献
