The suitability of micellar electrokinetic capillary chromatography (MEKC) with laser-induced fluorescence (LIF) detection for simultaneous determination of two arginine analogues (homocysteine and homoarginine) and five closely related metabolites (asymmetric dimethyl-l-arginine, dimethyl-l-arginine, monomethyl-l-arginine, citrullin, and ornithine) in fluids from type 2 diabetics with peptic ulcer bleeding (PUB) has been investigated. 5-Carboxyfluorescein succinimidyl ester (CFSE) was chosen as the fluorescent labeling reagent and non-endogenous phenylpropanolamine (PPA) as the internal standard. Conditions affecting derivatization and separation were optimized. Under the optimum conditions, maximum derivatization could be achieved in 20 min at room temperature. Complete baseline separation was achieved in 10 min, and the relative standard deviations (RSD) of migration times and corrected peak areas were <3 % for intra-day assay and <5 % for inter-day assay. Limits of detection (LODs) were 0.12–1.70 nM for the eight analytes, which are well below the concentrations expected in real fluids. Compared with previously reported methods, 5 to 600-fold improvements in sensitivity were achieved by use of LIF detection. Sample preparation and analysis time were short and the derivatives of the analytes were highly stable. The method was fully validated with real plasma and urine and recoveries of spiked compounds were 95–102 % with the RSD <4 %.

A gas chromatography–mass spectrometric method was developed in this study in order to determine fluoride in plasma and urine after derivatization with 2-(bromomethyl)naphthalene. 2-Fluoronaphthalene was chosen as the internal standard. The derivatization of fluoride was performed in the biological sample and the best reaction conditions (10.0 mg mL−1 of 2-(bromomethyl)naphthalene, 1.0 mg mL−1 of 15-crown-5-ether as a phase transfer catalyst, pH of 7.0, reaction temperature of 70 °C, and heating time of 70 min) were established. The organic derivative was extracted with dichloromethane and then measured by a gas chromatography–mass spectrometry. Under the established condition, the detection limits were 11 μg L−1 and 7 μg L−1 by using 0.2 mL of plasma or urine, respectively. The accuracy was in a range of 100.8–107.6%, and the precision of the assay was less than 4.3% in plasma or urine. Fluoride was detected in a concentration range of 0.12–0.53 mg L−1 in six urine samples after intake of natural mineral water containing 0.7 mg L−1 of fluoride.  相似文献

Capillary electrophoresis (CE) was used to separate the neptunium oxidation states Np(IV) and Np(V), which are the only oxidation states of Np that are stable under environmental conditions. The CE setup was coupled to an inductively coupled plasma mass spectrometer (Agilent 7500ce) using a Mira Mist CE nebulizer and a Scott-type spray chamber. The combination of the separation capacity of CE with the detection sensitivity of inductively coupled plasma mass spectrometry (ICP-MS) allows identification and quantification of Np(IV) and Np(V) at the trace levels expected in the far field of a nuclear waste repository. Limits of detection of 1?×?10-9 and 5?×?10-10 mol L-1 for Np(IV) and Np(V), respectively, were achieved, with a linear range from 10-9 to 10-6 mol L-1. The method was applied to study the redox speciation of the Np remaining in solution after interaction of 5?×?10-7 mol L-1 Np(V) with Opalinus Clay. Under mildly oxidizing conditions, a Np sorption of 31% was found, with all the Np remaining in solution being Np(V). A second sorption experiment performed in the presence of Fe2+ led to complete sorption of the Np onto the clay. After desorption with HClO4, a mixture of Np(IV) and Np(V) was found in solution by CE–ICP–MS, indicating that some of the sorbed Np had been reduced to Np(IV) by Fe2+.  相似文献

A sensitive method based on derivatization with pentafluorophenylhydrazine then headspace gas chromatography–mass spectrometry has been used for analysis of malondialdehyde in human urine. Preparation of urine sample by one-step derivatization/evaporation was performed by reaction of malondialdehyde with pentafluorophenylhydrazine in a headspace vial for 10 min; the derivatives were then injected in GC–MS analysis. The reaction was performed at pH 3, and total analysis time was 35 min. The method detection limit was 0.04 μg L?1. For MDA concentrations of 2.0 and 10.0 μg L?1 the relative standard deviation was less then 5%. The concentration of MDA in urine was measured to be 0.199 ± 0.252 μmol g?1 creatinine (0.022 ± 0.028 μmol mmol?1 creatinine).  相似文献

A liquid chromatographic tandem mass spectrometric validated method was developed for the detection of chemicals attributing color, flavor, taste and medicinal properties to saffron (Crocus sativus L. stigma). Ultrasonic extractions of saffron stigmas were followed by LC procedure with Pinnacle II Cyano (5 μm 150 × 2.1 mm) column and acetonitrile: water (70:30, v/v) as mobile phase. Deprotonated ions formed by a turbo ion spray in negative MS mode were used to detect the analytes. MS–MS detection was by monitoring precursors (m/z) fragmentations; of 149 → 113 (safranal), 327 → 283 (crocetin), 329 → 167 (picrocrocin), 355 → 327 (dimethyl crocetin), 489 → 327 (crocin E), 535 → 489 (carotenes), 651 → 327 (crocin C), 813 → 652 (crocin B), 975 → 651 (crocin A) and 1,137 → 813 (crocin F). The method was validated for linearity, precision, repeatability and specificity.  相似文献

Cistrans stereochemistry of 2-(2-chlorobenzylideneamino)phenol (1) and 2-(2-chlorophenyl imino)methyl)phenol (2) (Schiff bases) was studied by GC-MS spectrometry, and cistrans conversion of the two compounds in solution was investigated by GC. Thus, 2 exists as a sole trans configuration and its conversion to cis was unsuccessful in any circumstance, while compound 1 exists as a mixture of two configurations in solution, occurrence of which were dependent on the temperature, heating time, solvent, and acid catalyst. A liquid chromatography coupled with photodiode array detection and electrospray ionization tandem mass spectrometry method was first developed for a chemical fingerprint analysis of Euonymus alatus (Thuhb) siebold (EAS) and rapid identification of major compounds in the fingerprints. Fingerprint profiles were found to be consistent for the herbs acquired from different locations, but the relative abundance of peaks was varied. Twelve peaks were chosen as the common peaks. Quercetin and rutin were detected by comparing the retention times, MS and UV spectra with the standards. The relative retention time and relative peak area of the 12 peaks in the fingerprint were calculated by setting the quercetin as the reference peak. The experimental data were used for similarity calculation and hierarchical clustering analysis. By comparing the UV and MS spectra data with those of the authentic standards and literature, five main peaks in the fingerprints were identified. Finally, five medicinal portions of the herb (leaf, fruit, stem, pterygium and root) were also analyzed by this method. It was found that there were similar chemical components in different parts of this herb but the contents were very different. The developed fingerprint assay was specific and could be readily utilized for comprehensive evaluation of EAS, as well as to distinguish different medicinal portions.  相似文献

Abstract

A terbium-sensitized fluorescence spectrophotometry method using an anionic surfactant, sodium dodecyl benzene sulphonate (SDBS), was developed for the determination of prulifloxacin (PUFX). It was found that SDBS significantly enhanced the fluorescence intensity of the PUFX–Tb3+ complex (about 13-fold). The optimal experimental conditions were determined as follows: excitation and emission wavelengths of 290 nm and 545 nm, pH 8.0, 4.0 × 10?5 mol L?1 terbium(III), and 4.0 × 10?4 mol L?1 SDBS. The enhanced fluorescence intensity of the system (ΔF) showed a good linear relationship with the concentration of PUFX over the range 6.0 × 10?8 to 2.0 × 10?6mol L?1 with a correlation coefficient of 0.9991. The detection limit (S/N = 3) was determined as 8.5 × 10?9 mol L?1. This method has been successfully applied for the determination of PUFX in pharmaceuticals and human urine/serum samples. Compared with most other methods reported, the rapid and simple procedure proposed here offered higher sensitivity, wider linear range, and good stability. The luminescence mechanism of the system was also discussed in detail. In the fluorescence system of PUFX–Tb3+–SDBS, SDBS acted not only as the surfactant but also as the energy donor.  相似文献

Mismatch binding molecules (MBLs), strongly and selectively bound to the mismatched base pair in duplex DNA, were immobilized on Sepharose. Three MBL–Sepharose columns were prepared with three MBLs, naphthyridine dimer (ND), naphthyridine–azaquinolone (NA), and aminonaphthyridine dimer (amND), which exhibited different binding profiles to the mismatched base pairs. These three MBL–Sepharose columns showed characteristic elution profiles for DNA duplexes containing mismatched base pairs. The ND–Sepharose column separated the G–G and G–A mismatched DNA from fully matched duplexes. The NA–Sepharose column separated the A–A and G–A mismatched DNA from other DNA duplexes. The amND–Sepharose column separated the C–C mismatched DNA. These chromatographic profiles were very consistent with the binding preference of each MBL. By changing the elution conditions from sodium hydroxide to sodium chloride, MBL–Sepharose columns were also able to separate the mismatched DNA that weakly bound to the MBL from fully matched DNA duplex. Figure MBL-Sepharose affinity chromatography successfully separates the mismatched duplex DNA from fully matched duplex.  相似文献

The reaction of 2-hydroxy-2-polyfluoroalkylchroman-4-ones with hydrazine affords 5-hydroxy-3-(2-hydroxyaryl)-5-polyfluoroalkyl-2-pyrazolines, whereas 2-polyfluoroalkylchromones under similar conditions produce 3(5)-(2-hydroxyaryl)-5(3)-polyfluoroalkylpyrazoles. 5-(2-Hydroxyaryl)-1-methyl-3-polyfluoroalkylpyrazoles were synthesized in the reaction with methylhydrazine, and the reaction with phenylhydrazine afforded regioisomeric 3(5)-(2-hydroxyphenyl)-1-phenyl-5(3)-polyfluoroalkylpyrazoles.  相似文献

Abstract

A new sensitive extractive spectrophotometric method for the determination of nickel in sub-microgram level is discussed. 5-(2′-carboxyphenyl) azo-8-quinolinol reats with nickel at pH 5.2 to 6.1 to produce a wine red complex extractable in isomayl alcohol. The complex is stable for 24 hrs. Beer's law is obeyed in the range 0.04 to 0.60 ug of nickel per ml. The molar absorptivity and Sandell's sensitivity of the method are 5.5 × 1041. mol?1 cm?1 and 0.001 μg cm?2, respectively, at wavelength of maximum absorption (λ max 540 nm). Effects of diverse ions and variables have been studied in detail. The method has been applied to the determination of nickel in alloys like manganin, constantan, german silver, bronze, steel and in geological samples. Features of the method include high sensitivity, excelllent reproducibility and reliability.  相似文献

The binding interaction between (−)-epigallocatechin (EGC) with bovine β-lactoglobulin (βLG) was investigated by fluorescence, circular dichroism (CD) and Fourier transform infrared (FTIR) spectroscopy methods. The binding parameters were determined by Stern–Volmer equation and the thermodynamic parameters were calculated according to the van’t Hoff equation. The results suggested that βLG was bound by EGC, which resulted in change of native conformation of βLG. van der Waals interactions and hydrogen bonding probably played major roles in the binding process. Our study is helpful for further elucidation of binding interactions between catechins with milk proteins, which would contribute to the development of novel milk products.  相似文献

We found that alkoxalyl chloride (ClCOCO2R) did not carry out an acylation on 1-copper(I) alkyne in solvent without additives, but chemoselectively on 5-copper(I) 1,2,3-triazole (an intermediate in cycloaddition of 1-copper(I) alkyne and azide). Thus, a one-pot preparation of 1,4,5-trisubstituted 5-(2-alkoxy-1,2-dioxoethyl)-1,2,3-triazole was achieved by simply stirring the mixture of 1-copper(I) alkyne, azide, and alkoxalyl chloride at room temperature for 4 h.  相似文献
