Centella asiatica (L.) Urban is a traditional herbal medicine used in Asiatic countries, and is commonly used to treat various wounds, leprosy, tuberculosis and lupus diseases. In this work, a new method based on microwave assisted extraction followed by liquid chromatography–diode array detection–electrospray ionization multistage tandem mass spectrometry analysis has been developed for the identification and quantification of biologically active constituents in C. asiatica, including asiatic acid, asiaticoside and madecassoside. The separation was performed on an Agilent Eclipse C18 column (150 mm × 4.6 mm i.d., 5 μm) with gradient elution of acetonitrile and 0.1% aqueous acetic acid within 50 min. Detection was performed at 205 nm. The calibration curves showed good linearity (r 2 > 0.9992). The limits of detection ranged from 1.2 to 1.6 μg mL?1. The intra- and inter-day precision was less than 3% and the recovery of the assay was in the range of 95.4–106.8%. The method was successfully applied to the quantification of the three constituents in different samples of C. asiatica. The results indicated that the developed method could be considered to be a simple, rapid and reliable method for the quality evaluation of C. asiatica. The samples were also analyzed on a liquid chromatography–electrospray ionization–time-of-flight mass spectrometry system to confirm the identification results.  相似文献

Melamine is attracting much attention because of its toxicity. In the work discussed in this paper, microwave-assisted extraction in combination with ion-exchange high-performance liquid chromatography with diode-array detection was used for analysis of melamine in pet food. Trichloroacetic acid–N,N-dimethylformamide 10:1 was the best extractant, because of the strong polarity of melamine. Separation was performed on a 250 mm × 4.6 mm i.d., 5-μm particle, cation-exchange column; isocratic elution with a mixture of ammonium formate solution (pH 3.0) and acetonitrile was complete within 10 min. UV absorbance DAD detection was performed at 240 nm. Response was a linear function of melamine concentrations from 0.1 to 50 μg mL?1, with a detection limit of 1.0 mg kg?1. Intra-day and inter-day precision, as RSD, was <3% and the recovery of the assay was in the range 95.4–106.8%. In analysis of spiked pet food, the new method yielded satisfactory results. Because of its simplicity and accuracy this straightforward method is particularly suitable for routine melamine analysis.  相似文献

The purpose of this article was to develop a rapid and robust LC–MS–MS method for quantifying shikonin and deoxyshikonin simultaneously in rat plasma using emodin as internal standard. The LC system consisted of an Agilent ZORBAX SB-C18 (1.8 μm, 250 × 4.6 mm, 20 °C) column. Elution with an isocratic mobile phase consisted of methanol/10 mM ammonium acetate in water/acetonitrile containing 0.05% formic acid (45:10:45, v/v/v) at a flow rate of 0.8 mL min?1 yielded sharp, high-resolved peaks within 12 min. The lower limits of quantitation were 0.5 ng mL?1 for shikonin, and 8 ng mL?1 for deoxyshikonin. Correlation coefficient (r) values for the linear range of two analytes were greater than 0.99. Assay precision was <13% and accuracy was 87–99%. This newly developed method was used to the pharmacokinetic studies of the shikonin analogues in rats after intravenous administration (n = 4).  相似文献

A simple, sensitive, selective and rapid liquid chromatography–tandem mass spectrometry method was developed and validated for the simultaneous separation and quantitation of atenolol and chlorthalidone in human plasma using metoprolol and hydrochlorothiazide as internal standard. Following solid phase extraction, the analytes were separated by an isocratic mobile phase on a reversed-phase C18 column and analyzed by MS in the multiple reaction-monitoring mode (atenolol in positive and chlorthalidone in the negative ion mode). The limit of quantitation for this method was 10 and 15 ng mL?1 and the linear dynamic range was generally 10–2,050 ng mL?1 and 15–3,035 ng mL?1 for atenolol and chlorthalidone, respectively.  相似文献

A sensitive and selective liquid chromatographic tandem mass spectrometric (LC–MS–MS) method was developed for simultaneous identification and quantification of tamsulosin and dutasteride in human plasma, which was well applied to clinical study. The method was based on liquid–liquid extraction, followed by an LC procedure with a Gemini C-18, 50 mm × 2.0 mm (3 μm) column and using methanol:ammonium formate (97:3, v/v) as the mobile phase. Protonated ions formed by a turbo ionspray in positive mode were used to detect analytes and internal standard. MS–MS detection was by monitoring the fragmentation of 409.1 → 228.1 (m/z) for tamsulosin, 529.3 → 461.3 (m/z) for dutasteride and 373.2 → 305.3 (m/z) for finasteride (IS) on a triple quadrupole mass spectrometer. The lower limit of quantification for both tamsulosin and dutasteride was 1 ng mL?1. The proposed method enables the unambiguous identification and quantification of tamsulosin and dutasteride for clinical drug monitoring.  相似文献

In this work, an analytical multiresidue method using liquid chromatography tandem-mass spectrometry (LC–MS–MS) with triple quadrupole in selected reaction monitoring (SRM) mode for the simultaneous determination of 54 pesticides in vegetables (pepper and tomato) and citrus fruits (orange and lemon) has been developed. The procedure involves initial single phase extraction of sample with acetonitrile by agitation, followed by liquid–liquid partition aided by “salting out” process using NaCl. The average recovery by the LC–MS–MS method obtained for these compounds varied from 65.5 to 114.5% with a relative standard deviation between 2.3 and 8.3%. The method presents good linearity over the range assayed 10–500 μg L?1 (except famoxadone 50–1,000 μg L?1) and the detection limits for the pesticides studied varied from 0.03 to 14.9 μg kg?1. The proposed method was used to determine pesticide levels in vegetables and citrus fruit samples from different experimental orchards and greenhouses from the Region of Murcia.  相似文献

A new, rapid, and efficient method, multiple reaction monitoring liquid chromatography–tandem mass spectrometry, has been developed for analysis of nanafrocin in foodstuffs of animal origin. The researchers used a C18 stationary phase coupled with triple-quadrupole tandem mass spectrometry in negative-electrospray mode. The limits of detection (LOD) and quantification (LOQ) were 0.005 and 0.01 mg kg?1, respectively, in the matrixes. Detector response was found to be a linear function of concentration over the range 0.005–0.1 mg kg?1 in each matrix. Mean overall recovery (n = 10) of nanafrocin varied from 71 to 101%. The results show that identification and quantification of nanafrocin residues in foodstuffs of animal origin can be successfully achieved by use of the proposed LC–MS–MS method.  相似文献

A liquid chromatographic-tandem mass spectrometric method for the simultaneous determination of anabolic androgenic steroids and their esters in hair has been developed. The hair sample was treated with methanol to extract the esters, followed by alkaline digestion for optimum recovery of the anabolic androgenic steroids. After liquid–liquid extractions, the extract was dried, redissolved and analyzed by multiple reaction monitoring with a quadrupole mass spectrometer. The lower limits of detection ranged from 0.001 to 0.020 ng mg?1 for the 21 analytes. The applicability of the method was demonstrated using guinea pig hair samples gained from controlled experiments.  相似文献

A rapid, selective and convenient liquid chromatography–mass spectrometric method for the simultaneous determination of paracetamol and caffeine in human plasma was developed and validated. Analytes and theophylline [internal standard (I.S.)] were extracted from plasma samples with diethyl ether-dichloromethane (3:2, v/v) and separated on a C18 column (150 × 4.6 mm ID, 5 μm particle size, 100 Å pore size). The mobile phase consisted of 0.2% formic acid–methanol (60:40, v/v). The assay was linear in the concentration range between 0.05 and 25 μg mL?1 for paracetamol and 10–5,000 ng mL?1 for caffeine, with the lower limit of quantification of 0.05 μg mL?1 and 10 ng mL?1, respectively. The intra- and inter-day precision for both drugs was less than 8.1%, and the accuracy was within ±6.5%. The single chromatographic analysis of plasma samples was achieved within 4.5 min. This validated method was successfully applied to study the pharmacokinetics of paracetamol and caffeine in human plasma.  相似文献

Magnetic solid-phase extraction (MSPE) coupled with gas chromatography–mass spectrometry was applied for the analysis of organophosphorus pesticides (OPPs) in water samples. We chose C18-functionalized Fe3O4@mSiO2 microspheres as the magnetic sorbents to extract and enrich OPPs from water samples with the advantages of good solubility in water, large surface area and fast separation ability. In this study, six kinds of OPPs were analyzed and various parameters of MSPE procedure, including eluting solvent, the amount of magnetic absorbents and extraction time were optimized. Validation experiments showed that the optimized method had good linearity with correlation coefficients r 2 > 0.98 and satisfactory precision with the relative standard deviation ≤10.7 %. The limits of detection were 1.8–5.0 μg L?1 and the limits of quantification ranged from 6.1 to 16.7 μg L?1. We concluded that the proposed method was successfully applied to analyze OPPs in real water samples and the results indicated that it had the advantages of simplicity, convenience and efficiency.  相似文献

A new analytical approach, simultaneous derivatization and dispersive liquid–liquid microextraction followed by liquid chromatography–atmospheric-pressure chemical ionization tandem mass spectrometry, has been developed for analysis of hexanal and heptanal in human blood. In the derivatization and extraction procedure a solution of 2,4-dinitrophenylhydrazine (derivatization reagent) in 85 μL acetonitrile (dispersive solvent) and 50 μL tetrachloromethane (extraction solvent) was rapidly injected into the aqueous sample containing hexanal and heptanal. Within a few seconds the aldehydes were derivatized and simultaneously extracted. After centrifugation, the hydrazones in the sediment phase were analyzed by LC–APCI–MS–MS. Derivatization and extraction conditions were investigated systematically. Under the optimum conditions enrichment factors for hexanal and heptanal in a 1-mL sample were 63 and 73, respectively. The calibration plots were linear in the ranges 0.5–100 and 100–1,000 nmol L?1, respectively, and the respective limits of detection (LOD) were 0.17 and 0.076 nmol L?1. Reproducibility and recovery were good. The experimental results were compared with those obtained by use of solid-phase extraction and polymer monolithic microextraction. Because sample derivatization, extraction, and concentration were combined in a single step, the proposed method enabled simple, rapid, inexpensive, and efficient analysis of aldehydes in blood. The method has great potential for clinical analysis of biologically relevant aldehydes.  相似文献

Liquid chromatography–ion trap mass spectrometry was employed to investigate the metabolism of linarin in rats. Identification and structural elucidation of the metabolites were performed by comparing the differences in molecular masses, retention times, and full scan MS n spectra between linarin and its metabolites. Six metabolites (acacetin, apigenin, acacetin glucuronide, apigenin glucuronide, acacetin sulfate, apigenin sulfate) were detected in rat urine after oral administration of linarin at the dose of 50 mg kg?1. Furthermore, a selective and sensitive liquid chromatography–triple quadruple mass spectrometry assay was developed and validated for the simultaneous determination of linarin and acacetin (the major metabolite of linarin) in rat urine. Chromatographic separation was carried out on a C18 column, and mass spectrometric detection was performed using a triple-quadrupole mass spectrometer coupled with an electrospray ionization source in the positive ion mode. Quantitation of linarin and acacetin was performed using selected reaction monitoring of precursor–product ion transitions at m/z 593 → 285 for linarin, 285 → 242 for acacetin, and 303 → 153 for hesperitin (internal standard), respectively. The assay exhibited good linearity (r > 0.9900) for both linarin and acacetin. The intra- and inter-day precisions were <13.4 % and the accuracy was between ?8.1 and 3.1 %. The method was successfully applied to the urinary excretion study of linarin in rats after oral administration of linarin.  相似文献

For the first time a sensitive, specific and rapid LC–MS–MS assay is presented for the simultaneous determination of levodopa (L-DP), 3-O-methyldopa (3-OMD) and benserazide (BSZ) in human serum. The three compounds were extracted from human serum by protein precipitation followed by dilution of the supernatant with aqueous formic acid. In serum, linearity was observed between 50 and 1,000 ng mL?1 of L-DP, 3-OMD and BSZ, respectively. Intra-day and inter-day RSD values were below 10.56 and 6.22% at concentrations of 120, 360 and 720 ng mL?1. The presented method showed excellent specificity and sensitivity compared with other methods reported. It was applied to a pharmacokinetic study and demonstrated its applicability to pre-clinical and clinical pharmacological research.  相似文献

A selective, rapid and sensitive liquid chromatography tandem mass spectrometry method has been developed for the simultaneous determination of ramipril and ramiprilat in human plasma using enalapril as the internal standard via one-step extraction with ethyl acetate under acidic condition. The analysis was carried out on a Diamonsil C18 column (150 mm × 4.6 mm i.d., 5 μm) with a mobile phase consisting of 1% formic acid-acetonitrile (25:75, v/v) at a constant flow rate of 0.5 mL min?1. The detection was performed on a triple-quadruple tandem mass spectrometer by selective reaction monitoring mode via electrospray ionization. Linear calibration curves of ramipril and ramiprilat were obtained in the concentration range of 0.107–107.0 and 0.262–105.0 ng mL?1, respectively. The intra- and inter-day precision (RSD) values were below 8.2 and 4.8% for ramipril, 10.4 and 12.3% for ramiprilat, and accuracy (RE) were within ±5.5 and ±3.2%, respectively at all QC levels. The method was utilized to support clinical pharmacokinetic studies in healthy volunteers following oral administration of ramipril tablets.  相似文献

This paper describes a new and rapid method for accurate quantification of the six ergot alkaloids, ergometrine, ergotamine, ergosine, ergocristine, ergocryptine, and ergocornine, by liquid chromatography–tandem mass spectrometry (LC–MS–MS). The six ergot alkaloids studied have been defined by the European Food Safety Authority (EFSA) as among the most common and physiologically active ones. In addition, the method enables the quantification of the corresponding six epimers (ergo-inines) of these ergot alkaloids. This is of considerable importance in terms of the differences in toxicity of the isomeric forms. The method involves extraction under alkaline conditions using a mixture of acetonitrile and ammonium carbonate buffer followed by a rapid clean-up using dispersive solid-phase extraction with PSA (primary secondary amine) and a short chromatographic LC-run (21 min) with subsequent MS–MS detection. The method was developed and validated using ten different cereal and food samples. The major strength of the new method compared with previously published techniques is the simplicity of the clean-up procedure and the short analysis time. The limits of quantification were 0.17 to 2.78 μg kg−1 depending on the analyte and matrix. Recovery values for the 12 ergot alkaloids spiked into ten different matrices at levels of 5, 50, and 100 μg kg−1 were between 69 and 105% for 85 of 90 recovery measurements made over six days. Measurement uncertainty values were highly satisfactory. At a concentration level of 5 μg kg−1 the expanded measurement uncertainty ranged from ±0.56 to ±1.49 μg kg−1, at a concentration level of 100 μg kg−1 the expanded measurement uncertainty ranged from ±8.9 to ±20 μg kg−1. Both LOQs and measurement uncertainties were dependent on the analyte but almost independent of the matrix. The method performance was satisfactory when tested in a mini-intercomparison study between three laboratories from three different countries. A sensitive, selective, and simple liquid chromatography-tandem mass spectrometry method has been developed for the simultaneous separation and determination of isosorbide dinitrate and its active metabolite, isosorbide 5-mononitrate, in human plasma. Topiramate was used as the internal standard. Sample preparation consisted of a simple one-step liquid–liquid extraction with ethylacetate without pH adjustment. The method was fully validated with respect to linearity, sensitivity, specificity, recovery, accuracy, and precision. Isosorbide dinitrate and isosorbide 5-mononitrate were stable in standard solution and in plasma samples under storage and processing conditions. The assay was successfully applied to the pharmacokinetic study of isosorbide dinitrate and isosorbide 5-mononitrate in human plasma.  相似文献

This study describes an accurate, sensitive, and specific chromatographic method for the simultaneous quantitative determination of lamivudine and zidovudine in human blood plasma, using stavudine as an internal standard. The chromatographic separation was performed using a C8 column (150 × 4.6 mm, 5 μm), and ultraviolet absorbency detection at 270 nm with gradient elution. Two mobile phases were used. Phase A contained 10 mM potassium phosphate and 3% acetonitrile, whereas Phase B contained methanol. A linear gradient was used with a variability of A-B phase proportion from 98–2% to 72–28%, respectively. The drug extraction was performed with two 4 mL aliquots of ethyl acetate.  相似文献

An environmentally friendly method for the determination of fluoroquinolone (FQ) antibiotics including enrofloxacin, ciprofloxacin, levofloxacin, fleroxacin and sparfloxacin in four feeds is proposed. Disodium ethylenediaminetetraacetate (EDTA)?CMcllvaine buffer (0.1 mol L?1, pH 4.0) was used as an extracting solvent and the extraction process was accelerated by microwave irradiation. No organic solvents were used in the extraction procedure. The extract obtained was then cleaned up and concentrated by an Oasis hydrophilic?Clipophilic balance (HLB) solid-phase extraction cartridge. The relative intra- and inter-day standard deviations obtained were in the range of 3.7?C9.1 and 2.1?C11.4%, respectively. In the three fortified levels of blank feed sample (30, 100 and 500 ng g?1), recoveries of FQs ranging from 61.1 to 97.9% were obtained. The analytes desorbed from HLB were analyzed by liquid chromatography?Ctandem mass spectrometry. The limit of detection was in the range of 5.0?C9.1 ng g?1. The method presented here can be considered a promising alternative to traditional techniques using shaking or stirring for extraction, being more effective, and producing less pollution.  相似文献

Summary A sensitive and selective liquid chromatographic method coupled with electrospray ionization tandem mass spectrometry (LC–ESI–MS–MS) has been developed for quantification of felodipine in human and dog plasma. Compounds were separated on a 2.0 mm × 150 mm, 5.0 m particle, C8 column with 1 m m ammonium acetate–acetonitrile, 20:80, pH 6.0, as mobile phase at a flow rate of 200 L min–1. Nifedipine was used as internal standard. Plasma samples were extracted with diethyl ether, the centrifuged upper layer was evaporated, the residue was reconstituted with mobile phase, and the reconstituted samples were injected. The analytical column lasted for at least 1000 injections. By use of multiple reaction monitoring (MRM) mode in MS–MS felodipine and nifedipine were detected without severe interference from the human or dog plasma matrix. Felodipine produced a protonated precursor ion ([M + H]+) at m/z 384 and a corresponding product ion at m/z 338. And internal standard (nifedipine) produced a protonated precursor ion ([M + H]+) at m/z 347 and a corresponding product ion at m/z 315. Detection of felodipine in human and dog plasma was accurate and precise, with a limit of quantification of 0.05 ng mL–1. The method has been successfully applied to preliminary pharmacokinetic study of felodipine in human and dog plasma.  相似文献

Yupingfeng granules (YPFG) is commonly used in the treatment of immunological diseases, inflammations, and pulmonary diseases. Several studies have found that chromones, flavones, and saponins were the major bioactive compounds of YPFG. However, few studies have reported accurate quantification methods of these compounds. This study aimed to establish a simple and rapid method by using liquid chromatography–tandem mass spectrometry (LC–MS/MS) to determine 15 bioactive compounds in YPFG. The experimental parameters including extraction methods, extraction solvents, extraction time, solid–liquid ratio, and LC–MS/MS condition were optimized. The linearity, precision, repeatability, stability, and recovery of the established method were evaluated. The contents of 15 bioactive compounds in seven batches of YPFG samples were analyzed by the established method and the results were compared with the values determined by HPLC. The optimal extraction condition was to extract 0.1 g of YPFG by ultrasound with 50 mL 50% ethanol for 30 min. A Waters ACQUITY UPLCBEH C18 column using the 0.1% formic acid water solution and acetonitrile as mobile phase with a gradient elution was applied to the chromatographic separation. The linearity, precision, repeatability, stability, and recovery of the method were within acceptable ranges. Compared with HPLC analysis methods in Chinese Pharmacopoeia and literature, the established method was faster, simpler, more accurate, and more reliable. The method of simultaneous determination of 15 components in YPFG by LC–MS might provide a basis for the study of the bioactive compounds and the improvement of the quality standard of YPFG.  相似文献
