3-(5-Substituted 2-Methoxyphenyl)-4-Nitro-2-Phenyl-5-Thiophene-2-yl-Isoxazolidines - Synthesis and Conformational Studies

Abstract

The synthesis of a new set of 2,3,4,5-substituted isoxazolidines having a sulfur containing heterocyclic moiety has been described. The structural features of the compounds have been analyzed by ¹H and ¹³C NMR spectroscopy as well as by single crystal X-ray studies.

Synthesis of 1-phenyl-3-methyl-4-alkyl-4-[N-methylbutyro(capro)lactyl]-5-pyrazolones

The corresponding hydrazones, which on heating are cyclized to give 1-phenyl-3-methyl-4-alkyl-(benzyl)-4-(2-oxopyrrolidinomethyl)- and 1-phenyl-3-methyl-4-alkyl(benzyl)-4-(N-methylcaprolactyl)pyrazolones, are formed in the reaction of alkyl(benzyl) (2-oxopyrrolidinomethyl)- and alkyl-(benzyl) (N-methylcaprolactyl)acetoacetic esters with phenylhydrazine.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 11, pp. 1546–1548, November, 1976.

Transformation of 4-(1-dimethylaminoethylidene)-2-phenyl-5(4H)-oxazolone into methyl 2-benzoylamino-3-oxobutanoate. The synthesis of 1-substituted 4-benzoylamino-3-methyl-5(2H)-pyrazolones

Methyl 2-benzoylamino-3-oxobutanoate ( 3 ) was prepared from hippuric acid (1) which was converted with N,N-dimethylacetamide and phosphorus oxychloride into 4-(1-dimethylaminoethylidene)-2-phenyl-5(4H)-oxazolone ( 2 ) followed by hydrolysis with hydrochloric acid in methanol. Compound 3 was treated with hydrazines 4 to give 4-benzoylamino-3-methyl-1H-pyrazol-5(2H)-one ( 6a ) and its 1-substituted derivatives 6b-j . The corresponding hydrazones 5f, i, j were isolated as intermediates.

以氢氧化钾为催化剂,1-苯基-3-甲基-吡唑啉-5-酮与取代苯甲醛亚胺反应合成了新型1-苯基-3-甲基-吡唑啉-5-酮衍生物——1-苯基-3-甲基-4-(1-N-氯乙胺基-1-取代苯基)甲基-吡唑啉-5-酮,其结构经1H NMR,IR,MS和元素分析表征。     

铌(V)-1-苯基-3-甲基-4-苯甲酰基-吡唑啉酮-5络合物吸附波的研究

在pH=5.0的HAc-NaAc介质中,Nb(V)与1-苯基-3-甲基-4-苯甲酰基-吡唑啉酮-5(PMBP)生成络合物,于-0.96V(us.SCE)出现一尖锐的极谱波.在滴汞电极上用示波极谱仪或在悬汞电极上测吸附伏安曲线.峰电流与Nb(V)浓度分别在0.0075~0.80μg/mL及0.000075~0.0075μg/mL范围内呈线性关系.实验了30多种离子的干扰影响,经苯肿酸分离后用于钢中Nb(V)的测定.检出限为0.00003μg/mL.溶液可稳定96h以上.用多种电化学方法研究了电极反应机理.     

Structural Requirements of 1-(2-Pyridinyl)-5-pyrazolones for Disproportionation of Boronic Acids

We observed an unusual formation of four-coordinate boron(III) complexes from the reaction of 1-(2-pyridinyl)-5-pyrazolone derivatives with arylboronic acids in the basic media. The exact mechanism is not clear; however, the use of unprotected boronic acid and the presence of a bidentate ligand appeared to be the key structural requirements for the transformation. The results suggest that base-promoted disproportionation of arylboronic acid with the assistance of the [N,O]-bidentate ligation of 1-(2-pyridinyl)-5-pyrazolone should take place and facilitate the formation of pyrazole diarylborinate. Experiments to obtain a deeper understanding of its mechanism are currently underway.     

Organophosphorus Chemistry 34 1 . The Reaction of Alkyl Phosphites and Hexamethylphosphorus Triamide with 1-Phenyl-3-methyl-4-ylidene-5-pyrazolones

1-Phenyl-3-methyl-4-ylidene-5-pyrazolones ( 1a-c ) produce the respective 1:1 phosphonate adducts ( 4a-i ) upon reaction with the appropriate dialkyl phosphites ( 2a-c ). The reaction of 1a-c with trialkyl phosphites ( 3a-c ) proceeds only in presence of water, also to give phosphonates 4a-c almost exclusively. Hexamethylphosphorus triamide ( 3d ) was successfully utilized to introduce the bis(dimethylamino)-phosphoryl function in the f , g -unsaturated carbonyl moiety in 1a-c , producing phosphoramidates 15a-c . Possible reaction mechanisms were discussed. Compatible elemental and spectroscopic results were recorded for the new products.     

1-苯基-3-甲基-4-苯甲酰基-吡唑酮-5与中性磷(膦)萃取剂协同萃取镧B

研究了 1 苯基 3 甲基 4 苯甲酰基 -吡唑酮 5 (HPMBP)和中性有机磷 (膦 )类萃取剂Cyanex 4 71X(TIBPS ,B)在硝酸介质中对稀土元素La 的萃取 ,用斜率法和恒摩尔法探讨了萃取机理 ,确定了萃合物的组成为La(NO3 ) 2 ·PMBP·B ,计算了萃取平衡常数。比较了HPMBP与Cyanex 4 71X、Cyanex 92 1、Cyanex 92 3、Cyanex 92 5、DEH/EHP、P35 0及TBP的单独及混合体系萃取La 的性能 ,结果表明 :所有混合体系对La 均有协同效应 ,其中HPMBP与Cyanex 92 3、Cyanex 92 1、Cyanex 92 5的混合体系是萃取La 的有效体系。     

Synthesis and prototropic tautomerism of 1-n-alkyl-3-methyl-4-nitroso-5-pyrazolones

Nitrosation at C-4 of 1-n-alkyl-3-methyl-5-pyrazolones is achieved in about 50% yield with sodium nitrite in hydrochloric acid medium. A tautomeric equilibrium in solution with a proton moving from the OH at C-5 to N-2 in the nitrosated pyrazolones is proposed.     

Synthesis of 1-(2,4,6-trichlorophenyl)-3-(3-nitro- and 2-chloro-5-nitroanilino)-5-pyrazolones

A convenient one flask synthesis of 1-aryl-3-nitroanilino-5-pyrazolones in yields of 60–65% is described. Synthesis involves initial reaction of ethyl 3-ethoxy-3-iminopropionate hydrochloride and a nitroaniline in methanol producing an imidate ester. The imidate ester is then reacted with an arylhydrazine forming an amidine. The amidine is then cyclized with base.     

2-[3-Phenyl-5-(m-Chlorophenyl)-2-Pyrazolin-1-yl]-3-Methyl-4(3H)-Quinazolinone

There are two molecules in the asymmetric unit (I and II). The structure consists of a 2-pyrazoline ring and three aromatic rings two of which are free and the third one is condensed with a 3-methyl-4(3H)-pyrimidine ring (4(3H)-Quinazolinone). While the pyrazoline ring is in the distorted envelope conformation, the free aromatic rings are planar and the pyrimidine ring deviates from the planarity. The crystal structure is stabilized by C–HO inter and intramolecular bonds.

We have studied chlorination of 5-(4-methoxyphenyl)-3-phenyl-1,2-oxathiolane 2-oxide and we discuss possible mechanisms for this reaction.     

Synthesis of N-Substituted Derivatives of 2-(4-Amino-2-methyl-1H-indol-3-yl)- and 2-(6-Amino-2-methyl-1H-indol-3-yl)acetic Acids

A method was developed for the production of indole compounds containing an amino group at positions 4 and 6 of the benzene ring on the basis of the indolization of the 3-acetylaminophenylhydrazone of ethyl levulinate. A series of derivatives of 2-(4-amino-2-methyl-1H-indol-3-yl)- and 2-(6-amino-2-methyl-1H-indol-3-yl)acetic acids at the 4- and 6-amino group were synthesized.     

1-苯基-3-甲基-4-(6-氢-4-甲基氨基-5-硫杂-2,3-吡嗪)-吡唑啉酮的合成与晶体结构

通过1-苯基-3-甲基-4-氯乙酰基-吡唑啉酮-5 (PMCP) 和4-甲基氨基硫脲 (MTSC) 缩合, 形成了一种新的双杂环化合物 (PMCP-MTSC)。利用元素分析与单晶X-射线衍射进行了表征。 该化合物(C14H15N5OS)属单斜晶系, P21 空间群, 晶体学参数为 a = 7.634(1), b = 11.639(2), c = 8.150(2) ? ?= 98.13(1), V = 716.9(2) ?, Z = 2, Dc = 1.396 g/cm3, (MoKa) = 0.232 mm-1, F(000) = 316, Mr = 301.37。结构由直接法解出,共收集3114个衍射点,其中I > 2(I)的独立衍射点有2453个,用全矩阵最小二乘法修正,最终偏离因子:R = 0.0294, wR = 0.0738。结构分析表明: 该化合物以酮式存在,分子间通过氢键作用(N(5)H…O)连接起来并形成一维链状结构。     

2-(4-甲基苯基)-4-(4-甲氧基苯基)噻唑-5-甲酸乙酯的合成

以对甲氧基苯甲酸为原料,经5步反应合成了新化合物2-(4-甲基苯基)-4-(4-甲氧基苯基)噻唑-5-甲酸乙酯,其结构经1H NMR,MS及X-射线单晶衍射确证。
