o-Benzoylbenzenesulfonyl chlorides (I) were prepared conveniently from aminobenzophenones by diazotization followed by reaction with sulphur dioxide in the presence of Cu+, according to the general method of Meerwein. Reaction of the sulfonyl chlorides with hydrazine led to 4-phenyl-2H-1,2,3-benzothiadiazine-1,1-dioxides (II). The latter compounds could be methylated and acetylated readily in the 2-position. The 2-methyl derivative (III) could be prepared also by reaction of the sulfonyl chloride (Ia) with methylhydrazine. Catalytic hydrogenation of 6-chloro-4-phenyl-2H-1,2,3-benzothiadiazine-1,1-dioxide (IIa) gave the 3,4-dihydro derivative (V). Reaction of the sulfonyl chlorides (I) with o-phenylenediamine followed by cyclodehydration led to 11H-11,11a-dihydrobenzimidazo[1,2-b] [1,2]benzisothiazole-5,5-dioxides (VII). One of the latter compounds (VIIa) in sodium hydroxide solution in the presence of methyl iodide or benzyl chloride was transformed into 6-methyl- and 6-benzyl-5H-dibenzo[c,g] [1,2,6]thiadiazocine-5,5-dioxides (VIII), respectively. 5H-Dibenzo[c,g] [1,2,6] thiadiazocine-6,6-dioxides (XIV) were prepared also by cyclodehydration of 2-amino-2′-benzoylbenzenesulfonanilides (XIII).  相似文献

Abstract

We describe a rapid and convenient methodology for the preparation of diverse 3-aryl and 3-trifluoromethyl 4H-1,2,4-benzothiadiazine-1,1-dioxides in a one-pot microwave-promoted reaction between 2-aminobenzenesulfonamides and benzaldehydes, trifluoroacetic acid or benzoic acids.

The intermolecular reductive cyclization of o-nitrobenzene-sulfonamides with aliphatic ketones induced by TiCl4/Sm system was studied and a series of 3,3-disubstituted-3,4-dihydro-(2H)-1,2,4-benzothiadiazine-1,1-dioxides were synthesized in moderate to high yields under mild and neutral conditions.  相似文献

Variously substituted 2-phenyl-1,3-dioxolanes and 2-(2-bromophenyl)-1,3-dioxolanes, prepared from the corresponding benzaldehydes, were lithiated ortho to the acetal group. Reaction of the lithio derivatives with sulfur dioxide led to the lithium sulfinate salts, which gave, upon oxidative chlorination with sulfuryl chloride, the corresponding benzenesulfonyl chlorides. Then, depending on the aromatic substitution pattern of the molecule, several protocols were elaborated for the functional group transformations leading to the target compounds. Ring closure was performed with hydrazine hydrate or acetylhydrazine, in the latter case with one-pot removal of the acetyl group. The 4-unsubstituted 2H-1,2,3-benzothiadiazine 1,1-dioxides thus obtained are potential drug candidates based on their structural similarity to biologically active phthalazinones.  相似文献

We achieved efficient synthesis of imidazo- and pyrimido[1,2-b]benzo-1,2,4-thiadiazine-1,1-dioxides by the tandem aza-Wittig reaction/intramolecular NH-nucleophilic addition/NH-nucleophilic substitution cyclization methodology, involving sulfonamide ester-containing carbodiimides as the key intermediates. Similarly, imidazo[2,1-b]quinazolinones, imidazo[1,2-a]pyrimidinediones, and imidazo[1,2-a]imidazolidinediones were also synthesized through the aza-Wittig reaction-tandem cyclization strategy. When homochiral (l)-alanine methyl ester was incorporated as a building block into the corresponding iminophosphorane starting materials, we obtained optically active imidazo[1,2-b]benzo-1,2,4-thiadiazine and imidazo[2,1-b]quinazolinone derivatives without any racemization through the one-pot tandem methodology.  相似文献

Ring Enlargement of Six- to Nine-Membered Heterocycles: Reaction of 3-(Dimethylamino)-2,2-dimethyl-2H-azirine with 3,4-Dihydro-2H-1,2,4-benzothiadiazin-3-one 1,1-Dioxides Reaction of 3-(dimethylamino)-2,2-dimethyl-2H-azirine ( 1 ) and N-substituted 3,4-dihydro-2H-1,2,4-benzothiadiazin-3-one 1,1-dioxides ( 4 ) in CHCl3 yields 3-(dimethylamino)-4,5,6,7-tetrahydro-1,2,5,7-benzothiatriazonin-6-one 1,1-dioxides 5 , a novel nine-membered heterocyclic system, by ring enlargement (Schemes 2 and 4). In refluxing MeOH, the heterocycle 5a rearranges to give the N-[1-methyl-1-(1,1-dioxo-4H-1,2,4-benzothiadiazin-3-yl)ethyl]-N′, N′-dimethylurea 10 . The three isomeric 2-(methylamino)benzenesufonamides 8,9 , and 11 (Scheme 3) are obtained by naBH4 reduction of 5a and 10 , respectively. Mechanisms for the thermal isomerization 5a → 10 and the NaBH4 reduction of 5a are proposed in Schemes 5 and 6.  相似文献

Ammonium salts were found to be a convenient and inexpensive reagent in the Cu-catalyzed three-component reaction with terminal alkynes and sulfonyl or phosphoryl azides leading to N-unprotected amidines. Thus obtained amidines bearing 2-bromobenzenesulfonyl moiety were efficiently cyclized by the Cu-catalyzed intramolecular N-arylation to give an important pharmacophore skeleton of 2H-1,2,4-benzothiadiazine 1,1-dioxides. Conveniently, two tandem catalytic procedures could be readily operated in one pot.  相似文献

An interesting protocol for the synthesis of benzothiadiazine-1,1-dioxides through iodine-catalyzed one-pot dehomologative oxidation of styrenes and readily available 2-aminobenzenesulfonamide has been developed. Diverse benzothiadiazine-1,1-dioxides were prepared using I2 as a catalyst, TBHP as an oxidant and Na2CO3 as a base. This reliable, metal and ligand free conversion involves dehomologation of styrene to aromatic aldehyde which on subsequent cyclisation affords benzothiadiazine-1,1-dioxides in good yield.  相似文献

Diels-Alder reactions of thiophene-1,1-dioxides with strong electron withdrawing groups (EWG) were studied experimentally and theoretically. Thiophene-1,1-dioxides with two strong EWG behave as dienophiles and regio- and stereoselectively react with cyclopentadiene to give [2+4] cycloadducts 2a-c, which are derivatives of benzothiophene. In contrast, thiophene-1,1-dioxides with one EWG behave as dienes in the inverse electron demand Diels-Alder reaction yielding dihydro-1H-indenes derivatives. Cope [3,3]-sigmatropic rearrangement of adducts 2a-c was also demonstrated. MP2 calculations successfully rationalize the contrasting regioselectivities of these cycloaddition reactions.  相似文献

The synthesis of some novel 3-carboethoxy-4,11-dihydro-11-alkyl/phenyl-4-oxopyrimido[1,2-b][1,2,4]benzo-thiadiazine 6,6-dioxides ( IVa-c ) from 3-amino-4-alkyl/phenyl-4H-1,2,4-benzothiadiazine 1,1-dioxides ( 1a-c ) and diethyl ethoxymethylenemalonate ( II ) is described. The reaction of chlorosulfonie acid with propiophenones was found to give the 3-chloro-2-methylbenzothiophene-1, 1-dioxides (I) and (VIII); reaction of Mannich bases of aceto-phenone, with chlorosulfonie acid, gave the corresponding 3-chloro-2-substituted amino-methylbenzothiophene-1,1-dioxides (IX) and (X). The above structures were established by reaction with various nucleophiles and by hydrogenation. o-Hydroxypropiophenones, under the above conditions, are cyclized to the cyclic sulfones (XII) and (XIII). Ring opening with pyrrolidine gave the corresponding phenolic sulfonamides (XIV) and (XV).  相似文献

The hydrochlorothiazide, 6-chloro-3,4-dihydro-2H-1,2,4-benzothiadiazine-7-sulfonamide-1,1-dioxide, (HCTZ), widely used as a diuretic and anti-hypertensive drug, was transformed into its N-nitroso-derivative, 6-chloro-4-nitroso-3,4-dihydro-2H-1,2,4-benzothiadiazine-7-sulfonamide-1,1-dioxide (ON-HCTZ) by sodium nitrite in an acidic medium. The crystalline complexes of ON-HCTZ with 18-crown-6 (18C6) and cis-anti-cis-dicyclohexyl-18C6 (DCH6B) demonstrated different H-bonding modes from those present in the co-crystals of HCTZ with the same crown ethers. The influence of the nitroso-group on the binding mode and crystal packing is discussed.  相似文献

Partially hydrogenated 2-amino[1,2,4]triazolo[1,5-a]pyrimidines and 2-amino[1,2,4]triazolo[5,1-b]quinazolines react with the chlorides of chloroacetic, 3-chloropropanoic, and 4-chlorobutanoic acids at 0–5 °C to give amides through acylation of the 2-amino group. Heating the corresponding 3-chloropropanoyl derivatives at 80–90 °C in DMF leads to selective intramolecular alkylation at N-3 to form the chlorides of partially hydrogenated [1,2,4]triazolo[1,5-a:4,3-a′]dipyrimidin-5-ones and pyrimido[2′,1′:3,4][1,2,4]triazolo[5,1-b]quinazolin-12-ones. It may be more convenient to prepare such compounds through one-pot processes. Some reactions of the synthesized chlorides of polycondensed heterocycles have been studied. Conditions have been found to effect the selective synthesis of free bases, oxidative aromatization or hydrolysis of the dihydropyrimidine cycle, and the selective hydrolytic cleavage or elimination of the pyrimidone ring. Some of the resulting compounds represent new mesoionic heterocycles.  相似文献

We describe a practical and elegant method of constructing a thiazine ring fused with benzene under mild reaction conditions. A variety of 4-iodo-2H-benzo[e][1,2]thiazine-1,1-dioxides were prepared with high regioselectivity via a two-step process involving Pd/C-mediated C-C coupling of o-halobenzenesulfonamides with terminal alkynes, followed by iodocyclization of the resulting o-(1-alkynyl)arenesulfonamide using elemental iodine in acetonitrile. The coupling reaction was carried out using 10% Pd/C-PPh3-CuI as a catalyst system in the presence of Et3N. The process worked well for bromides and iodides, and a wide array of terminal alkynes containing alkyl and aryl substituents were employed. The iodocyclization step tolerated a variety of functional groups such as hydroxy, chloro, cyano, and methoxy, producing the six-membered heterocyclic ring selectively. The resulting 4-iodo-2H-benzo[e][1,2]thiazine-1,1-dioxides participated in Sonogashira, Heck, and Suzuki reactions producing a wide range of functionally substituted benzothiazines in good yields.  相似文献

Earlier1 it was reported that 4-aryl-4,5-dihydrothiophene-1,1-dioxides (I) are converted in aqueous and alcohol solution of bases to isomeric 3-aryl-2,5-dihydrothiophene dioxides (II) and 3-aryl-4,5-dihydrothiophene-1,1-dioxides (III) (Fig. 1).In the presence of electron-donor substituents in the p-position of the phenyl ring, IIb, c compounds are practically irreversibly isomerised to III b, c. For unsubstituted phenyldihydrothiophene-1,1-dioxides IIa and IIIa the reaction proceeds riversibly but equilibrium is shifted towards isomer IIIa indicating its high thermodynamic stability. It is known2 that 2,5-dihydrothiophene-1,1 -dioxide is more stable than 4,5-dihydrothiophene-1,1-dioxide. Consequently, the observed stability increase of 3-aryl-4,5-dihydrothiophene-1,1-dioxides (IIIa–c) is due to interaction of the substituent C6H4X with the sulfonyl group. The first one-pot copper-catalyzed highly regioselective C–N bond formation between aryl/alkyl amines and sulfonamide-substituted naphthoquinones was accomplished. Facile chemoselective routes obtained diverse ring-opening 6-amino/anilino-naphthalen-dione-1-sulfonamides and ring-fused 6-amino/aniline-5H-naphth[1,8-cd]isothiazol-5-one,1,1-dioxides with great functional group tolerance. The Suzuki reaction of both (E)- and (Z)-α-chloro-γ-hydroxyenamides with benzeneboronic acid by using Pd2(dba)3, tBu3P, and Cs2CO3 proceeded stereoselectively to afford only (E)-trisubstituted enamides. Also, palladium-catalyzed intramolecular cyclization of (E)- and (Z)-α-chloro-γ-hydroxyenamides bearing an N-arylsulfonyl group took place stereoselectively to give the corresponding (E)-2,3-dihydrobenzoisothiazole 1,1-dioxides. The γ-hyroxy group in α-chloroenamides was found to accelerate the present palladium-catalyzed carbon-carbon bond forming reactions.  相似文献

2,4-Bis(trimethylsilyloxy)pyrimidines 1/2 on reaction with o-chlorobenzyl chlorides in 1,2-dichloroethane in the presence of I2 undergo single step 1,3-dibenzylation to provide 1,3-bis(o-chlorobenzyl)pyrimidine-2,4-diones. The reactions of 1 with allyl/alkyl bromide followed by subsequent addition of o-chlorobenzyl chloride provide a simple one-pot synthesis of 1,3-unsymmetrical pyrimidine-2,4-diones. Amongst these, 1,3-bis(o-chlorobenzyl)uracil (6a) shows anti-HIV-1 activity.  相似文献
