The capability of human promyelocytic leukemia cells HL60 to be induced to differentiate to various stages along the monocytic or myelocytic pathway was exploited for investigation of the uptake of selected photo-sensitizers by diverse types of cells of the same origin. The results showed that there was no substantial difference in photofrin uptake between noninduced HL60 cells, immature monocytes, immature neutrophils and cells differentiated along the eosinophilic pathway. In contrast, HL60 cells differentiated into macrophages (HL609) exhibited markedly increased photofrin uptake, which was further enhanced by their pretreatment with bacterial lipopolysaccharide. Similar results were obtained with other photosensitizers tested: di-and tetrasulfonated aluminum phthalocyanines (AIPcS2 and AIPcS4), tetrasulfonated zinc phthalocyanine (ZnPcS4), tetraphenylporphine tetrasulfonate (TPPS4) and benzoporphyrin derivative monoacid (BPD). Despite marked differences in the state of self-aggregation and other chemical properties of these compounds, the degree of their preferential uptake by HL60 PH cells showed very little variation. In a typical experiment, the uptake of these photosensitizers by HL60 PH cells was four to five times higher than the uptake by noninduced HL60 cells. In addition to the fluorometric assay employed in most of the experiments, cellular concentration of AlPcS4 was determined by measurement of elementary aluminum using atomic absorption spectroscopy.  相似文献

Heavy metal cations Cd~(2+), Pb~(2+), and Hg~(2+)were added to substitute for Ca~(2+)in culture media to study their effect on the relationship between CaM and the proliferation of cultured rabbit aortic smooth muscle cells (ASMC). It was found that all the heavy metal cations studied stimulated the proliferation of ASMC in varying degrees, increased the CaM content in cells at late G_1 stage and decreased the activity of cAMP PDE. These results suggest that the adverse effect of heavy metals may be related to the pathogenesis of atherosclerosis and hypertensive disease.  相似文献

For the first time we have shown here that the constituent from tectal extract (Te) which can support and promote the survival and growth of the retinal ganglion cell is a 30 kD protein. (i) Using MTT colorimetric microassay to measure the optical density for the survival and growth of the cultured retinal neuron, it was found that the optical densities for the experimental cultures with either Te, or its 10-30 kD fraction, or its ≥30 kD fraction were 2-4 times that of the control culture without Te (P<0.01). This indicated that experimental cultures were more active in growth, (ii) The retinal neurons cultured on Te Phast gels showed that large retinal neurons (>18 μm) grew only on the gel region containing the 30 kD protein. The retrograde prelabelled with horseradish peroxidase (HRP) for retinal ganglion cells indicated that these surviving neurons grown on Te Phast gel were HRP positive, i.e. retinal ganglion cells, (iii) The retinal explants cultured with 30 kD gels at a close distance of 1 mm ap  相似文献

The sex expression of Lagenaria siceraria can be modified by phytohormones. This modification is related to "a certain bisexual stage" during floral bud differentiation. ACC (ethylene precursor) and STS (ethylene antagonist) can change the direction of sex differentiation of the floral buds in vitro. STS induces maleness by means of inhibiting ethylene action, directly promoting the development of the primordia of stamens, and then suppressing that of pistils. Hence, it can be deduced that ethylene induces femaleness by directly suppressing the development of the primordia of stamens and then promoting that of pistils. Such a regulation of the development of sexual organs is due to a change in the direction of assimilates to the pistils by ACC and to the stamens by STS. Therefore, ethylene induces femaleness by first limiting the sink strength of the stamens and then increasing that of the pistils.  相似文献

温敏梳状嵌段共聚物对PS微球阻抗蛋白吸附作用的研究   总被引:2,自引:0,他引:2  
采用可逆加成断裂链转移聚合(RAFT)方法和大分子单体技术,制备了温敏性聚N-异丙基丙烯酰胺(PNIPAM)-聚乙烯基吡咯烷酮(PVP)与PNIPAM-聚氧化乙烯(PEO)梳状嵌段共聚物,这些共聚物具有PVP或PEO支链.以溶菌酶为蛋白模型研究了所得共聚物对聚苯乙烯(PS)微球表面蛋白吸附的抑制作用.通过絮凝实验、激光散射法表观粒径测定、电泳迁移率测定及蛋白吸附量的定量数据比较了不同梳状结构的抗蛋白吸附效果.结果表明,预吸附梳状嵌段共聚物可有效阻抗蛋白吸附,亲水支链增加阻抗性能提高,即使环境温度高于PNIPAM的相转变温度也能阻抗蛋白吸附.透射电镜和共聚物胶体粒径测试表明,梳状嵌段共聚物阻抗蛋白吸附的机制是预吸附后PVP或PEO亲水支链在微球表面形成了阻隔层.通过PS微球的变温絮凝实验可评价预吸附聚合物的抗蛋白吸附性能,快速获得定性结果.  相似文献

This paper reveals the formation of annular lamellar body (ALB) in the ganglion of leech by means of in situ fixation and the lanthanum nitrate tracer technique. This formation involves both wrapping and internalization of the gap junctions between glial processes themselves, as well as between neuron and glial process. The results indicate that there is probably an active process of internalization of membrane structures involving gap junctions between neuron and glial cell in the central nervous system in leech. Abstract— Benoxaprofen [2-(4-chlorophenyl)-α-methyl-5-benzoxazoleacetic acid] is a phototoxic non-steroidal anti-inflammatory agent. Irradiation of human erythrocytes in the presence of benoxaprofen (8 μ M ) and oxygen resulted in rapid cell lysis which began after 10 min and was complete within 30 min. While photohemolysis was also observed under anerobic conditions, its onset was delayed for more than 20 min and it took nearly 100 min for complete lysis to occur. Photohemolysis was also delayed by butylated hydroxyanisole but was unaffected by reduced glutathione. 1,4-diazabicyclo[2.2.2]octane, D2O. β-carotene, or superoxide dismutase. The main photoproduct of benoxaprofen, 2-(4-chlorophenyl)-5-ethylbenzoxazole, was almost as effective in causing photohemolysis as benoxaprofen itself. In the presence of UV irradiation, benoxaprofen (10 (μ M ) caused the degranulation of rat peritoneal mast cells and the release of histamine. The release of mast cell histamine may provide a reasonable explanation for the urticarial response to benoxaprofen and irradiation seen in human subjects.  相似文献

In this study we demonstrate: (i) The GnRH agonist exerts a direct dose-dependet stimulative effect on the aromatase activity and progesterone production in cultured monkey granulosa cells; (ii)the stimulative effect on steroidogenesis can be completely blocked by concomitant treatment with a GnRH antagonist, suggesting that the actions of GnRH are mediated through stringent stereospecific recongnition sites; (iii) in addition to the stimulative effect, the GnRH agonist in the presence of gonadotropins also exerts an inhibitory effect, even though the peptide by itself is more effective in the stimulation of steroidogenesis, and the stimulation of gonadotropin on steroidogenesis could be gradually restored by decreasing the concentration of the GnRH agonist in the culture; and (iv) paradoxical effect can also be observed in the presence of cAMP-inducing agents, suggesting that the inhibitory action of the peptide on gonadotropin-induced steroidogenesis is localized at a step distal to the stringent reco  相似文献

Ketone was the optimum sensitizer selected from many additives. With ketonc as the additive, the yield of formaldehyde was more than 2.40% and the highest yield Was 3.58% over the silica gel-1 catalyst, and remained almost unchanged during the catalyst life test of 500 h. 选择 3种不同丙烯酸含量的乙烯 丙烯酸共聚物 (EAA)为原材料 ,二苯醚 (DPE)为稀释剂 ,研究了淬冷温度、粗化时间等影响液滴生长的动力学因素对热诱导相分离法 (TIPS)制备EAA DPE亲水性高分子微孔膜结构的影响 .淬冷温度的高低决定了EAA DPE体系是发生液 液相分离还是固 液相分离 ,而产生相分离的机理不同将影响稀释剂液滴的生长 ,最终影响微孔膜的孔径 .实验结果表明 ,在相同粗化时间的条件下 ,随着EAA1 41 0 DPE、EAA3 0 0 2 DPE、EAA3 0 0 3 DPE三体系冷却温度的逐渐升高 ,孔径逐渐变大 .在结晶温度以下 ( 0℃、3 0℃、60℃ )粗化时间相同时 ,温度对微孔膜的孔径影响较小 ,例如 0℃和 3 0℃的恒温条件粗化 1 0min,微孔膜的孔径在 1~ 3 μm之间 ;在 60℃的恒温条件粗化 1 0min ,微孔膜的孔径在 3~ 5 μm之间 .而在 90℃的恒温条件粗化相同的时间 ,由于体系始终处于结晶温度线以上 ,体系始终处在液 液相分离区域 ,最终得到微孔膜的孔径达到了 6~8μm .在结晶温度以下 ( 3 0℃ )进行恒温粗化 ,由于体系的过冷程度很大 ,液滴相的粗化过程被抑制住 ,所以粗化时间对微孔膜的孔径影响不大 ;而在结晶温度以上 ( 90℃ )进行恒温粗化时 ,则是随着粗化时间的延长 ,微孔膜的孔径逐渐变大  相似文献

In this paper, the following carbonyl-imino bridged compounds and related analogs havebeen synthesized:X= C1, m = 0); and the structural effect of the carbonyl-imino bridges has been studied by means of the UV and other properties of the compounds. A new conception of conjugative segment is put forward. The structural bases for each of the three π-π* bands of (C) are ascertained and verified by modification on structure of segments, such as eliminating, merging, lengthening or shortening of segments. It is demonstrated that both bridges -CO-and -NH- can block the conjugated polarization of the whole molecule so as to form three segments S, M and L of which the λmax each exhibits relative independence. The electronic absorption peaks resulting from different segments can be readily recognized and the existence of each segment can be ascertained. This may provide a new way for structural analysis of bridged compounds.  相似文献

利用流动注射分光光度法技术,考察了不同离子强度(NaCl)下,在水溶液中表面带负电的活性炭分别吸附3种阴离子染料的动力学行为.利用固-液相互作用方程,求取了活性炭-染料相互作用能.比较了无机盐中阳离子的种类对吸附影响的差异.结果表明:对于3种阴离子染料,离子强度的增大都起到加速吸附的作用:表现吸附速率常数、活性炭-染料相互作用能与离子强度三者之间存在密切的内在联系:在相同离子强度下,二价阳离子对吸附的加速作用要比一价阳离子的显著,但在同价阳离子之间这种作用的差别较小.  相似文献

The physical aging behavior of atactic polystyrene (aPS) films achievedunder different drawing conditions has been studied by optical birefringence and modulatedifferential scanning calorimeter (m.d.s.c.). The results show that on annealing at specifictemperature below glass transition temperatue (T_g), the enthalpy relaxation (△H) and T_gdecrease with increasing of birefringence (△n). On the other hand, the effect of moleculardraw ratio (MDR) is confusing: △H and T_g decrease with increasing of MDR in the earlystage of aging, but the MDR's effect become unobvious with the development of aging.It may be concluded that the ordered domain generated by the drawing below or aboveglass transition temperature will affect the development of physical aging behavior. Theviewpoint of cohesional entanglement is used to account for the above observations.  相似文献

The present study is designed to investigate the regulatory effect of mammalian erythroblasts, prior to naturally-occurring denucleation, on malignancy of mouse plasmocytoma cells and the possibility of reactivation of the pyknotic late erythroblast nuclei in hybrid cells crossed between rat intermediate or late erythroblasts of 15-day Wistar rat embryonic livers, and mouse plasmocytoma (SP~2/O) cell lines. Results indicated that:(ⅰ) Suppression of tumorigenicity and reversion of the malignant phenotype were observed in hybrid cells in a similar way as those of cybrid cells crossed between reticulocytes and myeloma cells as we reported previously, thus providing further evidence to support the hypothesis that some regulatory substances already existed in mammalian intermediate and late erythroblasts long before nuclear extrusion.
