A novel approach has been developed for the synthesis of β‐arylacyl/β‐heteroarylacyl‐β‐alkylidine malonates in moderate to good yields by the reaction of Stork aryl and heteroaryl enamine with β‐chloroalkylidene malonates. The reaction involves conjugate (Michael) addition of Stork enamine on β‐chloroalkylidene malonates and elimination of chloride ion. These Michael adducts were utilized as intermediates for the synthesis of highly substituted 1,4‐dialkyl‐2‐oxo‐6‐aryl/hetreoaryl‐1,2‐dihydro‐pyridine‐3‐carboxylic acid ethyl esters via 5 + 1 ring annulation protocol.  相似文献

A detailed investigation of the reactions of PhSO2CF2H and PhSO2CH2F with (E)‐chalcone (=(E)‐1,3‐diphenylprop‐2‐en‐1‐one) at low temperatures revealed that these two reactions were kinetically controlled, and the ratios of 1,2‐ vs. 1,4‐adducts, which did not change much over time at these temperatures, reflect the relative rates of the two reaction pathways. The controlled experiments of converting the PhSO2CF2‐ and PhSO2CHF‐substituted 1,2‐adducts to 1,4‐adducts showed that these isomerizations are not favored due to the low stability and hard‐soft nature of PhSO2CF and PhSO2CHF? anions. Moreover, taking advantage of the remarkable stability and softness of (PhSO2)2CF? anion, an efficient thermodynamically controlled isomerization of (PhSO2)2CF‐substituted 1,2‐adduct to 1,4‐adduct was achieved for the first time.  相似文献

A series of N‐substituted 1,4‐dihydro‐4‐oxo‐1,8‐naphthyridine‐3‐carboxylate esters has been prepared in two steps from ethyl 2‐(2‐chloronicotinoyl)acetate. Treatment of the β‐ketoester with N,N‐dimethylformamide dimethyl acetal in N,N‐dimethylformamide (DMF) gave a 95% yield of the 2‐dimethylaminomethylene derivative. Subsequent reaction of this β‐enaminone with primary amines in DMF at 120oC for 24 h then afforded the target compounds in 47–82% yields by a tandem SNAr‐addition‐elimination reaction. Synthetic and procedural details as well as a mechanistic rationale are presented.  相似文献

A series of twelve new 2‐[(o‐ and p‐substituted)aminophenyl]‐3H‐5‐[(o‐ and p‐substituted)phenyl]‐7‐chloro‐1,4‐benzodiazepines, which have possible pharmacological properties has been obtained. The synthesis was carried out following six steps. The structure of all products was corroborated by ir, 1H nmr, 13C nmr and ms. In addition for the compound 2‐(o‐chloroaminophenyl)‐3H‐5‐(o‐fluorophenyl)‐7‐chloro‐1,4‐benzodiazepine 7, its structure was confirmed by X‐ray diffraction.  相似文献

The crystal structure of the title compound, C20H23N3O7, consists of relatively isolated mol­ecules. The substituted 1,4‐di­hydro­pyridine ring adopts a flattened boat conformation. Both ester groups, at positions 3 and 5, have cis,cis geometry. The phenyl ring is nearly planar and is approximately perpendicular to the 1,4‐di­hydro­pyridine ring (dihedral angle 87.70°).  相似文献

In the title compound, C22H17Cl2NO3S, the mol­ecule is a substituted 3,4‐di­hydro‐2H‐1,4‐benzoxazine compound which has three phenyl rings which are essentially planar. The 3,4‐di­hydro‐2H‐oxazine part of the mol­ecule is fused to the benzo ring and has a half‐boat conformation; the dihedral angle between the planar part of the oxazine ring and the benzo ring is 10.2 (2)°. The (3‐chloro­phenyl)­methyl­idene substituent has a Z configuration in relation to the ring N atom of the oxazine moiety. Interestingly, the p‐toluenesulfonyl (p‐tosyl) substituent on the ring N atom protrudes away from the 3‐­chloro­phenyl substituent thus avoiding any steric interaction.  相似文献

Chiral rhodium catalysts comprising 2,5‐diaryl‐ substituted bicyclo[2.2.1]diene ligands L1 – L10 were utilized in the enantioselective 1,4‐addition reaction of arylboronic acids to N‐substituted maleimides. In the presence of 2.5 mol % of RhI/ L2 , enantioenriched conjugate addition adducts were isolated in 72–99 % yields with 86–98 % ee. This protocol offers a convenient method to access a variety of 3‐arylsuccinimides in a highly enantioselective manner. Maleimides with readily cleavable N‐protecting groups were tolerated enabling the synthesis of useful synthetic intermediates. Pyrrolidine 4 , a biologically active compound, and pyrrolidine 5 , an ent‐precursor to an HSD‐1 inhibitor, were synthesized to demonstrate the utility of this method.  相似文献

The starting material O‐protected glycosyl isothiocyanate ( 1?3 ) was refluxed with 1,4‐diaminobenzene in CHCl3 under nitrogen atmosphere to give 1,4‐bis(N‐glycosyl)thioureidobenzene ( 4?6 ). Then 1,4‐bis[N‐(4/6‐substituted benzothiazole‐2‐yl)‐N′‐glycosylguanidino]benzenes ( 8a?8e , 9a?9e , 10a?10e ) were obtained in good yield by reaction of compounds ( 4?6 ) with 2‐amino‐4/6‐benzothizoles ( 7a?7e ) and HgCl2 in the presence of TEA in DMF. The structures of all 18 new compounds were confirmed by IR, 1H NMR, LC‐MS and elemental analysis. The bioactivity of anti‐HIV‐1 protease (HIV‐1 PR) and against angiotensin converting enzyme (ACE) have been evaluated.  相似文献

Desymmetrization of the divinyl carbinol 1,4‐pentadien‐3‐ol was accomplished by the asymmetric 1,3‐dipolar cycloaddition of azomethine imines based on a magnesium‐mediated, multinucleating chiral reaction system utilizing diisopropyl (R,R)‐tartrate as the chiral auxiliary. The corresponding optically active trans‐pyrazolidines, each with three contiguous stereogenic centers, were obtained with excellent regio‐, diastereo‐, and enantioselectivity, with results as high as 99 % ee. This reaction was shown to be applicable to both aryl‐ and alkyl‐substituted azomethine imines. The use of a catalytic amount of diisopropyl (R,R)‐tartrate was also effective when accompanied by the addition of MgBr2.  相似文献

A facile and efficient method for the construction of 3‐alkyl/aryl substituted 1,4‐benzoxazine and benzoxazepine via AgNO3 catalyzed cyclization of propargyloxy sulfonamides and their anti‐tubercular activity against Mycobacterium tuberculosis H37RV is described. This cyclization proceeds through 6‐exo‐dig manner to generate the products in moderate to good yields.  相似文献

The ethyl 1,4‐dihydro‐4‐oxo‐3‐quinolinecarboxylate ring structure, important in several drug compounds, has been prepared in two steps from ethyl 2‐(2‐fluorobenzoyl)acetate. Treatment of this β‐ketoester with N,N‐dimethylformamide dimethyl acetal gives a 97% yield of the 2‐dimethylaminomethylene derivative. Reaction of this β‐enaminone with primary amines in N,N‐dimethylformamide at 140°C for 48 h then affords the 1,4‐dihydro‐4‐oxo‐3‐quinolinecarboxylate esters in 60–74% yields by a tandem addition‐elimination‐SNAr reaction. The condensation of 4‐amino‐3‐aryl‐5‐mercapto‐1, 2, 4‐triazoles (1a‐f) with 6‐/8‐substituted 1,4‐dihydro‐4‐oxo‐quinoline‐3‐carboxylic adds (2a‐d) in the presence of phosphorus oxychloride on refluxng or under microwave irradiation gave twenty four novel 3‐aryl‐6‐ (6‐/8‐substituted 4‐chloroquinoline‐3‐yl)‐s‐triazolo[3,4‐b]‐1, 3,4‐thiadiazoles (4a‐x), Considerable increase in the reaction rate has been observed with improved yields under microwave irradiation. The structures of the compounds synthesized were determined by elemental analyses, IR, 1H NMR and MS spectra. Their spectral properties and the reaction mechanism were also discussed. The preliminary biological test showed that some of compounds bad moderate antibacterial activities.  相似文献

A medium consisting of K2CO3 and H2O in [omim][BF4] ionic liquid (IL) was used to synthesize N‐substituted 2H‐benzo[b][1,4]oxazin‐3(4H)‐one derivatives from their corresponding o‐aminophenols and 2‐bromoalkanoates. As a result, chemoselective formation of benzoxazinones in high yields has been observed at room temperature. After the reactions and separation of the products, the IL was recovered and successfully reused in subsequent reactions without significant loss of activity.  相似文献

Twelve compounds unknown in the literature N‐(E)‐2‐stilbenyloxymethylenecarbonyl substituted hydrazones of 2‐, 3‐ and 4‐pyridinecarboxaldehydes, as well as methyl‐3‐pyridylketone have been prepared. The stereochemical behavior of these compounds in dimethyl‐d6 sulfoxide solution has been studied by 1H NMR technique. The E geometrical isomers and cis/trans amide conformers have been found for N‐substituted hydrazones 1–12. EI induced mass spectral fragmentation of these compounds were also investigated. The data obtained create the basis for distinguishing isomers.  相似文献

Three title compounds 4a—4c have been synthesized by the cyclodehydration of 1’-benzylidine-4’-(3β-substituted-5α-cholestane-6-yl)thiosemicarbazones 2a—2c with thioglycolic acid followed by the treatment with cold conc. H2SO4 in dioxane. The compounds 2a—2c were prepared by condensation of 3β-substituted-5α-cholestan- 6-one-thiosemicarbazones 1a—1c with benzaldehyde. These thiosemicarbazones 1a—1c were obtained by the reaction of corresponding 3β-substituted-5α-cholestan-6-ones with thiosemicarbazide in the presence of few drops of conc. HCl in methanol. The structures of the products have been established on the basis of their elemental, analytical and spectral data.  相似文献

The facile and convenient access by a conventional procedure in ethanol as solvent to a new series of succinyl‐spaced pyrazoles including 1,4‐bis[5‐(trichloromethyl)‐5‐hydroxy‐4,5‐dihydro‐1H‐pyrazol‐1‐yl]butane‐1,4‐diones (64–82%) and the respective dehydrated derivatives as 1,4‐bis[5‐(trichloromethyl)‐1H‐pyrazol‐1‐yl]butane‐1,4‐diones in 57–82% yields, from the regioselective cyclocondensation reactions of 4‐substituted 4‐methoxy‐1,1,1‐trichloroalk‐3‐en‐2‐ones with succinic acid dihydrazide, where the 4‐substituents are Me, Ph, 4‐FC6H4, 4‐ClC6H4, 4‐NO2C6H4, 2‐furyl, and 2‐thienyl, is reported. γ‐Chlorocarbanions of proper nucleophilicity, generated from 3‐chloropropyl pentachlorophenyl sulfone (=pentachloro[(3‐chloropropyl)sulfonyl]benzene; 1 ; Ar=C6Cl5), add to electron‐deficient formal imines 3a – l to produce anionic adducts that enter intramolecular substitution leading to substituted pyrrolidines. This new and simple synthesis of pyrrolidines mimics a 1,3‐dipolar cycloaddition, although it proceeds in two distinct steps.  相似文献

A cheap synthesis of the so‐called ‘decalin‐1,8‐diones’ started with the conjugate (1,4‐) addition of cyclohex‐2‐en‐1‐one derivatives to the γ‐position of the dilithium derivative (buta‐1,3‐diene‐1,1‐bis(olate)) of crotonic acid. Hydrogenation of these ‘1,4‐γ’ adducts and final cyclization afforded the enol tautomers of decalin‐1,8‐diones. Nucleophilic substitutions at these 3‐oxoenols by NH3 or primary amines created only monoamino products (namely, 3‐oxoenamines) whose reactions with OPCl3 yielded dihydro(1,3,2)oxazaphosphinin‐2‐one derivatives. The two regioisomers of a trimethyl‐3‐oxoenamine served as models for the constitutional assignments of the two rapidly interconverting (hence, individually NMR‐invisible), tautomeric trimethyl‐3‐oxoenols. Such methyl substitutions served to break the ‘pretended’ symmetry of ‘decalin‐1,8‐dione’. Hydrazine and 3‐oxoenols furnished oxygen‐free indazole derivatives whose N?H bonds exchanged with t1/2=ca. 0.00035 s at ca. ?58(9) °C.  相似文献

A series of some substituted diethyl 4‐(2‐chloroquinolin‐3‐yl)‐2,6‐dimethylpyridine‐3,5‐dicarboxylates has been synthesized from substituted diethyl4‐(2‐chloroquinolin‐3‐yl)‐1,4‐dihydro‐2,6‐dimethylpyridine‐3,5‐dicarboxylates (1,4‐DHPs) by treating the latter with SiO2–HNO3 which proved to be a better oxidant in terms of product yield, reaction time, and cost. Further, these compounds were screened for their antimicrobial activity. All the diethyl 4‐(2‐chloroquinolin‐3‐yl)‐2,6‐dimethylpyridine‐3,5‐dicarboxylates exhibited more potent activities than the corresponding 1,4‐DHPs. Further, docking simulation of the most active and least active compounds 3e and 2e into Escherichia coli topoisomerase II DNA Gyrase B was also performed.  相似文献
