The reaction of methyl α-diazoacetoacetate with enamines catalyzed by dirhodium and copper complexes underwent formal 1,3-dipolar addition to give 5-amino-4,5-dihydrofurans in moderate yields. The reaction was suggested to proceed via a nucleophilic addition of enamines to rnetal-carbenes and a subsequent intramolecular cyclization of the resulting zwitterionic intermediates.  相似文献

A general and efficient NH insertion reaction of rhodium pyridyl carbenes derived from pyridotriazoles was developed. Various NH‐containing compounds, including amides, anilines, enamines, and aliphatic amines, smoothly underwent the NH insertion reaction to afford 2‐picolylamine derivatives. The developed transformation was further utilized in a facile one‐pot synthesis of imidazo[1,5‐a]pyridines.  相似文献

Metal carbenes can undergo a myriad of synthetic transformations. Sulfur ylides are potential safe precursors of metal carbenes. Herein, we report cascade reactions that involve carbenoids derived from sulfoxonium ylides for the efficient and regioselective synthesis of indoles and pyrroles. The tandem action of iridium and Brønsted acid catalysts enables rapid assembly of the heterocycles from unmodified anilines or readily accessible enamines under microwave irradiation. The key mechanistic steps are the catalytic transformation of the sulfoxonium ylide into an iridium–carbene complex, followed by N−H or C−H functionalization of an aniline or enamine, respectively, and a final acid‐catalyzed cyclization. The present method was successfully applied to the synthesis of the densely functionalized pyrrole subunit of atorvastatin.  相似文献

The pyrrolidine and morpholine enamines of cyclic ketones such as cyclohexanone and cyclopentanone were successfully diacetylated at alpha- and alpha'-positions with trichloroacetyl chloride using zinc catalyst. Morpholine enamines of the cyclic ketones gave acetylated morpholine in good yields besides the corresponding diacetylated cyclic enamines. When the same reactions were performed by using triethylamine without using zinc, monoacetylation products of the same enamines were synthesized.  相似文献

This account summarizes the results that have been obtained by our group since 2000 in the area of stable singlet acyclic carbenes. It includes aryl- and alkyl-(phosphino)carbenes, aryl(amino)carbenes, and (amino)(phosphino)carbenes. Our most recent achievements, the transformation of stable carbenes into other stable carbenes are also discussed, along with preliminary results concerning the ligand properties of these species.  相似文献

Crystal structure analyses of five crystalline enamines together with recently published structural data for two other enamines reveal varying degree of pyramidality at the enamine nitrogen atom. The pyramidality appears to be most pronounced in enamines from piperidine and morpholine, less so in enamines derived from prolinoid amines. A pyrolidine enamine obtained from a derivative of cyclohexane-1,4-dione seems to have a virtually planar enamine group. The implications of these findings for current stereochemical problems in enamine chemistry are discussed.  相似文献

The isolation of stable carbenes, such as N-heterocyclic carbenes, cyclic alkyl amino carbenes, and abnormal N-heterocyclic carbenes, has encouraged synthetic chemists to use them as neutral ligands for stabilizing unusual compounds with main group elements. In this Minireview, an overview is provided of the recent developments in the chemistry of carbene-stabilized silicon halides, in which the silicon atoms are in low oxidation state. The structural diversities emanating from the differences in the HOMO–LUMO gaps of various carbenes will also be discussed.  相似文献

The cyclization of enamines derived from β-aminoesters and β-aminonitriles into bi- and tricyclic enaminoketones ( 6,9,13 and 24 ) has been investigated. The enamines derived from aminonitriles cyclize smoothly with magnesium perchlorate in benzene or toluene, whereas the enamines derived from aminoesters cyclize spontaneously during their formation. The partial protolysis of organotin imines—enamines leads to organic secondary enamines, which we found to be stable in completely aprotic media.Their structure was determined by IR and NMR methods, and their reactivity towards acrylonitrile studied. The results show a much greater reactivity for enamines, as compared to that of the corresponding imine tautomers.  相似文献

Nucleophilic fluoroalkylation reactions of imines and enamines with α-phenylthio, α-phenylsulfonyl, and α-diethylphosphoryl substituted fluorinated silanes have been investigated. The reactions are promoted by hydrofluoric acid generated in situ from potassium hydrodifluoride and trifluoroacetic acid. Sulfur reagents worked well with both imines and enamines, whereas phosphorus reagent efficiently coupled only with enamines.  相似文献

用核磁共振法研究了亚胺型杂卓1和烯胺型杂卓2在不同质子溶剂—甲醇、乙醇、叔丁醇、乙酸及CF3CO2D的DMSO-d6溶液中的稳定性及互变异构.结果表明,烯胺型杂卓2在上述溶剂中稳定,亚胺型杂卓1在酸性较弱的质子溶剂中(乙醇、叔丁醇)及较低的温度下(≤30℃)基本稳定,在酸性较强的质子溶剂中(甲醇、乙酸、CF3CO2D的DMSO-d6溶液)向烯胺转化,而且随着溶剂酸性的增强、温度的升高以及反应物与溶剂接触时间的延长,亚胺向烯胺的转化进一步加强.本文还以1a和1b为例,研究了在乙酸溶液中亚胺型杂卓向烯胺型杂卓转化的热力学和动力学特征.研究表明,在283~333 K范围内,该转化的焓变(△H)和熵变(△S)均大于零,吉布斯自由能变(△G)小于零,因此其是吸热、自发及熵增加的过程,该过程可用二级动力学方程来描述,质子对亚胺向烯胺的转化起催化作用.  相似文献

Equilibria between carbonyl compounds and their enamines (from O-TBDPS-derived prolinol) have been examined by NMR spectroscopy in DMSO-d(6). By comparing the exchange reactions between pairs (enamine A + carbonyl B → carbonyl A + enamine B), a quite general scale of the tendency of carbonyl groups to form enamines has been established. Aldehydes quickly give enamines that are relatively more stable than those of ketones, but there are exceptions to this expected rule; for example, 1,3-dihydroxyacetone acetals or 3,5-dioxacyclohexanones (2-phenyl-1,3-dioxan-5-one and 2,2-dimethyl-1,3-dioxan-5-one) show a greater tendency to afford enamines than many α-substituted aldehydes.  相似文献

Depending on the reaction conditions, the reactions of (E)-3,3,3-trichloro-1-nitropropene with cyclohexanone enamines led to bicyclo[4.2.0]octanes or trisubstituted enamines, which are the ring-chain tautomers capable of reversible transformations. Diastereoselectivity of the reactions of (E)-3,3,3-trichloro(trifluoro)-1-nitropropenes with cycloalkanone enamines were studied, a series of hitherto unknown CX3-containing nitroalkylated enamines and γ-nitro ketones were synthesized, the structures of novel compounds were determined by NMR spectroscopy and X-ray diffraction.  相似文献

为了了解芳酰基化的效率和双酰基衍生物的形成, 对几个典型醛和酮的吗啉烯胺进行了邻硝基苯甲酰化的探索。丁醛和异丁醛吗啉烯胺的酰化正常, 水解分别生成55%的2-(邻硝基苯甲酰基)丁醛(2)和40%的2-(邻硝基苯甲酰基)异丁醛(3)。3-戊酮和1, 3-二甲氧基丙酮的吗啉烯胺在酰化和水解时分别生成37%和14.8%相应的β-二酮的烯醇邻硝基苯甲酸酯(4)和(5)。4和5的酸水解发生意外的1, 5-和1,3-酰基转位, 分别生成3, 5-二甲基-2, 6-双(邻硝基苯基)吡喃-4-酮(6)和2-甲氧基-2-甲氧乙酰基-1, 3-双(邻硝基苯基)-1, 3-丙二酮(7)。环戊酮和环已酮吗啉烯胺的酰化分别产生9%和34%的双酰基烯胺(9a)和(9b),逐次酸水解得到2-(邻硝基苯甲酰基)环戊酮(11a)和-环已酮(11b)。在总酰化产物的直接水解中, 11a的产率可以达到81%, 而11b只有35%。对双酰化进行了一些讨论。  相似文献

The reaction between N-chloro-N'-aroyl-amidines and β,β-disubstituted enamines affords 1 - aroyl - 4 -amino - 4,5 - dihydro - imidazoles in low yields, the main reaction product being N - (2 - morpholino - 2,2 -disubstituted) - ethylidene - N' - aroyl - amidines. A similar reaction course was not observed with enamines bearing a hydrogen atom in the β-position. In this case only products derived from electrophilic chlorination on enamines were isolated.  相似文献

A study of the nucleophilic addition of β-lactam carbenes to 3,6-diphenyltetrazines is reported. Instead of the formation of pyrazole derivatives like most reactions between nucleophilic or ambiphilic carbenes and 3,6-disubstituted tetrazines, β-lactam carbenes reacted with 3,6-diphenyltetrazines to produce indeno[2,1-b]pyrrol-2-ones in good yields. The reaction proceeds most probably through a five-step cascade process. This work has not only provided a one-pot operation for the efficient construction of mutisubstituted indeno[2,1-b]pyrrol-2-ones but also revealed the nucleophilicity of β-lactam carbenes.  相似文献

The reaction of linear push-pull enamines bearing a methyl group at the α-position with a set of trifluoromethylated carbonyl compounds was investigated. It has been found that the reaction proceeds at the methyl group of the enamines. The first computational study of the reaction between push-pull enamines and strong electrophilic reagents was reported. Out of three pathways considered DFT and MP2 calculations support ene-mechanism previously suggested based on experimental results only.  相似文献

Although ketone enamines are widely used in organic synthesis, aldehyde enamines are rarely employed due to the limitations of their preparation using known methods (need for acid or base, excess of amine, and/or elevated temperature). We have successfully developed rapid and particularly mild condensation conditions (1 h, 0 degrees C, 1.2 equiv of amine) leading to di- and trisubstituted enamines with excellent conversion (84-100%). Remarkably high chemoselectivity was observed with complete discrimination between aldehyde and ketone, among other functional groups positively tested.  相似文献

An efficient solid-phase approach has been developed to prepare nitrogen heterocycles with a 1,2,3,4-tetrahydro-2-pyridone core via aza-annulation of enamines. Immobilized enamines were prepared from the reaction of primary amines with propynoic acid derivatives or ketones. Aza-annulation reactions were carried out by reacting resin-bound enamines with symmetrical alpha,beta-unsaturated acid anhydrides or alpha,beta-unsaturated acids in the presence of DPPA and TEA. The annulation products were isolated in good to high crude yields. Influence of sterically hindered amines as well as alpha- and beta-substituted acrylic acid derivatives on the annulation reaction was also investigated.  相似文献
