A novel and efficient regioselective synthesis of various arylated highly congested 7-aryl-5-methylsulfanylindan-4-carbonitriles (3a-f), methyl 7-aryl-5-methylsulfanylindan-4-carboxylates (10a-e) and 7-aryl-5-methylsulfanylindan-4-carboxylic acids (11a-e) through base-catalyzed reaction of 6-aryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carbonitriles (1a-f) and methyl 6-aryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carboxylates (9a-e) by cyclopentanone (2) has been delineated. The synthetic potential of 2-pyranone was explored further to generate molecular diversity using 6-aryl-4-sec-amino-2-oxo-2H-pyran-3-carbonitriles (7a-h), 5,6-diaryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carbonitriles (5a,b) and methyl 5,6-diaryl-4-methylsulfanyl-2-oxo-2H-pyran-3-carboxylates (12a,b) as precursors for the ring transformation by cyclopentanone to assess the effects of substituents on the course of the reaction to obtain highly congested indans, 6,7-diaryl-5-methylsulfanylindan-4-carbonitriles (6a,b), 7-aryl-5-(piperidin-1-yl)indan-4-carbonitriles (8a-h) and methyl 6,7-diaryl-5-methylsulfanylindan-4-carboxylates (13a,b).  相似文献

Substituent-controlled regioselective synthesis of highly functionalized 1,2-teraryls 3a-k has been achieved through ring transformation of 6-aryl-4-(pyrrolidin-1-yl/piperidin-1-yl)-2H-pyran-2-one-3-carbonitriles 1a-g by aryl acetones 2a-c in the presence of powdered KOH in DMF in very good yield. Under similar reaction conditions, 6-aryl-4-methylsulfanyl-2H-pyran-2-ones 5a-f afforded 1,7-diaryl-2-methyl-4H,5H-pyrano[3,4-c]pyran-4,5-diones 6a-j as major products and 3,4-diaryl-2-methyl-6-methylsulfanylbenzonitriles as minor constituents 7a-j.  相似文献

The (E)-2-aryl-1-[2-(methylthio)phenyl]-1-nitroethylenes 5 can easily be oxidized to the relevant sulfones 6 and effectively subjected to cyclization via an intramolecular Michael addition after metallation with lithium bis(trimethylsilyl)amide in THF. After quenching with ammonium chloride the 3-aryl-4-nitrothiochroman S,S-dioxides 2 are obtained as diastereomeric mixtures in good to excellent yields. Both yields and stereochemistry of the ring-closure step appear to be influenced by steric effects of the 3-aryl moiety. As sulfides 5 derive from an initial ring opening of 3-nitrobenzo[b]thiophene (1), the overall 1 to 2 process can be considered as an effective 5 to 6 ring enlargement of the sulfur heterocycle. A conformational 1H NMR and molecular-mechanics investigation on the isolated diastereomeric 2 has also been accomplished.  相似文献

An innovative synthesis of aryl tethered 1,3-dimethylimidazo[4,5-b]pyrazin-2-ones 4 and 6 has been delineated through base catalyzed ring transformation of 6-aryl-4-(piperidin-1-yl)-2H-pyran-2-one-3-carbonitriles 1 and methyl 6-aryl-4-methylsulfanyl-2H-pyran-2-one-3-carboxylates 5 with 7-acetyl-1,3-dimethyllumazine 2 with subsequent ring contraction of the fused pyrimidine to an imidazole ring. An additional product, methyl [6-(1,3-dimethyl-2-oxo-2,3-dihydro-1H-imidazo[4,5-b]pyrazin-5-yl)-4-thiophen-2-ylpyran-2-ylidene]acetate 8b, was also isolated from the reaction of 5 and 2, as a minor constituent.  相似文献

In this Letter, we described the synthesis of new 5-(5-amino-1-aryl-1H-pyrazole-4-yl)-1H-tetrazoles 2ac from 5-amino-1-aryl-1H-pyrazole-4-carbonitriles 1ac as well as the unexpected 1H-pyrazolo[3,4-d]pyrimidine derivatives 6ac from 5-amino-1-aryl-3-methyl-1H-pyrazole-4-carbonitriles 4ac, instead of 5-(5-amino-1-aryl-3-methyl-1H-pyrazole-4-yl)-1H-tetrazoles 5ac as desired. In an attempt to obtain these tetrazole derivatives containing the methyl group at C3-position in the pyrazole ring, the amino group in 5-amino-1-(4-methoxyphenyl)-3-methyl-1H-pyrazole-4-carbonitrile 4c was protected by the reaction with sodium hydride and di-tert-butyl-dicarbonate (Boc). The tetrazole derivative 5c was synthesized from the protected compound 7c using analogue methodology to obtain 2ac and 6ac.  相似文献

We present an easy and straightforward synthesis of 3-arylpyrrolines 4a-g by repeated treatment of 4-aryl-1,2,5,6-tetrahydropyridines 2a-g with m-chloroperoxybenzoic acid (MCPBA) and boron trifluoride etherate (BF3-OEt2). The transformation proceeds via epoxidation, ring contraction, Baeyer-Villiger oxidation and elimination reaction and affords 3-arylpyrrolines 4a-g with 61-70% yield. This facile strategy was also used to synthesize racemic baclofen (6).  相似文献

A novel synthesis of highly functionalized alkylsulfanylmethyl arenes 8a-m through the ring transformation of 6-aryl-4-methyl/ethylsulfanyl-2H-pyran-2-one-3-carbonitriles 1a-j, by reaction with methyl vinyl ketone 2, is delineated and illustrated. Syntheses of rac/meso-{PhP(3-t-Bu-C5H3)2}Zr{Me3SiN(CH2)3NSiMe3} (rac-3/meso-3) and rac/meso-{PhP(3-t-Bu-C5H3)2}Zr{PhN(CH2)3NPh} (rac-4/meso-4) were achieved by metallation of K2[PhP(3-t-Bu-C5H3)2] · 1.3 THF (2) with Zr{RN(CH2)3NR}Cl2(THF)2 (where R = SiMe3 or Ph, respectively) using ethereal solvent. These isomeric pairs were characterized by 1H, 13C{1H}, and 31P{1H} NMR spectroscopy; rac-3 and rac-4 were also examined via single crystal X-ray crystallography. The structures of rac-3 and rac-4 are notable in the tendency of the cyclopentadienyl rings towards η3 coordination. While isolated samples of rac-3/meso-3 and rac-4/meso-4 slowly isomerize in tetrahydrofuran-d8 to equilibrium ratios, the isomerization rate for 3 is more than 15-fold greater than that for 4. In addition, equilibrium ratios are rapidly reached when isolated samples of rac-3/meso-3 and rac-4/meso-4 are exposed to tetrabutylammonium chloride in tetrahydrofuran-d8 solvent. We propose that a nucleophile (either chloride or the phosphine interannular linker) brings about dissociation of one cyclopentadienyl ring, thus promoting the rac/meso isomerization mechanism.  相似文献

Palladium complexes of N-phenyl-2-pyridylamine (4) and dipyridylamine substrates (7, 11) have been studied. Due to the coordination ability of the pyridine-nitrogen atoms, the pyridyl substrates, 4, 7, 11 were subjected to Pd(OAc)2 complexations and a number of N-aryl-2-pyridylamine Pd complexes (13-17) were isolated and characterised, in particular by NMR and ESI-MS. A new method for the preparation of the acetato-bridged six-membered ring palladacycle complex (13) of 4 is reported. The dipyridyl amines 7, 11 formed cis/trans bis-dentate acetato-bridged dimeric Pd2Lig2(OAc)2 (14a,b/16a,b) and Pd3Lig2(OAc)4 complexes (15a,b/17a,b). The N-aryl-2-pyridylamine substrates (4, 7, 11) were prepared by oxidative nucleophilic substitution, by 1,3-cycloaddition reaction or by Buchwald amination.  相似文献

The C,N-(trimethylsilyliminodiphenylphosphoranyl)silylmethylmetal complexes [Fe(L)2] (3), [Co(L)2] (4), [ZrCl3(L)]·0.83CH2Cl2 (5), [Fe(L)3] (6), [Fe(L′)2] (7) and [Co(L′)2] (8) have been prepared from the lithium compound Li[CH(SiMe2R)P(Ph)2NSiMe3] [1a, (R = Me) {≡ Li(L)}; 1b, (R = NEt2) {≡ Li(L′)}] and the appropriate metal chloride (or for 7, FeCl3). From Li[N(SiMe3)C(Ph)C(H)P(Ph)2NSiMe3] [≡ Li(L″)] (2), prepared in situ from Li(L) (1a) and PhCN, and CoCl2 there was obtained bis(3-trimethylsilylimino- diphenylphosphoranyl-2-phenyl-N-trimethylsilyl-1-azaallyl-N,N)cobalt(II) (9). These crystalline complexes 3-9 were characterised by their mass spectra, microanalyses, high spin magnetic moments (not 5) and for 5 multinuclear NMR solution spectra. The X-ray structure of 3 showed it to be a pseudotetrahedral bis(chelate), the iron atom at the spiro junction.  相似文献

The asymmetric Friedel-Crafts reaction between methyl (E)-2-oxo-4-aryl-3-butenoates (1a-c) and activated benzenes (2a-d) has been efficiently catalyzed by the ScIII triflate complex of (4′S,5′S)-2,6-bis[4′-(triisopropylsilyl) oxymethyl-5′-phenyl-1′,3′-oxazolin-2′-yl]pyridine (pybox 3). The 4,4-diaryl-2-oxo-butyric acid methyl esters (4) are usually formed in good yields and the enantioselectivity is up to 99% ee. The sense of the stereoinduction can be rationalized with the same octahedral complex (10) between 1, pybox 3 and Sc triflate already proposed for other reactions involving pyruvates, and catalyzed by the same complex.  相似文献

A simple and straightforward synthesis of 1-substituted-4-aryl-5-fluoro-1,2,3,6-tetrahydropyridine (3) or 1-substituted 4-diarylmethanoyl-4-fluoropiperidine (6) by the treatment of piperidinyl exo- or endo-olefin with 1-chloromethyl-4-fluoro-1,4-diazoniabicyclo[2.2.2]octane bis(tetrafluoroborate (Selectfluor) is reported. Two transformations from endo-olefin 1 to allylic fluoride 3 and from exo-olefin 2 to fluorohydrin 6 proceed via allylic fluorination and fluorohydroxylation in moderate yields. It presents two novel reactions promoted by Selectfluor and broadens the scope of application.  相似文献

Two new diamagnetic, mononuclear and aminated porphyrin complexes of O,O-trans-Cd (3-trans) and O,S-cis-Cd (4-cis) have been synthesized and characterized by 1H, 13C NMR spectroscopy. The crystal structures of (acetato)(N-2-furancarboxamido-meso-tetraphenylporphyrinato)cadmium(II) [Cd(N-NHCO-2-C4H3O-tpp)(OAc); 3-trans] and (acetato)(N-2-thiophenecarboxamido-meso-tetraphenylporphyrinato)cadmium(II) [Cd(N-NHCO-2-C4H3S-tpp)(OAc); 4-cis] were determined. The coordination sphere around Cd2+ is a distorted square-based pyramid in which the apical site is occupied by a bidentate chelating OAc group for 3-trans and 4-cis. The plane of three pyrrole nitrogen atoms [i.e., N(1), N(2), N(4) for 3-trans and N(1), N(2), N(3) for 4-cis] strongly bonded to Cd2+ is adopted as a reference plane 3N. The N(3) and N(4) pyrrole rings bearing the 2-furancarboxamido (Fr) and 2-thiophenecarboxamido groups in 3-trans and 4-cis, respectively, deviate mostly from the 3N plane, thus orienting separately with a dihedral angle of 33.4° and of 31.0°. In 3-trans, Cd2+ and N(5) are located on different sides at 1.06 and −1.49 Å from its 3N plane, while in 4-cis, Cd2+ and N(5) are also located on different sides at 1.04 and −1.53 Å from its 3N plane. An attractive electrostatic interaction between the Cd2+ and O(4) atoms in furan stabilizes the O,O-trans conformer of 3. A repulsive electrostatic interaction between Cd2+ and S(1)+ destabilizes the O,S-trans conformer of 4. Both of these repulsive and the mutually attractive interactions between S(1)+ and O(3) atoms favor the O,S-cis rotamer of 4 both in the vapor phase and in low polarity solvents. NOE difference spectroscopy, HMQC and HMBC were employed for the unambiguous assignment of the 1H and 13C NMR resonances of 3-trans and 4-cis in CDCl3 at 20 and −50 °C.  相似文献

In this Letter we present a method for the synthesis of N-acyl-N′-arylhexahydropyrimidines 1, by ring closure of N-acyl-N′-aryl-1,3-propanediamines 3 with formaldehyde. Cyclodehydrations were performed in aqueous medium under microwave irradiation, and led to high yields of the desired compounds in remarkably short reaction times. The method also allowed for the synthesis of hitherto unreported N-acyl-N′-arylhexahydro-1,3-diazepines 2. The acyclic tetramethylenic precursors 4 were synthesized by selective functionalization of N-arylputrescines.  相似文献

Cyclocondensation of 1-aryl-3-ferrocenyl-2-propen-1-ones (1) with hetaryl hydrazines resulted in N-hetaryl-3-aryl-5-ferrocenyl pyrazolines (3, 4). The analogous 3-aryl-1-ferrocenyl-2-propen-1-ones (5) gave the isomeric N-hetaryl-5-aryl-3-ferrocenyl-pyrazolines (6, 10), but in lower yield. The reaction of aryl-chalcones (7) with 4-hydrazino-phthalazinone led to 3,5-bis-aryl-N-hetaryl-pyrazolines (8) or to the corresponding ene-hydrazones (9). The structure of the new compounds was established by IR, 1H and 13C NMR spectroscopy, including DNOE, HMQC, HMBC and DEPT methods. For compounds 1b, 3b and 8b the stereo structure was elucidated also by X-ray diffraction.  相似文献
