在DMF溶剂中,不外加催化剂使邻取代芳香醛(1)与5,5-二甲基-1,3-环己三酮(2)发生缩合和加成反应生成3,3,6,6-四甲基-4a-羟基-9-芳基-1,8二氧化-2,3,4,4a,5,6,7,8,9a-十氢化-1H氧杂蒽(3a~3d)。在同样条件下,邻羟基芳香醛与5,5-二甲基-1,3-环己三酮则发生缩合,加成和脱水反应生成3,3-二甲基-9-(5,5-二甲基-3-羟基-2-环己烯-1-酮-2-基)-1-氧代-2,3,4,9-四氢化-1 氧杂蒽(4a~4b)。用单晶X-射线分析法确定了产物3a和4a的晶体结构。  相似文献

报道了采用溴氧化3-异丙烯基(艹卓)酚酮和3-肉桂酰基(艹卓)酚酮合成杂环并(艹卓)酮化合物的新方法。3-异丙烯基(艹卓)酚酮5位偶联产物1a~1f和3-肉桂酰基(艹卓)酚酮5位偶联产物3a~3d分别在吡啶介质中与过量溴作用生成5-取代苯偶氮基-7-溴-3-甲基-8-氢环庚并呋喃-8-酮2a~2f和6-取代苯偶氮基-2-苯基-8-溴-4,9-二氢环庚并吡喃-4,9-二酮4a~4d。  相似文献

用KBH_4,CaCl_2为还原剂使D及L缬氨酸甲酯还原得到光学活性产物R-3-甲基-2-氨基丁醇(2a)及S-3-甲基-2-氨基丁醇(2b)。2a,2b同CS_2在KOH存在下反应得到(R)-4-异丙基四氢噻唑-2-硫酮(3a)及(S)-4-异丙基四氢噻唑-2-硫酮(3b)。肉桂酰氯分别同3a及3b在Et_3N存在下反应得到N-肉桂酰(R)-4-异丙基四氢噻唑-2-硫酮(4a)及N-肉桂酰(S)-4-异丙基四氢噻唑-2-硫酮(4b)。用半经验的量子化学PM3方法研究了反应物和产物的电子结构,得到了产物的最优构型和电荷键序分布以及反应焓变。  相似文献

用X射线衍射方法测定了4-苯甲酰基-3-(邻氯苯基)-1-乙酸乙酯基-1a-苯基-1,1a,2,3-四氢化-4H-环丙胺-[1,2-a][1,5」-苯并二氯化合物的晶体结构,晶体结构测定结果表明分子核心骨架二氮杂为类船式构象.用动力学模拟退火(Simulated Annealing)方法对分子进行了构象分析,结果表明分子中的二氮杂七元环具有三种较稳定的构象,其中以船式构象为最稳定,并存在着明显的芳环堆积作用.分子中七元环同时并接苯环和三元环,且环中的N(2)—C(12)—C(11)—N(1)有π键成分,大大限制了七元环构象的相互转换.  相似文献

利用均四氯乙及二氯甲烷的胺化反应,合成了联氢化嘧啶衍生物1~5,4a,8b-反-4,5,8a,9a-四氮杂全氢芴(6)和8b,8c-顺-3a,4a,7a,8a-四氮杂环戊并全氢芴(7)。化合物(6)和(7)的顺反异构体在酸催化下开环,异构生成较稳定的6b和7a。  相似文献

以氯化三乙基苄基铵作为相转移催化剂, 用2, 3, 4, 6-四-O-乙酰基-β-D-溴代吡喃葡萄糖和2, 3, 4, 6-四-O-乙酰基-β-D-溴代吡喃半乳糖与羧酸反应, 合成了相应的1-O-酰基-2, 3, 4, 6-四-O-乙酰基-β-D-吡喃葡萄糖和半乳糖。通过解析这些化合物的^1H NMR和红外光谱, 确证其糖甙键构型为β。该相转移催化法具有反应条件温和, 后处理简单和立体选择性强等优点。  相似文献

以L-半胱氨酸盐酸盐出发,经与二卤代烷偶联,成酯和格氏反应制得三种双手性β-氨基醇,1,2-双[R-2-氨基-3-羟基-3,3-二苯基丙硫基]乙烷(4a),1,3-双[R-2-氨基-3-羟基-3,3-二苯基丙硫基]丙烷(4b),1,4-双[R-2-氨基-3-羟基-3,3-二苯基丙硫基]丁烷(4c)。将此类双手性β-氨基醇与硼烷在THF溶液中反应,in situ制备双手性恶唑硼烷催化硼烷对芳酮的不对称还原,产物苯乙醇的光学收率达72.8%、α-溴代苯乙醇的光学收率达91.4%。  相似文献

Salicylaldehyde reacts with dimethyl acetylenedicarboxylate in benzene solution to give a mixture of dimethyl o-formylphenoxymaleate, dimethyl o-formylphenoxyfumarate, 2,3-dicarbomethoxychrom - 2 - en - 4 - ol, 2,3 - dicarbomethoxychrom - 3 - en - 2 - ol, dimethyl (2,3 - dicarbomethoxychrom - 2 - en - 4 - yl) - oxalacetate and dimethyl fumarate. 2,3 - Dicarbomethoxychrom - 3 - en - 2 - ol in this reaction is formed from 2,3 - dicarbomethoxychrom - 2 - en - 4 - ol through a benzopyrylium intermediate. The reaction of salicylaldehyde with excess of dimethyl acetylenedicarboxylate, however, gives a mixture of 2,3 - dicarbomethoxychrom - 3 - en - 2 - ol and dimethyl (2,3 - dicarbomethoxychrom - 3 - en) - 2 - oxyfumarate. 2,3 - Dicarbomethoxychrom - 3 - en - 2 - ol itself reacts further with dimethyl acetylenedicarboxylate to give 2,3,12 - tricarbomethoxychrom - 3,4 - eno[2,3 - b] pyrone. Similarly, the reaction of o-hydroxyacetophenone with dimethyl acetylendicarboxylate gives a mixture of dimethyl o-acetylphenoxymaleate, dimethyl o-acetylphenoxyfumarate, 2,3 - dicarbomethoxy - 4 - methylchrom - 2 - en - 4 - ol and 2,3 - dicarbomethoxy - 4 - methylchrom - 3 - en - 2 - ol. Both 2-hydroxychalcone and 2 - hydroxy - 4′ - methoxychalcone give mixtures of chalcone - 2 - oxymaleate and chalcone - 2 - oxyfumarate. The reaction of 2,2t?-dihydroxychalcone, however, gives 2′-hydroxyflavone, in addition to the expected maleate and fumarate. Similar reactions of 5 - chloro - 2,2′ - dihydroxychalcone and of 3,5 - dibromo - 2,2′ - dihydroxychalcone, on the other hand, give 2,3 - dicarbomethoxy - 4 - (o - hydroxyphenacyl) - 6 - chlorochrom - 2 - ene and 2,3 - dicarbomethoxy - 4 - (o - hydroxyphenacyl) 6,8 - dibromochrom - 2 - ene, respectively, together with the corresponding maleates and fumarates.  相似文献

研究了新的手性试剂,5-(l-孟氧基)-3-溴-呋喃酮(5a)的合成及其与亲核性醇发生的串联不对称双Michael加成/分子内亲取代反应。通过此反应,一举生成了四个新的手性中心,得到了一般方法难以合成的含有多个手性中心的丁内酯并螺-环丙烷类化合物8a~8d。详细报道了8a~8d的合成方法以及它们的[α]、IR、UV、^1^HNMR、^13^CNMR、MS、元素分析等结构分析数据。此不对称双Michael加成/分子内亲核取代反应可以为某些新的光学活性螺-环内丙烷类化合物以及某些复杂结构的分子提供合成策略。  相似文献

以3-芳基-5-巯基-1,2,4-三唑为原料合成了20个3-芳基-1,2,4-三唑-5-巯基乙酸乙酯(2a~e)、3-芳基-1,2,4-三唑-5-巯基乙酸(3a~e)、3-芳基-5,6-二氢噻唑并[2,3-c]均三唑(5a~e)和3-芳基-6,7-二氢均三唑并[3,4-b][1,3]噻嗪(6a~e)。研究了3a~e在微波辐射下的环化反应,合成了5个3-芳基-5-氧代-6H-噻唑[2,3-c]均三唑(4a~e)。产物经元素分析、红外、核磁共振以及质谱方法确定了结构。初步研究了代表化合物的生物活性。  相似文献

Bioactivity-guided fractionation led to the isolation of a new steroidal saponin, orbiculatoside B, together with a pair of furostanol saponins, protobioside and methyl protobioside, from the fresh rhizomes of Dioscorea deltoidea Wall var. orbiculate. The new compound was identified to be isonarthogenin 3-O-α-L-thamnopyranosyl(1→2)-[α-L-rhamnopyranosyl-(1→4)]-β-D-glucopyranoside by various NMR techniques in combination with chemical methods. The three saponins showed strong activity against Pyricularia oryzae,and were cytotoxic to cancer cell line K562,HCT-15,A549, and A2780a in vitro.  相似文献

以两个新型手性配体S-(+)-2-(2-吡啶酰胺基)-2'-(二苯基膦基)-1,1'-联苯(1a)和S-(+)-2-(6-甲基-2-吡啶酰胺基)-2'-(二苯基膦基)-1,1'-联萘(1b)的铜配合物催化的二乙基锌对开链烯酮的1,4-共轭加成反应的研究。以1a为标准配体,查耳酮为代表性底物,考察了溶剂、催化剂前体等因素对反应的影响。在优化条件下,系统研究了以[Cu(OTf)](C~6H~6)~1~/~2/1b为催化剂,甲苯-二氯乙烷为溶剂时进行了二乙基锌对七个开链烯酮的1,4-共轭加成反应,取得了突破性进展,获得最高达98%e.e.值的加成产物。  相似文献

本文通过Knorr合成法制备了四个3-位乙酰基取代的吡咯衍生物:1, 2-二甲基-4-异丙基-5-苯基-3-乙酰基-吡咯(5a); 1, 2, 4-三甲基-5-对甲氧苯基-3-乙酰基-吡咯(5b); 1, 2, 5-三甲基-4-苯基-3-乙酰基-吡咯(5c); 1, 2, 5-三甲基-4-对甲氧苯基-3-乙酰基-吡咯(5d)。通过红外, 质谱, 核磁等方法对其结构进行了表征。测定了其中三个化合物的晶体结构。对这类吡咯环上4或5-位有芳环取代基时化合物的晶体结构特征进行了扼要讨论, 晶体衍射实验结果表明,4, 5-位上的芳环与吡咯环本身处于非共平面结构。  相似文献

We report on a stabilizer of the interaction between 14-3-3ζ and the Estrogen Receptor alpha (ERα). ERα is a driver in the majority of breast cancers and 14-3-3 proteins are negative regulators of this nuclear receptor, making the stabilization of this protein-protein interaction (PPI) an interesting strategy. The stabilizer ( 1 ) consists of three symmetric peptidic arms containing an arginine mimetic, previously described as the GCP motif. 1 stabilizes the 14-3-3ζ/ERα interaction synergistically with the natural product Fusicoccin-A and was thus hypothesized to bind to a different site. This is supported by computational analysis of 1 binding to the binary complex of 14-3-3 and an ERα-derived phosphopeptide. Furthermore, 1 shows selectivity towards 14-3-3ζ/ERα interaction over other 14-3-3 client-derived phosphomotifs. These data provide a solid support of a new binding mode for a supramolecular 14-3-3ζ/ERα PPI stabilizer.  相似文献

The stereochemistry of the cyclobutanones 1-7, resulted from the reaction of t-butylcyanoketene with bicyclo[2.2.1]heptene, bicyclo[2.2.1]heptadiene, 1,4 - dihydro - 1,4 - methanonaphthalene, 1,4 - dihydro - 9 - (1 - methylethylidene) - 1,4 - methanonaphthalene, 1,4 - dihydro - 1,4 - epoxynaphthalene, l,4,4a,8b - tetrahydro - 1,4 - methanobiphenylene (l,4,4a,8b) and 1,4,4a,8b - tetrahydro - 1,4 - methanobiphenylene(1,4,4aβ,8bβ) was established as having the cyclobutanone ring exo and the t-Bu group in the configuration. These findings represent a stereochemical argument in favour of a π2s + π2a reaction mode of t-butylcyanoketene to the above mentioned bicyclo[2.2.1]heptene derivatives. Observations regarding preservation of the original configurations of alkenes as well as the geometrical distorsion of the cyclobutanones are shortly discussed.  相似文献

Novozym-435 has been found to be an effective biocatalyst for the kinetic resolution of a series of racemic 2,3-allenols, affording highly optically active (S)-(-)-2,3-allenols and (R)-(+)-2,3-allenyl acetates in high yields and with excellent ee values. The reaction of 3-(n-butyl)-3,4-pentadien-2-ol (1 a) was successfully performed on a 10 g scale to afford the corresponding (S)-(-)-2,3-allenol (1 a) and (R)-(+)-2,3-allenyl acetate (2 a) in synthetically useful amounts and with high ee values. The advantages of this reaction are the ready availability of the starting materials, high stereoselectivities for both (-)-2,3-allenols and (+)-2,3-allenyl acetates, the use of a relatively high substrate concentration, and a lower catalyst loading. The resulting (S)-(-)-2,3-allenol 1 a can be converted into the corresponding chiral 2,5-dihydrofuran and the vinylic epoxide.  相似文献

选取三层原子簇模型模拟Ag(110)表面,采用量子化学的电子密度泛函方法(DFT)研究了氧原子吸附在Ag(110)表面而引起银表面的多种重构现象。通过计算体系结合能优化得到吸附后体系的表面几何构型,并给出了相关的电离能、电子跃迁能。计算表明:氧原子吸附在银原子的长桥位上,位于银表面之上约0.4 处,氧原子的吸附引起银原子表面强烈弛豫。第一、二层银原子间距扩张;第二、三层银原子间距收缩。由于氧原子的吸附,第一层银原子出现丢失行现象,第二层银原子出现两两成对现象,而第三层银原子出现弯曲现象。  相似文献

三氯锗基丙酸(1a)和三苯锗基丙酸(2a)与过量苯基溴化镁起反应时,这两个分子中的羧基表现出一些奇特性质。当1a和2a与不同浓度的苯基溴化镁起反应时,都能生成相应的酮(3a)和醇(4a)。在稀盐酸作用下,4a极易脱水生成1,1-二苯基-3-三苯锗基丙烯-1(5a)。当1a与苯基溴化镁起反应,然后再用稀盐酸处理也同样得到5a。5a的Ge-C键能被LiAlH~4高选择性地切断,生成三苯基锗烷(6a)和1,1-二苯基丙烯(6b)。  相似文献
