STUDIES ON THE MOLECULAR STRUCTURE OF WT-12 DERIVATIVE OF PYRAZOLO [3,4-d] - 1,3,2-DIAZAPHOSPHORIN-6-ONE

<正> The title compound was obtained by reaction of 1-phenyl-5-benzy-lamino-pyrazole-4-(N-methyl) carboxamide with p-fluorophenyl phosphorus dichloride and its molecular structure determined by X-ray diffraction method. The crystal is mono-clinic,space group P21/c,with a=11. 874(2),b= 12. 986(2),c=14. 587(2) A ,β= 106. 21(1)°,V=2159. 6A3,Mr = 430. 43(C24H20N4OFP),Z=4, Dx = 1. 324g/cm-3,μ(MoKa) = 1. 53cm-1 ,and F(000) = 896. The atoms in the pyrazolo[3,4-d] - 1,3,2-di-azaphosphorin-6-one are coplanar except the P atom.     

SYNTHESIS OF SOME NEW HETEROCYCLIC COMPOUNDS FROM BENZOPYRANO[2,3-c]PYRAZOL-3-ONE DERIVATIVES

The reaction of benzopyrano[2,3-c]pyrazol-3-one (1) with some active halo compounds, afforded compounds 4, 8 and 12, respectively. The cyanoethyl derivative 19 was synthesized and treated with active methylenes and sulfur or benzoylisothiocyanate and phenacyl bromide, to give compounds 24_a,b and 27. Compounds 25_a,b and 28 were obtained through the reaction of compounds 24_a,b with acrylonitrile or compound 27 with maleic anhydride. Thiation of compound 1 afforded the corresponding thio derivative 29. The reaction of 4-benzylidene-2-methyloxazolin-5-one with compounds 1 or 29 gave products 30_a,b , respectively.     

A NOVEL SYNTHESIS OF 2-SUBSTITUTED-4H-THIENO [2,3-d][1,3] OXAZIN-4-ONE AND 2,3-DISUBSTITUTED THIENO [2,3-d]PYRIMIDIN-4(3H)-ONE DERIVATIVES

Abstract

The synthesis of acetic 2-{[1-methyl-2-(4-oxo-5,6,7,8-tetrahydro-4H-benzo [4,5]-thieno [2,3-d] [1,3-oxazin-2-yl)ethylidene]amino}-4,5,6,7-tetrahydrohenzo[b]thiophene ?3-carboxylic acid anhydride 5 and 2-(oxopropyl)-5,6,7,8-tetrahydro-4H-benzo-[4,5] thieno[2,3-d][1,3]oxazin-4-one 7, has been achieved in three steps from ethyl 2-amino-4,5,6,7-tetrahydrobenzo(b]thiophene-3-carboxlate 1 via the reaction with ethyl acetoacetate followed by hydrolysis and acetic anhydride-induced cyclization. The 2-substituent in compound 5 has two functional groups i.e. active methylene and acid anhydride which are suitably located for intramolecular transformation. Thermal and/or base catalyzed intramolecular cyclization of 5 afforded 2-(4-acetoxy-(hydroxyl)-2-methyl-5,6,7,8-tetrahydrobenzo[4,5] thieno[2,3-b]pyridin-3-yl)-5,6,7,8- tetrahydro-4H-benzo[4,5]thieno[2,3-d] [1,3]oxazin-4-one 10 and 9 respectively. Treatment of 5 with hydrazine hydrate, aromatic and/or heterocyclic amines induced the same intramolecular cyclization with a concomitant oxazine-pyrimidine interconversion to give 3-amino(aryl or heteryl)-2-(4-hydroxy-5,6,7,8-tetrdhydrobenzo-[4,5]thieno[2,3-b]pyridin-3-yl)-3,4,5,6,7,8-hexahydrobenzo[4,5] thieno[2,3-d] pyrimid in-4-one 11–14 respectively.     

AN UNUSUAL ADDITION AND RING-CLOSURE REACTION OF 1-(2-BROMOETHYL)-2,3-DIHYDRO- 3-PROPYL-1,3,2-BENZO-DIAZAPHOSPHORIN-4(1H)-ONE 2-OXIDE WITH CARBON DISULFIDE FOR A NEW AND CONVENIENT SYNTHESIS OF THE FUSED PHOSPHORUS HETEROCYCLIC COMPOUND

ABSTRACT

The reaction of 1-(2-bromoethyl)-2,3-dihydro-3-propyl-1,3,2-benzodiazaphosphorin-4(1H)-one 2-oxide with carbon disulfide takes an alternative pathway in the use of different bases. The sodium hydride mediated reaction leads to the formation of the tricyclic fused 1,2,3,4,4a,4b,5,6-octahydro-6-oxo-5-propyl-4-thia-3,4b,4a-thiazphosphaphenanthridine 4a-oxide via addition of H-P bond across the double bond of carbon disulfide followed by intramolecular cyclization. In the presence of triethylamine, refluxing a mixture of 1-(2-bromoethyl)-2,3-dihydro-3-propyl-1,3,2-benzodiazaphosphorin-4(1H)-one 2-oxide with carbon disulfide in benzene takes an unusual course with formation in excellent yield of the first example of fused phosphorus heterocyclic 4-[1′-(β-bromoethyl)-4′-oxo- 3′-propyl-1′,2′,3′,4′-tetrahydro-1,3,2-benzodiazaphosphorin-2′- sulfide]-1,2,3,4,4a,4b,5,6-octahydro-6-oxo-5-propyl-3,4b, 4a-thiazphosphaphenanthridine 4a,2′-dioxide, which was confirmed by spectroscopic methods, microanalyses and single crystal X-ray structure determination.     

SYNTHESIS AND SOME REACTIONS OF 2-MERCAPTO-3,5,7-TRIPHENYLPYRIDO[2,3-d]-PYRIMIDINE-4(3H)-ONE

Abstract

2-Mercapto-3,5,7-triphenylpyrido[2,3-d]pyrimidine-4(3H)-one (II) was synthesized by the interaction of ethyl-2-amino-4,6-diphenylincotinate (I) with phenyl isothiocyanate in pyridine. The reaction of II with different reagents was carried out to give some condensed heterocyclic systems.     

SYNTHESIS OF 4,6-DISUBSTITUTED 5-THIOXO-1,2,4-TRIAZIN-3-ONE FROM BENZOTHIOFORMANILIDE

A method of synthesis of 4,6-disubstituted 5-thioxo-1,2,4-triazin-3-ones from benzothioformanilides and semicarbazide is described.And six newcompounds were synthesized by this method.     

VERSATILE SYNTHESIS AND FUNGICIDAL ACTIVITIES OF 6-AMINO-3- ALKYLTHIO-1,5-DIHYDRO-1-PHENYL-PYRAZOLO [3,4-D]PYRIMIDIN-4-ONE DERIVATIVES

A series of new 6-alkylamino-3-alkylthio-1-phenyl-1H-pyrazolo[3,4-d] pyrimidin-4(5H)-one derivatives 5 and 6 have been rapidly synthesized by a novel solution-phase regioselective synthetic method. Treatment of pyrazole o-aminoester 1 with dibromotriphenylphosphorane gave iminophosphorane 2, which underwent a aza-Wittig reaction with phenyl ioscyanate to provide the carbodiimide 3. The latter intermediate reacted with alkylamines and regioselectively provided the 1,5-dihydro-pyrazolo[3,4-d]pyrimidin-4-one derivatives 5 and 6, some of which exhibited good fungicidal activity.

A CONVENIENT SYNTHESIS OF 2,3-DISUBSTITUTED DERIVATIVES OF 4H-PYRIMIDO[2,1-b]-BENZOTHIAZOLE-4-ONE

ABSTRACT

4H-Pyrimido[2,1-b ]benzothiazole-2-thiomethyl-3-cyano-4-one (3) has been prepared by the reaction of 2-amino benzothiazole (1) with ethyl-2-cyano-3,3-bismethyl thio acrylate (2) in the presence of dimethyl formamide and anhydrous potassium carbonate. Susceptibility of compound (3), towards condensation with different reagents like aryl amines, heteryl amines, phenols and compounds containing active methylene groups has been investigated.     

ACTIVATED NITRILES IN HETEROCYCLIC SYNTHESIS: NOVEL SYNTHESIS OF PYRROLO[1,2-c]IMIDAZOLE AND PYRANO[2,3-d]IMIDAZOLE DERIVATIVES

Abstract

Several new pyrrolo[1,2-ylidene ethyl cyanoacetate (lc, d) with each of (2) and (11) afforded the 5-ylidene-2-thiohydantoins (Sa, b), (Wn. b) respectively. Compounds Sn, b react with each of malononitrile and ethyl cyanoacet-ate to give the corresponding pyrroloimidazole derivatives (h,b) and (an, b) respectively. The structure of the isolated products were established by elemental analyse and spectral data studies.     

SYNTHESIS AND REACTIONS OF SOME NEW HETEROCYCLIC COMPOUNDS CONTAINING THE THIENYLTHIENO[2,3-B]PYRIDINE MOIETY

(4-Aryl-3-cyano-6-(2-thienyl)pyridin-2-ylthio)acethydrazides (5a–c), 3-amino-4-aryl-6-(2-thienyl)thieno[2,3-b]pyridine-2-carbohydrazides (6a–c) and 3-amino-4-phenyl-6-(2-thienyl)thieno[2,3-b]pyridine-2-carboxylic acid (30) were prepared and employed as key intermediates in the synthesis of the title compounds.

6,7-Methylenedioxy-3-thia-1(2H,4H)acridone and the related acridine-fused quinolines and pyridines,andalso their flavylium-perchlorates were synthesized.The structures of all these compounds prepared are characterizedby elemental analysis.IR and H-NMR spectra.

5H-[1,3,4]Thiadiazolo[2,3-d][1,2,4]triazin-5-ones were synthesized by cyclocondensation of 4-amino-5-mercapto-1,2,4-triazin-3-ones with a variety of carboxylic acids in the presence of phosphorus oxychloride.

<正> The title compound C20H20N2O4(Mr=352.39)belongs to mono-clinic space group C2/c with a=9.208(4),b=11.714(3),c=32.336(8)A,β=100.83(2)°,V=7146.2(6)A3,Z=16,Dc=1.35 g/cm3,μ(Moka)=1.03 cm-1 and final R=0.06 for 1781 reflections with F≥2.5(F).There are two independent molecules in an asymmetric unit.Neither pyridinone nor phenyl ring is coplanar with the ketonic carbonyl group in the molecule.     

SYNTHESIS OF 3-(2-MERCAPTO-4-THIAZOLYL)-2H-1-BENZOPYRAN-2-ONE AND ITS DERIVATIVES

Abstract

Treatment of 3-(2-bromoacetyl)coumarins with ammonium dithiocarbamate provides 3-2-mercapto-4-thiazolyl)-2H-1-benzopyran-2-one (11) but not 4-hydroxy-4-(3-coumaririnyl) thiazolidine-2-thione (I). II undergoes smooth condensation with alkyl, aralkyl, phenacyl and acid halides to give corresponding thioethers and thioesters (III) respectively. The structures of the newly synthesized compounds were established on the basis of spectral data (IR, PMR and MS).

3-Methylfuran is an important building block of terpenoid compounds¹⁾ and some of these compounds are biologically active, for example as a defence substance of animal.²⁾ Though many methods have been developed for the syntheses of 3-substituted furan derivatives,³⁾ most of them lack the generality as a synthetic method of (3-furanyl) methyl derivative. Tannis has recently reported the construction of 3-substituted furan derivative by using 3-(lithiomethyl) furan or 3-(chloromagnesium methyl) furan, and demonstrated the generality of his method.⁴⁾
