In this work, an automatic multi-channel ink-jet for chemiluminescence (CL) analysis was developed. The four-channel ink-jet device was controlled by a home-made circuit. Differing from the classic flow injection CL, the whole procedure for CL analysis was automatically completed on a hydrophobic glass side. CL reaction of luminal and hydrogen peroxide for the determination of horseradish peroxidase (HRP) was selected as an application to automatic CL analysis platform. All solutions delivered by different channels were precisely ejected to the same position of the glass slide for the CL analysis. The consumption of reaction solution was reduced to nanoliter level. The whole CL analysis could be completed in less than 4 min, which was benefited from the prompt solution mixing in small size of droplet. The CL intensity increased linearly with HRP concentration in the range from 0.01 to 0.5 μg mL−1. The limit of detection (LOD) (S/N = 3) was 0.005 μg mL−1. Finally, the automatic CL system could also be used for the detection of HRP in HRP–protein conjugates, which showed its practical application in immunoassay.  相似文献

In this study, the catalytic activity and stability of flowerlike hybrid horseradish peroxidase (HRP) nanobiocatalyst (HRP-Cu 2+ ) obtained from Cu 2+ ions and HRP enzyme in the polymerization reaction of guaiacol were analyzed. We demonstrated that HRP-Cu 2+ and hydrogen peroxide (H 2 O 2 ) initiator showed significantly increased catalytic activity and stability on the polymerization of guaiacol compared to that of free HRP enzyme. Poly(guaiacol) was observed with quite high yields (88%) and molecular weights (38,000 g/mol) under pH 7.4 phosphate-buffered saline (PBS) conditions at 60 °C with 5 weight% of HRP-Cu 2+ loading. HRP-Cu 2+ also shows very high thermal stability and works even at 70 °C reaction temperature; free HRP enzyme denatures at that temperature. Furthermore, HRP-Cu 2+ provided considerable repeated use and showed some degree of catalytic activity, even after the fourth recycle, in the polymerization of guaiacol.  相似文献

Direct electron transfer was found to be greatly facilitated for horseradish peroxidase (HRP) in a didodecyldimethylammonium bromide (DDAB) biomembrane-like film at a pyrolytic graphite (PG) electrode involving the FeIII FeII couple. The heterogeneous electron transfer rate constant ks was fitted as 9.0 s−1 using the non-linear regression analysis of the square wave voltammograms at a series of frequencies and pulse heights. The pH dependence of the formal potential for HRP in DDAB film at medium pH environments suggested one-proton transfer coupled with a one-electron transfer reaction. Scanning electron microscopy (SEM) showed different film morphology for HRP and HRP---DDAB films. UV–vis and reflectance absorption infrared (RAIR) spectra inferred that the heme state of HRP in DDAB film was similar to that in its native state. Circular dichroism (CD) results indicated slight perturbation of DDAB on the second structure of HRP. Thus, the embedded HRP in the biomembrane-like DDAB film showed nearly native structural properties and improved electrochemical characteristics. This has potential value for the basic and applied bioelectrochemistry of enzymes.  相似文献

The detection of hydrogen peroxide is detailed using horseradish peroxidase and anthraquinone. Both species are immobilized on a glassy carbon electrode substrate. This dual immobilization gives rise to lower detection limits compared with the situation when either of the species is immobilized. Detection limits of 40 nM are reported within physiologically-relevant media.  相似文献

A simple and versatile approach for covalent immobilization of redox protein on solid surface via self-assembled technique and click chemistry is reported. The alkynyl-terminated monolayers are obtained by self-assembled technique, then, azido-horseradish peroxidase (azido-HRP) was covalent immobilized onto the formed monolayers by click reaction. The modified process is characterized by reflection absorption infrared spectroscopy (RAIR), surface-enhanced Raman scattering spectroscopy (SERS) and electrochemical methods. All the experimental results suggest that HRP is immobilized onto the electrode surface successfully without denaturation. Furthermore, the immobilized HRP shows electrocatalytic reduction for H2O2, and the linear range is from 5.0 to 700 μM. The heterogeneous electron transfer rate constant ks is 1.11 s−1 and the apparent Michaelis-Menten constant is calculated to be 0.196 mM.  相似文献

The solution behaviour has been investigated for an alcohol ethoxylate terminated with a formic acid ester. This compound has previously been reported to be an important degradation product in the auto-oxidation of alcohol ethoxylates. In this work we have investigated the solution behaviour of the formic acid ester surfactant C12H25(OCH2CH2)4OCHO (C12E4---OCHO). The pure formate was found to be sparsely soluble in water with no clear point at 0.1%. The critical micelle concentration was found to be 129 μM at 35°C, compared to 50 μM for the parent surfactant C12H25(OCH2CH2)5OH (C12E5). To mimic the behaviour of the oxidised surfactant, the formate was mixed in different ratios with C12E5 and the cloud point, surface tension and critical micelle concentration (cmc) of these mixtures were studied. The gradual increase of formate was found to shift the cloud point and isotropic regions to lower temperatures. The cmc of the mixture was found to be lower than for the pure surfactant. The favourable interaction was analysed according to the non-ideal model by Rubingh and the interaction parameter, β, was determined to be −4±0.53, which is unusually large for a mixture of two non-ionic surfactants. These results indicate that the reduction of cloud point observed during oxidation of non-ionic surfactants can in part be attributed to the formation of formate esters.  相似文献

In this article tetrabutylammonium bromide (TBAB) was first added in buffer to compose a convenient and environmentally friendly system, and enzymatic polymerization of phenol catalyzed by horseradish peroxidase (HRP) could proceed efficiently in this system. When TBAB was added, the most conversion of phenol could reach 99.1%. The phenol polymer was considered to consist of a mixture of phenylene (Ph) and oxyphenylene (Ox) units by IR analysis, and the ratio of phenylene to oxyphenylene units (Ph/Ox) was measured by titration. Moreover, the effects of the dosage of horseradish peroxidase (HRP) and pH value on the conversion of phenol were investigated. The reaction performed very effectively in this novel system when the addition of HRP was only 0.2 mg. In all cases, the weight-average molecular weight calculated by GPC-SLS was in a range from 12000 Da to 30000 Da. The phenol polymer prepared in the present research possessed good thermal stability shown by TG analysis.  相似文献

One-step, diameter-selective dispersion of single-walled carbon nanotubes (SWCNTs) has been accomplished through noncovalent complexation of the nanotubes with a water-soluble, biocompatible polymer chitosan at room temperature. Such chitosan-wrapped individual SWCNTs can be used for the immobilization of horseradish peroxidase (HRP) and be used to construct an electrode for direct bioelectrochemical sensing without an electron mediator. The direct electron transfer between HRP and the electrode surface was observed with a formal potential of approximately −0.35 V (vs. saturated calomel electrode) in phosphate buffer solution and the calculated heterogeneous electron transfer rate constant is approximately 23.5 s−1. Experimental results indicate that the immobilized HRP retains its catalytic activity for the reduction of nitric oxide. Such an HRP–SWCNT–chitosan-based biosensor exhibited a rapid response time of less than 6 s and a good linear detection range for nitrite concentration, from 25 to 300 μM with a detection limit of 3 μM. The apparent Michaelis–Menten constant (K m) and the maximum electrode sensitivity (imax/K m) are found to be 7.0 mM and 0.16 μA mM−1, respectively. Both the unique electrical properties of SWCNTs and biocompatibility of chitosan enable the construction of an excellent biosensing platform for improved electrocatalysis of HRP, allowing, specifically, the detection of trace levels of nitric oxide.  相似文献

In this contribution the substitution of the central protoporphyrin IX iron complex of horseradish peroxidase by the respective osmium porphyrin complex is described. The direct electrochemical reduction of the Os containing horseradish peroxidase (OsHRP) was achieved at ITO and modified glassy carbon electrodes and in combination with spectroscopy revealed the three redox couples OsIIIHRP/OsIVHRP, OsIVHRP/OsVHRP and OsVHRP/OsVIHRP. The midpoint potentials differ dependent on the electrode material used with E1/2 (OsIII/IV) of − 0.4 V (ITO) and − 0.25 V (GC), E1/2 (OsIV/V) of − 0.16 V (ITO) and + 0.10 V (GC), and E1/2 (OsV/VI)of + 0.18 V (ITO), respectively. Moreover, with immobilised OsHRP the direct electrocatalytic reduction of hydrogen peroxide and tert-butyl hydroperoxide was observed. In comparison to electrodes modified with native HRP the sensitivity of the OsHRP-electrode for tert-butyl hydroperoxide is higher.  相似文献

A novel hydrogen peroxide biosensor has been fabricated based on covalently linked horseradish peroxidase (HRP) onto L- glutathione self-assembled monolayers (SAMs). The SAMs-based electrode was characterized by electrochemical methods, and direct electrochemistry of HRP can be achieved with formal potential of-0.242 V (vs. saturated Ag/AgCl) in pH 7 phosphate buffer solution (PBS), the redox peak current is linear to scan rate and rate constant can be calculated to be 0.042 s^-1. The HRP-SAMs- based biosensors show its better electrocatalysis to hydrogen peroxide in the concentration range of 1 × 10^-6 mol/L to 1.2 × 10^-3 mol/L with a detection limit of 4 × 10^-7 mol/L. The apparent Michealis-Menten constant is 3.12 mmol/L. The biosensor can effectively eliminate the interferences of dopamine, ascorbic acid, uric acid, catechol and p-acetaminophen.  相似文献

It has previously been shown that alcohol ethoxylates readily undergo autoxidation and that one of the major oxidation products is the surfactant aldehyde, i.e. an ethoxylate carrying a –CH2CHO group at the terminal end of the polyoxyethylene chain. In this work the cloud point, phase behavior and aggregation characteristics of the surfactant aldehyde produced by oxidation of C12H25(OCH2CH2)5OH (C12E5) are determined and compared with the values obtained with the parent surfactant. It was found that the physico–chemical behavior of the two species was very similar, which indicates that a considerable portion of the aldehyde group is in hydrated state, i.e. the surfactant aldehyde consists of a mixture of aldehyde in carbonyl form and the corresponding geminal diol. The cloud point of the surfactant aldehyde decreased rapidly with time, even when it was stored at low temperature. Also the parent surfactant and its homologue C12E6 exhibited a decrease in cloud point during storage. For instance, a 1% aqueous solution of C12E6 showed a cloud point decrease from 62 to 32°C after 4 months storage at 40°C. Such a change in solution behavior can have important practical implications.  相似文献

In this work, novel mesoporous silica hollow spheres (MSHS) were chosen as an immobilization matrix, to construct a mediator-free third-generation HRP biosensor. UV-vis spectroscopy revealed that horseradish peroxidase (HRP) entrapped in MSHS could retain its native structure. FTIR spectroscopy and nitrogen adsorption-desorption isotherms indicated that HRP are intercalated into the mesopores. The direct electron transfer of HRP entrapped in MSHS was observed. A pair of stable and well-defined redox peaks of HRP with a formal potential of about −0.150 V (vs. Ag/AgCl) in 0.1 M pH 7.0 phosphate-buffered solution (PBS) were obtained. The biosensor exhibited a fast amperometric response to H2O2 with a linear range of 3.9 × 10−6 to 1.4 × 10−4 M (R = 0.997, N = 20). The detection limit was 1.2 × 10−6 M based S/N = 3.  相似文献

A highly soluble poly(3,4-ethylenedioxythiophene)-poly(styrene sulfonic acid)/Au (PEDOT-PSS/Au) nanocomposite was prepared via one-step chemical synthesis and the matrix was characterized by UV-vis spectroscopy (UV-vis), Fourier transform infrared spectroscopy (FT-IR), scanning electron microscope (SEM) and transmission electron microscope (TEM). Due to the excellent aqueous compatibility and biocompatibility, the PEDOT/PSS-Au nanocomposite can be used as biomaterial for enzymes immobilization. In this system, redox enzyme, horseradish peroxidase (HRP) was integrated with PEDOT/PSS-Au nanocomposite and the direct electron transfer of HRP was observed. Moreover, we find that the HRP/PEDOT-PSS/Au modified electrode shows excellent electrocatalytic ability for H2O2 and the formal Michaelis-Menten constant was 0.78 mmol/L. The response currents have good linear relation with the concentrations of H2O2 with a linear range from 2.0 × 10−7 to 3.8 × 10−4 mol/L.  相似文献

The physical immobilization behavior of horseradish peroxidase (HRP) on cellulosic fiber surfaces was characterized using adsorption and inactivation isotherms measured by the depletion method followed by fitting of Langmuir’s and Freundlich’s models to the experimental data. The adsorption and inactivation behavior of simpler and relatively non-porous high and low crystalline cellulosic substrates (microcrystalline cellulose and regenerated cellulose) as well as more complex and porous cellulosic pulp fibers (bleached kraft softwood fibers) were investigated. The effect of the sorbent surface energy on HRP adsorption was demonstrated by increasing the hydrophobicity of the cellulosic fibers using an internal sizing agent. The influence of the fiber surface charge density on HRP adsorption was studied via modification of the cellulosic fibers using TEMPO (2,2,6,6-tetramethyl-1-piperidiniloxy radical)-mediated oxidation methods. Results showed that hydrophobic interactions had a much larger effect on HRP adsorption than electrostatic interactions. More hydrophobic fiber surfaces (lower polar surface energy) result in larger enzyme-fiber binding affinity constants and higher binding heterogeneity. It was also found that oxidation of the cellulosic fiber substrate reduces enzyme adsorption affinity but significantly increases the loading capacity per unit weight of the surface.  相似文献

Stabilization of Horseradish Peroxidase (HRP; EC 1.11.1.7) against temperature and pH via the formation of the conjugates obtained by multipoint covalent bonding of dextran aldehyde (DA) to the enzyme were studied. Hence, three different molar weighted dextrans (17.5 kD, 75 kD, 188 kD) were covalently bonded to purified enzyme with different molar ratios (nHRP/nDA 20/1, 10/1, 1/1, 1/5, 1/10, 1/15, 1/20). The thermal stabilities of the obtained conjugates were evaluated with the activities determined at different temperatures (25, 30, 35, 40, 50, 60, 70, 80°C) applying 60 minutes incubation time. Conjugates formed were characterized by gel-permeation chromatography (GPC) and fluorescence techniques. The conjugate synthesized using dextran 75 kDa with nHRP/nDA 1/10 molar ratio showed better thermal stability than other conjugates and purified enzyme at pH 7. This conjugate also has wider activity pH range than purified enzyme. In addition, mentioned conjugate at pH 7 had very long storage lifetime compared to purified enzyme at +4°C and room temperature; which is considered a favorable feature for usage in practice.   相似文献

A new kind of magnetic dextran microsphere (MDMS) with uniform shape and narrow diameter distribution has been prepared from magnetic iron nanoparticles and dextran. Horseradish peroxidase (HRP) was successfully immobilized on the surface of an MDMS-modified glassy-carbon electrode (GCE), and the immobilized HRP displayed excellent electrocatalytic activity in the reduction of H2O2 in the presence of the mediator hydroquinone (HQ). The effects of experimental variables such as the concentration of HQ, solution pH, and the working potential were investigated for optimum analytical performance. This biosensor had a fast response to H2O2 of less than 10 s and an excellent linear relationship was obtained in the concentration range 0.20 μmol L−1–0.68 mmol L−1, with a detection limit of 0.078 μmol L−1 (S/N = 3) under the optimum conditions. The response showed Michaelis–Menten behavior at larger H2O2 concentrations, and the apparent Michaelis–Menten constant was estimated to be 1.38 mmol L−1. Moreover, the selectivity, stability, and reproducibility of the biosensor were evaluated, with satisfactory results. A hydrophilic polyacrylonitrile (PAN) flat sheet membrane was aminated (8.5 μmol of NH2/mg of dry support) for covalent binding of horseradish peroxidase (HRP), mediated by the soluble carbodiimide 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC). Silica microbeads derivatized by silanization, to yield an aminated support, and commercial aminated glass microbeads were also coupled to HRP with EDC or activated with glutaraldehyde. The immobilized enzyme activities were determined in a batch enzyme reactor with an external loop, the highest specific immobilized HRP activity being obtained on the glass support (55.8U/mg of protein). Continuous operational stability studies showed that hydrophilic PAN membrane led to the highest retention of HRP activity after an overall period of 35 h, with a normalized productivity of 59.5 μmol of H2O2 reduced/(h·Uimmob HRP).  相似文献

Uric acid (UA) is determined using the UV-vis molecular absorption properties of peroxidase (HRP). The method as a whole involves UA oxidation in the presence of uricase (UOx), giving H2O2. The H2O2 then reacts with HRP forming the compound I species which returns to its initial form by reaction with UA and intramolecular reduction. The molecular absorption changes of HRP at 420 nm during the reaction enable the UA to be determined. A mathematical model relating the analytical signal to UA, UOx and HRP has been developed and experimentally validated. The possibility of carrying out both enzymatic reactions sequentially or simultaneously is discussed, the latter option producing better analytical performances. The method permits UA determination in the range 1.5 × 10−6-4.0 × 10−5 M, with an R.S.D. of about 3% (n = 5, 1.5 × 10−6 M UA). It has been applied to analyte determination in synthetic serum samples.  相似文献
