A new abscisic acid derivative, named litseaglutinan A ( 1 ), and a new arylnaphthalene‐type lignan, (7′S,8R,8′S)‐4,4′,9‐trihydroxy‐3,3′,5‐trimethoxy‐9′‐Oβ‐D ‐xylopyranosyl‐2,7′‐cyclolignan ( 2 ), were isolated from the AcOEt extract of Litsea glutinosa, together with nine known lignans. A new abietane diterpene, named abietopinoic acid, and a new podocarpane diterpene, named podopinoic acid, were isolated from the acetone extract of the heartwood of Pinus massoniana. Their structures were established as 12‐hydroxy‐7‐oxoabieta‐8,11,13‐trien‐18‐oic acid ( 1 ) and 13‐hydroxy‐7‐oxopodocarpane ( 2 ) by means of spectroscopic analyses including 2D‐NMR. To the best of our knowledge, this is the second report of a podocarpa‐8,11,13‐trien‐18‐oic acid diterpene, isolated from the genus Pinus.  相似文献

A new helvolic acid derivative named helvolic acid methyl ester (1), together with two known helvolic acid compounds, helvolic acid (2) and hydrohelvolic acid (3), were isolated from the fermentation of endophytic fungus Fusarium sp. in Ficus carica leaves. Their structures were elucidated and identified by spectroscopic methods. Compounds 1–3 showed potent antifungal and antibacterial activities.  相似文献

Successful application of matrix‐assisted laser desorption/ionization (MALDI) MS started with the introduction of efficient matrices such as cinnamic acid derivatives (i.e. 3,5‐dimethoxy‐4‐hydroxycinnamic acid, SA; α‐cyano‐4‐hydroxycinnamic acid). Since the empirical founding of these matrices, other commercial available cinnamic acids with different nature and location of substituents at benzene ring were attempted. Rational design and synthesis of new cinnamic acids have been recently described too. Because the presence of a rigid double bond in its molecule structure, cinnamic acids can exist as two different geometric isomers, the E‐form and Z‐form. Commercial available cinnamic acids currently used as matrices are the geometric isomers trans or E (E‐cinnamic and trans‐cinnamic acids). As a new rational design of MALDI matrices, Z‐cinnamic acids were synthesized, and their properties as matrices were studied. Their performance was compared with that of the corresponding E‐isomer and classical crystalline matrices (3,5‐dihydroxybenzoic acid; norharmane) in the analysis of neutral/sulfated carbohydrates. Herein, we demonstrate the outstanding performance for Z‐SA. Sulfated oligosaccharides were detected in negative ion mode, and the dissociation of sulfate groups was almost suppressed. Additionally, to better understand the quite different performance of each geometric isomer as matrix, the physical and morphological properties as well as the photochemical stability in solid state were studied. The influence of the E/Z photoisomerization of the matrix during MALDI was evaluated. Finally, molecular modeling (density functional theory study) of the optimized geometry and stereochemistry of E‐cinnamic and Z‐cinnamic acids revealed some factors governing the analyte–matrix interaction. The isolation and structure elucidation of a new triterpenic acid named floccosic acid ( 1 ) is reported on the basis of the 1D‐ and 2D‐NMR assignments. This secondary metabolite was isolated as a new constituent, along with the known triterpenoids, betulinic acid and β‐amyrin. All these compounds were purified by repeated column chromatography of the MeOH extract of Nepeta floccosa. The structure elucidation of the new compound was accomplished by the combined mass spectrometry (MS), infrared (IR) and ultraviolet (UV) absorption spectroscopy, one‐ (1H‐ and 13C‐) and two‐dimensional (H? C correlations; HMBC and HSQC) NMR techniques. The known compounds were identified by comparison of their physical and spectroscopic data with those reported in the literature.  相似文献

A new subergane‐type sesquiterpene, named epoxysubergorgic acid ( 1 ), along with seven known sesquiterpenes, were isolated from the gorgonian coral Subergorgia suberosa. The structure of the new compound was determined by extensive spectroscopic analyses. The previously uncertain absolute configuration of the known analogues 2 – 6 was determined on the basis of CD, Mosher's method, and through chemical conversions. A new caffeoylgluconic acid derivative, trans-caffeoyl-6-O-d-gluconic acid methyl ester (1), together with two known compounds named trans-caffeoyl-6-O-d-glucono-γ-lactone (2) and trans-caffeoyl-6-O-d-gluconic acid (3), was isolated from the nearly ripe fruits of Evodia rutaecarpa (Juss.) Benth.. These compounds were isolated by various separation methods associated with the UPLC-Q-TOF-MS technique. Aristolochic acids (AAs) are a mixture of structural-related compounds, in which aristolochic acid I (AA I) and aristolochic acid II (AA II) are reported to be correlated with Aristolochic acid nephropathy (AAN). In this work, a rapid and sensitive ultra-high-pressure liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) method was developed to determine AA I and AA II in herbal products and biological fluids. By using gradient elution with a mobile phase composed of a mixture of 10 mM ammonium formate buffer (pH 3.0) and acetonitrile, AAs could be determined within 10 min. Under optimum UHPLC-MS/MS conditions, the limit of detections was 0.14 and 0.26 ng mL−1 for AA I and AA II, respectively. Run-to-run repeatability and intermediate precision of peak area for AA I and AA II were less than 5.74% relative standard deviation (RSD). Accuracy was tested by spiking 10, 100 and 1000 ng mL−1 in rat serum and the recoveries were within 76.5-92.9%. Matrix effects were within 78.8-127.7%. The developed method was successfully applied to determine AA I and AA II in several herbal products and to investigate their pharmacokinetic behavior in female Wister rats. The result shows that the developed UHPLC-MS/MS method is efficient, sensitive, and accurate for the determination of AA I and AA II in herbal products and biological samples.  相似文献

The phytochemical investigation of Xylopia langsdorffiana A.St.‐Hil. & Tul . led to the isolation of eight diterpenes, i.e., of the four new compounds (5β,7β,8α,9β,10α,12α)‐atisane‐7,16‐diol 7‐acetate ( 1 ), named xylodiol 7‐acetate, (5β,8α,9β,10α,12α)‐16‐hydroxyatisan‐7‐one ( 2 ), named xylopinone, (3α,12Z)‐3‐hydroxy‐ent‐labda‐8(20),12,14‐trien‐18‐oic acid ( 3 ), named labdorffianic acid A, and 8,20‐epoxy‐13‐hydroxy‐ent‐labd‐14‐en‐18‐oic acid ( 4 ), named labdorffianic acid B, and of the four known compounds 5 – 8 , i.e., ent‐kauran‐16‐ol, ent‐kaur‐16‐en‐19‐oic acid, ent‐kaur‐16‐en‐19‐ol, and ent‐trachyloban‐18‐oic acid. The structures were established by IR, HR‐ESI‐MS, and NMR data analysis with the aid of 2D techniques.  相似文献

A new and fast sample preparation technique based on three‐phase hollow fiber liquid‐phase microextraction with a magnetofluid was developed and successfully used to quantify the aristolochic acid I (AA‐I) and AA‐II in plasma after oral administration of Caulis akebiae extract. Analysis was accomplished by reversed‐phase high‐performance liquid chromatography with fluorescence detection. Parameters that affect the hollow fiber liquid‐phase microextraction processes, such as the solvent type, pH of donor and acceptor phases, content of magnetofluid, salt content, stirring speed, hollow fiber length, extraction temperature, and extraction time, were investigated and optimized. Under the optimized conditions, the preconcentration factors for AA‐I and AA‐II were >627. The calibration curve for two AAs was linear in the range of 0.1–10 ng/mL with the correlation coefficients >0.9997. The intraday and interday precision was <5.71% and the LODs were 11 pg/mL for AA‐I and 13 pg/mL for AA‐II (S/N = 3). The separation and determination of the two AAs in plasma after oral administration of C. akebiae extract were completed by the validated method.  相似文献

A new cycloartane triterpenoid, named gardenolic acid C (1), a new ursane triterpenoid, named 3β,16β,21β,23,24-pentahydroxy urs-12,18,20-trien-28-oic acid γ-lactone (2), together with three know triterpenoids, gardenolic acid A (3), gardenolic acid B (4), and 3α,16β,23,24-tetrahydroxy-28-nor-ursane-12,17,19,21-tetraen (5) were isolated from the fruits of Gardenia jasminoides Ellis. The structures of these compounds were elucidated by analyses of spectroscopic data. All isolates were evaluated for their neuroprotective effects in vitro.

Studies on the chemical constituents of the aerial parts of Kalidium foliatum led to the isolation of three new olean‐12‐ene‐23,28‐dioic acids in their either partly or fully esterified forms (Me or glucosyl (Glc) esters), which were named kalidiumoside A ( 1 ), kalidiumin ( 2 ), and kalidiumoside B ( 3 ). Their structures were elucidated through spectral studies including 2D‐NMR experiments (HMQC, HMBC, COSY, NOESY) and J‐resolved spectra. Also isolated were the two known compounds dianic acid and dianoside F, which were identified through comparison of their spectroscopic data with those reported in the literature.  相似文献

Aristolochic acids (AA) are toxic components of Aristolochia plants which result in diseases of the kidney such as urothelial cancer. It is, therefore, essential to monitor the amount of aristolochic acid in herbal medicines. In this study a reversed-phase high-performance liquid-chromatographic (HPLC) method has been developed for rapid determination of aristolochic acids I and II. Baseline separation was achieved within five minutes by use of an ODS C18 column with methanol–water, 60:40, as mobile phase. Two kinds of aristolochic acid were successfully determined in 31 herbal samples of Aristolochia fangchi Wu and Caulis Aristolochiae Manshuriensis. The results indicated that in most samples the aristolochic acid I content is much higher than that of aristolochic acid II. The two kinds of aristolochic acid were not detected in Aristolochia fangchi from the Guangdong region, so Aristolochia fangchi from this region is recommended for use in herbal remedies.  相似文献

By degradation and nitration of the reaction product three new compounds, a dinitrophenanthrene, a nitro- and a dinitrophenanthrene acid were obtained and their structures were established.

Two new iridoids, 10‐O‐benzoyl‐6′‐Oα‐L ‐arabino(1→6)‐β‐D ‐glucopyranosylgeniposidic acid ( 1 ), deacetyl‐6‐ethoxyasperulosidic acid methyl ester ( 2 ), along with 14 other known iridoids, were isolated from the whole plant of Hedyotis diffusa Willd. Their structures were determined on the basis of spectroscopic analysis. The minor unidentified compounds of the acetic acid/perchloric acid dehydration of elemol ( 1 ) were fully characterized. The structure and relative configuration of the less polar fragrant compound 2 , named elemoxide, was deduced by 1D‐ and 2D‐NMR data including C,C‐connectivity, NOE, and NOESY experiments. The absolute configuration was established as (3S,3aR,7aR)‐1,3,3a,4,7,7a‐hexahydro‐6‐isopropyl‐1,1,3,3a‐tetramethylisobenzofuran ( 2 ) on the basis of its preparation from elemol ( 1 ). (+)‐β‐cyperone ( 3 ), a known sesquiterpene, was also identified as a minor product of the reaction. A plausible mechanistic explanation for the formation of elemoxide ( 2 ) and (+)‐β‐cyperone ( 3 ) is presented.  相似文献

A new furan derivative named 5-acetoxymethylfuran-3-carboxylic acid (2), together with a known furan compound, 5-hydroxymethylfuran-3-carboxylic acid (1), were isolated from the fermentation of Aspergillus flavus, endophytic fungi in Cephalotaxus fortunei. The structures of 1 and 2 were elucidated by NMR, IR, UV and MS data, as well as compared with literature data. The compounds 1 and 2 exhibited potent antibacterial activity against Staphylococcus aureus with MIC values of 31.3 and 15.6 μg/mL, respectively. The crystal structures of cis‐3‐iodoacrylic acid (1), trans‐3‐iodoacrylic acid (2), trans‐3‐iodoacrylic acid methyl ester (3), 3,3‐diiodopropanoic acid (4), and trans‐2,3‐diiodoacrylic acid (5) are reported. Compounds 1 and 2 are the kinetic and thermodynamic products, respectively, of the hydroiodination of propiolic acid. Compound 4 results from addition of a second equivalent of hydroiodic acid to 1 or 2, whereas 5 results from the addition of trace elemental iodine to propiolic acid.  相似文献
