2-Amino-2H-1,4-benzoxazin-3(4H)-one 3 , 2-amino-2H-1,4-benzothiazin-3(4H)-one 4 , 2-mercapto-2H-1,4-benzoxazin-3(4H)-one 7 , and 2-mercapto-2H-1,4-benzothiazin-3(4H)-one 8 representing aza and thio analogues of the natural product's aglucone Blepharigenin (2-hydroxy-2H-1,4-benzoxazin-3(4H)-one) from Gramineae and Acantnaceae species have been synthesized for the first time from their 2-bromo precursors 1 and 2 . Attempts to similarly prepare the 4-hydroxy derivatives of 7 and 8 , which would represent new thio analogues of the naturally occurring cyclic hydroxamic acid, 2,4-dihydroxy-2H-1,4-benzoxazin-3(4H)-one, have failed.  相似文献

A new representative of benzothiazines, (2Z)-2-(nitromethylene)-2H-1,4-benzothiazin-3(4H)-one, was synthesized, and its structure was studied. According to 1H NMR, IR, and UV spectral and X-ray diffraction data, the molecules are planar highly conjugated structures that exist in crystal as centrosymmetric dimers; the latter form layers overlapping with each other and held together by strong π-π interaction between the carbonyl groups of neighboring layers.  相似文献

The present work reports an experimental and computational study of the energetics of 1,2-benzisothiazol-3(2H)-one and 1,4-benzothiazin-3(2H, 4H)-one. The standard (p° = 0.1 MPa) massic energy of combustion, at T = 298.15 K, of each compound was measured by rotating bomb combustion calorimetry, in oxygen that allowed the calculation of the respective standard molar enthalpy of formation, in the condensed phase, at T = 298.15 K. The standard molar enthalpies of sublimation, at T = 298.15 K, were measured by high-temperature Calvet microcalorimetry. From the combination of data obtained by both techniques we have calculated the standard molar enthalpies of formation, in the gaseous phase, at T = 298.15 K. In addition, computational calculations were carried using the density functional theory with the B3LYP functional and the 6-31G1 basis set and some correlations between structure and energetics were obtained for the keto and enol forms of both compounds. Using the G3(MP2)//B3LYP composite method and various appropriate reactions, the standard molar enthalpies of formation of 1,2-benzisothiazol-3(2H)-one and 1,4-benzothiazin-3(2H, 4H)-one, at T = 298.15 K, were computationally derived and compared with the experimental data. The aromaticity of 1,2-benzisothiazol-3(2H)-one, 1,4-benzothiazin-3(2H, 4H)-one and that of some related species was evaluated by analysis of nucleus independent chemical shifts (NICS).  相似文献

Reductive cyclizations of methyl 2-(2-nitrophenylthio)acetate 2 by means of electrochemistry or by catalytic hydrogenation have been used as key steps in the syntheses of the hemiacetals 2-hydroxy-2H-1,4-benzothiazin-3(4H)-one 10 and its 4-hydroxy derivative 9 representing thio analogues of allelo chemicals found in Gramineae and Acanthaceae. In contrast to its natural counterpart 2,4-dihydroxy-2H-1,4-benzoxa-zin-3(4H)-one hydroxamic acid 9 did not undergo degradation by extrusion of formic acid.  相似文献

The synthesis of 1,4-thiazine 1-oxide and 1,1-dioxide analogs of the antibiotic emimycin is described. Reaction of methylthioglycolate with 1-bromo-2,2-diethoxyethane gave methyl (2,2-diethoxyethylthio)acetate ( 2 ). Treatment of 2 with methanolic ammonia followed by cyclization furnished 2H-1,4-thiazin-3(4H)-one ( 5 ). Oxidation of 5 with m-chloroperoxybenzoic acid converted it to 2H-1,4-thiazin-3(4H)-one 1-oxide ( 6 ). Oxidation of 2 with potassium permanganate, followed by treatment with methanolic ammonia, and cyclization gave 2H-1,4-thiazin-3(4H)-one 1,1-dioxide.  相似文献

The methods of synthesis of 1,4-benzodioxin-2(3H)-one have been reexamined. Frequently quoted in the literature the method of Ghosh has been found to give impure (2-hydroxyphenoxy)acetic acid rather than the lactone. Using various methods some simple derivatives of the title lactone substituted in the benzene ring have been prepared and characterized. The bromination of the lactone with NBS gives predominantly 7-bromo-1,4-benzodioxin-2(3H)-one under electrophilic conditions, while 6-bromo-1,4-benzodioxin-2(3H)-one is obtained as a major isomer under photolytic conditions.  相似文献

标题化合物C14H10ON2是由邻硝基苯甲酰苯胺与原甲酸三乙酯在低价钛试剂作用下反应而得。结构通过单晶X-射线衍射分析确定, 其晶体属于单斜晶系, 空间群P21/c, a = 12.080(2), b = 7.793(1), c = 11.599(1) ? b = 97.56(1), Mr = 222.24, V = 1082.4(2) ?, Dc = 1.364 g/cm3, Z = 4, m(MoKa) = 0.88 mm-1, F(000) = 464, R = 0.0385, wR = 0.0851。X-衍射分析表明: 平面I (C(1)~C(6))与平面II (C(9)~C(14))之间的夹角为124.38; 平面I与平面III (ON(1)?N(2)C(7)C(8)C(9)C(14))之间的两面角为125.47; 平面II与平面III之间的两面角为2.76。  相似文献

Anthranilic acid and its 5-bromo and 4-chloro derivatives react with pinanoic and pinonoic acid chlorides to give the corresponding N-acyl derivatives. The pinanoyl derivatives give the corresponding 2-(3-ethyl-2,2-dimethyl-cyclobutylmethyl)-4-(3H)-quinazolinones when refluxed in formamide. Pinanoylanthranilic acid reacts with dicyclohexylcarbodiimide to give 2-(3-ethyl-2,2-dimethylcyclobutylmethyl)benz-3,1-oxazin-4(H)-one and subsequently with hydrazine hydrate to give 3-amino-2-(3-ethyl-2,2-dimethylcyclobutylmethyl)-4(3H)-quinazolinone. 2H-Imidazole-4(3H)-thiones (a), available from methyl alkyl and methyl aryl ketones with sulfur and ammonia, react via their corresponding N-sodium compounds or in presence of tert. amines with alkyl and aryl carboxylic acid chlorides to give the corresponding intensely coloured (orange to violett) cryst. 3-acyl-2H-imidazole-4(3H)-thiones4 a-q and6–26. With dicarboxylic acid dichlorides the colourless cryst. N,N′-diacyl-bis-3-imidazoline-5-thiones5 a-d and27–32 are obtained. With carbamic acid chlorides and chloroformic acid esters the corresponding urea (33–35) and urethane derivatives36, 37 are formed. In an analogous way 2H-imidazol-4(3H)-ones react with acid chlorides to 3-acyl-2-imidazol-4(3H)-ones (44–50), which can also be obtained by treating the corresponding 3-acyl-2H-imidazole-4(3H)-thione with KMnO4.  相似文献

Direct, chemo-specific arylation at C-2 or N-4 of 1,4-benzothiazin-3-one with aryl halides, based on Pd or Cu catalyst system, respectively, provided easy entry to arylated derivatives, a class of molecules not easily accessible via existing methods. Under Pd-catalysis conditions with LiHMDS as the base, N-arylation of 1,4-benzothiazin-3-one was inhibited leading to Cα-arylation of a secondary amide without the need for protection and de-protection of more acidic amido NH.  相似文献

2,7-Dihydroxy-2H-1,4-benzoxazin-3(4H)-one 11 (DHBOA) and 2,4,7-trihydroxy-2H-1,4-benzoxazin-3(4H)-one 14 (TRIBOA) representing aglucones of naturally occurring acetal glucoside type allelo chemicals found in Gramineae have been for the first time synthesized by two pathways both involving selective reductive cyclizations of appropriate 7-benzyloxy-2-nitrophenol derivatives as precursors. TRIBOA 14 and its bioactive naturally occurring 7-methyl ether DIMBOA have been found to undergo a hitherto unknown transformation to the corresponding 2,6-dibromo substituted lactam forms 20 and 21 in the presence of hydrogen bromide in acetic acid, which is of value in a better understanding of the possible mode of bioactivity.  相似文献
