Gynostemma pentaphyllum, a traditional herbal tea, and its flavonoids possess antioxidant activities. This study was carried out to isolate and identify these flavonoids from G. pentaphyllum and determinate the contents of the main flavonoids and their antioxidant activities. Nine flavonoids were determinated rapidly from G. pentaphyllum using medium-pressure liquid chromatography combined with other simple chromatographic methods. And nine flavonoids were identified as rutin (1), 4′-O-methyl-kaempferol-3-O-rutinoside (2), ombuoside (3), kaempferol-3-β-D-O-rutinoside (4), isorhamnetin-3-O-β-D-rutinoside (5), quercetin-3-O-β-D-glucoside (6), isorhamnetin (7), kaempferol (8), and quercetin (9) by liquid chromatography mass spectrometry-ion trap time of flight and 13C nuclear magnetic resonance spectroscopy, respectively. All of them displayed various potent antioxidant effects against the DPPH radical and A549 cell injury by using H2O2-generate peroxyl radicals in vitro . A sensitive and specific ultra-performance liquid chromatography coupled with multiple-reaction monitoring mode was utilized to rapidly analyze the main six flavonoids of the ethanol extract of G. pentaphyllum within less than 8?min using a Waters Acquity UPLC BEH C18 column. The contents of them were from 57.11 to 12907.74?µg/g in G. pentaphyllum.  相似文献

Abstract

Using thin layer chromatography in flat chamber or liquid column chromatography on the micropreparative scale the separation and isolation of furocoumarins from Angelica Archangelica off.(L) and Pastinaca sativa (L) fruit extracts was carried out.

The best results were obtained using chromatography and rechromatography in different systems (e.g., different selectivity of reversed and normal phase systems). Close analogy of polarity and molecular structure of furocoumarins limited the loading of the system to 0.001 g per 1 g of the adsorbent.  相似文献

Abstract

Twelve N-acetyl or NH2-free D-glucosaminyl bolaamphiphiles have been synthesized by the intermediate of N-allyloxycarbonyl-d-glucosaminyl precursors. Thus, glycosylation of α,ω-diols with 1,3,4,6-tetra-O-acetyl-2-allyloxycarbonylamino-2-deoxy-β-d-glucopyranose (1) gave the bis(glycosides) 2a-h in good yields and without column chromatography. Alkaline treatment of these derivatives followed by acetylation gave the peracetylated N-acetyl compounds 3a-h which were further deprotected by the Zemplén deacetylation procedure to the N-acetyl-d-glucosaminyl bolaamphiphiles 4a-h. The bis(glycosides) 2c,d,g were also transformed into the O-acetylated amino-free derivatives 5c,d,g by chemospecific deprotection of the N-allyloxycarbonyl groups with palladium (0). Further deprotection of the ester functions led to the completely deprotected bolaamphiphiles 7c,d,g with high yields. Fully deprotected compounds 7a,d,g,h were also obtained from 2a,d,g,h by alkaline treatment and purification by column chromatography. Surface tension measurements were realized for aqueous solutions containing the soluble bolaamphiphiles.  相似文献

Eleven constituents were characterised by gas chromatography–mass spectrometry analysis, and five molecules were isolated using column chromatography. The in vitro study of the extract and isolated molecules against KB and SiHa cell lines revealed oleanolic acid (1) and oleic acid (2) as potent cytotoxic molecules with potential anticancer activity. The IC50 values of n-hexane extract (CPHF), oleanolic acid (1) and oleic acid (2) were >300, 56.08 and 70.7 μg/mL (μM), respectively, against KB cell lines and >300, 47.24 and 80.2 μg/mL (μM), respectively, against SiHa cell lines.  相似文献

Melampyrum bihariense A. Kern. (Scrophulariaceae), a plant species used in traditional medicine for the treatment of rheumatic disorders and skin infections, was investigated with regard to its antioxidant activity and identification of its bioactive chemical constituents. The crude methanolic extract of the aerial parts of M. bihariense was examined by the spectrophotometric DPPH (1,1-diphenyl-2-picrylhydrazyl) and ferric reducing antioxidant power methods. The free radical scavenging capacity (SC50) of the extract was found by the DPPH method to be 27.10 mg mL−1, and the ferric reducing ability equivalent to ascorbic acid at 50 mg mL−1 was 0.709 μg mL−1. The chemical composition of this highly effective in the methanolic extract was analysed, and the main compounds were isolated through solvent–solvent partition, and multiple chromatographic separations, including column chromatography, vacuum liquid chromatography, centrifugal planar chromatography and preparative thin-layer chromatography. The structures were established by one- and two-dimensional NMR and liquid chromatography-mass spectrometry. The iridoids aucubin (1), 8-epi-loganin (2) and mussaenoside (3), the flavones apigenin and luteolin and the triterpene acids ursolic acid and oleanolic acid were identified; components 2, 3, ursolic acid and oleanolic acid for the first time in this species. The present study reveals that M. bihariense exerts antioxidant activity, and the iridoids, flavonoids and triterpene acids may be the main bioactive constituents of its methanolic extract.

Optically active telluroxides 1 and 2 were isolated for the first time by means of medium-pressure liquid chromatography using an optically active column. Absolute configuration of the telluroxides (+)-1 and (+)-2 was determined to be R based on their specific rotations and circular dichroism spectra. The configurational lability and mechanism for racemization via an achiral hydrate were clarified by kinetic study and isotope tracer experiment.  相似文献

Summary Several chiral mono- and disubstituted 9,10-dihydro-9,10-ethanoanthracenes have been prepared from the corresponding anthracenes. Most of them were separated into enantiomers by chromatography on cellulose triacetate (CTA) and their absolute chiralities established by chiroptical comparison (via their CD spectra) with key compounds of known configuration. From the laevorotatory 1,5-dibromo derivative16 the dextrorotatory dideuterio hydrocarbon (+)(9S, 10S)-20 was obtained.Complexation of 2,6-dimethyl 9,10-dihydro-9,10-ethanoanthracene (+)-25, obtained by enantio-selective chromatography onCTA [with its chirality (9R, 10R) deduced from optical comparison with the 2-monomethyl derivative of known configuration], with Cr(CO)6 afforded two mono tricarbonyl-chromium complexes [endo(+)-26 andexo(+)-27] as well as the bis-exo,endo-complex (+)-28. Abstract

One new capsaicinoid, N-vanillyl-4E,6E-dien-8-methylnonanamide (4), along with nine known capsaicinoids, capsaicin (1), dihydrocapsaicin (2), N-vanillyloctanamide (3), nordihydrocapsaicin (5), N-vanillyldecanamide (6), homocapsaicin (7), N-vanillyl-4,8-dimethylnonanamide (8), homodihydrocapsaicin II (9), and homodihydrocapsaicin I (10) were isolated from the fruits of Capsicum annuum using semi-preparative high-performance liquid chromatography. The structural characterizations of the isolated compounds were elucidated by spectroscopic data and comparison with the literatures. Bioassays showed that the isolated capsaicinoids significantly reduced the radical length of Lactuca sativa seedling, this inhibition being dose-dependent.  相似文献

A practical synthetic route to N-Boc protected or Boc-amino acid coupled ω-aminoalkanoic acids is reported and exemplified by the preparation of 8-(t-butoxycarbonylamino)caprylic acid 2 and (N-t-butoxycarbonylphenylalanyl)-8-aminocaprylic acid 3. The sequence does not involve column chromatography, hydrogenation, azide or bromine related rearrangements, and therefore is amenable to scale-up. Homologues of the ω-aminoalkanoic acid derivatives may also be prepared by using different cycloalkanones.  相似文献

Summary Reinvestigation by HPLC of the petals ofLilium tigrinum and the isolation of some minor compounds is reported. Using HPLC-controlled, preparative-column chromatography, 5,6-diakarpoxanthin (6), 6-epikarpoxanthin (2), 5,6-diacapsokarpoxanthin (8), and 9Z-antheraxanthin (9Z-13) were isolated and characterized. Based on spectroscopic data the absolute configurations of6 and8 were identical with those originating from paprika, thus the 5,6-diakarpoxanthin (6) and 5,6-diacapsokarpoxanthin (8) have the 3S, 5S, 6S configuration and 6-epikarpoxanthin (2) has the 3S, 5R, 6S configuration. 利用HPLC法考察了六、七元瓜环(Q[6], Q[7])与邻苯二胺(g1)、间苯二胺(g2)、对苯二胺(g3)、邻硝基苯胺(g4)、间硝基苯胺(g5)、对硝基苯胺(g6)的相互作用. 实验结果表明: Q[6]可与客体g1g3, g5形成1∶1的包结配合物; Q[7]与客体g1g6形成1∶1包结配合物, 同时计算了包结配合物的包结稳定常数, 探讨了主-客体的相互作用模式, 并利用1H NMR、紫外吸收光谱法进行了佐证.  相似文献

ABSTRACT

A sensitive and selective method was developed to determine pesticides in carrots by gas chromatography–mass spectrometry following the development of an optimized extraction procedure. The method was validated for 30 organochlorine pesticides for gas chromatography with electron capture detection obtaining limit of detection from 0.18 to 0.92?µg/kg except for cis- and trans-permenthrin. Twenty-six carrot samples were analyzed and six pesticides were detected. The results compared with the accepted maximum residue levels in correlation to crop origin.  相似文献

The four diastereomeric 2-(hydroxyphenylmethyl)cyclohexanols2a-2d are prepared. By reaction with benzaldehyde each2a-2c gives stereospecifically only one of the two possible configurationally correlated 2,4-diphenylhexahydro-4H-1,3-benzodioxins3a-3c. However, from (1RS, 2SR)-2 [(SR)-hydroxyphenylmethyl]cyclohexanol (2d) both dioxins3d and3e are obtained. The configurational assignments are based on1H-NMR spectroscopy and synthetic correlations. Conformational aspects are discussed. As is shown by chromatography, inPrins reactions of cyclohexene with benzaldehyde only one dioxin3c is formed.  相似文献

A new sesquiterpenoid, 10α-methoxyartemisinic acid (1) was isolated from Saussurea lappa roots along with two known compounds, costunolide (2) and dehydrocostus lactone (3) by high-speed countercurrent chromatography with purities of 85, 95 and 98%, respectively. A new procedure was developed for the synthesis of 2-aminothiazolo[3,2-a]pyridinium salts 8 by the reaction of 2-halo-N-phenacylpyridinium salts with KSCN. The anion compositions of salts 8 were studied by ion chromatography. Acylation of salts 8 afforded representatives of the previously unknown bicyclic mesoionic thiazolo[3,2-a]pyridinium 2-imidate system 9. The three-dimensional structures of 2-amino-3-(p-bromobenzoyl)thiazolo[3,2-a]pyridinium thiocyanate and N-trifluoroacetyl-3-(p-nitrobenzoyl)thiazolo[3,2-a]pyridinium 2-imidate were established by X-ray diffraction analysis.__________Published in Russian in Izvestiya Akademii Nauk. A practical, nine-step protocol is described for the preparation of synthetically useful N-acetyllactosamine (LacNAc) derivatives as well as LacNAc itself from lactose using the benzoylated oxime of lactos-2-ulosyl bromide 2 as the key intermediate. This paper reports on the first phytochemical analysis ever performed on Jasminum tortuosum Willd. This analysis, mainly carried out by means of column chromatography separation, NMR spectroscopy and mass spectrometry, led to the isolation and the identification of four compounds, namely the lignans ginkgool (1) and olivil-4′-O-β-glucopyranoside (2) and the secoiridoids oleoside dimethyl ester (3) and oleoside 11-methyl ester (4). The presence of these compounds is significant from a chemotaxonomic point of view, confirming the correct botanical classification of the species and, from a phytochemical standpoint, may suggest its possible use in the ethno-medicinal field.  相似文献

The fruit body of Lasiosphaera fenzlii was found to show cytotoxicity on cancer cells during a preliminary screening. Repeated column chromatography of the fungal methanol extract resulted in the isolation of six compounds identified as 5α,8α-epidioxy-ergosta-6,22-dien-3β-ol (1), 5α,8α-epidioxy-ergosta-6,9(11),22-trien-3β-ol (2), 5α-ergosta-7,22-dien-3β-ol (3), 5α-ergosta-7,22-dien-3-one (4), ergosta-7,22-dien-3β,5α,6β-triol (5) and 6-dihydroxy-2,3-dihydro-1H-isoindol-1-one (6). The two peroxide compounds, 1 and 2, showed cytotoxic activity and compound 1 was selectively cytotoxic to cancer cells. Furthermore, compound 1 synergised the cytotoxicity of paclitaxel on Hela cells by increasing intracellular accumulation of paclitaxel in cancer cells but not in normal cells.  相似文献

Abstract

Thermal and catalytic reactions between (2E)-4,5,6-tri-O-acetyl-2,3-dideoxy- aldehydo- d-erythro-hex-2-enose (1) and cyclopentadiene led, in each case, to the four stereoisomeric 5-formyl-6-(1,2,3-tri-O- acetyl-d-erythro-triol-1-yl)bicyclo[2.2.1]hept-2-enes (2-5). The role of the catalysts is discussed, and the face selectivity is rationalized in terms of both steric and electrostatic arguments. Upon subsequent sodium borohydride reduction, iodination, and zinc reduction, the adducts 2-5 were converted to their respective primary alcohols 6-9, that could be separated by column chromatography. Sequential basic deacetylation, oxidative cleavage of the sugar side-chain with sodium metaperiodate, and reduction with sodium borohydride, yielded the previously known (5R,6R)- or (5S, 6S)-bis(hydroxymethyl)norbornene-diols 12 or 13, thus establishing the C-5 and C-6 absolute configurations of their synthetic precursors.  相似文献
