Abstract

The effect of instrumental parameters, mobile phase composition, and flow rate on high performance liquid chromatography separations using 3 cm, 5 cm, and 10 cm columns packed with 3y spherical ODS materials is discussed. The results indicate that when instrumental parameters were optimized, the 10 cm column gave better separation than the shorter columns under the same experimental conditions. However, when the mobile phase composition was adjusted so that the solute residence time was comparable in the three columns, short and long columns gave comparable results.  相似文献

Abstract

In a comparative study of the separation of a mixture of polycyclic aromatic hydrocarbrons on 5 cm and 10 cm columns packed with 3μ and 5μ ODS spherical particles, the separations achieved on the 10 cm column were not significantly better than those on the 5 cm column. Although, columns packed with 3μ ODS spherical particles gave slightly better resolution than those packed with 5μ ODS spherical particles having the same physical properties, differences were observed when columns packed with ODS spherical particles were compared with columns packed with ODS irregular shaped particles of the same size.

The results show that separations on a 5μ or 3μ packed columns takes place at the first few centimeters.  相似文献

Abstract

Two narrow-bore columns packed with octadecylsilane of various particle sizes were used to compare their efficiency for the separation of organic acids in apple and cider with that of ordinary columns. The best simultaneous resolution of quinic, malic, shikimic, lactic, acetic, citric and succinic acid was accomplished by using a 100 × 2.1 mm ID, 3-μm Spherisorb ODS-2 column and a phosphate buffer as the mobile phase. This chromatographic system provided a separation efficiency comparable to that afforded by an ordinary 250 × 4.6 mm ID, 5-μm Spherisorb ODS-2 column, plus greater rapidity (30%) and economy, all of which allowed the accurate, precise determination (CV=3%) of the above-mentioned compounds. Finally, the performance of an ordinary UV detector and that of a rapid spectral detector in this type of determination were critically compared.  相似文献

A procedure for packing 15 cm × 1 mm id reverse-phase microbore columns with 3 μm silicas obtained from different manufacturers is described. The speed of analysis and detection limits are compared to those obtained with a 50 cm × 1 mm id column packed with 10 μm ODS. The effect of detector time constant on the system, and flow rates on column efficiency are also examined.  相似文献

Three mixed‐mode high‐performance liquid chromatography columns packed with superficially porous carbon/nanodiamond/amine‐polymer particles were used to separate mixtures of cannabinoids. Columns evaluated included: (i) reversed phase (C18), weak anion exchange, 4.6 × 33 mm, 3.6 μm, and 4.6 × 100 mm, 3.6 μm, (ii) reversed phase, strong anion exchange (quaternary amine), 4.6×33 mm, 3.6 μm, and (iii) hydrophilic interaction liquid chromatography, 4.6 × 150 mm, 3.6 μm. Different selectivities were achieved under various mobile phase and stationary phase conditions. Efficiencies and peak capacities were as high as 54 000 N/m and 56, respectively. The reversed phase mixed‐mode column (C18) retained tetrahydrocannabinolic acid strongly under acidic conditions and weakly under basic conditions. Tetrahydrocannabinolic acid was retained strongly on the reversed phase, strong anion exchange mixed‐mode column under basic polar organic mobile phase conditions. The hydrophilic interaction liquid chromatography column retained polar cannabinoids better than the (more) neutral ones under basic conditions. A longer reversed phase (C18) mixed‐mode column (4.6 × 100 mm) showed better resolution for analytes (and a contaminant) than a shorter column. Fast separations were achieved in less than 5 min and sometimes 2 min. A real world sample (bubble hash extract) was also analyzed by gradient elution.  相似文献

A set of fifteen 2-naphthol-derived atropisomers were evaluated on three different cyclofructan-based chiral stationary phases (CSP). The cyclofructan CSPs were a dimethylphenyl-derivatized cyclofructan 7 (CF7-DMP), a isopropyl (CF6-P) and a R-naphthylethyl cyclofructan 6 (CF6-RN) derivative, all bonded to 5-µm spherical fully porous silica particles packed into three 25?cm?×?4.6?mm columns (commercially available as Larihc columns). The 15 atropisomers were analyzed in the normal-phase mode with heptane/alcohol mobile phases. The CF7-DMP column was the most effective partially or fully separating 14 of the 15 compounds (93%). All 15 compounds could be separated by at least one cyclofructan column. Five compounds could be partially or fully separated by all three CSPs. The effect of ethanol, 2-propanol and butanol as 5 and 10% v/v polar modifiers in heptane was studied. A prototype 15?cm?×?4.6?mm column packed with superficially porous 2.7?µm CF6-P bonded particles was tested with the same set of compounds and a standard HPLC system. The increased efficiency and solvent saving were significant.  相似文献

Polypropylene (PP) capillary‐channeled polymer (C‐CP) fiber stationary phases are investigated for applications in HPLC. Specifically, the roles that fiber size and shape, linear velocity, interstitial fraction, and column inner diameter play in separation efficiency were evaluated using a uracil and butylparaben mixture eluted under isocratic conditions. Four fiber types, having nominal diameters ranging from 30 to 65 μm, were used in 250 mm × 2.1 mm columns. Optimum flow characteristics, as judged by plate height and resolution, were observed for 40 μm diameter PP C‐CP fibers packed at an interstitial fraction of ~0.63, over a broad range of linear velocities (~2 to 37 mm/s). The influence of column inner diameter was studied on 1.5, 2.1, and 4.6 mm columns packed at the optimal interstitial fraction. The best performing column in terms of plate height and resolution was the 2.1 mm inner diameter. C‐CP columns were also evaluated for the separation of a protein mixture composed of ribonuclease A, cytochrome c, and transferrin. Results obtained with the biomacromolecules mixture validate the optimal structural and operative conditions determined with the small solutes, laying the groundwork towards biomacromolecule applications, focusing more on the chemical aspects of separations.  相似文献

Abstract

The effect of column dimension on resolution, sample capacity, retention time, efficiency and mobile phase composition were studied, using both constant flow rate and constant linear velocity. The four columns selected (A = 238 × 3.2 mm, B = 153 × 4.0 mm, C = 116 × 4.6 mm and D = 50 × 7 mm) had the same volume. K1 values were found to be constant, within experimental error, for all columns. At constant linear velocity, the retention time was found to be a linear function of column length, while at constant flow rate retention time was constant for all columns. The longest column (A) generated the largest N values while columns 3 and C gave the lowest H values, for dilute solutions, while they decreased with decreasing column length. On the other hand, it was observed that as the sample size increased, N generated by column A decreased more rapidly and eventually fell below the values generated by columns B and C. These two columns (B & C) can tolerate a larger sample size with less reduction in N value than the longest column. It is important to note that although there were minor differences in performance between columns B and C, there were significant differences between them (B and C) and the other two columns (A and D). Column A offered the highest sensitivity (narrower peaks) for dilute solutions, while columns B and C offered higher loadability. The volume of organic modifier in the mobile phase affected the retention equally in the four columns. It was also found that equal separation (a) was obtained for each column at constant flow rate and constant linear velocity, except with the latter the retention times were longer.  相似文献

The potential of high-speed analyses by rapid resolution liquid chromatography (RRLC) and RRLC/MS on 1.8-microm porous particles packed into short columns operated at high flow-rate was investigated and compared with the performance of 5-microm porous particles packed into conventional columns. Using similar chemistries, the ease of conversion from conventional HPLC to an RRLC method was demonstrated. In order to display the practicality of RRLC separations, the analysis of pesticides in crops and catechins in Japanese green tea was selected. Using the Japanese Food Hygiene Law method, which employs a conventional 5-microm RP column (250 mm x 4.6 mm) for quantification of pesticides in crops, the analysis time was 25 min under isocratic conditions. Using the RRLC method on the short (50 mm x 4.6 mm) column packed with 1.8-microm porous particles, the same separation could be performed in 0.8 min with the RRLC/MS method without a loss in resolution. At the highest flow rate, compared to the conventional method, the time could be reduced by a factor of 31. In gradient elution, the fastest separation of catechins in Japanese green tea was achieved by RRLC on 50-mm x 4.6-mm id or 50-mm x 2.1-mm id RRLC columns packed with 1.8-microm particles. The analysis time at 5 mL/min was less than 1 min. Compared to the conventional HPLC method on a 150-mm column packed with 5-microm particles, time was reduced by a factor of 15. The effect of other experimental parameters such as the column temperature, acquisition rate of the detector and the influence of cell volume on chromatographic performance was also investigated. After the optimization, the analysis precision under the fastest RRLC conditions was examined. RSDs of retention time and peak area were 0.2% and 0.47%, respectively.  相似文献

High plate numbers were obtained in conventional LC by coupling columns and by using temperature to reduce the viscosity of the mobile phase. At 80 degrees C up to eight columns of 25 cm x 4.6 mm I.D. packed with 5 microm ODS particles could be coupled generating 180,000 effective plates while the pressure drop was only 350bar. For routine work, a set of four columns is preferred. The analysis times on one column operated at 30 degrees C and 1 mL/min flow rate and on four columns at 80 degrees C and 2 mL/min flow rate are the same in isoeluotropic conditions while the resolution is doubled. Multicolumn systems were successfully applied in isocratic and gradient mode for the analysis of pharmaceutical and environmental samples.  相似文献

High efficiency separations (200 000 plates) were obtained on conventional LC equipment by coupling 8 x 25 cm x 2.1 (or 4.6) mm id x 5 microm d(p) ODS columns (total length 2 m) and operation at 60 degrees C using a dedicated LC oven. The peak capacity in this 1-D set-up was 900 for the separation of human serum tryptic peptides analyzed after depletion of six highly abundant proteins. The chromatographic performance of an elevated temperature-extended column length conventional LC is highlighted.  相似文献

Abstract

Polystyrene gels of a particle diameter 10 ± 2 μm for the use in oligomer separation were packed into 1.5 mm i.d. × 25 cm length columns by the balanced density slurry-packing technique under a constant flow rate of 500 μL/min. The slurry solvent was a mixture of toluene and chloroform (50.5/49.5, v/v). The example of the number of theoretical plates (N) of these columns was 8600 plates/25 cm (HETP = 29.1 μm) at flow rate of 40 μL/min by injecting 1 μL of 0.5% benzene solution. Sixteen columns were connected and the overall value of N was 103000 plates/4 m. A typical example of oligomer separation was demonstrated. A constant-flow technique is preferable to a constant-pressure technique. When two or three column blanks were packed together, the columns located at the outlet of the packer-column assembly had higher values of N. Optimum flow rate of the slurry solvent when three column blanks were packed together lay between 400 and 500 μL/min. The packing efficiency, that is, the probability of getting valid columns was about 60%. Viscous slurry solvents were not effective to get efficient columns. To pack gels in the less swollen state gave sometimes efficient columns. Pressure monitoring in progress of packing was very effective to foresee the column efficiency.  相似文献

The impact of column inner diameter on chromatographic performance in temperature gradient liquid chromatography has been investigated in the present study. Columns with inner diameters of 0.32, 0.53, 3.2 and 4.6 mm were compared with respect to retention and efficiency characteristics using temperature gradients from 30 to 90 degrees C with temperature ramps of 1, 5, 10 and 20 degrees C min(-1). The columns were all of 15 cm length and were packed with 3 microm Hypersil ODS particles. Alkylbenzenes served as model compounds, and the mobile phase consisted of acetonitrile-water (50:50, v/v). The study revealed that the column ID is not a critical limiting factor when performing temperature programming in LC, at least for columns narrower than 4.6 mm inner diameter in the temperature interval 30-90 degrees C. The retention times for all components on all columns were highly comparable, with similar peak profiles without any signs of peak splitting. The use of mobile phase pre-heating when using the larger bore columns was avoided by starting the temperature gradients close to ambient. However, the relative apparent efficiency was inversely proportional to column inner diameter, making the capillary columns generally more functional towards temperature gradients than the larger bore columns with respect to chromatographic efficiency. In addition, the capillary columns possessed higher robustness towards temperature programming than the conventional columns.  相似文献

Ultra high-performance liquid chromatographic (UHPLC) systems on columns packed with materials ranging from 1.9 to 2.7 μm average particle size were assessed for the fast and sensitive analysis of porphyrins in clinical materials. The fastest separation was achieved on an Agilent Poroshell C(18) column (2.7 μm particle size, 50 × 4.6 mm i.d.), followed by a Thermo Hypersil Gold C(18) column (1.9 μm particle size, 50 × 2.1 mm i.d.) and the Thermo Hypersil BDS C(18) column (2.4 μm particle size, 100 × 2.1 mm i.d.). All columns required a mobile phase containing 1 m ammonium acetate buffer, pH 5.16, with a mixture of acetonitrile and methanol as the organic modifiers for optimum resolution of the type I and III isomers, particularly for uroporphyrin I and III isomers. All UHPLC columns were suitable and superior to conventional HPLC columns packed with 5 μm average particle size materials for clinical sample analysis.  相似文献

Monolithic silica capillary columns were successfully prepared in a fused silica capillary of 530 microm inner diameter and evaluated in HPLC after octadecylsilylation (ODS). Their efficiency and permeability were compared with those of columns pakked with 5-microm and 3-microm ODS-silica particles. The monolithic silica columns having different domain sizes (combined size of through-pore and skeleton) showed 2.5-4.0-times higher permeability (K= 5.2-8.4 x 10(-14) m2) than capillary columns packed with 3-mm particles, while giving similar column efficiency. The monolithic silica capillary columns gave a plate height of about 11-13 microm, or 11 200-13 400 theoretical plates/150 mm column length, in 80% methanol at a linear mobile phase velocity of 1.0 mm/s. The monolithic column having a smaller domain size showed higher column efficiency and higher pressure drop, although the monolithic column with a larger domain size showed better overall column performance, or smaller separation impedance (E value). The larger-diameter (530 microm id) monolithic silica capillary column afforded a good peak shape in gradient elution of proteins at a flow rate of up to 100 microL/min and an injection volume of up to 10 microL.  相似文献

Summary In this paper, practical considerations of column efficiency, separation speed, thermal stability, and column polarity of capillary columns packed with polybutadiene-coated zirconia were investigated under solvating gas chromatography (SGC) conditions using carbon dioxide as mobile phase. When compared with results obtained from conventional porous octadecyl obtained from conventional porous octadecyl bonded silica (ODS) particles, PBD-zirconia particles produced greater change in mobile phase linear velocity with pressure than conventional ODS particles under the same conditions. The maximum plate number per second (Nt) obtained with a 30 cm PBD-zirconia column was approximately 1.5 times higher than that obtained with an ODS column at 100 °C. Therefore, the PBD-zirconia phase is more suitable for fast separations than conventional ODS particles in SGC. Maximum plate numbers per meter of 76,900 and 63,300 were obtained using a 57 cm×250 μm i.d. fused silica capillary column packed with 3 μm PBD-zirconia at 50 °C and 100 °C, respectively. The PBD-zirconia phase was stable at temperatures up to 320 °C under SGC conditions using carbon dioxide as mobile phase. Polarizable aromatic compounds and low molecular weight ketones and aldehydes were eluted with symmetrical peaks from a 10 cm column packed with 3 μm PBD-zirconia. Zirconia phases with greater inertness are required for the analysis of more polar compounds by SGC.  相似文献

Summary Fast separations of perfluorinated polyethers and polymethylsiloxanes that are composed of 50–80 oligomers were demonstrated in packed capillary column supercritical fluid chromatography (SFC) using a carbon dioxide mobile phase. Separations were accomplished within 10 min using a 13 cm×250 μm i.d. column packed with 2 μm porous octadecyl bonded silica (ODS) particles. Effects of particle diameter of the packing material and pressure programming on separation were investigated, and packed column SFC was compared with open tubular column SFC. Results show that as the particle diameter was decreased from 5 to 3 to 2 μm and the column length was reduced from 85 to 43 to 13 cm, the separation time could be reduced from 70 to 20 to 10 min while still maintaining similar separation (resolution). Short columns packed with small porous particles are very suitable for fast SFC separations of polymers.  相似文献

将10° 锥角台锥型液相色谱柱放大至150 mm长、入口直径54 mm、出口直径27 mm,容积为200 mL,填料为粒径40~75 μm、孔径11 nm的C18球形硅胶。流动相在锥型柱内呈现塞子状流形。系统地评价了该柱的分离性能,结果表明: 在最佳流速为6 mL/min时,以萘峰计,锥型柱的折合理论塔板高度为2.11,柱效下降10%时的样品质量和体积载样量分别为2.1 mg和1.7 mL,与同长度同体积圆柱型柱相比,柱效提高了20%,质量载样量提高了16%以上,体积载样量提高了19%以上。当进样质量由2.4 mg增加到12 mg时,对羟基苯甲酸乙酯峰与对羟基苯甲酸丁酯峰的分离度(Rs2)由2.14降到1.71,对羟基苯甲酸丁酯峰与萘峰的分离度(Rs3)由2.91降到2.52;当进样体积由3 mL增加到19 mL,Rs2由2.23降到1.28,Rs3由2.95降到2.30,但此时的色谱峰峰形仍然高度对称,没有拖尾,有利于从基质中制备分离微量组分。实验结果表明锥型液相色谱柱将具有广泛的应用前景。  相似文献

Abstract

Sixteen columns of 1.5 mm i.d. × 25 cm length packed with polystyrene gels of a particle diameter 10 ± 2 μm and having the exclusion limit of 8000 molecular weight as polystyrene were connected in series and used for the separation of oligomers such as oligostyrenes, epoxy resins, methylated melamine-formaldehyde resins, and phenol-formaldehyde resins of novolac and resol types. The observed overall value of the number of theoretical plates was 103000 plates/4 m. Separation results were compared with the conventional SEC which used two SEC columns of 8 mm i.d. × 30 cm length packed with PS gels of a particle diameter 6 ± 2 μm and having the same exclusion limit of the semimicro SEC. The value of N of this column was 17500 plates/30 cm. Oligostyrenes up to n = 11 (undecamer) were separated. Methyl ether derivatives of polynuclear methylol melamines up to penta-nuclear methylol melamine were separated. The possibility for separation of overlapping peaks by SEC having clearly higher efficiency was discussed.  相似文献

Small columns packed with core-shell and sub-2 μm totally porous particles and monolith columns are very popular to conduct fast and efficient chromatographic separations. In order to carry out fast separations, short (2-5 cm) and narrow-bore (2-2.1 mm) columns are used to decrease the analyte retention volume. Beside the column efficiency, another significant issue is the extra-column band-spreading. The extra-column dispersion of a given LC system can dramatically decrease the performance of a small very efficient column. The aim of this study was to compare the extra-column peak variance contribution of several commercially available LC systems. The efficiency loss of three different type 5 cm long narrow bore, very efficient columns (monolith, sub-2 μm fully porous and sub-2 μm core-shell packing) as a function of extra-column peak variance, and as a function of flow rate and also kinetic plots (analysis time versus apparent column efficiency) are presented.  相似文献
