Capillary electrochromatography (CEC) continues to captivate many separation scientists. A remarkable activity is apparent from the numerous publications in the literature using CEC. A review of the most recent progress in CEC is presented herein, covering an extensive fraction of the literature on CEC published from the year 1997 until the beginning of 2000. This review brings a survey of studies on analytical ITP published since 2016 until the first quarter of 2018 and includes chapters about theory and principles, instrumentation and techniques, and analytical applications of ITP. It shows the position of analytical ITP among contemporary separation techniques, where particularly its unique concentrating capabilities keep the interest to include it into novel high‐sensitivity analytical procedures. The reviewed papers are considered according to their nature, techniques used, and instrumentation employed. Bioanalysis is a fast growing domain with new technological developments in the field of mass spectrometry, separation science, and bioinformatics. The central role of separation prior to detection and evaluation of analytes is indisputable. In fact, since the decipherment of the human genome via multichannel capillary electrophoresis in combination with laser-induced fluorescence [1], the analysis of the proteome, i.e., the type and amount of all proteins expressed at a certain time by the genome in a cell or tissue [2] became of central interest. Therefore, new fast and sensitive capillary liquid chromatographic, electrophoretic, and electrochromatographic separation techniques, suitable for high-throughput analysis of proteins, peptides and nucleic acids, are required.  相似文献

近年来,蛋白质组学技术在样品前处理、分离技术和质谱检测技术方面获得了快速发展,已经可以实现在几小时内对上万种蛋白的同时定性和定量分析。然而,目前的主流蛋白质组学技术仍无法满足极微量生物样品,尤其是单细胞样品的组学分析需求。毛细管电泳分离技术具有峰宽窄、柱效高、样品用量少等优势,是与高分辨质谱在线联用的理想选择之一。该文评述了集成化和在线样品前处理以及主流的纳升液相色谱-质谱联用系统在高灵敏度蛋白质组学分析领域的发展现状和挑战,认为该领域的重要技术挑战之一在于目前的纳升液相色谱分离已经无法完全匹配现代高分辨质谱超过40 Hz的超高扫描速度,从而导致质谱使用效率的降低。针对上述技术挑战,该文重点探讨了毛细管电泳-质谱联用技术的独特技术优势和潜在发展机遇,主要包括:(1)面向微量酶解多肽样品的高柱效毛细管电泳分离。通过采用毛细管电色谱可以进一步改善毛细管电泳柱容量不足的局限;(2)面向高灵敏度分析的无鞘液/鞘液接口开发;(3)高效毛细管电泳分离与高扫描速度质谱检测的协同化使用。总之,我们预期毛细管电泳-质谱联用技术的进一步发展有望在针对单细胞等超微量生物学样品的蛋白质组学分析中获得更广泛的应...  相似文献

A number of recent developments in DNA analysis by capillary electrophoresis are here reviewed. They include capillary arrays for fast, parallel DNA sequencing as well as microfabricated capillary arrays. Microfluidic chips for DNA sizing and quantitation are also covered, as well as microdevices containing arrays of regular obstacles acting as size-separators during DNA migration. Screening of DNA point mutations by two much improved techniques is also reported: in one case, such mutations are detected (but only on relative short, ca. 60-70 base-long fragments) by free electrophoresis in rather acidic (pH ca. 3) buffers; in the case of single-strand chain polymorphism, an improved technique is described based on near-neutral pH buffers with mixtures of Tris/MES cations/zwitterions. When studying the behavior of inorganic and organic cations in the Debye-Hückel layer of DNA, it was found that the latter (especially a large number of Good's buffers and other zwitterions, such as His) would bind to the DNA filament not only via charge interaction, but also via additional bonds, notably hydrogen bonds, thus altering the electrophoretic (and possibly the biological) behavior of DNA molecules. However, whether or not borate ions would bind to DNA remains still an unsettled question. Finally, capillary electrophoresis was found to be instrumental in measuring fine physicochemical parameters pertaining to DNA polyelectrolytes, such as their free mobility and their translational diffusion coefficients.  相似文献

This review is a continuation of the previous reviews (Electrophoresis 1999, 20, 3259-3268; Electrophoresis 2001, 22, 4244-4248) to update the recent publications from 2001 to 2003 on pharmacokinetic studies using capillary electrophoresis (CE). During this period of time, CE remains as a unique analytical method for some studies, which would otherwise be limited by many factors, such as the sample volume, detection sensitivity, or separation power. It is particularly noticeable that the separation of chiral drugs in biological sample and the use of solid-phase extraction (SPE) as a simple and convenient means of sample preparation appear to become popular for CE-based assays. The use of CE for assessing complete pharmacokinetic information, however, did not show a significant growth during this period of time. In order to provide a broad range of view on how biological samples are analyzed by CE, this review will cover publications during the past two years on the use of CE for the analysis of drugs in biological fluids for general pharmacokinetic applications including drug monitoring and bioavailability studies.  相似文献

Recent progress in enantiomeric separations by capillary electrochromatography (CEC) is reviewed. The development of simple and robust CEC column technologies plays an important role for popularization of CEC. During the last several years, various approaches for the preparation of enantioselective columns have been reported. Currently, the monolithic column technology (continuous beds) represents the most advanced approach for the preparation of CEC columns. The development of new chiral stationary phase used for CEC is another important issue in this field. Fundamental investigations on electrochromatographic behaviors of various CSPs are necessary in order to understand the separation mechanism and thus improve the separation performance. Some chiral stationary phases performed better under nonaqueous CEC conditions than reversed-phase conditions. Coupling CEC with mass spectrometry (MS) provides a powerful tool for enantiomeric separation. Finally, some applications of enantiomeric separation by CEC are summarized.  相似文献

Advances in the fundamental studies and methodology of capillary electrophoresis (CE) as applied to metal ion analysis over the last two years are reviewed, with the objective of providing the interested reader with a state-of-the-art picture of technique's potentialities in the area. In particular, novel strategies for separation selectivity control and CE system innovations designed to enhance the detection sensitivity are described. In addition, a brief overview of the primary metal analytes and samples for which the technique appears to be best suited is given. The current limitations of the technique regarding most of all the implementation for routine use are considered along with the approaches on how they could be addressed. Finally, some pointers as to the likely trends in the future research are discussed.  相似文献

Enantiomer separation by electrochromatography (CEC) can be performed in three modes: (i) open-tubular capillary electrochromatography (o-CEC), in which the chiral selector is physically adsorbed coated, and thermally immobilized or covalently attached to the internal capillary wall; (ii) packed capillary electrochromatography (p-CEC), in which the capillary is either filled with chiral modified silica particles or with an achiral packing material, and a chiral selector is added to the mobile phase; and (iii) monolithic (rod)-capillary electrochromatography (rod-CEC) in which the chiral stationary phase (CSP) consists of a single piece of porous solid. We present an overview on methods and new trends in the field of electrochromatographic enantiomer separation such as CEC with either nonaqueous mobile phases or stationary phases with incorporated permanent charges, or with packing beds consisting of nonporous silica particles or particles with very small internal diameters.  相似文献

This overview focuses on the development of new chiral selectors and the introduction of new techniques rather than applications of already established selectors and methods. This review surveys the recent progress in the adsorbed stationary phases for capillary electrochromatography (CEC). Adsorption-based methods for preparation of stationary phase are novel approaches in CEC, which allow rapid and facile preparing stationary phases with desirable selectivity onto an open-tubular fused-silica capillary, a bare-silica or ion-exchange packed column or a monolithic silica or polymer column. A variety of adsorbing agents have been developed as adsorbed stationary phases, including ionic long-chain surfactant, protein, peptide, amino acid, charged cyclodextrin (CD), basic compound, aliphatic ionene, and ion-exchange latex particle. The adsorbed stationary phases have been applied to separation of neutral, basic and acidic organic compounds, inorganic anions and enantiomers. They have also been applied to on-line sample concentration, fast separation and study of the competitive binding of enantiomers with protein.  相似文献

In this review research papers on the application of CEC are summarized that have been published between May 2003 and May 2005. First, a short overview is given of trends and developments in CEC that may increase the applicability of the separation technique. Electrophoretic sample stacking comprises a group of capillary electrophoretic techniques where trace analytes from the sample are concentrated into a short zone (stack). This paper is a continuation of our previous reviews on the topic and brings a survey of more than 120 papers published approximately since the second quarter of 2016 till the first quarter of 2018. It is organized according to the particular stacking principles and includes chapters on concentration adjustment (Kohlrausch) stacking, on stacking techniques based on pH changes, on stacking in electrokinetic chromatography and on other stacking techniques. Where available, explicit information is given about the procedure, electrolyte(s) used, detector employed and sensitivity reached. An overview of major developments in capillary electrophoresis and capillary electrochromatography systems in the environmental field is presented, covering relevant publications between the second half of 1999 and early 2001. Contributions are reviewed in relation to developments in detection, sample preparation/preconcentration, precision and applications.
