Ethyl vis‐ and trans‐2‐isothiocyanato‐1‐cyclopentanecarboxylates 2 and 7 were prepared by the reaction of the corresponding alicyclic ethyl 2‐amino‐1‐carboxylates and thiophosgene. The cis‐isothiocyanato compound 2 underwent ring closure with amines in one or two steps, resulting in 3‐substituted‐cis‐2‐thioxocyclopenta[d]pyrimidin‐4‐ones 3a‐g. The trans isomer 7 failed to cyclize, but gave carboxamide 8a,b or thiourea ester derivatives 9a,b. 相似文献

New high yield preparation methods were developed for the pharmaceutically interesting compounds, 1‐benzyl‐, 1‐methyl‐, and 1H‐5‐[(2‐oxo‐2‐phenyl)ethyl]imidazoles 1a‐c , respectively. The title compounds were synthesized by four different methods using various starting materials. Two of the methods involved transformation reactions of the key intermediates, 1‐substituted‐5‐[(2‐nitro‐2‐phenyl)ethenyl]imidazoles 2a‐c and 1‐substituted‐5‐[(2‐nitro‐2‐phenyl)ethyl]imidazoles 3a‐c , while the other two utilized the oxidation of 1‐substituted‐5‐[(2‐hydroxy‐2‐phenyl)ethyl]imidazoles 4a‐c , with chromic oxide, and the umpolung reaction of benzaldehyde followed by a condensation reaction of the umpolung intermediate with imidazolecarboxaldehydes 6a‐c. 相似文献

Bicycle ring closure on a mixture of (4aS,8aR)‐ and (4aR,8aS)‐ethyl 2‐oxodecahydro‐1,6‐naphthyridine‐6‐carboxylate, followed by conversion of the separated cis and trans isomers to the corresponding thioamide derivatives, gave (4aSR,8aRS)‐ethyl 2‐sulfanylidenedecahydro‐1,6‐naphthyridine‐6‐carboxylate, C11H18N2O2S. Structural analysis of this thioamide revealed a structure with two crystallographically independent conformers per asymmetric unit (Z′ = 2). The reciprocal bicycle ring closure on (3aRS,7aRS)‐ethyl 2‐oxooctahydro‐1H‐pyrrolo[3,2‐c]pyridine‐5‐carboxylate, C10H16N2O3, was also accomplished in good overall yield. Here the five‐membered ring is disordered over two positions, so that both enantiomers are represented in the asymmetric unit. The compounds act as key intermediates towards the synthesis of potential new polycyclic medicinal chemical structures. 相似文献

The gas‐phase elimination kinetics of the above‐mentioned compounds were determined in a static reaction system over the temperature range of 369–450.3°C and pressure range of 29–103.5 Torr. The reactions are homogeneous, unimolecular, and obey a first‐order rate law. The rate coefficients are given by the following Arrhenius expressions: ethyl 3‐(piperidin‐1‐yl) propionate, log k1(s?1) = (12.79 ± 0.16) ? (199.7 ± 2.0) kJ mol?1 (2.303 RT)?1; ethyl 1‐methylpiperidine‐3‐carboxylate, log k1(s?1) = (13.07 ± 0.12)–(212.8 ± 1.6) kJ mol?1 (2.303 RT)?1; ethyl piperidine‐3‐carboxylate, log k1(s?1) = (13.12 ± 0.13) ? (210.4 ± 1.7) kJ mol?1 (2.303 RT)?1; and 3‐piperidine carboxylic acid, log k1(s?1) = (14.24 ± 0.17) ? (234.4 ± 2.2) kJ mol?1 (2.303 RT)?1. The first step of decomposition of these esters is the formation of the corresponding carboxylic acids and ethylene through a concerted six‐membered cyclic transition state type of mechanism. Ethyl 3,4‐dihydro‐2H‐1,4‐benzoxazine‐3‐carboxylate derivatives 2 were obtained and isolated in low yields from the condensation of 2‐aminophenol and ethyl 2,3‐dibromopropanoate. They can be obtained by hydrogenation of ethyl 2H‐1,4‐benzoxazine‐3‐carboxylate in satisfactory yield. Using 2‐iminophenol did not direct the condensation with ethyl 2,3‐dibromopropanoate towards 2 but was fruitfull for the preparation of ethyl 2‐(4‐benzyl‐3,4‐dihydro‐2H‐1,4‐benzoxazin‐3‐yl)acetate from ethyl bromocrotonate. 相似文献

The crystal structures of (1R,1′S)‐2′,2′‐dichloro‐N‐(1‐phenylethyl)cyclopropane‐1′‐carboxamide, C12H13Cl2NO, (I), and (1R,1′R)‐2′,2′‐difluoro‐N‐(1‐phenylethyl)cyclopropane‐1′‐carboxamide, C12H13F2NO, (II), have been determined. Both crystals contain two independent molecules with different conformations of the phenylethyl groups. In the crystals of both compounds, the molecules are linked together by N—H⃛O hydrogen bonds, thus forming chains in the a direction. 相似文献

A series of novel 1‐[(1R)‐1‐(6‐fluoro‐1,3‐benzothiazol‐2‐yl)ethyl]‐3‐substituted phenyl ureas were synthesized by the condensation of (1R)‐1‐(6‐fluoro‐1,3‐benzothiazol‐2‐yl)ethanamine with substituted phenyl isocyanates under mild conditions. Their structures were confirmed 1H, 13C, and 19F NMR spectra, and elemental analyses. The optical activities were confirmed by optical rotation measurements. The inhibition activity of 1‐[(1R)‐1‐(6‐fluoro‐1,3‐benzothiazol‐2‐yl)ethyl]‐3‐substituted phenyl ureas to acetylcholinesterase (ACHE) and butyrylcholinesterase (BCHE) was also tested. Preliminary bioassay indicated that the target ureas displayed excellent acetylcholinesterase and butyrylcholinesterase inhibition activity. J. Heterocyclic Chem., 2011. 相似文献

The gas‐phase elimination kinetics of ethyl 2‐furoate and 2‐ethyl 2‐thiophenecarboxylate was carried out in a static reaction system over the temperature range of 623.15–683.15 K (350–410°C) and pressure range of 30–113 Torr. The reactions proved to be homogeneous, unimolecular, and obey a first‐order rate law. The rate coefficients are expressed by the following Arrhenius equations: ethyl 2‐furoate, log k1 (s?1) = (11.51 ± 0.17)–(185.6 ± 2.2) kJ mol?1 (2.303 RT)?1; ethyl 2‐thiophenecarboxylate, log k1 (s?1) = (11.59 ± 0.19)–(183.8 ± 2.4) kJ mol?1 (2.303 RT)?1. The elimination products are ethylene and the corresponding heteroaromatic 2‐carboxylic acid. However, as the reaction temperature increases, the intermediate heteroaromatic carboxylic acid products slowly decarboxylate to give the corresponding heteroaromatic furan and thiophene, respectively. The regioselective reactions of luminol with 1,3‐cyclohexanedione (or malononitrile) and aromatic aldehydes catalyzed by 2‐1′‐methylimidazolium‐3‐yl‐1‐ethyl sulfate were developed to synthesize 7‐amino‐3,4‐dihydro‐2H‐indazolo[2,1‐b]phthalazine‐1,6,11(13H)‐triones and 3,9‐diamino‐5,10‐dihydro‐5,10‐dioxo‐1H‐pyrazolo[1,2‐b]phthalazine‐2‐carbonitriles in good to excellent yields in short times. 相似文献

The title compound, C16H23N5O3S, ethyl 5-amino-1-(5‘-methyl-1‘-t-butyl-4‘-pyrazolyl)carbonyl-3-methylthio-1H-pyrazole-4-carboxylate (5) has been synthesized by the treatment of ethyl 2-cyano-3,3-dimethylthioacrylate with 1-t-butyl-5-methyl-4-hydrazinocarbonylpyrazole (4) in refluxed ethanol. The possible mechanism of the above reaction was also discussed. The results of biological test show that the title compound has fungicidal and plant growth regulation activities. 相似文献

Novel polystyrene derivatives comprising [1‐(3‐isopropenyl‐phenyl)‐1‐methyl‐ethyl]‐carbamate in the side chain were synthesized as photoreactive copolymers. Poly(4‐vinylphenol) was made to react with 1‐(1‐isocyanato‐1‐methyl‐ethyl)‐3‐isopropenyl‐benzene (m‐TMI) and the unreacted hydroxyl groups were protected with acetyl chloride. The copolymers are highly sensitive to the radical photoinitiators that can be activated by irradiation of UV light (λ = 300–365 nm). FTIR spectroscopy was employed to monitor the structural changes in the copolymers exposed to UV irradiation. The dielectric properties of the copolymers were investigated by measuring the capacitance and calculating the permittivity as a function of frequency, along with the I–V characteristics. Their properties were compared with those of thermally crosslinkable poly(4‐vinylphenol) blended with poly(melamine‐co‐formaldehyde), which is frequently used as a dielectric layer in organic field‐effect transistors (OFETs). No significant dielectric dispersion was observed in the frequency range of 1 kHz–1 MHz. The dielectric constant was determined to be in the range of 4.2–6.0, which offers a potential for the application of these copolymers to OFET gate insulators. These soluble dielectrics exhibit good film uniformity and can also be patterned using a standard photolithographic technique. © 2008 Wiley Periodicals, Inc. Two tricarbonyl complexes of rhenium(I) and manganese(I) coordinated by the ligand 2‐{[2‐(1H‐imidazol‐4‐yl)ethyl]iminomethyl}‐5‐methylphenolate are reported, viz. fac‐tricarbonyl(2‐{[2‐(1H‐imidazol‐4‐yl‐κN3)ethyl]iminomethyl‐κN}‐5‐methylphenolato‐κO)rhenium(I) methanol monosolvate, [Re(C16H14N3O4)(CO)3]·CH3OH, (I), and fac‐tricarbonyl(2‐{[2‐(1H‐imidazol‐4‐yl‐κN3)ethyl]iminomethyl‐κN}‐5‐methylphenolato‐κO)manganese(I), fac‐[Mn(C16H14N3O4)(CO)3], (II), display facial coordination in a distorted octahedral environment. The crystal structure of (I) is stabilized by O—H...O, N—H...O and C—H...O hydrogen‐bond interactions, while that of (II) is stabilized by N—H...O hydrogen‐bond interactions only. An efficient general route to the synthesis of 5‐substituted 1H‐1,2,4‐triazole‐3‐carboxylates was developed. N‐acylamidrazones were obtained from carboxylic acid hydrazides and ethyl thiooxamate or ethyl 2‐ethoxy‐2‐iminoacetate hydrochloride and then were reacted with chloroanhydride of the same carboxylic acid. As part of the structure‐activity relationship of the dopamine D2 and serotonin 5‐HT3 receptors antagonist 1, which is a clinical candidate with a broad antiemetic activity, the synthesis and dopamine D2 and serotonin 5‐HT3 receptors binding affinity of (R)‐5‐bromo‐N‐(1‐ethyl‐3‐methylhexahydro‐1,3‐diazin‐5‐yl)‐ and (R)‐5‐bromo‐N‐(1‐ethyl‐5‐methyloctahydro‐1,5‐diazocin‐3‐yl)‐2‐methoxy‐6‐methylaminopyridine‐3‐carboxam‐ides ( 2 and 3 ) are described. Treatment of 1‐ethyl‐2‐(p‐toluenesulfonyl)amino‐3‐methylaminopropane dihy‐drochloride ( 4a ) with paraformaldehyde and successive deprotection gave the 5‐aminohexahydro‐1,3‐diazine 6 in excellent yield. 3‐Amino‐1‐ethyl‐5‐methyloctahydro‐1,5‐diazocine ( 15 ) was prepared from 2‐(benzyloxycarbonyl)amino‐3‐[[N‐(tert‐butoxycarbonyl)‐N‐methyl]amino]‐1‐ethylaminopropane ( 9 ) through the intramolecular amidation of (R)‐3‐[N‐[(2‐benzyloxycarbonylamino‐3‐methylamino)propyl]‐N‐ethyl]aminopropionic acid trifluoroacetate ( 12 ), followed by lithium aluminum hydride reduction of the resulting 6‐oxo‐1‐ethyl‐5‐methyloctahydrodiazocine ( 13 ) in 41% yield. Reaction of the amines 6 and 15 with 5‐bromo‐2‐methoxy‐6‐methylaminopyridine‐3‐carboxylic acid furnished the desired 2 and 3 , which showed much less potent affinity for dopamine D2 receptors than 1 . 相似文献

A number of coumarino[6,7‐d]oxazoles (nitrogen analogs of psoralens) have been synthesized from (7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl) acetic acid ethyl ester 1 . The synthetic route began with the nitration of 1 with nitric acid in acetic acid to give (6‐nitro‐7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl) acetic acid ethyl ester 2 ; (3,6‐dinitro‐7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl) acetic acid ethyl ester 3 and (3,6,8‐trinitro‐7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl) acetic acid ethyl ester 4 . The reduction of 2 was accomplished with tin(II) chloride, tin, and concentrated hydrochloric acid in ethanol giving (6‐amino‐7‐hydroxy‐2‐oxo‐2H‐chromen‐4‐yl) acetic acid ethyl ester 5 . After the condensation of aminocoumarin 5 with aromatic aldehyde in glacial acetic acid medium, followed the dehydrocyclization to coumarino[6,7‐d]oxazoles 7a‐k . The intermediate Schiff's bases 6a‐k have been obtained from 5 with aromatic aldehyde in ethanol. Antibacterial and antifungal activities of the compounds have been evaluated. 相似文献

The reaction of 1 with dibenzoylmethane, a dicarbonyl compound, provided ethyl 3‐benzoyl‐4‐oxo‐2,6‐diphenylpyran‐5‐carboxylate derivative 9 . Compound 9 was converted into the corresponding ethyl 3‐benzoyl‐4‐hydroxy‐2,6‐diphenylpyridine‐5‐carboxylate derivative 10 via its reaction with ammonium hydroxyde solution in 1 ‐butanol. 相似文献

Two‐step synthesis of N‐aryl 4‐[(ethoxycarbonyl) oxy]‐1H‐pyrazole‐3‐carboxylates is achieved starting from the commercially available ethyl 4‐chloroacetoacetate and aromatic amines. Azo coupling followed by cyclization with ethyl chloroformate–DMAP pair resulted in the formation of new 4‐oxy‐1H‐pyrazole derivatives in high yields. 相似文献
