It has been shown that dimethylamino-substituted 2,5-diaryloxazoles and 2,5-diaryl-1,3,4-oxadiazoles undergo heterocyclization under Vilsmeier-Haack conditions with the participation of the dimethylamino group to form quinazolinium salts. The oxazole ring can also be formylated at the free 4 position. In alkaline medium the quinazolinium ring is readily hydrolyzed with desalkylation.  相似文献

The reactions of trichloromethylarenes with excess hydrazine hydrate in ethanol gives symmetrical 2,5-diaryl-1,3,4-oxadiazoles in 68–96% yields. The reaction of 1,4-bis(trichloromethyl)benzene with acylhydrazines in an ethanol-pyridine mixture gives the corresponding substituted or unsubstituted 1,4-phenylenebis-1,3,4-oxadiazoles in 35–51% yields. The mass spectra of 2,5-diaryl-1,3,4-oxadiazoles and 1,4-phenylenebis-1,3,4-oxadiazoles were studied. Translated fromIzvestiya Akademii Nauk. A series of novel 2,5-disubstituted-1,3,4-oxadiazoles have been synthesized from long-chain alkanoic and alkenoic acids. The structures of these compounds have been elucidated by elemental and spectral (IR, ^1H NMR, ^13C NMR, MS) analysis. Furthermore, compounds were screened for in vitro antibacterial activity against the representative panel of two Gram-positive and two Gram- negative bacteria. All the synthesized compounds were also tested for their inhibitory action against five strains of fungus. The various compounds show potent inhibitory action against test organisms. 2007 Abdul Rauf. The effect of the solvent on the course of the reaction between trichloromethylarenes and thioacylhydrazines or acylhydrazines has been considered. In alcohols as solvents, alkyl arenecarboxylates form as a result of alcoholysis, while 2,5-disubstituted 1,3,4-thiadiazoles (1,3,4-oxadiazoles) form as minor products. In pyridine solutions, the major or sole products are those of reductive condensation,i.e., the correspondingN-substituted hydrazones of arenecarbaldehydes. 1,3,4-Thiadiazole or 1,3,4-oxadiazole derivatives are obtained in good yield when the reaction is carried out in a pyridine-alkanol mixture.Translated fromIzvestiya Akademii Nauk. A series of new symmetrical 2,5-dialkyl-1,3,4-oxadiazoles containing substituted alkyl groups at the terminal positions with substituents, such as bromine, isopropyloxycarbonylmethylamino, and carboxymethylamino, were successfully synthesized. The developed multistep method employed commercially available acid chlorides differing in alkyl chain length and terminal substituent, hydrazine hydrate, and phosphorus oxychloride. The intermediate bromine-containing 2,5-dialkyl-1,3,4-oxadiazoles were easily substituted with diisopropyl iminodiacetate, followed by hydrolysis in aqueous methanol solution giving the corresponding 1,3,4-oxadiazoles bearing carboxymethylamino substituents. The structure of all products was confirmed by conventional spectroscopic methods including 1H NMR, 13C NMR, and HRMS.  相似文献

Four series of symmetrically 2,5-disubstituted 1,3,4-oxadiazoles with different lateral substituents and terminal alkoxy chain length were synthesised and characterised. Then, all these bent-shaped compounds were separately doped into the blue phase (BP) liquid crystal host and their effects on the BP range of the host were investigated, which shows that the BP ranges varied greatly with the difference in the structure of 2,5-disubstituted 1,3,4-oxadiazoles and their doped concentration. Moreover, in order to explore the influence of the inherent molecular geometry and properties of the bent-shaped molecules on the stability of BPs, the molecular structures of all the compounds were studied by using the theory of density functional theory, and the optimised structural parameters of the molecules were calculated at the B3LYP/6–31G* level.  相似文献

Reaction of 2,5-dimercapto-1,3,4-thiadiazole with ammonia or pyridine gives monoammonium or monopyridinium slats, and the reaction with hydrazine hydrate gives both mono- and dihydrazine salts, which was confirmed by alkylation of the salts obtained. Abstract

Ceric ammonium nitrate in polyethylene glycol has been used as a sustainable, non-volatile, and ecofriendly catalytic medium for the green synthesis of 2,5-disubstituted 1,3,4-oxadiazoles. This protocol is effective toward various substrates having different functionalities. The easy recyclability of the reaction medium makes the reaction economically and potentially viable for commercial applications.  相似文献

The desired 1,3,4-thiadiazole compounds bearing different substituents were obtained by the cyclization of the corresponding thiosemicarbazide followed by the reaction with electrophilic reagents, such as aromatic aldehydes, isatin, phenyl isothiocyanate, and carbon disulfide. The newly synthesized 2,5-disubstituted 1,3,4-thiadiazoles were obtained in good yields and their structures were elucidated by spectral data and elemental analysis.

A one-pot synthesis of some unsymmetrical 2,5-disubstituted-1,3,4-oxadiazoles via cyclocondensation of benzoylhydrazines with orthoesters under solvent-free and microwave conditions are described here. The reaction is efficiently catalyzed by silica- supported sulfuric acid as it provided the title compounds in high yields and relatively short times. The catalyst is reusable and can be applied several times without considerable decrease in the yields and rates of the reactions.  相似文献

The behavior of dimethylamino-substituted 7H-benzo[de]pyrazolo[5,1-a]isoquinoline-7-ones, synthesized for the first time, under conditions of the Vilsmeier-Haack reaction. It has been shown that, on heating with POCl3 and DMF, they are converted by electrophilic substitution at the position ortho to the dimethylamino group, followed by cyclization of the iminium adduct to a quinazolinium salt. When an acetyl group is present, the Arnold reaction, leading to the formation of a chloroaryl, accompanies the heterocyclization. The rates and proportion of the reaction products depend on the position of the dimethyl groups relative to the pyrazole ring.  相似文献

An X-ray structural study of 2,5-bis(fluorodinitromethyl)-1,3,4-oxadiazole (1), one of the representatives of azoles with strong electron-withdrawing substituents, has been carried out. It has been found that in the orthorhombic modification of1 the conformations of the fluorodinitromethyl groups are different. Intermolecular contacts in the crystal are realized through the heterocycles arranged at large angles to each other. The presence of strong electron-withdrawing substituents results in shortening of the C-O bonds and in a certain increase in the OCN angle in the 1,3,4-oxadiazole ring.Translated fromIzvestiya Akademii Nauk. A kinetic investigation has been carried out, using electronic absorption spectra, of the alkylation with benzyl chloride of the dihydrazine, monohydrazine, and the disodium salts of 2,5-dimercapto-1,3,4-thiadiazole, and also of the hydrazine and sodium salts of 5-benzylthio-2-mercapto-1,3,4-thiadiazole. Nine novel symmetrical and asymmetrical 2,5-disubstituted 1,3,4-oxadiazoles have been synthesized by a facile and mild method with high yield. Meantime, it was found that the fluorine was easy substituted by hydrazine in polyhalogen-substituted aroyl hydrazine. The preliminary bioassay tests show that two of the compounds (D5 and D6) exhibited a significant insecticidal activity (LC50=116.02 and 70.93 mg l−1) on armyworm, Leucania separata Walker. Reactions of symmetrical 1,1′-diphenylthiodiacetic acid dihydrazide and triethyl orthoesters in the presence of catalytic amount of glacial acetic acid resulted in the formation of three heterocyclic products: the appropriate bis(1,3,4-oxadiazol-2-yl-phenylmethyl) sulfides, 2-benzyl-1,3,4-oxadiazoles and 2-benzoyl-1,3,4-oxadiazoles. The presence of the latter two compounds is connected with carbon-sulfur fission in the molecule of the starting hydrazide. The identity of the unexpected fission products was confirmed by parallel syntheses of the model 1,3,4-oxadiazoles from phenylacetic acid hydrazide and 2-hydroxymethyl-1,3,4-oxadiazole derivatives.  相似文献

New interesting class of new polyhydrazides having inherent viscosities in the range 0.45–0.71 dI/g were prepared by polymerizing a series of diacid chlorides, e.g., sebacoyl, isophthaloyl or terphthaloyl with 9,10-dihydro-9,10-ethanoanthracene-11,12-dihydrazide I in polar aprotic solvent and by the low-temperature polycondensation technique. In order to characterize the polymers, a model compound II was synthesized from I and benzoyl chloride. All the hydrazide polymers are semi crystalline in nature and are readily soluble in various polar solvents such as N-methyl pyrrolidine (NMP) and dimethylsulfoxide (DMSO). Their Tgs were recorded in the range of 78–95 °C and could be thermally dehydrated into the corresponding polyoxadiazoles in the region of 310–20 °C, as evidenced by the DTA thermograms. The oxadiazole polymers showed a dramatically decreased in solubility and higher Tg when compared to their respective hydrazide prepolymers. The morphology of polyhydrazide V was examined by SEM.  相似文献
