This study compared phenolic contents and antioxidant activity in different organs of Acacia albida (leaves and bark) and focuses on identification of phenolic compounds of leaves by HPLC-DAD. The analysed organs exhibited differences in total polyphenol contents (100 and 59.5 mg GAE g? 1 DW). Phenolic contents of leaves were two times higher than those in bark. Ethanolic extracts exhibited good antioxidant activities with IC50 = 26 μg mL? 1 for DPPH and EC50 = 50 μg mL? 1 for FRAP. Identification by HPLC-DAD revealed the presence of nine phenolic compounds known for their high antioxidant activity. The results suggested that this species can be used as source of natural antioxidants.  相似文献

Diarylheptanoid-rich extracts (DAHEs) isolated from grey and black alder bark with pressurized ethyl acetate were tested in mayonnaise as a dietary antioxidant using the Oxipres method. DAHE contains >700 mg g?1 of oregonin possessing quite similar structure to the well-known dietary antioxidant curcumin, which is widely used as a bioactive constituent of food supplements. Strong radical scavenging capacity of oregonin was clearly demonstrated by the online HPLC–DPPH? scavenging assay. The antioxidative effect of DAHEs in mayonnaise was dose dependent; however, even the lowest concentration (0.5 mg g?1) of DAHE additive increased the induction period of oxidation more than two times. Consequently, DAHE may be considered as a promising natural antioxidant for commercial applications.  相似文献

Background

In this study Aesculus indica fruit was subjected to isolation of phytochemicals. Two antioxidants quercetin and Mandelic acid were isolated in pure state. The free radical scavenging and acetyl choline esterase inhibitory potential of the crude extract and sub fractions were also determined.

Results

The antioxidant capacity of crude extract, fractions and isolated compounds were determined by DPPH and ABTS methods. Folin-Ciocalteu reagent method was used to estimate the total phenolic contents and were found to be 78.34?±?0.96, 44.16?±?1.05, 65.45?±?1.29, 37.85?±?1.44 and 50.23?±?2.431 (mg/g of gallic acid) in crude extract, ethyl acetate, chloroform, n-hexane and aqueous fractions respectively. The flavonoid concentration in crude extract, ethyl acetate, chloroform, n-hexane and aqueous fraction were; 85.30?±?1.20, 53.80?±?1.07, 77.50?±?1.12, 26.30?±?1.35 and 37.78?±?1.25 (mg/g of quercetin) respectively. The chloroform fraction was more potent against enzymes, acetyl choline esterase and butyryl choline esterase (IC50?=?85 and 160 μg/ml respectively). The phenolic compounds in the crude extract and fractions were determined using HPLC standard method. Chlorogenic acid, quercetin, phloroglucinol, rutin, mandelic acid and hydroxy benzoic acid were detected at retention times 6.005, 10.062, 22.623, 30.597, 35.490 and 36.211 in crude extract and different fractions. The ethyl acetate fraction was rich in the targeted compounds and was therefore subjected to column isolation. The HPLC chromatogram of isolated compounds showed single peak at specified retention times which confirms their isolation in pure state. The isolated compounds were then characterized by FTIR and NMR spectrophotometric techniques.

Conclusion

The Aesculus indica fruit extracts showed antioxidant and anticholine esterase inhibitory potentials. Two bioactive compounds were isolated in the pure form ethyl acetate fraction. From results it was concluded that the fruit of this plant could be used to minimize oxidative stress caused by reactive oxygen species.

Methanolic extracts from in vitro grown Scutellaria lateriflora shoots cultured on five Murashige and Skoog (MS) medium variants supplemented with different combinations of 6-benzylaminopurine (BAP) and α-naphthaleneacetic acid (NAA) under different light conditions (monochromatic light, white light and no light) were analysed by HPLC for three groups of metabolites: flavonoids (26 compounds), phenolic acids and their precursors (19+2) and phenylethanoid glycosides (2). The analyses revealed the presence of baicalein, baicalin, wogonin, wogonoside, 3,4-dihydroxyphenylacetic acid and verbascoside. There was clear evidence of the influence of plant growth regulators and light conditions on the accumulation of the analysed groups of secondary metabolites. The amounts of the compounds changed within a wide range—for the total flavonoid content, 30.2-fold (max. 1204.3 mg·100 g?1 dry weight (DW)); for 3,4-dihydroxyphenylacetic acid, 5.5-fold (max. 33.56 mg·100 g?1 DW); and for verbascoside, 1.5-fold (169.15 max. mg·100 g?1 DW). The best medium for the production of most of the compounds was the Murashige and Skoog variant with 1 mg l?1 BAP and 1 mg l?1 NAA. For verbascoside, the best ‘productive’ medium was the MS variant supplemented with 0.5 mg l?1 BAP and 2 mg l?1 NAA. The accumulation of the metabolites was stimulated to the greatest extent by blue light, under which the extracts were found to contain the highest total amount of flavonoids and the highest amounts of flavonoid glucuronides, baicalin and wogonoside, as well as of verbascoside. Their amounts were, respectively, 1.54-, 1.49-, 2.05- and 1.86-fold higher than under the control white light.  相似文献

Mycelial growth in a defined medium by submerged fermentation is a rapid and alternative method for obtaining fungal biomass of consistent quality. Biomass, exopolysaccharides (EPS) and intracellular polysaccharides (IPS) production were optimised by response surface methodology in Lentinula edodes strain LeS (NCBI JX915793). The optimised conditions were pH 5.0, temperature 26°C, incubation period of 25 days and agitation rate of 52 r/min for L. edodes strain LeS. Under the calculated optimal culture conditions, biomass production (5.88 mg mL? 1), EPS production (0.40 mg mL? 1) and IPS production (12.45 mg g? 1) were in agreement with the predicted values for biomass (5.93 mg mL? 1), EPS (0.55 mg mL? 1) and IPS production (12.64 mg g? 1). Crude lentinan exhibited highest antibacterial effects followed by alcoholic, crude and aqueous extracts. The results obtained may be useful for highly effective yield of biomass and bioactive metabolites.  相似文献

Clematis gouriana (Ranunculaceae), a perennial herb, is used by the local inhabitants of the western Himalayan region for its medicinal properties. Major bioconstituents of C. gouriana leaves using different solvent extracts were obtained and analysed. The results revealed promising contents of phenolics (from 18.19 ± 0.10 to 22.17 ± 0.10 mg g? 1) as gallic acid and flavonoids (from 2.83 ± 0.01 to 6.52 ± 0.08 mg g? 1) as quercetin equivalent in different extracts. Aqueous acetone extract showed higher antioxidant activity with IC50 value of 129.11 and 25.35 μg mL? 1 against DPPH and ABTS free radicals, respectively. Antioxidant yield ranged from 16.87 ± 0.27 to 24.48 ± 0.13 mg g? 1 of Trolox equivalent in different extracts as measured by the FRAP assay. Furthermore, ethylacetate extract exhibited strong in vitro cytotoxicity against Chinese hamster ovary and glioma cell lines. Proximate composition (proteins, fats, ash and minerals) of C. gouriana leaves was also assessed. Results demonstrated the potential of C. gouriana bioconstituents as nutraceuticals.  相似文献

Taking into account the importance of natural antioxidants in the preservation of oils and fats, the present study evaluated the antioxidant action of five plant extracts in the control of soybean oil stability, by means of the accelerated techniques Rancimat and PDSC. These plants are rosemary (Rosmarinus officinalis L.), chamomile (Matricaria recutita L.), coriander (Coriandrum sativum L.), fennel (Foeniculum vulgare), and senna (Cassia angustifolia Vahl). The plant extracts and also the synthetic antioxidant BHT were added to the samples of crude soybean oil at the concentration of 1,000 mg kg?1. The values of total phenolic contents ranged from 8.7 ± 0.4 to 63.0 ± 2.3 mg GAE g?1 extract and a strong positive correlation was observed between the total phenolic contents and the overall antioxidant activity of the plant extracts. Such high values indicate a good protection of the analyzed soybean oil, moreover for the Rosemary extract that was superior to the remaining extracts. In the Rancimat technique the rosemary extract was more effective than the synthetic BHT antioxidant. The OIT values of Rosemary extract and the BHT antioxidant were equivalent, and the former, showed the highest phenolic contents among the extracts, for all the performed tests, confirming that it is a powerful natural source of antioxidants.  相似文献

The objective of this study was to develop a simple, efficient and reliable method for routine quantitative analysis for Psoralea corylifolia L. An ultra performance liquid chromatography with DAD detector system was employed for simultaneous quantification of ten compounds. The chromatographic analysis was performed by UPLC with C18 column and gradient elution of 0.05% formic acid aqueous solution and acetonitrile in 16 min. All calibration curves were linear (R 2 ≥ 0.9990) over the tested ranges. The LOD and LOQ were lower to 13.07 and 39.22 ng mL?1 with 2 μL of injection volume, respectively. The intra- and inter-day precisions as determined from sample solutions were below 4.1 and 4.2%. The average recoveries were ranged from 94.2 to 108.8% with RSDs ≤ 4.6%. This validated method was applied for the analysis of ten analytes in P. corylifolia L. from different origins. The variation of the content of ten compounds was remarkable among the tested samples: psoralenoside increased from 7.42 to 17.04 mg g?1, isopsoralenoside from 6.05 to 14.34 mg g?1, psoralen from 2.37 to 3.90 mg g?1, isopsoralen from 2.53 to 3.65 mg g?1, neobavaisoflavone from 1.59 to 2.96 mg g?1, bavachin from 1.02 to 2.35 mg g?1, psoralidin from 0.45 to 1.91 mg g?1, isobavachalcone from 1.33 to 4.71 mg g?1, corylifol A from 1.02 to 2.40 mg g?1 and bakuchiol from 28.10 to 63.89 mg g?1.  相似文献

Solid phase extraction (SPE) using Sep-Pak? cartridges is one of the techniques used for fractionation of antioxidant compounds in waste of dabai oil extraction (defatted dabai parts). The aim of this study was to determine the phenolic compounds and antioxidant capacity in crude extracts and several SPE fractions from methanolic extract of defatted dabai pulp and peel. Based on SPE, Sep-Pak? cyanopropyl and C18 cartridges were used to fractionate the antioxidant-rich crude extracts into water and methanolic fractions. Analyzed using LC-MS, flavonoids, anthocyanins, saponin derivatives and other unknown antioxidative compounds were detected in the defatted dabai crude extracts and their SPE fractions. Anthocyanins were the major phenolic compounds identified in the defatted dabai peel and detected in most of the SPE fractions. Methanolic fractions of defatted dabai parts embraced higher total phenolics and antioxidant capacity than water fractions. This finding also revealed the crude extracts of defatted dabai peel have the most significant antioxidant properties compared to the methanolic and water fractions studied. The crude extract of defatted dabai parts remain as the most potent antioxidant as it contains mixture of flavonoids, anthocyanins and other potential antioxidants.  相似文献

Two anthracene molecularly imprinted nanoparticle polymers namely; An–MINP1 and An–MINP2, were synthesized using the precipitation polymerization method. An–MINPs were used for selective separation of anthracene from aqueous solutions. The data revealed that the maximum binding capacity of An–MINPs for anthracene were 320.8 and 374.3 mg g?1 for An–MINP1 and An–MIPN2, respectively, compared with 2.8 and 4 µg g?1 obtained by using their corresponding non-imprinted polymers NIP1 and NIP2, respectively. Under optimized conditions, An–MINPs give high selectivity and sensitivity of anthracene separation. The anthracene uptake percentage from aqueous solutions ranged from 90.3 to 99.9%.  相似文献

The use of natural antioxidants has been widely promoted in the food industry, because it is economically viable and very attractive with consumers. In this study, we determined the extractable total phenolic content (FET) of crude ethanol extracts of the leaves (EL), flowers, and seed pods of Moringa oleifera Lam. We also evaluated the antioxidant effect of the extracts on oxidative stability of soybean oil, through the accelerated PDSC and Rancimat® techniques using the synthetic antioxidants butylated hydroxytoluene (BHT) and tert-butylhydroquinone (TBHQ) as positive control. The values of FET in the extracts ranged from 41.75 ± 3.35 to 53.69 ± 1.00 mg GAE g?1, and EL extract exhibited best result. The results of oxidative stability in both techniques showed that the EL extract provided greater protection to the oil, indicating a correlation between the amount of FET and the protective effect. Comparison made between the synthetic antioxidants and extracts revealed that in the Rancimat technique, extracts were less effective than the synthetic antioxidants TBHQ and BHT, however, in the PDSC technique the EL extract was more effective than BHT, proving to be a good alternative for applications in the soybean oil, replacing this synthetic antioxidant.  相似文献

This paper reports the evaluation of the antioxidant potential of the ethanolic extracts of the leaves (EL), flowers (EF), seed pods (EP) and seeds (ES) from Moringa oleifera Lam. The antioxidant potential was assessed, upon the addition of the extracts to fish oil, by means of the total extractable phenol content (TEP), the DPPH· free radical scavenging efficiency and using pressurized differential scanning calorimetry (P-DSC). The results of TEP and DPPH· showed that the ES extract does not present a potential to be used as an antioxidant additive, and it was thus discarded from the remaining analyses. Thus, the following treatments were prepared: pure fish oil, fish oil with BHT (100 mg kg?1), fish oil with TBHQ (100 mg kg?1) and fish oil with EL, EF and EP—all at the concentration of 100 mg kg?1, in relation to the total extractable phenolics contained in each one of the extracts. The leaf and flower extracts displayed a protecting effect, with an increase in about 20 and 11 % of the OIT values, respectively. However, such protection was smaller than that conferred by the synthetic antioxidants utilized. As for the thermal analysis results, it was noticed that EL presented the highest thermal stability among the extracts.  相似文献

In this study, the use of the organic fraction of municipal solid waste as an abundant and low-cost raw material for producing activated carbon was investigated. For this purpose, ZnCl2 was used as a chemical activation agent and the carbonization process took place at 800 °C in N2 atmosphere. Seven sorbents were prepared by chemical activation (pyrolysis under N2 atmosphere at temperature of 800 °C after impregnation with ZnCl2) with different ratios of ZnCl2. The optimum ratio of organic fraction of municipal solid waste to ZnCl2 was inspected via methylene blue number and iodine number (ASTM Designation: D4607–94). The results showed that the adsorbent with 60 % ZnCl2/raw material was the most appropriate one with a satisfactory adsorption capacity, 112.4 mg g?1 for methylene blue and 134.0 mg g?1 for iodine. In addition, the structural analysis of this sorbent was performed using FT-IR, BET surface area, SEM–EDX and thermal analysis. Application of this sorbent to remove Cr(VI) from wastewater was studied to find an adsorption capacity of 66.7 mg g?1. The experimental adsorption equilibrium data were fitted to Langmuir adsorption model with an acceptable adsorption capacity of 66.7 mg g?1.  相似文献

The present study investigates the adsorption capability of raw and biochar forms of Chrysanthemum indicum flowers biomass to remove cobalt ions from aqueous solution in a fixed-bed column. Column adsorption experiments were conducted by varying the bed height (1.0, 2.0, 3.0 cm), flow rate (1.0, 2.5, 5.0 mL min?1) and initial cobalt ion concentration (25, 50, 75 mg L?1) to obtain the experimental breakthrough curves. The adsorption capacity of the raw and biochar forms of C. indicum flowers were found to be 14.84 and 28.34 mg g?1, respectively, for an initial ion concentration of 50 mg L?1 at 1.0 cm bed height and 1.0 mL min?1 flow rate for Co (II) ion adsorption. Adam–Bohart, Thomas and Yoon–Nelson models were applied to the experimental column data to analyze the column performance. The Thomas model was found to best represent the column data with the predicted and experimental uptake capacity values correlating well and with higher R 2 values for all the varying process parameters. Desorption studies revealed the suitability of the adsorbents for repeated use up to four adsorption–desorption cycles without significant loss in its efficiency. It can thus be inferred from the fixed-bed column studies that C. indicum flowers can suitably be used as an effective adsorbent for Co (II) ion removal from aqueous solution on a higher scale.  相似文献

Uranium (VI)-containing water has been recognized as a potential longer-term radiological health hazard. In this work, the sorptive potential of sunflower straw for U (VI) from aqueous solution was investigated in detail, including the effect of initial solution pH, adsorbent dosage, temperature, contact time and initial U (VI) concentration. A dose of 2.0 g L?1 of sunflower straw in an initial U (VI) concentration of 20 mg L?1 with an initial pH of 5.0 and a contact time of 10 h resulted in the maximum U (VI) uptake (about 6.96 mg g?1) at 298 K. The isotherm adsorption data was modeled best by the nonlinear Langmuir–Freundlich equation. The equilibrium sorption capacity of sunflower straw was observed to be approximately seven times higher than that of coconut-shell activated carbon as 251.52 and 32.37 mg g?1 under optimal conditions, respectively. The positive enthalpy and negative free energy suggested the endothermic and spontaneous nature of sorption, respectively. The kinetic data conformed successfully to the pseudo-second-order equation. Furthermore, energy dispersive X-ray, fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy demonstrated that U (VI) adsorption onto sunflower straw was predominantly controlled by ion exchange as well as complexation mechanism. The study revealed that sunflower straw could be exploited for uranium remediation of aqueous streams as a promising adsorbent.  相似文献

The objective of this study was to investigate the biological activities of Algerian algae, Sargassum vulgare, Cladostephus hirsutus and Rissoella verruculosa. Antimicrobial activity of the crude extracts and their fractions was assessed using the disc diffusion assay, the minimum inhibitory concentration and the minimum bactericidal concentration. Antiparasitic activity was studied in vitro against the blood stream forms of Trypanosoma brucei brucei and the intraerythrocytic stages of Plasmodium falciparum. Ethyl acetate (EA) fractions of the three tested algae showed more potent antimicrobial activity against S. aureus (7–14.5 mm) and B. cereus (7–10.75 mm), MIC values ranged from 0.9375 to 7.5 mg mL?1 and MBC values > 15 mg mL?1. Concerning the antiparasitic activity, EA factions of S. vulgare (IC50 = 9.3 μg mL?1) and R. verruculosa (IC50 = 11.0 μg mL?1) were found to be more effective against T. brucei brucei, whereas the three EA fractions were little active against P. falciparum.  相似文献

Cytokinins are phytohormones that play multiple roles to control plant growth and development. In this study, leaf biomass and the production of andrographolide compounds in a medicinal plant Andrographis paniculata were significantly increased after exogenously treating with the synthetic cytokinin cytokinin-1-(2-chloro-4-pyridyl)-3-phenylurea (CPPU) at 0 (water), 5, or 10 mg L?1 and observed the results for 24 h, 48 h, and 7 days of treatment. It was found that CPPU could significantly enhance new axillary bud formation and further promote branching 4.6–5.6-fold higher, resulting in higher fresh weight (FW) and dry weight (DW) than the control. Application of CPPU at 5 mg L?1 significantly promoted the highest contents of total reducing sugar at 2.5-fold in leaves and at 1.5-fold in roots. Although treatments of CPPU significantly affected the increasing contents of chlorophyll and carotenoid (1.2–1.6-fold), CPPU at 10 mg L?1 slightly caused leaf stress and chlorophyll reduction. Interestingly, 5 mg L?1 CPPU could enhance andrographolide content, an active anti-infectious compound in Andrographis paniculata (2.2-fold higher than the control) that reached the highest content at 24 h after treatment. This study suggested that CPPU should be suitable for field application to promote leaf yields and induce the production of useful pharmaceutical compounds in Andrographis paniculata.  相似文献

The mucilage in Lepidium sativum L. is considered a biologically active compound with diverse medicinal properties. Different explants (hypocotyls and leaf) were transferred to Murashige and Skoog (MS) medium supplemented with twelve different plant growth regulator combinations under two different incubations (light and dark). The best mucilage production from callus (36.76% g g?1 dry weight) was obtained in the MS medium supplemented with 1 mg L?1 of 2, 4-D and 2 mg L?1 of BAP under the light condition. The mucilage produced by callus culture was nearly three times more than the mucilage yield of the seeds. The glucose, arabinose + mannose and galactose were 43.4 (mg g?1 DW), 195.3 (mg g?1 DW) and 86.2 (mg g?1 DW) in the mucilage originated from seed, callus leaf and callus hypocotyl, respectively. The present study proposes an efficient method for producing large scales of mucilage with a favorable sugar aimed at food or pharmaceutical industries.  相似文献

This study evaluates the antidepressant-like effect and analysed the qualitative and quantitative 74 phenolic standards of ethyl acetate fraction from Tabernaemontana catharinensis leaves. Acute administration of fraction in mice reduced the immobility time in forced swimming and tail suspension tests confirming its antidepressant-like activity. The anti-immobility effect elicited by this fraction was prevented by the pretreatment of mice with PCPA (100 mg kg?1), ketanserin (5 mg kg?1), SCH 23,390 (0.05 mg kg?1) or yohimbine (1 mg kg?1). A sub effective dose of the fraction produced a synergistic effect with fluoxetine (5 mg kg?1). Chromatographic analysis identified 4-hydroxybenzoic and p-coumaric acids in the ethyl acetate fraction from T. catharinensis. Capillary electrophoresis presented 7.34 ± 0.02 mg g?1 of p-coumaric acid concentration in the fraction. Therefore, it is possible that antidepressant-like effect elicited by ethyl acetate fraction from T. catharinensis be dependent on the p-coumaric acid.  相似文献

This work aimed to further investigate the phenolic profiles and antioxidant capacities of the crude extracts and the subsequent fractions of Potentilla fruticosa leaves. Result showed that P. fruticosa leaves contained high amounts for hyperoside, ellagic acid and (+)-catechin contents, and the highest amount being registered for hyperoside (17.67 mg g?1). Nine sub-fractions were obtained after column chromatographic separation. EF-3, EF-4, EF-5 and BF-2 presented higher values for their total phenolic or flavonoid, (+)-catechin, ellagic acid and hyperoside content. Besides, EF-3, EF-4, BF-2 and BF-3 showed significant in vitro antioxidant capacities and protective effects on Escherichia coli under peroxide stress. The correlation between chromatograms and antioxidant activity showed that (+)-catechin, ellagic acid and hyperoside may play crucial roles in the antioxidant capacities of P. fruticosa and could be used as chemical markers for its quality assessment. Moreover, this is the first time P. fruticosa leaves have been systematically studied.  相似文献
