The present work describes the individual, selective and simultaneous quantification of acetaminophen (ACP) and tramadol hydrochloride (TRA) using a modification‐free boron‐doped diamond (BDD) electrode. Cyclic voltammetric measurements revealed that the profile of the binary mixtures of ACP and TRA were manifested by two irreversible oxidation peaks at about +1.04 V (for ACP) and +1.61 V (for TRA) in Britton‐Robinson (BR) buffer pH 3.0. TRA oxidation peak was significantly improved in the presence of anionic surfactant, sodium dodecyl sulfate (SDS), while ACP signal did not change. By employing square‐wave stripping mode in BR buffer pH 3.0 containing 8×10?4 mol L?1 SDS after 30 s accumulation under open‐circuit voltage, the BDD electrode could be used for quantification of ACP and TRA simultaneously in the ranges 1.0–70 μg mL?1 (6.6×10?6–4.6×10?4 mol L?1) and 1.0–70 μg mL?1 (3.3×10?6–2.3×10?4 mol L?1), with detection limits of 0.11 μg mL?1 (7.3×10?7 mol L?1) and 0.13 μg mL?1 (4.3×10?7 mol L?1), respectively. The practical applicability of the proposed approach was tested for the individual and simultaneous quantification of ACP and/or TRA in the pharmaceutical dosage forms.  相似文献

A new reagent system has been reported for the extractive separation and simultaneous spectrophotometric de termination of vanadium (V). The method is based on the formation of a water in soluble blue‐violet V(V) complex with N‐hydroxy‐N‐m‐tolyl‐N′‐phenylbenzamidine (HTPBA), and neutral surfactant Triton X‐100 into chloroform over an acidity range of 1.0–10.0 M acetic acid. The complex shows a broad absorption maximum at 570 nm, when measured against a chloroform blank. The λmax (570 nm) of the complex and that of re agent (313 nm) are well separated, hence the excess of the reagent does not interfere in the spectrophotometric de termination of the metal. The molar absorptivity (?) of the complex is (4.74) × 103 1 mol?1 cm?1. The linearity of the calibration curve is followed between 0.5–12.0 μg mL?1 with slope, intercept and correlation coefficient of 9.16× 10?2, 4.5 × 10?3 and 0.999, respectively. The detection limit of the method is 45 μgl?1. The proposed re agent system provides significantly higher tolerance limit for iron (500 μg mL?1) and also various metalions commonly associated with vanadium did not interfere. The method was applied for the deter mi nation of vanadium content of three samples i.e. Spirogyra, Puccinia and Riccia.  相似文献

A quick and inexpensive validated method, based on sample treatment by liquid–liquid microextraction followed by liquid chromatography (LC) coupled with ultraviolet tandem fluorescence detection is proposed for the determination of 15 multiclass pollutants both in serum and in saliva, as a simple and easy to draw matrix. The method was set up and validated according to European guidelines. The compounds of interest include some endocrine‐disrupting chemicals (i.e. bisphenol A, bisphenol B, bisphenol E, bisphenol F, bisphenol AF, bisphenol A diglycidyl ether, bisphenol M, diethylhexyl phthalate, monoethylhexyl phthalate, triclosan and 4‐nonylphenol), as well as other pollutants belonging to the class of volatile organic compounds (2‐chlorophenol, 1,2 dichlorobenzene, 1,2,4,5‐tetrachlorobenzene). The limits of quantifications ranged from 2.28 × 10?3 μg mL?1 (bisphenol A diglycidyl ether) to 6.29 μg mL?1 (diethylhexyl phthalate), while those of detection ranged from 0.068 × 10?3 μg mL?1 (bisphenol A diglycidyl ether) to 1.031 μg mL?1 (diethylhexyl phthalate). To test method suitability, it was applied to real saliva and serum samples of healthy human volunteers and was found to meet the demands of the laboratories handling simple and relatively inexpensive equipment for screening oriented at rapid and reliable contamination assessment of a population.  相似文献

Piroxicam is a drug with analgesic and anti‐inflammatory properties. It is present in numerous pharmaceutical preparations. Injectable forms usually contain benzyl alcohol as an excipient, which is used as a blocking anesthetic (4%) and an antiseptic (4–10%). In this work, spectrophotometric methodology was used in order to determine benzyl alcohol in piroxicam injectable formulations by applying the fourth derivative method adopting the zero‐crossing technique. The results obtained show that the method has significant advantages over other reported methods and is appropriate for routine pharmaceutical analysis. The method showed excellent linearity in the range of 2–100 μg mL?1 with limit of detection (S/N = 3) 0.07 μg mL?1 (6.47 × 10?7 M). The proposed method could be applied successfully for the determination of benzyl alcohol in injectable formulations with average % recovery of 100 ± 0.61.  相似文献

A kinetic spectrophotometric method has been described for the determination of metoprolol tartrate in commercial dosage forms. The procedure is based on the reaction of the drug with 1‐chloro‐2, 4‐dinitrobenzene (CDNB) in dimethylsulfoxide (DMSO) at 100 ± 1 °C. The reaction is investigated by measuring the change in absorbance with time at 420 nm. Fixed‐time (ΔA) and equilibrium methods are chosen for obtaining the calibration curves. Both calibration curves were found to be linear over the concentration range of 5‐60 μg mL?1. The regression analysis of calibration data resulted in the linear regression equations of ΔA = ?1.608 × 10?4 + 3.96 × 10?3 C and A = 7.31 × 10?4 + 1.90 × 10?2 C for fixed time (ΔA) and equilibrium methods, respectively. The limit of detection (LOD) for fixed time and equilibrium methods are 1.16 and 0.415 μg mL?1, respectively. The method has been successfully applied to the quantitation of metoprolol tartrate in commercial dosage forms. Statistical comparison of the results shows that there is no significant difference between the proposed methods and El‐Ries's spectrophotometric method.  相似文献

To the determination of trace amount of Cd(II) present in food and water samples, a selective and extractive spectrophotometric method were developed with 2,6‐diacetylpyridine‐bis‐4‐phenyl‐3‐thiosemicarbazone as a complexing agent. The yellowish orange colored metal complex, Cd(II)‐2,6‐DAPBPTSC with 1:1 (M:L) composition was extracted in to cyclohexanol at pH 9.5 and was shows maximum absorbance at λmax 390 nm. This method obeys Beer's law in the range of 1.12‐11.25 ppm with 0.972 correlation coefficient of Cd(II)‐2,6‐DAPBPTSC complex, which is indicates linearity between the two variables. The molar absorptivity and sandell's sensitivity were found to be 6.088 × 104 L mol?1 cm?1 and 0.0018 μg cm?2, respectively. The instability constant calculated from Asmus' method (1.447 × 10?4)at room temperature. The precision and accuracy of the method were checked by relative standard deviation (n = 5), 0.929 and its detection limit, 0.0060 μg mL?1. The interfering effects of various cations and anions were also studied. The proposed method was successfully applied to the determination of Cd(II) in foods and water samples, and was evaluated its performance in terms of Student ‘t’ test and Variance ‘f’ test, which indicates the significance of present method. The inter comparison of the experimental values, using atomic absorption spectrometer (AAS), was also repoted.  相似文献

The simultaneous voltammetric determination of melatonin (MT) and pyridoxine (PY) has been carried out at a cathodically pretreated boron‐doped diamond electrode. By using cyclic voltammetry, a separation of the oxidation peak potentials of both compounds present in mixture was about 0.47 V in Britton‐Robinson buffer, pH 2. The results obtained by square‐wave voltammetry allowed a method to be developed for determination of MT and PY simultaneously in the ranges 1–100 μg mL−1 (4.3×10−6–4.3×10−4 mol L−1) and 10–175 μg mL−1 (4.9×10−5–8.5×10−4 mol L−1), with detection limits of 0.14 μg mL−1 (6.0×10−7 mol L−1) and 1.35 μg mL−1 (6.6×10−6 mol L−1), respectively. The proposed method was successfully to the dietary supplements samples containing these compounds for health‐caring purposes.  相似文献

In the present study, a simple, cheap and sensitive electrochemical method based on a cathodically pretreated boron-doped diamond (CPT-BDD) electrode is described for the detection of triclosan with the cationic surfactant (cetyltrimethylammonium bromide, CTAB) media. The oxidation of triclosan was irreversible and exhibited an adsorption controlled process. The sensitivity of the adsorptive stripping voltammetric measurements was significantly improved with addition of CTAB. Using square-wave stripping mode, a linear response was obtained for triclosan determination in Britton-Robinson buffer solution at pH 9.0 containing 2.5 × 10?4 M CTAB at around + 0.67 V (vs. Ag/AgCl) (after 30 s accumulation at open-circuit condition). The method could be used in the range of 0.01–1.0 μg mL?1 (3.5 × 10?8–3.5 × 10?6 M), with a detection limit of 0.0023 μg mL?1 (7.9 × 10?9 M). The feasibility of the proposed method for the determination of triclosan in water samples was checked in spiked tap water.  相似文献

Abstract

A rapid and sensitive flow‐injection chemiluminescence (FI‐CL) method, which is based on the CL intensity that generated from the redox reaction of Ce(IV)‐rhodamine B in H2SO4 medium, for the determination of acyclovir and gancyclovir is described. For acyclovir, the determination range is 3×10?8 g mL?1–7×10?5 g mL?1, with 1.56×10?8 g mL?1 as its determination limit. During 11 repeated measurements for 1×10?6 g mL?1 acyclovir, the relative standard deviation was 2.08%. For gancyclovir, the determination range was 5×10?8 g mL?1–7×10?5 g mL?1, with 2.35×10?8 g mL?1 as its determination limit. The relative standard deviation is 2.83% with 11 repeated measurements of 1×10?6 g mL?1 gancyclovir. This method can be successfully used to determine the content of acyclovir and gancyclovir in injections, acyclovir in eye drops, and, maybe, also for other ciclovirs.  相似文献

A highly sensitive automated sequential‐injection chemiluminescence (SIA‐CL) method for determination of glucosamine sulphate (GLS) was developed. The goal of the present work is the evaluation of the enhancement effect of the investigated drug glucosamine sulphate on the chemiluminescence reaction between luminol and H2O2 in alkaline medium of 1.0 × 10?2 mol L?1 sodium hydroxide at pH 11. The experimental conditions affecting the CL reaction such as the sequence of the reagents, concentrations, flow rate and aspirated volumes of reactants were systematically investigated and optimized. Under optimum conditions 50 μL of 1.0 × 10?3 mol L?1 luminol, 30 μL of a GLS test solution and 50 μL of 1.0 × 10?2 mol L?1 H2O2 were used and the luminescing zone was pushed into the detector at a flow rate 100 μL s?1. The proposed method recorded high sensitivity, accuracy and simplicity that could be clarified as linear concentration range 1.0‐2000 ng mL?1 with rectilinear part (r = 0.9992, n = 9) and limit of detection 0.3 ng mL?1, along with relative standard deviation 1.3%. It was found that the developed method can be used directly to determine the investigated drug GLS in its pharmaceutical dosage forms and in spiked serum and urine by diluting the samples for a 1000 fold. The obtained results were statistically analyzed and compared with those obtained by the reported method.  相似文献

Coated‐wire (CW) and tubular (Tu) type membrane sensors for creatinine are developed. These consist of creatinine tungstophosphate(CTP), creatinine molybdophosphate (CMP) and creatinine picrolonate (CPC) ion‐pair complexes as electroactive materials dispersed in plasticized poly(vinyl chloride) matrix membranes. Electrochemical evaluation of these sensors under static (batch) mode of operation reveals near‐Nernstian response with slopes of 62.9, 58.1, and 55.2 mV decade?1 over the concentration range 1×10?2–5.0×10?6, 1×10?2–7.5×10?5, and 1×10?2?3.1×10?5 mol L?1. The lower detection limits are 0.39, 3.49, and 2.20 μg mL?1 creatinine with CTP, CMP and CPC membrane based sensors plasticized with o‐NPOE, respectively. Tubular and coated wire CTP membrane sensors are incorporated in flow‐through cells and used as detectors for flow injection analysis (FIA) of creatinine. The intrinsic characteristics of the detectors under hydrodynamic mode of operation in a low dispersion manifold are determined and compared with data obtained under static mode of operation. With 10?2 mol L?1 phosphate buffer of pH 4.5 as a carrier solution, the tubular and coated wire CTP detectors exhibit rapid response of 58.9 and 50.7 mV decade?1 over the concentration range 1×10?2–1×10?5 mol L?1 and detection limits of 0.39 μg mL?1 and 0.85 μg mL?1, respectively. Validation of the assay methods with the proposed sensors by measuring the lower detection limit, range, accuracy, precision, repeatability and between‐day‐variability reveals good performance characteristics confirming applicability for continuous determination of creatinine. The sensors are used for determining creatinine in human blood serum at an input rate of 40 samples per hour. No interferences are caused by creatine, most common anions, cations and organic species normally present in biological fluids. The results favorably compare with data obtained using the standard spectrophotometric method.  相似文献

A validated, selective and sensitive spectrophotometric method has been developed for the determination of labetalol hydrochloride in commercial dosage forms. The method is based on the coupling reaction of positive diazonium ion of 4‐aminobenzenesulfonic acid with phenolate ion of labetalol to form a colored azo compound which absorbs maximally at 395 nm. Under the optimized experimental conditions, the color is stable up to 2 h and Beer's law is obeyed in the concentration range of 0.8–17.6 μg mL?1 with a linear regression equation of A = 4.84 × 10?4 + 7.864 × 10?2 C and coefficient of correlation, r = 0.9999. The molar absorptivity and Sandell's sensitivity are found to be 2.874 × 104 L mol?1 cm?1 and 0.013 μg cm?2 per 0.001‐absorbance unit, respectively. The limits of detection and quantitation of the proposed method are 0.08 and 0.23 μg mL?1, respectively. The intra‐day and inter‐day precision variation and accuracy of the proposed method is acceptable with low values of standard analytical error. The recovery results obtained by the proposed method in drug formulations are acceptable with mean percent recovery ± RSD of 99.97 ± 0.52 ‐ 100.03 ± 0.63%. The results of the proposed method compared with those of Bilal's spectrophotometric method indicated excellent agreement with acceptable true bias of all samples within ± 2.0%.  相似文献

A selective and sensitive derivative method has been proposed for the simultaneous determination of trace amounts of Co(II) and Ni(II) with morpholinedithiocarbamate (MDTC) in the presence of sodium lauryl sulphate (SLS). The molar absorption coefficients of the 1:2 complex of Co(II) and Ni(II) at 326 nm and 322 nm are 2.248 × 104 and 2.505 × 104 L mol?1 cm?1 for zero order. The analytical sensitivity for the second derivative of Co(II) and Ni(II) complexes are 0.0044 μg mL?1 and 0.0060 μg mL?1. The developed derivative procedure, using the zero‐crossing technique, has been successfully applied for the analysis of Co(II) and Ni(II) simultaneously in different alloy samples.  相似文献

Cobalt (II) phthalocyanine tetracarboxylate [Co (II)Pc-COOH] has been prepared and used in aqueous solutions as a novel chromogenic reagent for the spectrophotometric determination of cyanide ion. The method is based on measuring the increase in the intensity of the monomer peak in the reagent absorbance at 682 nm due to the formation of a 1 : 2 [Co (II)Pc-COOH] : [CN] complex. The complex exhibits a molar absorptivity (ε) of 7.7 × 104 L mol?1 cm?1 and a formation constant (Kf ) of 5.4 ± 0.01 × 106 at 25 ± 0.1°C. Beer's law is obeyed over the concentration range 0.15–15 µg mL?1 (5.8 × 10?6–5.8 × 10?4 M) of cyanide ion, the detection limit is 20 ng mL?1 (7.7 × 10?7 M) the relative standard deviation is ±0.7% (n = 6) and the method accuracy is 98.6 ± 0.9%. Interference by most common ions is negligible, except that by sulphite. The proposed method is used for determining cyanide concentration in gold, silver and chromium electroplating wastewater bath solutions after a prior distillation with 1 : 1 H2SO4 and collection of the volatile cyanide in 1 M NaOH solution containing lead carbonate as recommended by ASTM, USEPA, ISO and APAHE separation procedures. The results agree fairly well with potentiometric data obtained using the solid state cyanide ion selective electrode.  相似文献

We report an innovative supramolecular architecture for bienzymatic glucose biosensing based on the non‐covalently functionalization of multi‐walled carbon nanotubes (MWCNTs) with two proteins, glucose oxidase (GOx) (to recognize glucose) and avidin (to allow the specific anchoring of biotinylated horseradish peroxidase (b‐HRP)). The optimum functionalization was obtained by sonicating for 10 min 0.50 mg mL?1 MWCNTs in a solution of 2.00 mg mL?1 GOx+1.00 mg mL?1avidin prepared in 50 : 50 v/v ethanol/water. The sensitivity to glucose for glassy carbon electrodes (GCE) modified with MWCNTs‐GOx‐avidin dispersion and b‐HRP (GCE/MWCNTs‐GOx‐avidin/b‐HRP), obtained from amperometric experiments performed at ?0.100 V in the presence of 5.0×10?4 M hydroquinone, was (4.8±0.3) μA mM?1 (r2=0.9986) and the detection limit was 1.2 μM. The reproducibility for 5 electrodes using the same MWCNTs/GOx‐avidin dispersion was 4.0 %, while the reproducibility for 3 different dispersions and 9 electrodes was 6.0 %. The GCE/MWCNT‐GOx‐avidin/b‐HRP was successfully used for the quantification of glucose in a pharmaceutical product and milk.  相似文献

A validated kinetic spectrophotometric method has been developed for the determination of losartan potassium in pure and dosage forms. The method is based on oxidation of the losartan potassium with alkaline potassium permanganate at room temperature (25 ± 1 °C). The reaction is followed spectrophotometrically by measuring the increase in absorbance with time at 603 nm, and the initial rate, fixed time (at 12.0 min) and equilibrium time (at 90.0 min) methods are adopted for constructing the calibration graphs. All the calibration graphs are linear in the concentration range of 7.5–60.0 μg mL?1 and the calibration data resulted in the linear regression equations of n? = ?6.422 × 10?7 + 1.173 × 10?5 C, A =3.30 × 10?4 + 5.28 × 10?3 C and A = ?2.09 × 10?2 + 1.05 × 10?1 C for initial‐rate, fixed time and equilibrium time methods, respectively. The limits of detection for initial rate, fixed time and equilibrium time methods are 0.71, 0.21 and 0.19 μg mL?1, respectively. The activation parameters such as Ea, ΔH?, ΔS?, and ΔG? are also determined for the reaction and found to be 87.34 KJ mol?1, 84.86 KJ mol?1, 50.96 JK?1 mol?1 and ?15.10 KJ mol?1, respectively. The variables are optimized and the proposed methods are validated as per ICH guidelines. The method has been applied successfully to the estimation of losartan potassium in commercial tablets. The performance of the proposed methods was judged by calculating paired t‐ and F‐ values. The analytical results of the proposed methods when compared with those of the reference method show no significant difference in accuracy and precision and have acceptable bias.  相似文献

Two simple and sensitive extractive spectrophotometric methods have been described for the analysis of clarithromycin in pure form and in pharmaceutical formulations. The methods involved formation of yellow colored chloroform extractable ion‐association complexes of clarithromycin with bromothymol blue (BTB) and cresol red (CR) in buffered aqueous solution at pH 4. The extracted complexes showed maximum absorbance at 410 and 415 nm for BTB and CR, respectively. Beer's law is obeyed in the concentration ranges 0.1–20 μg mL?1 and 2.0–20 μg mL?1 of clarithromycin with molar absorptivity of 2.01 × 104 and 4.378 × 103 for BTB and CR, respectively. The composition ratio of the ion‐association complex was clarithromycin: BTB and CR = 1:1 as established by Job's method. The methods have been applied to the determination of drug in commercial formulations. The results of analysis were validated statistically and through recovery studies.  相似文献

A novel and simple spectrophotometric method for the determination of phenformin hydrochloride using sodium nitroprusside as chromogenic reagent is developed in this paper. The experiment indicates that the sodium nitroprusside can react with the phenformin hydrochloride in a basic solution to form a product with colored N‐nitrosamines. The stoichiometric ratio of phenformin hydrochloride and sodium nitroprusside is 1:3 in the product. The maximal absorption wavelength (λmax) of the product is 520 nm, ?520 = 2.2 × 103 Lmol?1cm?1. A Good linear relationship is obtained between the absorbance (A) and the concentration (C) of phenformin hydrochloride in a wide range of 0.20‐400 μg mL?1. The equation of the linear regression is A = 0.03461 + 0.00907C (μg mL?1) with a linear correlation coefficient of 0.9991. The detection limit is 0.13 μg mL?1, and the relative standard deviation (R.S.D.) is 0.19%. The method is successfully applied to the determination of phenformin hydrochloride in tablet forms, and satisfactory recoveries are obtained.  相似文献

Two simple, sensitive and economical spectrophotometric methods have been developed for the determination of esomeprazole magnesium in commercial dosage forms. Method A is based on the reaction of esomeprazole magnesium with 5‐sulfosalicylic acid in methanol to form a yellow product, which absorbs maximally at 365 nm. Method B utilizes the reaction of esomeprazole magnesium with N‐bromosuccinimide in acetone‐chloroform medium to form α‐bromo derivative of the drug peaking at 380 nm. Under the optimized experimental conditions, Beer's law is obeyed in the concentration ranges of 2‐48 and 10‐100 μg mL?1 with molar absorptivity of 2.11 × 104 and 4.57 × 104L mol?1 cm?1 for methods A and B, respectively. The limits of detection for methods A and B are 0.35 and 0.46 μg mL?1, respectively. No interference was observed from excipients commonly present in tablet formulations. Methods A and B are successfully applied to the commercial tablets for the estimation of esomeprazole magnesium with good accuracy and precision. The results compare favorably with the reference spectrophotometric method indicating no significant difference between the methods compared.  相似文献

The main aim of this work is to develop and validate a spectrophotometric method for the determination of nicorandil in commercial tablets. The method is based on the reduction of the nitroxy ethyl group of nicorandil into carbonyl compound and nitrite ion by NH4Cl and Zn dust. The nitrite ion thus formed reacts with potassium iodide and starch in dilute HCl medium to form a blue product, which absorbs maximally at 550 nm. Beer's law is obeyed in the concentration range 0.4‐4.0 μg mL?1 with molar absorptivity of 7.92 × 104L mol?1 cm?1. The detection limit is 0.017 μg mL?1. The reaction conditions are optimized and validated as per the International Conference on Harmonisation guidelines (USA). The proposed method has been applied successfully for the determination of nicorandil in commercial tablets. The results of analyses are compared statistically with those of the author's spectrophotometric method, which confirmed that there is no significant difference between the methods compared.  相似文献
