Monoester‐diterpenoid alkaloids are the main bioactive components of Sini decoction, which is a well‐known traditional Chinese medicine formula for the treatment of myocardial infarction (MI) and heart failure in China. In this work, an ultra‐high‐performance liquid chromatography–mass spectrometry combined with microdialysis method was successfully established and applied for investigating for the first time comparative plasma pharmacokinetics of three monoester‐diterpenoid alkaloids (benzoylmesaconitine, benzoylaconitine and benzoylhypacoitine) in normal and MI rats after oral administration of Sini decoction. The statistical results of pharmacokinetic parameters demonstrated that benzoylmesaconitine, benzoylaconitine and benzoylhypacoitine showed lower peak concentration, longer half‐life, smaller area under the concentration–time curve, slower clearance, time to peak concentration and mean residence time in MI rats than in normal rats (p < 0.05), which indicated that monoester‐diterpenoid alkaloids exhibited lower systemic exposure and slower elimination in the MI rats. The results provided the experimental basis for understanding the metabolic fate and therapeutic effects of Sini decoction.  相似文献

Phytolacca acinosa is an herb for treatment of ascites and tumor. Two forms of P. acinosa, i.e. raw and vinegar‐processed herb, have been used in clinic. However, pharmacokinetic difference between the two forms of P. acinosa has not been fully understood. Herein, a comparative pharmacokinetic method based on liquid chromatography with tandem mass spectrometry was developed for quantification of six bioactive triterpenoids, including esculentoside H, esculentoside T, esculentoside A, esculentoside B, phytolaccagenic acid, and phytolaccagenin in rat plasma after oral administration of different forms of P. acinosa. Separation was performed on an Acquity BEH C18 column (1.7 µm, 2.1 mm × 50 mm). The method was validated over a linear range of 2.0–5000 ng/mL. Intraday and interday bias were within ±5%. Besides, all triterpenoids were stable in plasma during different storage conditions. The described method was successfully applied to a comparative pharmacokinetic study of raw and vinegar‐processed P. acinosa in rats. Notably, double peak phenomenon for six triterpenoids of P. acinosa was observed for the first time. AUC0→t and Cmax values of esculentoside H, esculentoside T, phytolaccagenic acid, and phytolaccagenin were significantly lower in vinegar‐processed group than that of raw group, indicating the oral bioavailability of the four triterpenoids was decreased after vinegar processing.  相似文献

Shaoyao‐Gancao decoction, a Chinese herbal formula, is composed of Paeoniae Radix alba and Glycyrrhiza Radix et rhizoma . It has been widely used to treat muscle spasms and asthma. However, little is known about the bioactive components of Shaoyao‐Gancao decoction. In the present study, the bioactive compounds in water‐extract of Shaoyao‐Gancao decoction were separated by the immobilized β2‐adrenoceptor affinity column and identified using quadrupole time‐of‐flight mass spectrometry. The affinity constants of the separated compounds that bind to β2‐adrenoceptor were determined by frontal analysis. Compound bioactivity was tested in a rat tracheal smooth muscle relaxation assay. We identified the bioactive compounds in the water extract of Shaoyao‐Gancao decoction that bound to the β2‐adrenoceptor as paeoniflorin and liquiritin. Paeoniflorin and liquiritin had only one binding site on the immobilized β2‐adrenoceptor, and the affinity constants were (2.16 ± 0.10) × 104 M−1 and (2.95 ± 0.15) × 104 M−1, respectively. Both compounds induced a concentration‐dependent relaxation of tracheal smooth muscle following K+‐stimulated contraction, and the relaxation effects were abrogated by the β2‐adrenoceptor antagonist, ICI 118551. Therefore, paeoniflorin and liquiritin are bioactive compounds in Shaoyao‐Gancao decoction and the β2‐adrenoceptor affinity chromatography is a useful tool for identifying potential β2‐adrenoceptor ligands in natural products used in traditional Chinese medicine.  相似文献

A rapid and sensitive ultra high performance liquid chromatography with tandem mass spectrometry method was established and validated for simultaneous determination of thirteen bioactive components (gallic acid, protocatechuic acid, puerarin, p‐hydroxycinnamic acid, daidzin, ononin, daidzein, naringenin, genistein, apigenin, formononetin, biochanin A, and β‐sitosterol) of Radix Puerariae extract in rat plasma and tissues. The plasma and tissues samples were pretreated by protein precipitation extraction, and umbelliferone and rutin were used as internal standards. Sample separation was performed on a ZORBAX RRHD Eclipse plus C18 column (2.1 mm × 50 mm, 1.8 µm, Agilent) with a mobile phase consisting of methanol–water (containing 0.1% formic acid). The mass spectrometry analysis was conducted in positive and negative ionization modes with multiple reaction monitoring. The lower limit of quantitation range for the 13 analytes was 0.2?35 ng/mL. The intra‐ and inter‐day precision of all the analytes were less than 10.92%, with an accuracy ranging from ?13.10 to 11.96%. Both the recovery and matrix effect were within acceptable limits. This method was successfully applied to pharmacokinetic and tissue distribution study of the 13 bioactive components in rats after oral administration of R. Puerariae extract.  相似文献

Many foods and herbs are experientially classified into different commodity grades in commercial circulation. Regarding the hypertoxic herb aconite, large samples are considered to be of better quality. However, this experiential classification lacks a scientific basis. In this study, we focused on the quality diversity among different grades and studied it using the minimum lethal dose assay and a novel ultra high performance liquid chromatography coupled with time‐of‐flight mass spectrometry method. Toxicity assay result suggested grade I aconite had the lowest toxicity (p < 0.05). Using this method with partial least squares‐discriminant analysis, we discovered nine chemomarkers, including neoline, songorine, fuziline, mesaconitine, talatizidine, dexyaconitine, talatisamine, hypaconitine, and fuzitine. Considering their toxicity and activity, we found the levels of toxic ingredients hypaconitine, dexyaconitine, and mesaconitine in grade I were lower than those in grade II (p < 0.01), while the levels of efficacy ingredients songorine, talatisamine, and neoline were the highest in grade I (p < 0.01). Further study demonstrated that the quality variation was associated with plant tissue development and toxic ingredient distribution law. Our results provide scientific evidence for the experiential quality evaluation of aconite, and it will be of great utility for other foods and herbs.  相似文献

Acotoxinine ( 1 ), a new norditerpene alkaloid, was isolated from the roots of Aconitum toxicum Rchb ., together with the structurally related C19 diterpene alkaloids neoline ( 2 ) and aconitine ( 3 ) and C20 diterpene alkaloids songorine ( 4 ) and songoramine ( 5 ). The structures were elucidated by HR‐MS and advanced NMR methods, including 1H‐NMR, JMOD, 1H,1H‐COSY, HSQC, and HMBC experiments. Artemisia annua L. (A. annua) has been used as herbal medicine in China for thousands of years for clearing deficiency heat, treating malaria and removing jaundice. A rapid, sensitive and specific liquid chromatography coupled with electrospray ionization tandem mass spectrometry (LC–ESI–MS/MS) method was developed, validated, and successfully used for simultaneous quantification of the active components in rat plasma after oral administration of A. annua extract. Molecular docking of each component with drug metabolizing enzymes was carried out to explore the effect of each component on CYP-mediated drug metabolism. Two coumarins (scopolin (SPL) and scopoletin (SPLT)), three flavonoids (rutin (RUT), chrysosplenol D (CHD), casticin (CAS)) and three sesquiterpenes (arteannuin B (ARN), dihydroartemisinic acid (DARM) and artemisinic acid (ARM)) were detected in rat plasma after oral administration. CHD and CAS were rapidly absorbed into rat blood with the Tmax values of 0.11 ± 0.04 h and 0.13 ± 0.05 h, respectively. Their half-lives (t1/2 2.68 ± 3.62 h and 0.33 ± 0.07 h) were shorter. SPLT were also rapidly absorbed into the blood (Tmax 0.15 ± 0.03 h), but exhibited a longer half-life (t1/2 6.53 ± 1.84 h), indicating that it could be effective in vivo for a longer period of time. The peak time of SPL, RUT, DARM and ARM ranged from 1 ~ 4 h, demonstrating that they could maintain considerable concentrations for a longer time. ARN showed strong enterohepatic circulation in rats, leading to slower onset time and longer effect. A few components including SPLT, CHD, CAS and ARN could be metabolized into their corresponding II phase metabolites combining with glucuronic acid or sulfuric acid. RUT could decompose its glycosyl to generate genin. The molecular docking results indicated that those flavonoids and coumarins of A. annua interacting with CYPs mainly through hydrogen bonding and π-π stacking had better CYP450 enzyme binding ability than the sesquiterpenoids, which were easier to induce drug interactions. This study presented an integrated strategy for investigating the pharmacokinetic behaviors of eight components in A. annua and laid the foundation for revealing the mechanism of action of A. annua in the organism.  相似文献

As a main source for the recognition and identification of lead compounds, traditional Chinese medicine plays a pivotal role in preventing diseases for years. However, screening bioactive compounds from traditional Chinese medicine remains challenging because of the complexity of the systems and the occurrence of the synergic effect of the compounds. The infructescence of Platycarya strobilacea Sieb. et Zucc is prescribed for allergic rhinitis treatment with unknown bioactive compounds and unclear mechanisms. Herein, we immobilized the β2-adrenoceptor and muscarine-3 acetylcholine receptor onto the silica gel surface to prepare the stationary phase in a covalent bond through one step. The feasibility of the columns was investigated by the chromatographic method. Ellagic acid and catechin were identified as the bioactive compounds targeting the receptors. The binding constants of ellagic acid were calculated to be (1.56 ± 0.23)×107 M−1 for muscarine-3 acetylcholine receptor and (2.93 ± 0.15)×107 M−1 for β2-adrenoceptor by frontal analysis. While catechin can bind with muscarine-3 acetylcholine receptor with an affinity of (3.21 ± 0.05)×105 M−1. Hydrogen bonds and van der Waals’ force were the main driving forces for the two compounds with the receptors. The established method provides an alternative for multi-target bioactive compound screening in complex matrices.  相似文献

A method of high performance liquid chromatography with a Hamilton PRP‐X100 ion‐exchange column (250 × 4.1 mm id, 10 μm) coupled to inductively coupled plasma mass spectrometry was employed to generate a full concentration–time profile of arsenic speciation after oral administration. The results exhibited good linearity and revealed that, in the pills, the average arsenic concentration was 10105.4 ± 380.7 mg/kg, and in the water extraction solution, the inorganic As(III) and As(V) concentrations were 220.1 ± 12.6 and 45.5 ± 2.3 mg/kg, respectively. No trace of monomethyl arsenic acid was detected in any of the plasma samples. We then successfully applied the established methodology to examine the pharmacokinetics of arsenic speciation. The resulting data revealed that, after oral administration in rats, the plasma concentration of each arsenic species reached Cmax shortly after initial dosing, and that the distribution and elimination of As(V) was faster than that of As(III) and dimethyl arsenic acid. Additionally, the t1/2 values of As(V), As(III), and dimethyl arsenic acid were 3.4 ± 1.6, 14.3 ± 4.0, and 19.9 ± 1.6 h, respectively. This study provides references for the determination of arsenic speciation in mineral‐containing medicines and could serve as a useful tool in measuring the true toxicity in traditional medicines that contain them.  相似文献

Radix Scrophulariae (Xuanshen) is one of the famous Chinese herbal medicines widely used to treat rheumatism, tussis, pharyngalgia, arthritis, constipation, and conjunctival congestion. Harpagoside and cinnamic acid are the main bioactive components of Xuanshen. The purpose of this study was to develop an HPLC–UV method for simultaneous determination of harpagoside and cinnamic acid in rat plasma and investigate pharmacokinetic parameters of harpagoside and cinnamic acid after oral administration of Xuanshen extract (760 mg kg−1). After addition of syringin as internal standard, the analytes were isolated from plasma by liquid–liquid extraction. Separation was achieved on a Kromasil C18 column, and detection was by UV absorption at 272 nm. The described assay was validated in terms of linearity, accuracy, precision, recovery, and limit of quantification according to the FDA validation guidelines. Calibration curves for both analytes were linear with the coefficient of variation (r) for both was greater than 0.999. Accuracy for harpagoside and cinnamic acid ranged from 100.7–103.5% and 96.9–102.9%, respectively, and precision for both analytes were less than 8.5%. The main pharmacokinetic parameters found for harpagoside and cinnamic acid after oral infusion of Xuanshen extract were as follows: C max 1488.7 ± 205.9 and 556.8 ± 94.2 ng mL−1, T max 2.09 ± 0.31 and (1.48 ± 0.14 h, AUC0–24 10336.4 ± 1426.8 and 3653.1 ± 456.4 ng h mL−1, 11276.8 ± 1321.4 and 3704.5 ± 398.8 ng h mL−1, and t 1/2 4.9 ± 1.3 and 2.5 ± 0.9 h, respectively. Qi-Shen-Ke-Li (QSKL), a traditional Chinese formula prepared from six herbs, has long been used for the treatment of coronary heart disease and chronic heart failure. However, the herbal combination mechanism and underlying material basis of this multi-herbal formula are not clear. In this study, an ultra-high-performance liquid chromatography–tandem mass spectrometry (UHPLC–MS/MS) method to simultaneously determine multiple bioactive compounds in QSKL was established and validated. Using the developed method, 18 bioactive components in rat plasma after oral administration of QSKL formula and its single herb extracts were quantified. Based on these results, pharmacokinetic (PK) parameters (T1/2, Tmax, Cmax, AUC0–48h, and AUC0–∞) of the 18 bioactive components were analyzed and compared using PKSlover 2.0 PK software. The experimental data suggested that significant changes in PK profiles were observed between the QSKL formula and its single-herb extracts. The herbal combination in QSKL significantly influences the system exposure and the PK behaviors of the 18 bioactive components, indicating multicomponent interactions among the herbs. This study provides insight into the herbal combination mechanism and underlying material basis of the QSKL formula.  相似文献

The first part of the study assessed the equivalence of practical salinity measurement results of a slightly diluted seawater sample from the North Atlantic, which were traceable to the SI. The study shows that the practical salinity reference value S RV (here S RV = 34.967) can be determined with a relative standard uncertainty of 3×10−4. This quantifies the uncertainty range, in which long-term comparability of practical salinity measurement results can be guaranteed currently. In the second part of the study, eight laboratories determined the mass fractions of five seawater components to quantify the equivalence of these measurement results. The results were: Na+ (10.726 ± 0.134) g/kg, Mg2+ (1.288 ± 0.018) g/kg, Sr2+ (0.00755 ± 0.00011) g/kg, Cl (19.360 ± 0.047) g/kg, SO4 2− (2.650 ± 0.025) g/kg. The investigation confirmed the reference composition mass fractions of sodium, magnesium and chloride ions, but it showed a disagreement for strontium and sulphate ions.  相似文献

The purpose of this study was to evaluate percutaneous penetration and arrhythmogenic effects of aconitine after transdermal administration, compared with the oral route. Skin penetration of aconitine was tested by a microdialysis technique in rats and in vivo recovery was determined by retrodialysis. After oral and transdermal administration of aconitine, dialysate was sampled at 20 min intervals until the end of the experiment for the determination of concentration of aconitine in skin. Blood samples were collected and analyzed using a validated HPLC‐MS/MS method. In addition, we concurrently recorded the electrocardiogram (ECG). The in vivo recovery of aconitine in the skin was calculated to be 39.59%. The Cmax values for aconitine absorbed into the skin after oral and transdermal administration were 1.51 ± 0.53 and 2723.8 ± 848.8 ng/mL, respectively, and within the plasma, 215.86 ± 79.29 and 20.92 ± 3.15 ng/mL. The Cmax value for the plasma concentration of aconitine after oral administration was approximately 10 times higher than with the transdermal route. For oral administration, the ECG revealed various types of arrhythmias at a period of Tmax, which is normal in transdermal gel administration. These results indicate that transdermal aconitine gel is a safe formulation that can deliver the drug in sufficient amounts and safe concentrations to produce therapeutic action in rats. Radiosynthesis of 99mTc-sitafloxacin (99mTc-STF) complex and its efficacy as a potential infection imaging agent was evaluated. Effect of sitafloxacin (STF) concentration, sodium pertechnetate (Na99mTcO4), stannous chloride dihydrate (SnCl2·2H2O), and pH on the % radiochemical purity yield (RCP) of 99mTc-STF complex was studied. A stable 99mTc-STF complex up to 120 min with maximum %RCP yield was observed by mixing 2 mg of STF with 3 mCi of Na99mTcO4 and 150 μL of SnCl2·2H2O (1 μg/μL in 0.01 N HCl) at a pH 5.5. Artificially infected rats with Staphylococcus aureus were used for studying the biodistribution behavior of the 99mTc-STF complex. After 30 min of the intravenous (I.V.) administration of the 99mTc-STF complex, 7.50 ± 0.10% was absorbed in the infected thigh of the rats and the uptake gradually increased to 18.50 ± 0.20% within 90 min. Rabbits with artificially induced infection were used for evaluating the scintigraphic accuracy. Higher uptake in the infected thigh was observed after 2 h of I.V. administration of the 99mTc-STF complex. Target to non-target organ ratio of the % absorbed dose incase of infected/normal muscle was 6.82 ± 0.40, 17.11 ± 0.60, and 23.13 ± 1.00% at 30, 60 and 90 min of administration. Stable and higher %RCP, higher uptake in the infected thigh, and spectral studies, recommend the 99mTc-STF for routine infection imaging.  相似文献

A new diterpene alkaloid, 12‐epi‐15‐O‐acetyl‐17‐benzoyl‐16‐hydroxy‐16,17‐dihydronapelline ( 1 ), along with nine known diterpene alkaloids including yunaconitine ( 2 ), neoline ( 3 ), mesaconitine ( 4 ), beiwutine ( 5 ), chasmanine ( 6 ), songorine ( 7 ), 12‐epi‐napelline ( 8 ), foresticine ( 9 ), and 15α‐hydroxyneoline ( 10 ) were isolated from the roots of Aconitum carmichaeli Debx. An electrospray ionization tandem mass spectrometric (ESI-MS-MS) method has been developed for the determination of cyanide (CN) in blood. Five microliters of blood was hemolyzed with 50 μL of water, then 5 μL of 1 M tetramethylammonium hydroxide solution was added to raise the pH of the hemolysate and to liberate CN from methemoglobin. CN was then reacted with NaAuCl4 to produce dicyanogold, Au(CN)2, that was extracted with 75 μL of methyl isobutyl ketone. Ten microliters of the extract was injected directly into an ESI-MS-MS instrument and quantification of CN was performed by selected reaction monitoring of the product ion CN at m/z 26, derived from the precursor ion Au(CN)2 at m/z 249. CN could be measured in the quantification range of 2.60 to 260 μg/L with the limit of detection at 0.56 μg/L in blood. This method was applied to the analysis of clinical samples and the concentrations of CN in the blood were as follows: 7.13 ± 2.41 μg/L for six healthy non-smokers, 3.08 ± 1.12 μg/L for six CO gas victims, 730 ± 867 μg for 21 house fire victims, and 3,030 ± 97 μg/L for a victim who ingested NaCN. The increase of CN in the blood of a victim who ingested NaN3 was confirmed using MS-MS for the first time, and the concentrations of CN in the blood, gastric content and urine were 78.5 ± 5.5, 11.8 ± 0.5, and 11.4 ± 0.8 μg/L, respectively.  相似文献
