The development of biodegradable inorganic nanoparticles with a tumor microenvironment‐activated therapeutic mode of action is urgently needed for precision cancer medicine. Herein, the synthesis of ultrathin lanthanide nanoscrolls (Gd2O3 NSs) is reported, which biodegrade upon encountering the tumor microenvironment. The Gd2O3 NSs showed highly controlled magnetic properties, which enabled their high‐resolution magnetic resonance imaging (MRI). Importantly, Gd2O3 NSs degrade in a pH‐responsive manner and selectively penetrate tumor tissue, enabling the targeted release of anti‐cancer drugs. Gd2O3 NSs can be efficiently loaded with an anti‐cancer drug (DOX, 80 %) and significantly inhibit tumor growth with negligible cellular and tissue toxicity both in vitro and in vivo. This study may provide a novel strategy to design tumor microenvironment‐responsive inorganic nanomaterials for biocompatible bioimaging and biodegradation‐enhanced cancer therapy.  相似文献

Various lanthanide oxides (Sm2O3 and Gd2O3) nanostructures were synthesized by a facile hydrothermal method. The loss of surfactants on the nanocrystals surface, followed by the resultant assembly is responsible for the formation of ultrathin nanosheets. Owing to strong surface effects, the different morphologies of the Sm2O3:5 % Eu and Gd2O3:5 % Eu nanocrystals present unique photoluminescence properties. As a proof‐of‐concept application, the as‐obtained Sm2O3 and Gd2O3 ultrathin nanosheets exhibit promising pH‐controlled anticancer drug‐delivery behavior.  相似文献

Activatable photosensitizers (PSs) have been widely used for the simultaneous fluorescence imaging and photodynamic ablation of cancer cells. However, the ready aggregation of traditional PSs in aqueous media can lead to fluorescence quenching as well as reduced phototoxicity even in the activated form. We have developed a series of PSs that show aggregation‐enhanced emission and phototoxicity and thus the exact opposite behavior to that of previously reported PSs. We further developed a dual‐targeted enzyme‐activatable bioprobe based on the optimized photosensitizer and describe simultaneous light‐up fluorescence imaging and activated photodynamic therapy for specific cancer cells. The design of smart probes should thus open new opportunities for targeted and image‐guided photodynamic therapy.  相似文献

RNA interference (RNAi) gene silencing technologies have shown significant potential for treating various diseases, including cancer. However, clinical success in cancer therapy remains elusive, mainly owing to suboptimal in vivo delivery of RNAi therapeutics such as small interference RNA (siRNA) to tumors. Herein, we developed a library of polymers that respond to a narrow pH change (ultra‐pH‐responsive), and demonstrated the utility of these materials in targeted and deep tumor‐penetrating nanoparticle (NP) for in vivo RNAi. The new NP platform is mainly composed of the following key components: i) internalizing RGD (iRGD) to enhance tumor targeting and tissue penetration; ii) polyethylene glycol (PEG) chains to prolong blood circulation; and iii) sharp pH‐responsive hydrophobic polymer to improve endosome escape. Through systematic studies of structure–function relationship, the optimized RNAi NPs (<70 nm) showed efficient gene silencing and significant inhibition of tumor growth with negligible toxicities in vivo.  相似文献

Understanding the nature of interactions of targeted drug‐delivery vehicles, such as functionalized carbon nanotubes (f‐CNTs) and their composites, with a cell or its organelles or DNA, where water is a major constituent, requires molecular‐level understanding of f‐CNTs with analogous chemical systems. The nature of interaction has not yet been explored within the scope of formation of giant aggregates by self‐assembly processes. Crystals of platinum(II) dithiolene [Pt(mnt)2][PPh4]2 ( 1 ) and gadolinium(III) dithiolene [Gd(mnt)3][PPh4]3 ( 2 ) (mnt=maleonitrile dithiolate) form nanospheres (diameter 88 nm) and nanoflowers (400–600 nm) in acetonitrile/water and DMF/water solvent mixtures, respectively. The formation of nanospheres or nanoflowers is proposed to be a water‐induced phenomenon. These nanospheres and nanoflowers interact with f‐CNTs by forming either spherical supramolecular assemblies ( 3 , diameter up to 45. 5 μm) in the case of platinum(II) dithiolene or composite flowers ( 4 ) with CNT buckling for gadolinium(III) dithiolene. Both nanostructures, ( 3 ) and ( 4 ), show emission upon excitation at a range of wavelengths (λex=385–560 nm). The fluorescence emissions of the composite materials 3 and 4 are proposed to be due to separation of energy states of the nanospheres of 1 or the nanoflowers of 2 by the energy states of the f‐CNTs, leading to the possibility of new electronic transitions.  相似文献

A facile strategy is presented to synthesize hyaluronic acid (HA) and a fluorescein isothiocyanate (FITC)‐conjugated mesoporous silica nanocomposite (MSN) with multiple functions of fluorescence, tumor‐cell targeting, pH‐triggered gelation, and enzyme‐responsive drug release. This injectable nanocomposite is able to indicate the entire tumor location and provides a microenvironment with rich anticancer drugs in and around tumor tissue for a long time, to avoid recrudescence. In this design, the mesoporous silica serves as the drug container, the FITC serves as a fluorescent probe, and the anchored HA plays multiple roles as drug‐release cap, tumor‐targeting points, and responsive gel matrix. Owing to the specific affinity between the HA on MSNs and the CD44 antigen over‐expressed on tumor cells, the MSNs can selectively attach to tumor cells. The nanocomposites then exploit the pH‐responsive interactions (hydrogen bonds) among the HA to self‐assemble in situ into a hydrogel around the tumor tissue. The resulting hydrogel gradually releases its payload (doxorubicin, anticancer drugs)‐loaded MSNs upon HA degradation in the presence of hyaluronidase‐1 (Hyal‐1), followed by endocytosis and intracellular drug release. All these properties have distinct benefits for tumor treatment, demonstrating that this device is a promising candidate for oncotherapy applications.

Lanthanide‐doped upconversion nanoparticles (UCNPs) have attracted considerable attention for their application in biomedicine. Here, silica‐coated NaGdF4:Yb,Er/NaGdF4 nanoparticles with a tetrasubstituted carboxy aluminum phthalocyanine (AlC4Pc) photosensitizer covalently incorporated inside the silica shells were prepared and applied in the photodynamic therapy (PDT) and magnetic resonance imaging (MRI) of cancer cells. These UCNP@SiO2(AlC4Pc) nanoparticles were uniform in size, stable against photosensitizer leaching, and highly efficient in photogenerating cytotoxic singlet oxygen under near‐infrared (NIR) light. In vitro studies indicated that these nanoparticles could effectively kill cancer cells upon NIR irradiation. Moreover, the nanoparticles also demonstrated good MR contrast, both in aqueous solution and inside cells. This is the first time that NaGdF4:Yb,Er/NaGdF4 upconversion‐nanocrystal‐based multifunctional nanomaterials have been synthesized and applied in PDT. Our results show that these multifunctional nanoparticles are very promising for applications in versatile imaging diagnosis and as a therapy tool in biomedical engineering.  相似文献

以氧化石墨烯(GO)为原料, 利用温和方法制备了3种不同还原程度的部分还原氧化石墨烯pRGO1, pRGO2和pRGO3(pRGO1—3); 利用傅里叶变换红外光谱(FTIR)、 拉曼光谱(Raman)、 X 射线光电子能谱(XPS)、 紫外-可见光谱(UV-Vis)、 透射电子显微镜(TEM)和 EDS能谱对其结构和形貌进行了表征. 细胞实验结果表明, 无激光照射下pRGO1—3本身的细胞毒性较低; 近红外(NIR)激光照射下pRGO1—3通过光热和光毒性双重作用杀伤肿瘤细胞. 实验结果显示了pRGO 在肿瘤光热疗法和光动力疗法领域的应用潜力.  相似文献

Discriminative detection of invasive and noninvasive breast cancers is crucial for their effective treatment and prognosis. However, activatable probes able to do so in vivo are rare. Herein, we report an activatable polymeric reporter (P‐Dex) that specifically turns on near‐infrared (NIR) fluorescent and photoacoustic (PA) signals in response to the urokinase‐type plasminogen activator (uPA) overexpressed in invasive breast cancer. P‐Dex has a renal‐clearable dextran backbone that is linked with a NIR dye caged with an uPA‐cleavable peptide substrate. Such a molecular design allows P‐Dex to passively target tumors, activate NIR fluorescence and PA signals to effectively distinguish invasive MDA‐MB‐231 breast cancer from noninvasive MCF‐7 breast cancer, and ultimately undergo renal clearance to minimize the toxicity potential. Thus, this polymeric reporter holds great promise for the early detection of malignant breast cancer.  相似文献

The development of stimuli‐responsive polymeric nanocarriers could significantly enhance drug bioavailability due to improved pharmacokinetics and biodistribution. However, in the drug delivery process, the poor cell uptake of drug‐loaded carriers has greatly limited the therapeutic efficiency for anti‐cancer applications. Herein, 2,3‐dimethylmaleic anhydride (DMMA) is engineered into the well‐defined biodegradable amphiphilic block copolymer poly(D,L‐lactide)‐block‐poly(2‐aminoethyl methacrylate) (PLA‐b‐PAEMA) to construct a tumor‐acidity activated nanocarrier (PLA‐b‐PAEMA/DMMA) for potential tumor therapy. After the loading of positively charged DOX·HCl into the negatively charged corona structure through electrostatic attraction, this carrier is expected to prolong the blood circulation time and smartly convert surface charge from negative to positive for enhanced tumor cell uptake and targeted drug release. Furthermore, this carrier exhibits additional cytotoxicity for tumor cells after the tumor‐acidity activated surface charge‐conversion from negative to positive. Thus, this smart carrier is a feasible candidate for potential cancer therapy.

Photothermal therapy (PTT), as a noninvasive and local treatment, has emerged as a promising anti-tumor strategy with minimal damage to normal tissue under spatiotemporally controllable irradiation. However, the necrosis of cancer cells during PTT will induce an inflammatory reaction, which may motivate tumor regeneration and resistance to therapy. In this study, polyoxometalates and a chloroquine diphosphate (CQ) co-loaded metal–organic framework nanoplatform with hyaluronic acid coating was constructed for efficient ovarian cancer therapy and anti-inflammation. Our results demonstrated that this nanoplatform not only displayed considerable photothermal therapeutic capacity under 808 nm near-infrared laser, but also had an impressive anti-inflammatory capacity by scavenging reactive oxygen species in the tumor microenvironment. CQ with pH dependence was used for the deacidification of lysosomes and the inhibition of autophagy, cutting off a self-protection pathway induced by cell necrosis–autophagy, and achieving the synergistic treatment of tumors. Therefore, we combined the excellent properties of these materials to synthesize a nanoplatform and explored its therapeutic effects in various aspects. This work provides a promising novel prospect for PTT/anti-inflammation/anti-autophagy combinations for efficient ovarian cancer treatment through the fine tuning of material design.  相似文献

A label‐free DNA biosensor based on three‐dimensional reduced graphene oxide (3D‐rGO) and polyaniline (PANI) nanofibers modified glassy carbon electrode (GCE) was successfully developed for supersensitive detection of breast cancer BRCA1. The results demonstrated that 3D‐rGO and PANI nanofibers had synergic effects for reducing the charge transfer resistance (Rct), meaning a huge enhancement in electrochemical activity of 3D‐rGO‐PANI/GCE. Probe DNA could be immobilized on 3D‐rGO‐PANI/GCE for special and sensitive recognition of target DNA (1.0×10?15–1.0×10?7 M) with a theoretical LOD of 3.01×10?16 M (3S/m). Furthermore, this proposed nano‐biosensor could directly detect BRCA1 in real blood samples.  相似文献

Photothermal therapy has attracted much interest for use in cancer treatment in recent years. In this study, Cu2Se nanoparticles as a novel photothermal agent modified by chitosan (CS‐Cu2SeNPs) were successfully synthesized through a facile route at room temperature. The as‐synthesized CS‐Cu2SeNPs exhibited good water solubility and significant stability. CS‐Cu2SeNPs can efficiently convert near‐infrared (NIR) light into heat and exhibit excellent thermostability. In vitro experiments showed that CS‐Cu2SeNPs had selective cellular uptake between cancer and normal cells and expressed clear anticancer activity on A375 and HeLa human cancer cells. In addition, the anticancer activity was increased to about 400 % by combination with a laser at 808 nm, which acted through induction of apoptosis with the involvement of intrinsic and extrinsic pathways. CS‐Cu2SeNPs irradiated with a laser effectively triggered the intracellular reactive oxygen species (ROS) overproduction that promoted cell apoptosis. Therefore, the developed CS‐Cu2SeNPs could be used as a novel phototherapeutic agent for the photothermal therapy of human cancers.  相似文献

An efficient and safe method to deliver active proteins into the cytosol of targeted cells is highly desirable to advance protein‐based therapeutics. A novel protein delivery platform has been created by combinatorial design of cationic lipid‐like materials (termed “lipidoids”), coupled with a reversible chemical protein engineering approach. Using ribonuclease A (RNase A) and saporin as two representative cytotoxic proteins, the combinatorial lipidoids efficiently deliver proteins into cancer cells and inhibit cell proliferation. A study of the structure–function relationship reveals that the electrostatic and hydrophobic interactions between the lipidoids and the protein play a vital role in the formation of protein–lipidoid nanocomplexes and intracellular delivery. A representative lipidoid (EC16‐1) protein nanoparticle formulation inhibits cell proliferation in vitro and suppresses tumor growth in a murine breast cancer model.  相似文献
