Substituted phenyl iodides or diiodides reacted with ethyl iodotetrafluoroproponylate ICF2CF2CO2Et, 1 in the presence of copper powder to give the coupled products 2 or 3 in good yields. Addition of 1 to ethylene and allyl acetate proceeded smoothly under thermal and radical conditions to give the corresponding adducts, which underwent elimination reaction to give β-vinyl and β-allyl α,α,β,β-tetrafluoroesters, CH2CHCF2CF2CO2Et, 4 and CH2CHCH2CF2CF2CO2Et, 5, respectively. 1 also readily reacted with 1,5-hexadiene and 1-hexene with copper or palladium complex, followed by reduction to remove iodine to produce ω-alkenyl-α,α,β,β-tetrafluoroester CH2CH(CH2)4CF2CF2CO2Et 6 and α,α,β,β-tetrafluoroester C4H9CH2CHICF2CF2CO2Et.  相似文献

A convenient and effective method for the synthesis of 6-demethoxycapillarisin was described.The highlight was involved in asimple access to the key intermediate 2-ethylthio-5,7-dimethoxy-4H-chromen-4-one from the inexpensive 2,4,6-trihydroxyacetophenone,using AlCl3 as the demethylation reagent.  相似文献

Condensation of α-phthaloylaminoacetophenones 1a-c with N,N-dimethylformamide dimethyl acetal afforded the novel enamines 3a-c. Cyclization of 3 with hydrazine, alkylhydrazine or phenylhydrazine salts (4a-d) gave 4-phthaloylamino-3-arylpyrazoles 7–9 in high yields. Deprotection of 7–9 was accomplished with hydrazine to provide 4-amino-3-arylpyrazoles 5 in good yields.  相似文献

In recent years, considerable efforts have been expended to the development ofenantioselective synthesis of α-amino acids and the use of α-amino acids as chiralbuilding blocks for the synthesis of complex molecules. Numerous unnatural aminoacids which are often the characteristic units of biologically active peptides, have alsobeen discovered. The alkylation of the diastereomeric mixture of the chiral morpholinone derivatives 5 and 6 occurs with good yield and a prevalence of the cis isomer. Cleavage of the alkylated intermediates 7b,c yields enantiomerically pure sterically constrained uncommon α,α′-dialkylated α-amino acids. The absolute configuration of the new stereocentres has been assigned on the basis of the 1H NMR spectra and NOE measurements. An enzymatic strategy for the preparation of optically pure α-alkyl-α,β-dihydroxyketones is reported. Homo- and cross-coupling reactions of α-diketones catalyzed by acetylacetoin synthase (AAS) produce a set of α-alkyl-α-hydroxy-β-diketones (30-60%, ee 67-90%), which in turn are reduced regio-, diastereo-, and enantioselectively to the corresponding chiral α-alkyl-α,β-dihydroxyketones (60-70%, ee >95%) using acetylacetoin reductase (AAR) as catalyst. Both enzymes are obtained from Bacillus licheniformis and used in a crude form. The relative syn stereochemistry of the enantiopure α,β-dihydroxy products is assigned by NOE experiments, whereas their absolute configuration is determined by conversion of the selected 3,4-dihydroxy-3-methyl-pentan-2-one to the natural product (+)-citreodiol.  相似文献

The synthesis of some α,α,ω,ω-tetrachloro and tetrabromo-derivatives of the hypolipidemic 3,3,14,14-tetramethylhexadecanedioic acid from α,ω-dihalo-esters, carbon tetrahalides and lithium diisopropyl amide has been described. Interaction of α,ω-dichloro esters with CBr4 was shown to give the expected α,ω-dibromo-α,ω-dichloro compounds, but treatment of bis(l-methylethyl) 2,15-dibromo-3,3,14,14-tetramethylhexa-decanedioatewith CCl4 was found to result in halogen exchange yielding the tetrachloro-diester as the major product.  相似文献

Herein we report the asymmetric synthesis of α,α-disubstituted α-amino alcohol derivatives 22, 25 and 26, key intermediates of a novel immunomodulator, using Seebach’s method. We report here that the reactions of in situ generated unprotected α,α-difluoroenolates with various N-sulfonyl imines take place under operationally convenient conditions affording a novel type of fluorinated sulfonamides of high pharmaceutical potential. With the aim to find an efficient synthetic procedure for the construction of 2,3-diamino-2,3-dideoxy-β-D-mannuronic acids, we evaluated three mannosyl donors: (S)-phenyl 4,6-di-O-acetyl-2,3-diazido mannopyranoside, (S)-phenyl 2,3-diazido-4,6-O-benzylidene mannopyranoside, and (S)-phenyl 2,3-diazido mannopyranosyl methyl uronate. The first two mannosylating agents are rather unselective or slightly α-selective in their condensation with three different acceptors. The mannuronic acid donor on the other hand reliably provides the desired β-mannosidic linkage. A mechanistic rationale is put forward to account for the different behavior of the three donor types. Suitably protected 2,3-diazido mannuronic acids were employed to construct the all-cis-linked tetrasaccharide repeating unit of the capsular polysaccharide of Bacillus stearothermophilus , featuring two 2,3-diacetamido-2,3-dideoxy-β-D-mannuronic acids.  相似文献

A visible light-mediated approach for the preparation of α-bromo-α,β-unsaturated ketones and aldehydes was developed. In comparison to traditional methods that generally take two steps to afford the above compounds, this protocol was highlighted by its operational simplicity, avoiding using hazardous bromine and mild reaction conditions.  相似文献
