Preparation of 1-[ω-(1,4,5,6-Tetrahydropyrimidin-2-Yl)Alkyl]Thymines

One-stage preparation of 1-[ω-(1,4,5,6-tetrahydropyrimidin-2-yl)alkyl]-thymines from 1-(ω-cyanoalkyl)thymines and a 1:1-mixture of 1,3-diaminopropane and ethanol saturated with hydrogen sulfide is described. 1-(2-Cyanoethyl)-thymine gave a facile reversal of the Michael reaction to thymine under the conditions used. The reaction of 1,3-bis(3-cyanobenzyl)thymine with the above reagent produced 1,3-bis[3-(1,4,5,6-tetrahydropyrimidin-2-yl)benzyl]thymine.     

New ylidene derivatives of pyrrolo[2,1-α]isoquinoline-2,3-dione in the synthesis of dispiropyrrolidines

Chiral α,ω-diaminoethers derived from d-mannitol and l-treitol as building blocks for the synthesis of macrocyclic compounds possessing 1,3-benzenedicarboxamide or 2,6-pyridinedicarboxamide subunits

Three new chiral α,ω-diaminoethers, derivatives of d-mannitol and l-treitol, possessing C₂ symmetry are prepared. The α,ω-diaminoethers were applied to the macrocyclization reaction under non-high-dilution conditions, which afforded chiral macrocyclic diamides possessing either 2,6-pyridinedicarboxamide or 1,3-benzenedicarboxamide moieties.     

2-Azidoethoxy derivatives of 2-aminocyclohexanecarboxylic acids (ACHC): interesting building blocks for the synthesis of cyclic β-peptide conjugates

Oligomers of cyclic-β-aminoacids possess a high resistance to peptidase-catalyzed hydrolysis and display a high intrinsic tendency to adopt regular secondary structures. These characteristics are attractive to develop new biologically active substances. However, cyclic-β-peptides often show limited solubility in water and cannot be conjugated to biologically relevant fragments, such as oligosaccharides, which are often essential for full biological activity of natural α-peptides. In this article, we report the synthesis of one trans- and one cis-2-aminocyclohexanecarboxylic acid (ACHC), both functionalized with a hydroxy group, to increase the solubility in water, and an azidoethoxy group to allow the synthesis of cyclic-β-peptide conjugates by a ‘click reaction’.     

Efficient synthesis of benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-ω-iodoalkanoates

The efficient synthesis of four benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-ω-iodoalkanoates {benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-3-iodopropanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-4-iodobutanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-5-iodopentanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-6-iodohexanoate} from natural or protected l-amino acids is described.     

Engaging thieno[2,3-b]indole-2,3-dione for the efficient synthesis of spiro[indoline-3,4′-thiopyrano[2,3-b]indole] by reaction with N-substituted isatilidenes

A simple and efficient method, proceeding through a new mechanistic pathway, for the synthesis of spiro[indoline-3,4-thiopyrano[2.3-b]indole derivatives have been developed by exploiting the reaction of thieno[2,3-b]indole-2,3-dione with N-substituted isatilidenes. The compounds synthesized have been screened for antibacterial activity. The generality of the reaction and mechanistic rationale are presented.     

Stereoselectivity of the captodative alkenes 1-acetylvinyl arenecarboxylates in Diels-Alder reactions with cyclic dienes and stereospecific rearrangement of their bicyclo[2.2.n] α-ketol adducts

Captodative alkene 1-acetylvinyl p-nitrobenzenecarboxylate 1a was evaluated for its reactivity and stereoselectivity with cyclohexadiene (10) in Diels-Alder reactions, showing exclusive endo preference. The two hydrolyzed products of the endo and the exo adducts obtained from the Diels-Alder cycloaddition between 1a and cyclopentadiene (7) were 8b and 9b. When treated with mCPBA, a ring expansion took place to stereospecifically yield the novel 3-oxatricyclo[3.3.1.0^2,4]nonanone acyloins 15 and 16, respectively. In the case of the α-ketol bicyclo[2.2.2]octanes 11b and 12b, the epoxidation/Baeyer-Villiger cascade process was preferred, resulting in the syn ketoepoxide 19b from each isomer. A synthetic application of this kind of transformation was carried out by reacting ketols 8b and 9b with an excess of mCPBA through a five-step cascade process to yield the bicyclic lactone 27 as a potential precursor of racemic β-carbaxylose.

Transfer of the chelating ligands in the systems [LAuCl2]+-[PdCl4]2−: Synthesis and structure of the salt (NH4)0.20[(Bipy)AuCl2]1.04[(Bipy)PdCl2]0.96[AuCl4]0.76[PdCl4]0.24

The formation of binary complex salts containing gold(III) in the cation and palladium(II) in the anion in the systems [(Bipy)AuCl₂]⁺-[PdCl₄]^2? occurs by transfer of the N,N-electron-donating chelating ligand bipyridine and the chloride ligands between the gold-containing cation and the palladium-containing anion. The resulting neutral salt [(Bipy)PdCl₂] crystallizes together with the anion [AuCl₄]^? from acetonitrile-water (1 : 1-1 : 2, v/v) to give the complex salt (NH ₄ ⁺ )_0.20[(Bipy)AuCl ₂ ⁺ ]_1.04[(Bipy)PdCl₂]_0.96[AuCl ₄ ^? ]_0.76PdCl ₄ ^2? ]_0.24 with a total Au : Pd ratio of 3 : 2. The ammonium cation is formed from acetonitrile upon its hydrolysis most likely catalyzed by Pd complexes. Quantum-chemical calculations were performed to study the transfer of the chelating ligand theoretically.     

Synthesis and characterization ofα-{3-[(2,3-dihydroxy) propoxy]propyl}-ω-butylpolydimethylsiloxanes

Hydrosilylation has been carried out between Si-H terminated polydimethylsiloxanes with narrow molecular weight distribution and protected 3-allyloxy-1,2-propanediol,and after subsequently alcoholyzed,α-{3-[(2,3-dihydroxy)propoxy]propyl}-ω-butyl -polydimethylsiloxanes with varying number of((Si(CH_3)_2-O) unit were obtained.At each step,the produced compounds were carefully characterized.The results showed that each step was successfully carried out and target products were achieved.     

Investigation of regioselectivity in the synthesis of spiro [pyrrolidine-2,3′-oxindoles] by use of the Huisgen reaction

The Huisgen reaction has been used to synthesize five-membered heterocyclic compounds in high yield and with high regio and stereoselectivity. In the synthesis of spiro [pyrrolidine-2,3′-oxindole] derivatives from isatin, α-amino acids, and (E)-β-phenyl nitroolefins, two regioisomers were obtained in each reaction. The regioselectivity of the major product was found to be different from that in reported work, and was investigated at the B3LYP/6-311G* level of theory. On the basis of this new finding, several conditions, for example molar ratio, solvent, and temperature, which affect the regioselectivity of this reaction were investigated; the results obtained are discussed. It was found that the regioselectivity of this reaction was affected by solvent and temperature, irrespective of the ratio of the reactants. Low temperature and high solvent polarity leads to high regioselectivity, and protic solvents result in higher yield and regioselectivity. These results are of benefit for regioselective synthesis of some compounds.     

Practicable synthesis of (1R,4R)-5-(ℓ-menthoxy)-2-azabicyclo[2.2.0]-hex-5-en-3-one and its derivatives: New building blocks for carbapenem nuclei

Enantioselective synthesis of (1R,4R)-2-azabicyclo[2.2.0]-hexane-3,5-dione and (1R, 4R,5S)-5-hydroxy-2-azabicyclo[2.2.0] hexan-3-one (new building blocks for carbapenems) from 4-(ℓ-menthoxy)-pyridin-2(1H)-one via photopyridone formation is reported.     

Selective syntheses of vinylenedithiathiophenes (VDTTs) and dithieno[2,3-b;2′,3′-d]thiophenes (DTTs); building blocks for π-conjugated systems

Selective syntheses of 3,4-vinylenedithiathiophenes (VDTTs) and dithieno[2,3-b;2′,3′-d]thiophenes (DTTs), having Ph, 4-CH₃OC₆H₄, 4-BrC₆H₄, 4-NO₂C₆H₄ and 4-(CH₃)₂NC₆H₄ groups, were achieved through the reaction of 1,8-diketone with phosphorus decasulfide (P₄S₁₀). The reaction could be shifted between VDTT and DTT by adding base (sodium bicarbonate) or acid (para-toluenesulfonic acid), respectively, to the reaction mixture. While the VDTTs were obtained in moderate yields, an important achievement has been made with the syntheses of the DTTs, obtaining them in higher yields compared with the previous report. Polymers of the VDTTs, which are the analogues of ethylenedioxythiophene, EDOT, were prepared using FeCl₃. Unfortunately, all attempts for their electropolymerization failed. Spin density calculations revealed that none of the VDTTs had a significant positive spin density at the ‘α’ carbon atoms of the thiophene ring. Considering their solubility and functional groups, which could be further derivatized, they are useful building blocks for the preparation of new organic materials.

The preparation of a bridged bicyloalkane via an intramolecular ketene-olefin cycloaddition reaction in the context of a total synthesis of clovene is described.
