In this paper, the effect of increasing amounts of lysozyme (Lyso) and bovine serum albumin (BSA) on the behaviour of lecithin (DMPC) and cephalin (DMPE) liposomes was investigated by means of Raman and DSC techniques. The results showed that both proteins affected, but in a different way, both lecithin and cephalin liposomes. In the samples with lower Lyso concentrations (up to 2 % w/w), a small decrease on the main transition temperature (T m) was observed, whereas T m increased by further addition of Lyso (up to 15.0 % w/w). At the same time, an increase of about 20 % in the ΔH of the transition was observed. Pre-transition was also affected in a greater extent by protein presence. When BSA interacted with liposomes, a smaller increase in the T m values was observed with a contemporary increase of about 8 % in the associated ΔH. The data suggested that the BSA–liposomes interaction involves only the external surface of the bilayer, excluding thus any penetration into the liposomal hydrophobic core. On the contrary, a partial penetration into the bilayer is suggested when Lyso is added to liposomes. Both considered proteins strengthened the overall bilayer structure of DMPC liposomes, suggesting a decrease in the membrane permeability. Moreover, Lyso secondary structure changed by interaction with liposomes, as demonstrated by the Raman spectra behaviour, in particular in the case of DMPE.  相似文献

The effect of adsorption of bovine serum albumin (BSA) on the membrane characteristics of liposomes at pH 7.4 was examined in terms of zeta potential, micropolarity, microfluidity and permeability of liposomal bilayer membranes, where negatively charged L-alpha-dipalmitoylphosphatidylglycerol (DPPG)/L-alpha-dipalmitoylphosphatidylcholine (DPPC), negatively charged dicetylphosphate (DCP)/DPPC and positively charged stearylamine (SA)/DPPC mixed liposomes were used. BSA with negative charges adsorbed on negatively charged DPPG/DPPC mixed liposomes but did not adsorb on negatively charged DCP/DPPC and positively charged SA/DPPC mixed liposomes. Furthermore, the adsorption amount of BSA on the mixed DPPG/DPPC liposomes increased with increasing the mole fraction of DPPG in spite of a possible electrostatic repulsion between BSA and DPPG. Thus, the adsorption of BSA on liposomes was likely to be related to the hydrophobic interaction between BSA and liposomes. The microfluidity of liposomal bilayer membranes near the bilayer center decreased by the adsorption of BSA, while the permeability of liposomal bilayer membranes increased by the adsorption of BSA on liposomes. These results are considered to be due to that the adsorption of BSA brings about a phase separation in liposomes and that a temporary gap is consequently formed in the liposomal bilayer membranes, thereby the permeability of liposomal bilayer membranes increases by the adsorption of BSA.  相似文献

The penetration of bovine serum albumin (BSA) into dipalmitoylphosphatidylglycerol (DPPG) monolayers was observed using atomic force microscopy (AFM) and surface pressure measurements. The effects of surface pressure, amount of BSA and the addition of ganglioside GM1 (GM1) were investigated. The surface pressure of the DPPG monolayer was increased by the penetration of BSA, and the increase in surface pressure was greater in the liquid-expanded film than that in the liquid-condensed film. The AFM images indicated that BSA penetrated into the DPPG monolayer. The amount of BSA that penetrated into the DPPG monolayer increased with time and with the amount of BSA added. On the contrary, the AFM image showed that BSA penetration into the mixed DPPG/GM1 (9 : 1) monolayer scarcely occurred. GM1 inhibited the penetration of BSA into the DPPG monolayer.  相似文献

采用反应条件温和的原子转移自由基聚合法(ATRP),以N-异丙基丙烯酰胺(NIPAAm)和丙烯酰胺(AAm)为功能单体,以N-(3-二甲氨基丙基)甲基丙烯酰胺(DMAPMA)为辅助功能单体,制备了以牛血清白蛋白(BSA)和溶菌酶(Lyz)为双模板蛋白质的表面印迹聚合物(Bi-MIP)。对印迹过程中辅助功能单体的量进行了考察,结果表明,在单一蛋白质溶液和混合蛋白质溶液中,当DMAPMA为20 μL时,制备的Bi-MIP对模板蛋白质BSA和Lyz有较好的吸附容量与选择性。通过静态吸附实验考察了Bi-MIP的吸附性能,并结合Langmuir吸附模型得到聚合物对模板蛋白质BSA和Lyz的最大吸附容量分别为10.2 mg/g和19.2 mg/g。且该印迹聚合物在实际样品中对模板蛋白质也表现出较强的吸附能力和较高的选择性。该方法将为复杂生物样品中同时对双/多种目标蛋白质的识别提供一种新的途径。  相似文献

Summary The stability of liposomes (2∶1 egg yolk lecithin:cholesterol, mole ratio; diameter about 100 nm) at increasing bovine serum albumin (BSA) concentration was study. The influence of introducing positive or negative charge to the liposomal bilayers was tested. The results indicated appreciable destructive effects of serum albumin on the liposomal membranes of neutral and negatively charged liposomes. Two classes of binding sites, a single high-affinity site with an association constant of 4·8×106 M−1 and two low-affinity sites with association constant of about 0·05×106 M−1 have been observed in the interaction of Naproxen with bovine serum albumin (BSA). Chemical modification of two tryptophan residues in BSA with 2-hydroxy-5-nitrobenzyl bromide has led to a reduction in the association constant of the high-affinity site by 89% and its number of binding sites by 66% suggesting the involvement of tryptophan residues in the high-affinity site. In contrast, the two low-affinity sites were not affected by the modification. Binding of Naproxen to the low-affinity sites of BSA induces microdisorganisation of the albumin structure leading to conformational changes as evident from fluorescence measurements with 1-anilino-8-naphthalenesulphonic acid as the probe.  相似文献

The binding reactions of the fluoroquinolone with bovine serum albumin (BSA) were investigated by microcalorimetry. The thermodynamic parameters were measured with the help of spectroscopy in a Tris–HCl buffer solution (pH 7.0, made isotonic with sodium chloride) at T = 298 K. Microcalorimetric measurements show that the molar change of enthalpy ΔrHm is insignificant for the reaction, which may suggest that the interaction is governed mainly by entropy, and the interaction between the protein and the drugs is stronger. The results also reveal an entropy–enthalpy compensation relationship of the interaction.  相似文献

Micrometer-sized silica spheres with bimodal macromesoporous structures are synthesized continuously by a modified gelation technology, and a novel coaxial microfluidic device is applied to control the size of the silica spheres. These spheres are used as a new protein adsorbent to realize fast adsorption and separation of protein mixture. BSA and lysozyme (LYS) are as model proteins with different sizes and pIs. High protein adsorption capacity and rapid adsorption rate have been achieved. Fast separation of a binary mixture of BSA and LYS through a short packed column (50 mm x 4.6 mm id) has also been successfully realized. LYS was selectively adsorbed while BSA flowed through the column in 12 s at a flow rate of up to 5.0 mL/min. As a result, an ultrafast adsorption and separation of BSA and LYS was obtained, showing great potential for applications in fast, large-scale protein separation processes of these monodispersed silica spheres.  相似文献

A series of ion-exchange equilibrium experiments were completed and analyzed by a mass action model. The isotherms at various counter-ion concentrations are regressed to determine the average number of binding sites, Z, the mass action constant, K, and the effective capacity of the exchanger, Pm. The estimates for Pm are 1-10 mmol of protein adsorbed per liter of packed Sepharose exchanger, in agreement with values determined from fitting the Langmuir isotherms. A geometric model, developed to relate the charge distribution of the exchanger and the size of the protein to the protein capacity, also confirms the estimates of Pm. The Z values estimated (and confirmed with an independent slope method) range from 0.8 to 3.6, indicating that most of the ionized sites on the protein do not undergo ion exchange. Geometric model estimates of exchanger sites covered by a protein at maximum surface coverage show that only 1-32% of the sites actually undergo ion exchange. This result indicates that adsorbed protein molecules "block" many exchanger sites, preventing other molecules from ion exchanging at those sites. This high degree of blocking by proteins also indicates that there is an excess of exchanger sites, as evidenced by Z showing no dependence on charge separation distance over the range 6-10 A. Scatchard pots of the equilibrium data indicate that there is competition between at least two different binding forms of each protein studied. For the high-affinity systems, cooperative binding was observed at low exchanger loading; the character then changed to heterogeneous competitive binding as the exchanger loading increased. The effect on an isotherm plot is a subtle but systematic deviation from the Langmuir and mass action models. More important, the competition means that Z represents an average for competing binding forms and, as such, Z can be fractional.  相似文献

Magnetic particles about 10 nm in size were prepared by chemical precipitation under nitrogen and used for the selective and sequential adsorption of bovine serum albumin (BSA) (pI = 4.7) and lysozyme (LSZ) (pI = 1.1) under different conditions, such as pH and initial protein concentration. The separation ratio of BSA over LSZ at pH 4.6 is about 5, which is about 1.5 times the separation ratio of LSZ over BSA at pH 11.0. Only 10% of the preadsorbed BSA could be displaced by the sequential adsorption of LSZ at pH 11.0. On the other hand, 60% of the preadsorbed LSZ was desorbed due to the sequential adsorption of BSA at pH 4.6. Over 50% desorption of BSA or LSZ could be achieved either by 0.5 M Na(2)HPO(4) or 0.5 M NaH(2)PO(4) after 2 h. Over 80% of the enzymatic activity of LSZ was preserved when it was desorbed from magnetic particles.  相似文献

The distribution of urocanic acid (UCA) isomers between aqueous solutions and n-octanol, egg yolk phosphatidylcholine (eggPC) liposomes or bovine serum albumin (BSA) has been evaluated. Regarding its partitioning between water and n-octanol, the behaviour of both isomers is very similar, and the amount incorporated to the organic solvent is mostly determined by the fraction of the compound that, in the aqueous phase, is present as uncharged species. This implies that the highest hydrophobicity occurs near the isoelectric point. cis- and trans-UCA are readily incorporated into eggPC unilamellar liposomes. A simple pseudophase treatment of ultrafiltration data renders a binding constant of 0.20+/-0.04mL/mg for the trans isomer at pH 7.4. The binding constant decreases, by a factor two, at pH 5.0, suggesting that the negatively charged species is more favourably bound to the liposomes than the neutral species, which is mostly present as zwitterions. The cis-isomer, at both pHs, is less incorporated to the bilayers. trans-UCA and cis-UCA readily bind to BSA at pH 7.4, with binding constants of 3400M(-1) and 6900M(-1), respectively. This result suggests that, as in the octanol/water partitioning, hydrophobic interactions predominate and the degree of binding is determined by the fraction present as uncharged species. A smaller binding constant at pH 5.0 indicates that the charge of the protein is also plying a relevant role.  相似文献

The amounts of negatively charged bovine serum albumin and positively charged lysozyme adsorbed on alumina, silica, titania, and zirconia particles (diameters 73 to 271 nm) in aqueous suspensions are measured. The adsorbed proteins change the zeta potentials and the isoelectric points (IEP) of the oxide particles. The added to adsorbed protein ratios at pH 7.5 are compared with the protein treated particle zeta potentials. It is found that the amounts of adsorbed proteins on the alumina, silica, and titania (but not on the zirconia) particle surfaces are highly correlated with the zeta potential. For the slightly less hydrophilic zirconia particles high amounts of protein adsorption are observed even under repulsive electrostatic conditions. One reason could be that the hydrophobic effect plays a more important role for zirconia than electrostatic interaction.  相似文献

Structural characteristics of a series of MAST carbons were studied using scanning electron microscopy images and the nitrogen adsorption isotherms analyzed with several models of pores and different adsorption equations. A developed model of pores as a mixture of gaps between spherical nanoparticles and slitlike pores was found appropriate for MAST carbons. Adsorption of ibuprofen [2-(4-isobutylphenyl)propionic acid] on activated carbons possessing different pore size distributions in protein-free and bovine serum albumin (BSA)-containing aqueous solutions reveals the importance of the contribution of mesopores to the total porosity of adsorbents. The influence of the mesoporosity increases when considering the removal of the drug from the protein-containing solution. Cellulose-coated microporous carbon Norit RBX adsorbs significantly smaller amounts of ibuprofen than uncoated micro/mesoporous MAST carbons whose adsorption capability increases with increasing mesoporosity and specific surface area, burnoff dependent variable. A similar effect of broad pores is observed on adsorption of fibrinogen on the same carbons. Analysis of the ibuprofen adsorption data using Langmuir and D'Arcy-Watt equations as the kernel of the Fredholm integral equation shows that the nonuniformity of ibuprofen adsorption complexes diminishes with the presence of BSA. This effect may be explained by a partial adsorption of ibuprofen onto protein molecules immobilized on carbon particles and blocking of a portion of narrow pores.  相似文献

Remote loading of insulin and bovine serum albumin (BSA) into neutral or positively-charged liposomes by incubation under a transmembrane pH gradient or non-pH gradient was investigated. Trapping efficiencies in several incubation conditions were compared with those of the conventional reverse-phase evaporation vesicle method (c-REV method). For neutral liposomes, insulin could not be effectively loaded into the liposomes by incubation, regardless of the incubation conditions. The trapping efficiency of insulin into positively-charged liposomes was higher than that of neutral liposomes, especially by the pH-gradient method. Insulin could be loaded into positively-charged liposomes about twofold more efficiently than by the pH-gradient method, compared with the c-REV method. Insulin distributed more on the surface of liposomes by the pH-gradient method than by the c-REV method. BSA showed significantly higher affinity to positively-charged liposomes than to neutral ones by various methods. However, the transmembrane pH-gradient method did not increase BSA loading into liposomes, compared with the c-REV and the non-pH-gradient methods. Our results suggest that the pH-gradient method, combining electrostatic interactions, may be useful for preparation of liposomal insulin and that the high hydrophobicity of BSA may not increase the remote loading of BSA into liposomes by the pH-gradient method.  相似文献

The presence of dimers in commercial bovine serum albumin (BSA) samples of nominal high purity is investigated along with their effects on the frontal analysis behavior of preparative anion-exchange media. Size-exclusion and analytical anion-exchange chromatography are used to determine the relative amounts of monomer and dimer for two samples of BSA. While the amount of dimer was generally low, its impact on adsorptive behavior is significant. Equilibrium experiments demonstrate that the anion-exchange media binds the dimer more strongly, leading to an unequal distribution of the monomer and dimer between the two phases. Analysis of the breakthrough behavior of BSA reveals that the monomer breaks through prior to the dimer. This leads to the characteristic tailing breakthrough curve often seen with BSA. Breakthrough experiments carried out using dimer-free BSA confirm that the extreme tailing observed with the commercial samples is curtailed by removing the dimer.  相似文献

The binding of vanadium(V) to bovine serum albumin (BSA), human serum albumin (HSA), and bovine pancreatic trypsin in the absence and presence of urea has been studied at different pH values and temperatures by spectrophotometric and equilibrium dialysis methods. The binding data were found to be pH and temperature dependent. The binding data at pH 5.57, studied by the absorbance method, were found approximately identical with those obtained from the equilibrium dialysis method at this pH. The enthalpy change at pH 5.57 for vanadium(V)-protein was −368.4 cal Mole−1 for BSA, −328.8 cal Mole−1 for HSA and −1372 cal Mole−1 for trypsin respectively. The association constants and the number of binding sites were calculated from Scatchard plots and found to be at maximum at lower pH and at lower temperature. The free energy of the combining sites was lowest at higher pH and highest at low pH. Therefore, a lower temperature and a lower pH offered more sites in the protein molecule for interaction with vanadium(V) ions. Statistical effects seem to be more significant at lower vanadium(V) ion concentrations, and electrostatic effects more significant at higher concentrations.  相似文献
