Summary. Cyanomethanesulfonyl chloride was reacted with amines yielding cyanomethanesulfonamides which could be transformed into alkoxymethylidene and aminomethylidene derivatives. The reaction of alkoxymethylidene derivatives with phenylhydrazine resulted in the formation of 5-aminopyrazol-4-sulfonamides, whereas from cyanomethanesulfonamides via the N-hydroxyamidine derivatives and their reaction with esters 1,2,4-oxadiazol-3-methanesulfonamides became accessible. Nitrosation of cyanomethanesulfonamides yielded 2-hydroxyimino derivatives which were then transformed into 2-hydroxyimino N-hydroxyamidine derivatives, and finally cyclized into 4-amino-1,2,5-oxadiazol-3-sulfonamides. On the other hand diazotation of cyanomethanesulfonamides gave the 2-arylhydrazono derivatives, which after transformation into N-hydroxyamidine derivatives gave by reaction with POCl3 5-amino-1,2,3-triazol-4-sulfonamides. Finally, the reaction between cyanomethanesulfonamides and formamidinium acetate opened an easy access to 4-aminopyrimidine-5-sulfonamides, which could be transformed by trialkyl orthoformiates into substituted pyrimidino[4,5-e][1,2,4]thiadiazine derivatives. All intermediates as well as transformation products of the heterocyclic systems were isolated and well characterized. Mechanisms were discussed, and the stereochemistry, when necessary and possible, was elucidated.  相似文献

An efficient synthetic route is reported for the preparation of thieno[2,3-e]furan-2-sulfonamides bearing oxygenated functionality at C-5. The method employs conversion of furan-3-carboxaldehyde to an intermediate that, in the key step, undergoes facile cyclodehydration to the fused [5,5] heterocycle under mild Knoevenagel conditions.  相似文献

A series of the title compounds were prepared for evaluation as inhibitors of carbonic anhydrase II. Oxidation of 5-substituted thieno[2,3-b]thiophene-2-sulfonamides provided the first examples of thiophene[2,3-b]thiophene-2-sulfonarnide 6,6-dioxides. These cyclic vinyl sulfones readily underwent addi tion to give predominately that 4,5-cis addition product.  相似文献

The reaction of the 5-unsubstituted pyrazoles 2a-k with lithium diisopropylamide or n-butyllithium gave intermediary 5-lithiopyrazoles, whose reaction with sulfur dioxide afforded the lithium pyrazole-5-sulfinates 3a-k . Subsequent reaction of 3a-k with N-chlorosuccinimide followed by ammonolysis provided the pyrazole-5-sulfonamides 5a-k .  相似文献

The 1,3-dipolar cycloaddition reaction of 1-substituted-1,2,3,4-tetrahydropyridines 4a-d with organic azides 5 afforded the respective 1-substituted-piperidylidene-2-sulfonamides 6 . In contrast, the reaction of (E)-1-(1-propenyl)-1,2,3,4-tetrahydropyridine ( 4e ) with 4-chlorobenzenesulfonyl azide yielded 6m as well as 1-(4-chlorophenyl)sulfonimine-1,2,3,4-tetrahydropyridine ( 7 ) arising from addition of the azide to the (E)-1-(1-propenyl) substituent of 4e .  相似文献

Oxidative chlorination of 1-aryl-4-benzylsulfanyl-1H-imidazole-5-carbaldehydes gave 1-aryl-5-formyl-1H-imidazole-4-sulfonyl chlorides which reacted with secondary amines and phenols to produce the corresponding N,N-disubstituted 5-formylimidazole-4-sulfonamides and aryl sulfonates. The reaction of 1-aryl-5-formyl-1H-imidazole-4-sulfonyl chlorides with sodium azide, followed by reduction of the resulting sulfonyl azides, led to the formation of N-unsubstituted 5-formylimidazole-4-sulfonamides, and the reaction with alcohols, to 5-formylimidazole-4-sulfonic acids.  相似文献

exo- and endo-5-Aminomethylbicyclo[2.2.1]hept-2-enes have been obtained from stereoisomeric exo- and endo-5-cyanobicyclo[2.2.1]hept-2-enes and the corresponding sulfonamides were obtained through reaction of amines with methyl-, n-propyl-, n-butyl-, benzyl-, and cyclohexylsulfonyl chlorides. From the stereoisomeric sulfonamides with peroxy acids, various products were obtained: exo-sulfonamides were transformed into epoxy derivatives, and, in contrast, most of the endo-stereoisomers underwent heterocyclization resulting in substituted exo-2-hydroxy-4-azatricyclo[4.2.1.03,7]nonanes. The type of the products obtained did not depend on the type of peroxy acid used (peroxyacetic, peroxyphthalic, and m-chloroperoxybenzoic one). In contrast to other endo-sulfonamides, N-(cyclohexylsulfonyl)-endo-5-aminomethylbicyclo[2.2.1]hept-2-ene in reaction with peroxyacetic acid did not undergo heterocyclization, probably, due to steric factors. The structure and stereochemical homogeneity of the sulfonamides and the structure of the products of their oxidation with peroxy acids were confirmed by spectroscopic methods. The molecular structure of N-(cyclohexylsulfonyl)-endo-5-aminomethyl-exo-2,3-epoxybicyclo[2.2.1]heptane was determined by X-ray diffraction analysis. The mechanism of the intramolecular heterocyclization reaction of N-substituted endo-5-aminomethyl-exo-2,3-epoxybicyclo[2.2.1]heptanes was studied at the BHandHLYP/6-31G(d) level of theory.  相似文献

The first one-pot copper-catalyzed highly regioselective C–N bond formation between aryl/alkyl amines and sulfonamide-substituted naphthoquinones was accomplished. Facile chemoselective routes obtained diverse ring-opening 6-amino/anilino-naphthalen-dione-1-sulfonamides and ring-fused 6-amino/aniline-5H-naphth[1,8-cd]isothiazol-5-one,1,1-dioxides with great functional group tolerance. Regiochemistry was confirmed by 1D- and 2D-NMRs.  相似文献

A series of new N-sulfonamides of substituted 3-hydroxy-2-azetidinones were prepared by the [2+2] cycloaddition of aldoximes, formed from the condensation of D-(R)-glyceraldehyde acetonide and the potent p-aminophenyl-N-substituted sulfonamides, with benzyloxyacetyl chloride followed by removal of the protecting groups. Obtaining both diastereoisomeric forms of β-lactams by resorting to kinetic resolution starting from racemic imino analogue is also described. The GC-EIMS, spectroscopic and analytical analyses for some of these derivatives are reported.  相似文献

A series of novel N-aryl-7-chloro-4-methyl-2-oxo-1,2-dihydroquinoline-6-sulfonamides were obtained. The structure-activity relationship (SAR) approach was used to study their diuretic effect on rats. Diuretic activity was exhibited by all the test compounds, with 7-chloro-4-methyl-2-oxo-1,2-dihydroquinoline-6-sulfonanilide being most active. A SAR analysis revealed that the substituted (especially ortho-substituted in the benzene ring) arylamides diminish diuresis compared to the most active compound. For the most and least active compounds, their effects on the electrolyte excretion (Na+, K+, and Cl?) and creatinine level were also studied.  相似文献

Naphth[1,2-d]oxazole-5-sulfonic acid ( 1 ) has been prepared by the fusion of 4-amino-3-hydroxynaphthalene-1-sulfonic acid with formamide. Interaction of 1 with a number of arenesulfonyl chlorides, aryloxyacetyl chlorides, 1-naphthyloxyacetyl chloride, and chloroacetyl chloride gave 2-(arylsulfonyl)-, 2-(aryloxyacetyl)-, 2-(1-naphthyloxyacetyl)- and 2-(chloroacetyl)naphth[1,2-d]oxaxole-5-sulfonic acids ( 2, 3, 4 and 5 ), respectively. The corresponding sulfonyl chloride of 2 was condensed with amines giving the expected 2-(arylsulfonyl)-naphth[1,2-d]oxazole-5-sulfonamides ( 6 ). Interaction of 5 with hydrazine gave 2-hydrazinoacetyl and disubstituted hydrazine derivatives 7 and 8 . Condensation of 7 with aromatic aldehydes yielded substituted hydrazonoacetyl derivatives 9 . Two moles of 5 react with one mole of hydroquinone in dry acetone in the presence of anhydrous potassium carbonate and potassium iodide gave 1,4-bis[5-sulfonaphth[1,2-d]oxazol-2-ylcarbonyl-methoxy]benzene ( 10 ).  相似文献

Reactions of bicyclo[2.2.1]hept-5-en-exo- and -endo-2-ylmethanamines, exo-5,6-epoxybicyclo-[2.2.1]hept-exo-2-ylmethanamine, 1-(bicyclo[2.2.1]hept-2-yl)ethanamine, and 1-(1-adamantyl)ethanamine with camphor-10-sulfonyl chloride in chloroform in the presence of triethylamine gave the corresponding sulfonamides having two cage-like fragments. Stereoisomeric N-(bicyclo[2.2.1]hept-5-en-2-ylmethyl)camphor-10-sulfonamides were oxidized with peroxyphthalic acid generated in situ from phthalic anhydride and 50% hydrogen peroxide. The exo stereoisomer was thus converted into the corresponding 5,6-epoxy derivative, while the endo isomer gave rise to 4-(7,7-dimethyl-2-oxobicyclo[2.2.1]hept-1-ylmethyl)-4-azatricyclo[4.2.1.03,7]-nonan-exo-2-ol (substituted azabrendane). The structure of the synthesized camphor-10-sulfonamides was confirmed by IR and 1H and 13C NMR spectra with the use of homo- (COSY) and heteronuclear 1H-13C correlation techniques (HMQC, HMBC). Heterocyclization of sulfonamides of the norbornene series was also simulated by quantum-chemical calculations at the PM3 and BHandHLYP/6-31G(d) levels of theory.  相似文献

A series of novel α-carboline derivatives such as 3-aryl-9H-pyrido[2,3-b]indole-6-sulfonamides have been synthesized from the readily available low-cost raw material, benzotriazole which is subjected to nucleophilic aromatic substitution with 5-bromo-2-chloropyridine followed by sequential steps of cyclization, sulfonation, amidation, and finally Suzuki–Miyaura coupling reactions. The C–C bond formation between 3-bromo-9H-pyrido[2.3-b]indole-6-sulfonamide and various boronic acids was achieved with more accessible palladium pre-catalyst, Pd-PEPPSI-IPr via Suzuki coupling under microwave condition in a short reaction time with excellent yields.  相似文献
