The present work describes a method based on solid-phase extraction (SPE) and ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) for the simultaneous determination of three quinolones (pipemidic acid, oxolinic acid and flumequine) and twelve fluoroquinolones (marbofloxacin, fleroxacin, pefloxacin, levofloxacin, norfloxacin, ciprofloxacin, enrofloxacin, danofloxacin, lomefloxacin, difloxacin, sarafloxacin, and moxifloxacin) in different infant and young children powdered milks. After suitable deproteination of the reconstituted powdered samples, a SPE procedure was developed providing recovery values higher than 84% (RSDs lower than 13%) for all the analytes, with limits of detection between 0.04 and 0.52 μg/kg. UPLC-MS/MS analyses were carried out in less than 10 min. Sixteen infant and young children powdered milk samples of different origin, type and composition bought at Spanish markets were analyzed. Residues of the selected antibiotics were not detected in any of the analyzed samples. 相似文献

Trace determination of bisphenol A and phytoestrogens in infant formula powders by gas chromatography-mass spectrometry 总被引:2,自引:0,他引:2
This investigation describes a reliable and sensitive method for simultaneously determining bisphenol A (BPA) and two major phytoestrogens, daidzein and genistein, in powdered milks and infant formulas by gas chromatography-mass spectrometric analysis after trimethylsilylation. To reduce the matrix interference associated with the constituents of the formulas, the dissolved formula solutions were firstly ultra-centrifuged and the analytes in the supernatant were then extracted using a C18 solid-phase extraction column. The accuracy and precision of the method were determined and the technique was successfully employed to measure trace concentrations of BPA, daidzein and genistein in powdered formulas. The results show that BPA, daidzein and genistein were detected in all the testing samples (n = 6) at concentrations from 45 to 113 ng/g (except one infant formula), 20 to 2050 ng/g and 21 to 6510 ng/g, respectively. The highest concentrations of daidzein and genistein (i.e., 2050 and 6510 ng/g) were detected in a soy-based powdered infant formula. The quantitation limits were 1.0 ng/g for BPA, and 10 ng/g for daidzein and genistein using 0.5 g powdered milk samples. 相似文献

A simple and sensitive method has been developed for species selective determination of chromium(III) and chromium(VI) in water by electrothermal atomic absorption spectrometry. The procedure is based on selective absorption of Cr(III) on a cellulose micro column (pH 11, 0.5 mol L(-1) NaCl). Total chromium was subsequently determined after appropriate reduction of Cr(VI) to Cr(III). Recoveries of more than 97% were found. A concentration factor of 100 was achieved. The relative standard deviations (n=10) at the 40 ng L(-1) level for chromium(III) and chromium(VI) were 2.3% and 1.8% and corresponding limits of detection (based on 36) were 1.8 ng L(-1) and 5.1 ng L(-1), respectively. No interference effects have been observed from other investigated species and the method has been successfully applied to natural water samples. 相似文献

In this work, a simple and environmental friendly methodology has been developed for the analysis of a group of six mycotoxins with estrogenic activity produced by Fusarium species (i.e. zearalanone, zearalenone, α‐zearalanol, β‐zearalanol, α‐zearalenol, and β‐zearalenol), using microdispersive SPE (μ‐dSPE) with multiwalled carbon nanotubes as sorbent. Separation, determination, and quantification were achieved by HPLC coupled to ion trap MS with an ESI interface. Parameters affecting the extraction efficiency of µ‐dSPE such as pH of the sample, amount of multiwalled carbon nanotubes, and type and volume of elution solvent, were studied and optimized. The methodology was validated for mineral, pond, and wastewater as well as for powdered infant milk using 17β‐estradiol‐2,4,16,16,17‐d5 (17β‐E2‐D5) as internal standard, obtaining recoveries ranging from 85 to 120% for the three types of water samples and from 77 to 115% for powdered infant milk. RSD values were lower than 10%. The LOQs achieved were in the range 0.05–2.90 μg/L for water samples and 2.02–31.9 μg/L for powdered infant milk samples. 相似文献

Biomolecular interaction analysis was evaluated for the automated determination of vitamin B12 in a range of foods. The analytical technique was configured as a biosensor-based, nonlabeled inhibition protein-binding assay using nonintrinsic R-protein. Sample extraction conditions were optimized, and both ligand specificity and nonspecific binding considerations were evaluated. Performance parameters included a quantitation range of 0.08-2.40 ng/mL, recoveries of 89-106%, agreement against assigned reference values for 3 independent certified food reference materials, and a mean between-laboratory reproducibility relative standard deviation of 4.9%. The proposed method was compared with reference microbiological and radioisotope protein-binding methods for a range of food samples. A wide selection of milks, infant formulas, meats, and liver were evaluated for their vitamin B12 content. The influence of season was studied in herd milk, early lactation was followed for a single animal, and the cobalamin content of bovine, caprine, and ovine milks was compared. 相似文献

建立了采用快速溶剂萃取-离子色谱同时测定塑料中三价铬和六价铬的方法。三价铬和六价铬分别以吡啶-2,6-二羧酸(PDCA)和1,5-二苯卡巴肼(DPC)作为络合剂在柱前和柱后进行衍生化,分别在紫外和可见波长下采用紫外检测器进行检测,灵敏度高,基体干扰小。本方法对三价铬和六价铬的检出限分别为5.0 μg/L和0.5 μg/L;分别在50~1000 μg/L和5.0~100 μg/L范围内呈现良好的线性关系,线性相关系数分别为0.9994和0.9998;三价铬和六价铬的回收率范围为90.7%~101.1%,相对标准偏差(RSD)为1.7%~4.4%。该方法分析速度快、灵敏度高、重现性好,可用于塑料中三价铬和六价铬的同时测定。 相似文献

The determination of bioactive peptides derived from food sources is gaining special attention in last years, due to their ability to promote health and their potential to reduce the risk of chronic diseases. In this work, a new analytical methodology has been developed enabling for the first time the determination of soymetide, a new immunostimulating peptide derived from soybean, in different soybean derived foodstuffs. Capillary-HPLC was employed for peptide separation after accelerated tryptic digestion of soybean proteins. Two different capillary-HPLC columns were tested and chromatographic separation was optimized in each case. The use of a 300 μm fused-core technology C18 column enabled a suitable separation of soymetide from the other peptides in less than 18 min. Different analytical characteristics of the method were evaluated: selectivity, linearity, accuracy, precision, limit of detection and quantitation, and stability. The developed method was applied to the determination of soymetide content in different soybean dairy-like products for human consumption (powdered milks and infant formulas). 相似文献

A method was developed for high-throughput speciation analysis of chromium by on-line coupling of short-column capillary electrophoresis (SC-CE) and inductively coupled plasma mass spectrometry (ICP-MS). Baseline separation of Cr(III) and Cr(VI) was achieved within 1 min by SC-CE in a 15 cm x 75 microm id fused-silica capillary at 6 kV using 15 mM HNO(3) as running electrolyte. The precisions (RSD, n = 5) of migration time and peak area for Cr(III) and Cr(VI) were in the range of 1.8-2.4% and 2.2-5.7%, respectively. The limits of detection (3sigma) of Cr(III) and Cr(VI) were 1.8 and 1.9 microg/L, respectively. The synthesized samples containing Cr(III) and Cr(VI) species were determined by the developed SC-CE-ICP-MS hyphenated technique, and the recoveries of Cr(III) and Cr(VI) in the synthesized samples were in the range of 103-110% and 90-108%, respectively. 相似文献

Choline was determined in infant formula by ion chromatography with suppressed conductivity detection. Samples were digested with 1M hydrochloric acid, filtered, diluted, and injected into the chromatographic system. Choline and the alkali and alkaline earth metals were separated on a high-resolution cation-exchange column and detected by suppressed conductivity. The method was linear between 2 and 200 mg/L (r2 = 0.9999), the concentration range of the diluted samples. This method accurately determined choline in powdered, concentrated, and ready-to-feed infant formulas. Recoveries of choline spikes into powdered infant formula at approximately 1, 0.8, 0.5, and 0.2 times the labeled value ranged from 85 to 114%. This method had good agreement for 8 blind duplicates. The values determined for these samples, which were used in an AOAC collaborative study of an enzymatic method, were consistent with the values determined by the enzymatic method. 相似文献

A novel method for the separation and preconcentration of Cr(III)/Cr(VI) with Lemna minor and determination by slurry atomization electrothermal atomic absorption spectrometry (ETAAS) was developed. A sample solution was added to a polyethylene beaker containing 10 mg of 160 mesh pre-treated Lemna minor, adjusted to pH 1.0, stirred for 8 min for selective absorption of Cr(III) and then centrifuged. The upper layer of solution was transferred into another polyethylene beaker containing 10 mg of 160 mesh pre-treated Lemna minor, adjusted to pH 5.0, stirred for 12 min for adsorption of the residual Cr(VI) and centrifuged. The two residues in two centrifuge tubes were washed twice with water, 2 ml of agar solution added, stirred for 2 min, then two slurries were prepared and used for the determination of Cr(III) and Cr(VI) by ETAAS. Detection limits (3sigma) of 0.01 microg L(-1) for Cr(III) and 0.03 microg L(-1) for Cr(VI) were obtained. The relative standard deviation was 2.8% for Cr(III) and 3.3% for Cr(VI) at the 1 microg L(-1) level. The method was applied to the determination of Cr(III)/Cr(VI) in water samples. The analytical recoveries of Cr(III) and Cr(VI) added to samples were 97-102 and 96-103%, respectively. 相似文献

A simple and rapid method is developed for the simultaneous determination of Cr(VI) and Cr(III) based on the formation of their different complexes with ammonium pyrrolidine-dithiocarbamate (APDC). Separation is performed using reversed-phase high-performance liquid chromatography coupled with UV detection. The conditions for complex formation and speciation are determined, such as solution pH, amount of APDC, temperature, and type of mobile phase. In order to substantially reduce the analysis time, the separation is carried out without extraction of chromium-APDC complexes from the mother liquor. Under the optimum analysis conditions, the chromatograms obtained show good peak separation, and the absolute detection limits (3s) are 2.2 microg/L for Cr(VI) and 4.5 microg/L for Cr(III). The calibration curves are linear from 3 to 5000 microg/L for Cr(VI) and 5 to 3000 microg/L for Cr(III). The relative standard deviations of peak areas in five measurements using a sample solution of 200 microg/L are less than 2% for Cr(VI) and 4% for Cr(III), indicating good reproducibility for this analytical method. Furthermore, simultaneous determination of Cr(VI) and Cr(III) is successful with the application of the proposed procedure in the synthetic wastewaters containing common heavy metal ions: Fe(III), Pb(II), Cd(II), Cu(II), and Zn(II). 相似文献

A rapid and sensitive method for the direct determination of hexavalent chromium in natural waters has been developed. Anion exchange chromatography was used for the fast separation of chromates from matrix anions, within 15 minutes, using a 35 mM KOH eluant. Mobile phase suppression was employed prior to direct conductivity (CD) detection, comparing two different electrolytic suppressor models and achieving a 10 times lower detection limit. Post column derivatization of Cr(VI) with diphenylcarbazide was used for further selective diode array (PDA) detection. Possible Cr(III) interference was evaluated with the addition of concentrations up to 10 mg/L of Cr(III) in a Cr(VI) positive natural water sample resulting in negligible interference. Both detection techniques gave instrumental LOD of 0.05 µg/L and method LOD of 0.08 µg/L for CD and 0.05 µg/L for PDA detection in underground water. Average repeatability and reproducibility (%RSD) was 1.3% and 5.4% for CD and 1.3% and 6.1% for PDA detection for two concentration levels. Recovery ranged from 70% to 103% for CD and from 90% to 100% for PDA. Method accuracy was additionally confirmed by analyzing positive samples with LC-ICP-MS. The method was successfully applied to the determination of Cr(VI) in several tap, spring and groundwater samples from the wider area of Asopos River, Greece. High concentrations where confirmed to the majority of the analyzed samples, revealing the serious and expanded problem of industrial pollution. 相似文献

A method for speciation of Cr(III) and Cr(VI) in real samples has been developed. Cr(VI) has been separated from Cr(III) and preconcentrated as its pyrrolidinedithiocarbamate (APDC) complex by using a column containing Amberlite XAD–2000 resin and determined by FAAS. Total chromium has also been determined by FAAS after conversion of Cr(III) to Cr(VI) by oxidation with KMnO4. Cr(III) has been calculated by subtracting Cr(VI) from the total. The effect of pH, flow‐rate, adsorption and batch capacity and effect of various metal cations and salt anions on the sorption onto the resin were investigated. The adsorption is quantitative in the pH range of 1.5–2.5, and Cr(VI) ion was desorbed by using H2SO4 in acetone. The recovery of Cr(VI) was 97 ± 4 at a 95% confidence level. The highest preconcentration factor was 80 for a 200 mL sample volume. The adsorption and batch capacity of sorbent were 7.4 and 8.0 mg g?1 Cr(VI), respectively, and loading half time was 5.0 min. The detection limit of Cr(VI) is 0.6 μg/L. The procedure has been applied to the determination and speciation of chromium in stream water, tap water, mineral spring water and spring water. Also, the proposed method was applied to total chromium preconcentration in microwave digested moss and rock samples with satisfactory results. The developed method was validated with CRM‐TMDW‐500 (Certified Reference Material Trace Metals in Drinking Water) and BCR‐CRM 144R s (Certified Reference Material Sewage Sludge, Domestic Origin) and the results obtained were in good agreement with the certified values. An improved method has been developed for the determination of acrylamide in infant powdered milk and baby foods in jars, a particular class of foodstuffs which represent an important source of nutrition for young infants and babies. This method uses isotope dilution liquid chromatography coupled to a tandem mass spectrometer with electrospray ionization and is significantly more sensitive than previous published methods with a limit of quantification estimated at 1 microg kg(-1). The new method offers effective sample preparation procedures including defatting with petroleum ether, extraction with aqueous solution of sodium chloride, further liquid-liquid extraction with ethyl acetate and clean-up by solid-phase extraction (SPE) with HLB 200 mg cartridges. The analytical method was well validated and good results were obtained with respect to repeatability (RSD < 5%) and recovery (86-97%) which fulfilled the requirements defined by European Union (EU) legislation. The acrylamide level in infant powdered milk and baby foods in jars were 3.01-9.06 microg kg(-1) and 6.80-124.93 microg kg(-1), respectively. Especially, this new method is successfully applied to the trace quantification of acrylamide in infant/baby foods, the content of which is less than 10 microg kg(-1). 相似文献
