A novel immunosensing strategy based on surface‐initiated atom‐transfer radical polymerization (SI‐ATRP) in combination with electrochemical detection is proposed. Specifically, 4‐acetoxystyrene (AS) has been chosen as a monomer for ATRP due to its ability to provide acetoxyl groups, which can be converted into phenolic hydroxyl groups for electrochemical detection in the presence of tyrosinase. A controlled radical polymerization reaction of 4‐acetoxystyrene at 60 °C was triggered after immobilization of initiator molecules on an electrode surface. The growth of long‐chain polymeric materials increased the concentration of phenolic hydroxyl groups, which in turn significantly enhanced the electrochemical signal output. Polymerization conditions, such as temperature and duration, monomer concentration, and the catalyst/monomer ratio have been optimized. The in situ surface‐initiated ATRP was confirmed by scanning electron microscope (SEM) images and X‐ray photoelectron spectroscopy (XPS) analysis. Cyclic voltammetric investigation revealed a pair of well‐defined oxidation and reduction peaks at 0.232 and 0.055 V, which corresponded to the redox behavior of catechol/o‐quinone on the electrode surface. The proposed approach has been successfully extended to immune recognition. A detection limit of 0.3 ng mL?1 for rabbit immunoglobulin G (IgG) as a model antigen has been achieved. Despite the limited availability of the IgG antibody, this technology might also be expanded to the detection of other proteins and DNA.  相似文献

The ability to manipulate and control the surface properties of nylons is of crucial importance to their widespread applications. In this work, surface-initiated atom-transfer radical polymerization (ATRP) is employed to tailor the functionality of the nylon membrane and pore surfaces in a well-controlled manner. A simple two-step method, involving the activation of surface amide groups with formaldehyde and the reaction of the resulting N-methylol polyamide with 2-bromoisobutyryl bromide, was first developed for the covalent immobilization of ATRP initiators on the nylon membrane and its pore surfaces. Functional polymer brushes of 2-hydroxyethyl methacrylate (HEMA) and poly(ethylene glycol)monomethacrylate (PEGMA) were prepared via surface-initiated ATRP from the nylon membranes. A kinetics study revealed that the chain growth from the membranes was consistent with a "controlled" process. The dormant chain ends of the grafted HEMA polymer (P(HEMA)) and PEGMA polymer (P(PEGMA)) on the nylon membranes could be reactivated for the consecutive surface-initiated ATRP to produce the corresponding nylon membranes functionalized by P(HEMA)-b-P(PEGMA) and P(PEGMA)-b-P(HEMA) diblock copolymer brushes. In addition, membranes with grafted P(HEMA) and P(PEGMA) brushes exhibited good resistance to protein adsorption and fouling under continuous-flow conditions.  相似文献

Novel copolymer brushes have been synthesized by a two-step "grafting from" method that consists of the electrografting of poly(2-phenyl-2-(2,2,6,6-tetramethyl-piperidin-1-yloxy)-ethylacrylate) onto stainless steel, followed by the nitroxide-mediated radical polymerization of 2-(dimethylamino ethyl)acrylate and styrene or n-butyl acrylate, initiated from the electrografted polyacrylate chains. The grafted copolymers were quaternized in order to endow them with antibacterial properties. Peeling tests have confirmed the strong adhesion of the grafted copolymer onto the stainless steel substrate. Quartz crystal microbalance experiments have proven that fibrinogen adhesion is lower on the hydrophilic quaternized films compared to the nonionic counterpart. Such quaternized copolymers exhibit significant antibacterial activity against the Gram-positive bacteria S. aureus and the Gram-negative bacteria E. coli.  相似文献

A new technique for polymer microchannel surface modification, called in-channel atom-transfer radical polymerization, has been developed and applied in the surface derivatization of thermoset polyester (TPE) microdevices with poly(ethylene glycol) (PEG). X-ray photoelectron spectroscopy, electroosmotic flow (EOF), and contact angle measurements indicate that PEG has been grafted on the TPE surface. Moreover, PEG-modified microchannels have much lower and more pH-stable EOF, more hydrophilic surfaces and reduced nonspecific protein adsorption. Capillary electrophoresis separation of amino acid and peptide mixtures in these PEG-modified TPE microchips had good reproducibility. Phosducin-like protein and phosphorylated phosducin-like protein were also separated to measure the phosphorylation efficiency. Our results indicate that PEG-grafted TPE microchips have broad potential application in biomolecular analysis.  相似文献

Surface-initiated atom-transfer radical polymerization (ATRP) of poly(ethylene glycol) monomethacrylate (PEGMA) was carried out on the hydrogen-terminated Si(100) substrates with surface-tethered alpha-bromoester initiator. Kinetic studies confirmed an approximately linear increase in polymer film thickness with reaction time, indicating that chain growth from the surface was a controlled "living" process. The "living" character of the surface-grafted PEGMA chains was further ascertained by the subsequent extension of these graft chains, and thus the graft layer. Well-defined polymer brushes of near 100 nm in thickness were grafted on the Si(100) surface in 8 h under ambient temperature in an aqueous medium. The hydroxyl end groups of the poly(ethylene glycol) (PEG) side chains of the grafted PEGMA polymer were derivatized into various functional groups, including chloride, amine, aldehyde, and carboxylic acid groups. The surface-functionalized silicon substrates were characterized by reflectance FT-IR spectroscopy and X-ray photoelectron spectroscopy (XPS). Covalent attachment and derivatization of the well-defined PEGMA polymer brushes can broaden considerably the functionality of single-crystal silicon surfaces.  相似文献

The synthesis of poly(methyl acrylate)-block-poly(gamma-benzyl-L-glutamate) (PMA-b-PBLG) diblock copolymers, using atom-transfer radical polymerization (ATRP) of methyl acrylate and living polymerization of gamma-benzyl-L-glutamate-N-carboxyanhydride (Glu-NCA) is described. Amido-amidate nickelacycle end groups were incorporated onto amino-terminated poly(methyl acrylates), and the resulting complexes were successfully used as macroinitiators for the growth of polypeptide segments. This method allows the controlled preparation of polypeptide-block-poly(methyl acrylate) diblock architectures with control over polypeptide chain length and without the formation of homopolypeptide contaminants.  相似文献

Human blood plasma and serum pose significant challenges to implanted devices because of highly unfavorable nonspecific protein adsorption on the surface. In this work, we introduce an improved two-step method to immobilize initiator thiols on a gold substrate for the surface-initiated atom-transfer radical polymerization (SI-ATRP) of hydroxypropyl methacrylate (HPMA). We investigate protein adsorption from a single-protein solution, diluted (10%) and undiluted (100%) human blood plasma, and serum on the poly(HPMA) brushes with different film thicknesses using surface plasmon resonance (SPR) sensors. SPR results show a correlation between antifouling properties and film thickness; that is, the poly(HPMA) brushes exhibit high protein resistance at medium film thicknesses of ~25-40 nm (e.g. <0.3 ng/cm(2) for single-protein adsorption and 10% human blood plasma and serum, ~24.5 ng/cm(2) for 100% human serum, and ~52.8 ng/cm(2) for 100% human plasma at a thickness of ~29 nm). With an optimal film thickness and surface roughness, the poly(HPMA) brush also demonstrates its high resistance to fibroblast adhesion. This work provides an alternative surface polymerization approach to preparing effective antifouling poly(HPMA) materials for potential applications in blood-contacting medical devices.  相似文献

Surface-tethered oppositely charged weak polyelectrolyte block copolymer brushes composed of poly(2-vinyl pyridine) (P2VP) and poly(acrylic acid) (PAA) were grown from the Si wafer by atom-transfer radical polymerization. The P2VP-b-PAA brushes were prepared through hydrolysis of the second PtBA block to the corresponding acrylic acid. The P2VP-b-PAA brushes with different PAA block length were obtained. The P2VP-b-PAA brushes revealed a unique reversible wetting behavior with pH. The difference between the solubility parameters for P2VP and PAA, the changes of surface chemical composition and surface roughness, and the reversible wetting behavior illustrated that the surface rearrangement occurred during treatment of the P2VP-b-PAA brushes by aqueous solution with different pH value. The reversible properties of the P2VP-b-PAA brushes can be used to regulate the adsorption of the sulfonated PS nanoparticles.  相似文献

Polymeric 1-vinyl-3-octylimidazolium hexafluorophosphate was synthesized in situ on stainless steel wire by surface radical chain-transfer polymerization and used as sensitive coatings in solid-phase microextraction. The outer surface of the stainless steel wire was firstly coated with microstructured silver layer via silver mirror reaction and then functionalized with self-assembled monolayers of 1,8-octanedithiol, which acted as chain transfer agent in the polymerization. Coupled to gas chromatography, extraction performance of the fiber was studied with both headspace and direct-immersion modes using benzene, toluene, ethylbenzene and xylenes (BTEX), phenols and polycyclic aromatic hydrocarbon (PAHs) as model analytes. In combination with the microstructured silver layer, the PIL-coated fiber exhibited high extraction efficiency. Linear ranges for BTEX with headspace mode were in the range of 0.2-1000 μg L(-1) for benzene, and 0.1-1000 μg L(-1) for toluene, ethylbenzene and xylenes. Limits of detection (LODs) were from 0.02 to 0.05 μg L(-1). Wide linear ranges of direct-immersion mode for the extraction of several phenols and PAHs were also obtained with correlation coefficients (R) from 0.9943 to 0.9997. The proposed fiber showed good durability with long lifetime. RSDs of 56 times extraction were still in an acceptable range, from 8.85 to 22.8%.  相似文献

In this paper, the electro-analysis and spectrophotometric analysis methods were used to study the antibacterial ability of copper and stainless steel materials. When Escherichia coli (E. coli) and photo-bacteria were used as samples, the antibacterial effect of stainless steel was very weak, while the percentage of bacteria dying from exposure to metallic copper for 30 min was over 90%. The antibacterial ability of copper has a potential application in the field of disinfection, food packaging and piping of drinking water. Barnacle cement (BC) was beneficially applied on stainless steel (SS) to serve as the initiator anchor for surface-initiated polymerization. The amine and hydroxyl moieties of barnacle cement reacted with 2-bromoisobutyryl bromide to provide the alkyl halide initiator for the surface-initiated atom transfer radical polymerization (ATRP) of 2-hydroxyethyl methacrylate (HEMA). The hydroxyl groups of HEMA polymer (PHEMA) were then converted to carboxyl groups for coupling of chitosan (CS) to impart the SS surface with both antifouling and antibacterial properties. The surface-functionalized SS reduced bovine serum albumin adsorption, bacterial adhesion, and exhibited antibacterial efficacy against Escherichia coli (E. coli). The effectiveness of barnacle cement as an initiator anchor was compared to that of dopamine, a marine mussel inspired biomimetic anchor previously used in surface-initiated polymerization. The results indicate that the barnacle cement is a stable and effective anchor for functional surface coatings and polymer brushes.  相似文献

The rapid development of new applications of photoredox catalysis has so far outpaced the mechanistic studies important for rational design of new classes of catalysts. Here, we report the use of ultrafast transient absorption spectroscopic methods to reveal both mechanistic and kinetic details of multiple sequential steps involved in an organocatalyzed atom transfer radical polymerization reaction. The polymerization system studied involves a N,N-diaryl dihydrophenazine photocatalyst, a radical initiator (methyl 2-bromopropionate) and a monomer (isoprene). Time-resolved spectroscopic measurements spanning sub-picosecond to microseconds (i.e., almost 8 orders of magnitude of time) track the formation and loss of key reactive intermediates. These measurements identify both the excited state of the photocatalyst responsible for electron transfer and the radical intermediates participating in propagation reactions, as well as quantifying their lifetimes. The outcomes connect the properties of N,N-diaryl dihydrophenazine organic photocatalysts with the rates of sequential steps in the catalytic cycle.

Short-lived intermediates are tracked in real-time by transient absorption spectroscopy during a multi-step photoredox catalysed polymerization reaction.  相似文献

The purpose of this work is to investigate the electropolishing of medical-grade 316 L stainless steel to obtain a clean, smooth, and defect-free surface in preparation for surface nano-texturing. Electropolishing of steel was conducted under stationary conditions in four electrolyte mixtures: (A) 4.5 M H2SO4 + 11 M H3PO4, (B) 7.2 M H2SO4 + 6.5 M H3PO4, (C) 6.4 M glycerol + 6.1 M H3PO4, and (D) 6.1 M H3PO4. The influence of electrolyte composition and concentration, temperature, and electropolishing time, in conjunction with linear sweep voltammetry and chronoamperometry, on the stainless steel surface was studied. The resulting surfaces of unpolished and optimally polished stainless steel were characterized in terms of contamination, defects, topography, roughness, hydrophilicity, and chemical composition by optical and atomic force microscopies, contact angle goniometry, and X-ray photoelectron spectroscopy. It was found that the optimally polished surfaces were obtained with the following parameters: electrolyte mixture A at 2.1 V of applied potential at 80 °C for 10 min. This corresponded to the diffusion-limited dissolution of the surface. The root mean square surface roughness of the electropolished surface achieved was 0.4 nm over 2 × 2 μm2. Surface analysis showed that electropolishing led to ultraclean surfaces with reduced roughness and contamination thickness and with Cr, P, S, Mo, Ni, and O enrichment compared to untreated surfaces.  相似文献

In this paper we report a new method for the electrochemical deposition of a metal/polymer composite layer on a conducting substrate. The electrochemical solution is a mixture of an acrylate (ethyl acrylate, EA; 2-phenyl-2-(2,2,6,6-tetramethylpiperidin-1-yloxy)ethyl acrylate, PTEA; 8-quinolinyl acrylate, 8QA), a metallic salt (silver(I) acetate), and a conducting salt in dimethylformamide. The process has been first studied with EA as the polymer precursor and then extended to PTEA and 8QA, respectively, with the purpose to prepare antibacterial surfaces. The final coating has been characterized by attenuated total reflection Fourier transform infrared spectroscopy, energy-dispersive X-ray analysis, environmental scanning electron microscopy, and atomic force microscopy. All the silver-containing coatings were effective against Gram-negative bacteria Escherichia coli. Bacteria Staphylococcus aureus could not adhere to the Ag(0)/polyacrylate films deposited on stainless steel.  相似文献

We report on the fabrication and performances of a solid-phase microextraction (SPME) fiber based on a stainless steel wire coated with a covalently attached polyacrylonitrile (PAN)/multi-walled carbon nanotubes (MWCNTs) composite. This new coating is obtained by atom transfer radical polymerization (ATRP) of acrylonitrile mixed with MWCNTs. ATRP is initiated from 11-(2-bromo-2-methylpropionyloxy)-undecyl-phosphonic acid molecules grafted on the wire surface via the phosphonic acid group. The extraction performances of the fibers are assessed on different classes of compounds (polar, non-polar, aromatic, etc.) from water solutions by headspace extraction. The optimization of the parameters affecting the extraction efficiency of the target compounds was studied as well as the reproducibility and the repeatability of the fiber. The fibers sustain more than 200 extractions during which they remain chemically stable and maintain good performances (detection limits lower than 2 μg/l, repeatability, etc.). Considering their robustness together with their easy and inexpensive fabrication, these fibers could constitute promising alternatives to existing products.  相似文献

Compound Al(OiPr)3 is shown to catalyze the halide-exchange process leading from [Mo(Cp)Cl2(iPrN=CH-CH=NiPr)] and CH3CH(X)COOEt (X=Br, I) to the mixed-halide complexes [Mo(Cp)ClX(iPrN=CH-CH=NiPr)]. On the other hand, no significant acceleration is observed for the related exchange between [MoX3(PMe3)3] (X=Cl, I) and PhCH(Br)CH3, by analogy to a previous report dealing with the Ru(II) complex [RuCl2(PPh3)3]. A DFT computation study, carried out on the model complexes [Mo(Cp)Cl2(PH3)2], [MoCl3(PH3)3], and [RuCl2(PH3)3], and on the model initiators CH3CH(Cl)COOCH3, CH3Cl, and CH3Br, reveals that the 16-electron Ru(II) complex is able to coordinate the organic halide RX in a slightly exothermic process to yield saturated, diamagnetic [RuCl2(PH3)3(RX)] adducts. The 15-electron [MoCl3(PH3)3] complex is equally capable of forming an adduct, that is, the 17-electron [MoCl3(PH3)3(CH3Cl)] complex with a spin doublet configuration, although the process is endothermic, because it requires an energetically costly electron-pairing process. The interaction between the 17-electron [Mo(Cp)Cl2(PH3)2] complex and CH3Cl, on the other hand, is repulsive and does not lead to a stable 19-electron adduct. The [RuCl2(PH3)3(CH3X)] system leads to an isomeric complex [RuClX(PH3)3(CH3Cl)] by internal nucleophilic substitution at the carbon atom. The transition state of this process for X=Cl (degenerate exchange) is located at lower energy than the transition state required for halogen-atom transfer leading to [RuCl3(PH3)3] and the free radical CH3. On the basis of these results, the uncatalyzed halide exchange is interpreted as the result of a competitive S(N)i process, whose feasibility depends on the electronic configuration of the transition-metal complex. The catalytic action of Al(OiPr)3 on atom-transfer radical polymerization (and on halide exchange for the 17-electron half-sandwich Mo(III) complex) results from a more favorable Lewis acid-base interaction with the oxidized metal complex, in which the transferred halogen atom is bound to a more electropositive element. This conclusion derives from DFT studies of the model [Al(OCH3)3]n (n=1,2,3,4) compounds, and on the interaction of Al(OCH3)3 with CH3Cl and with the [Mo(Cp)Cl3(PH3)2] and [RuCl3(PH3)3] complexes.  相似文献
