MULVADO is a newly developed software package for DOSY NMR data processing, based on multivariate curve resolution (MCR), one of the principal multivariate methods for processing DOSY data. This paper will evaluate this software package by using real-life data of materials used in the printing industry: two data sets from the same ink sample but of different quality. Also a sample of an organic photoconductor and a toner sample are analysed. Compared with the routine DOSY output from monoexponential fitting, one of the single channel algorithms in the commercial Bruker software, MULVADO provides several advantages. The key advantage of MCR is that it overcomes the fluctuation problem (non-consistent diffusion coefficient of the same component). The combination of non-linear regression (NLR) and MCR can yield more accurate resolution of a complex mixture. In addition, the data pre-processing techniques in MULVADO minimise the negative effects of experimental artefacts on the results of the data. In this paper, the challenges for analysing polymer samples and other more complex samples will also be discussed.  相似文献

DOSY-NMR experiments were used to characterize two structural isomers that have different shapes, but identical mass fragmentation patterns, from an organic mixture. It is shown that the spherical molecule diffuses faster than the ellipsoidal one. This distinct behaviour is tentatively explained in terms of microfriction effects from the solvent. Finally, to increase the resolution and efficiency of the 2D DOSY map construction, the data were treated with the regularized resolvent transform method. This method provides good separation for mildly overlapped peaks and requires fewer points than other classical DOSY processing methods.  相似文献

A new experiment for the single‐scan measurement of diffusion coefficients is presented. The principle is to introduce a spatial variation in the parameters of a conventional pulse sequence, so that all of the scans required to determine some physical parameter can be recorded simultaneously from different parts of the sample. The spectrum is acquired in the presence of a weak read gradient so that the resulting lineshapes contain the information required. The pulse sequence is described in detail and demonstrated on a sample containing three components; its advantages and limitations are discussed in relation to those of existing techniques. For uncrowded spectra with high signal‐to‐noise ratio, this experiment provides an order of magnitude reduction in experiment time compared with conventional methods and is likely to be of most benefit where samples are changing rapidly with time or where a long period of polarization, which may be difficult to reproduce accurately, prohibits the use of multiple‐scan techniques. Herein, we present the results obtained from our studies on supramolecular self‐assembly and molecular mobility of low‐molecular‐weight gelators (LMWGs) in organic solvents using pulsed field gradient (PFG) diffusion ordered spectroscopy (DOSY) NMR. A series of concentration‐dependent DOSY NMR experiments were performed on selected LMWGs to determine the critical gelation concentration (CGC) as well as to understand the behaviour of the gelator molecules in the gel state. In addition, variable‐temperature DOSY NMR experiments were performed to determine the gel‐to‐sol transition. The PFG NMR experiments performed as a function of gradient strength were further analyzed using monoexponential DOSY processing, and the results were compared with the automated Bayesian DOSY transformation to obtain 2D plots. Our results provide useful information on the stepwise self‐assembly of small molecules leading to gelation. We believe that the results obtained from these experiments are applicable in determining the CGC and gel melting temperatures of supramolecular gels. We present a simple and an efficient approach using spatially selective NMR to investigate solvation and diffusion of CO2 in ionic liquids. The techniques demonstrated here are shown as novel and effective means of studying solvated gas dynamics under non‐equilibrium conditions without the need for conventional high power gradients. Diffusion-ordered spectroscopy (DOSY) is a powerful tool for investigating mixtures and identifying peaks of chemical components. However, similar diffusion coefficients of the components, particularly for complex mixtures that contain crowded resonances, limit resolution and restrict application of the DOSY technique. In this paper, matrix-assisted DOSY were used to explore whether the diffusion resolution of a complex model involving indole alkaloid mixtures can be realized. Furthermore, we investigated the influence of different factors on the separation effect. The results showed that the changes in diffusion coefficient differences were achieved more obviously when using sodium dodecyl sulfate (SDS) micelles as the matrix. In addition, we also found that increasing the concentration of SDS can improve the resolution of the DOSY spectrum. Finally, after investigating the influence factors and NMR conditions, we demonstrated the applications of the SDS-assisted DOSY on analyzing the total alkaloid extract of Alstonia Mairei, and the virtual separation of mixtures was achieved.  相似文献

This article demonstrates the utility of DOSY NMR for the determination of the optimal conditions for the efficient covalent, reversible cross‐linking of macromolecules in water for hydrogel formation. The studied model system was hyperbranched polyglycidol (HbPGL) containing numerous diol groups in peripheral regions and two types of boronic acids, that is, B(OH)4? and benzene‐1,4‐boronic diacid, as cross‐linking agents. Diffusion coefficient changes of a polymer in solution, under the influence of various concentrations of cross‐linking agent and pH, which influences the equilibrium of the reaction between boronic acids and diols, were recorded. These data are consistent with the rheological properties, namely the Gmax(ω) of hydrogels prepared under analogous conditions, from more concentrated solutions of HbPGL. This approach appears to be promising as it facilitates avoiding the loss of a large amount of polymer that is necessary for the elaboration of appropriate conditions for network formation in aqueous media. © 2016 Wiley Periodicals, Inc. Diffusion-ordered spectroscopy (DOSY) NMR is based on a pulse-field gradient spin-echo NMR experiment, in which components experience diffusion. Consequently, the signal of each component decays with different diffusion rates as the gradient strength increases, constructing a bilinear NMR data set of a mixture. By calculating the diffusion coefficient for each component, it is possible to obtain a two-dimensional NMR spectrum: one dimension is for the conventional chemical shift and the other for the diffusion coefficient. The most interesting point is that this two-dimensional NMR allows non-invasive “chromatography” to obtain the pure spectrum for each component, providing a possible alternative for LC-NMR that is more expensive and time-consuming. Potential applications of DOSY NMR include identification of the components and impurities in complex mixtures, such as body fluids, or reaction mixtures, and technical or commercial products, e.g. comprising polymers or surfactants.

Data processing is the most important step to interpret DOSY NMR. Single channel methods and multivariate methods have been proposed for the data processing but all of them have difficulties when applied to real-world cases. The big challenge appears when dealing with more complex samples, e.g. components with small differences in diffusion coefficients, or severely overlapping in the chemical shift dimension. Two single channel methods, including SPLMOD and continuous diffusion coefficient (CONTIN), and two multivariate methods, called direct exponential curve resolution algorithm (DECRA) and multivariate curve resolution (MCR), are critically evaluated by simulated and real DOSY data sets. The assessments in this paper indicate the possible improvement of the DOSY data processing by applying iterative principal component analysis (IPCA) followed by MCR-alternating least square (MCR-ALS).  相似文献

2D DOSY 1H NMR has proved to be a useful technique in the identification of the molecular skeleton of the four major compounds of ethyl acetate extract of aerial parts of Bidens sulphurea (Asteraceae). The combination of this technique with HPLC, mass spectrometry and other NMR techniques enabled the identification of four flavonoid glycosides: quercetin‐3‐O‐β‐D ‐galactopyranoside, quercetin‐3‐O‐β‐D ‐glycopyranoside, quercetin‐3‐O‐α‐L ‐arabinofuranoside and quercetin‐3‐O‐β‐D ‐rhamnopyranoside. Diffusion‐ordered multidimensional NMR spectroscopy is a valuable technique for the analysis of complex chemical mixtures. However, this method is very time‐consuming because of the costly sampling of a multidimensional signal. Various sparse sampling techniques have been proposed to accelerate such measurements, but they have always been limited to frequency dimensions of NMR spectra. This study proposes a new correlation equation between surfactant diffusion coefficients and molecular mass using 2D DOSY 1H NMR experiment. Indeed, DOSY is a suitable NMR technique to discriminate the different components in a mixture. Using Inverse Laplace Transform (ILT), all chemical shifts in the direct dimension are dispersed along the second dimension in relation to the diffusion coefficient. This approach does not make any assumption about the number of exponential components. The empiric equation was determined for anionic and non-ionic surfactants dissolved in dimethylsulfoxide (DMSO). The parameters, K = Cr = 5.18 ± 0.70 and a shape factor α = 0.45 ± 0.02 that is related to the molecular family fractal dimension constant δF = 2.21 ± 0.11, have been extracted from the data. The equation validity was checked for simple commercial raw material and a simple mixture composed of three surfactants classically used in detergent formulations. This equation, which describes quite satisfactorily the diffusion phenomenon of surfactants, could be used for deformulation purpose to check commercial products in case of quality control or adulteration assessment.  相似文献

Macrolide antibiotics, such as azithromycin and erythromycin, are in widespread use for the treatment of bacterial infections. Macrolides are taken up and excreted mainly by bile. Additionally, they have been implicated in biliary system diseases and to modify the excretion of other drugs through bile. Despite mounting evidence for the interplay between macrolide antibiotics and bile acids, the molecular details of this interaction remain unknown. Herein, we show by NMR measurements that macrolides directly bind to bile acid micelles. The topology of this interaction has been determined by solvent paramagnetic relaxation enhancements (solvent PREs). The macrolides were found to be bound close to the surface of the micelle. Increasing hydrophobicity of both the macrolide and the bile acid strengthen this interaction. Both bile acid and macrolide molecules show similar solvent PREs across their whole structures, indicating that there are no preferred orientations of them in the bile micelle aggregates. The binding to bile aggregates does not impede macrolide antibiotics from targeting bacteria. In fact, the toxicity of azithromycin towards enterotoxic E. coli (ETEC) is even slightly increased in the presence of bile, as was shown by effective concentration (EC50) values.  相似文献

Inorganic polyphosphates (polyPs) have been identified in eukaryotic and prokaryotic cells alike. Various extraction methods have been optimized as a necessary step before identification and measurement of these polymers. Three commercially available sodium polyP glasses were either dissolved or dissolved and extracted by two commonly used polyP extraction techniques – perchloric acid or buffered phenol–chloroform. The products were separated by polyacrylamide gel electrophoresis (PAGE), stained with toluidine blue O, and the migration results quantitatively compared. Both extraction processes reduced the relative migration distances of the peak and leading edges, and the stained band lengths, suggesting reduced polyP migration and dispersion. 31P diffusion-ordered spectroscopy nuclear magnetic resonance confirmed that polyP extraction by perchloric acid or phenol–chloroform processes reduced polyP diffusion coefficients and suggested hydrolytic degradation with stronger end-chain signals. Reduced polyP diffusivity after extraction makes possible an overestimation of synthetic polyP chain length assignment when compared to unextracted polyP ladders with PAGE. The mechanism(s) for reduced synthetic polyP diffusion after extraction and intracellular chemical environment effects on migration are not known.  相似文献

The translational diffusion coefficients of various helicates have been determined by using NMR diffusion spectroscopy (Diffusion Ordered SpectroscopY, DOSY), in order to investigate the individual behaviour of the helicates according to their length (different nuclearities; 1-5 metals), to the nature of the metal involved (CuI or AgI), as well to their bulkiness due to the presence of substituents on the periphery of the assembly. Furthermore, the spectrum of a mixture of helicates belonging to the same series, but with different lengths and nuclearities, showed the signals of each component, with no observable cross-linking, confirming the self-recognition properties of the helicates.  相似文献

Single scan longitudinal relaxation measurement experiments enable rapid estimation of the spin‐lattice relaxation time (T1) as the time series of spin relaxation is encoded spatially in the sample at different slices resulting in an order of magnitude saving in time. We consider here a single scan inversion recovery pulse sequence that incorporates a gradient echo sequence. The proposed pulse sequence provides spectra with significantly enhanced signal to noise ratio leading to an accurate estimation of T1 values. The method is applicable for measuring a range of T1 values, thus indicating the possibility of routine use of the method for several systems. A comparative study of different single scan methods currently available is presented, and the advantage of the proposed sequence is highlighted. The possibility of the use of the method for the study of cross‐correlation effects for the case of fluorine in a single shot is also demonstrated. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献
