Summary An ICP-MS detector in combination with HPLC has been evaluated for the analysis of six arsenic compounds. The influence of the presence of an organic modifier in the mobile phase on arsenic response and the quality parameters of the analysis are discussed. Detection limits for arsenic species under study range from 10 to 30 pg. The determination of arsenic compounds in solutions simulating fish or sediment extracts has been used to demonstrate the applicability of this technique.  相似文献

It is known that arsenic has different toxicological properties dependent upon both its oxidation state for inorganic compounds, as well as the different toxicity levels exhibited for organic arsenic compounds. The field of arsenic speciation analysis has grown rapidly in recent years, especially with the utilization of high-performance liquid chromatography (HPLC) coupled to inductively coupled plasma mass spectrometry (ICP-MS), a highly sensitive and robust detector system. Complete characterization of arsenic compounds is necessary to understand intake, accumulation, transport, storage, detoxification and activation of this element in the natural environment and living systems. This review describes the essential background and toxicity of arsenic in the environment, and more importantly, some currently used chromatographic applications and sample handling procedures necessary to accurately detect and quantify arsenic in its various chemical forms. Applications and work using only HPLC-ICP-MS for arsenic speciation of environmental and biological samples are presented in this review.  相似文献

There is considerable evidence that toxicity and physiological behavior of arsenic depends on its chemical forms. Arsenic speciation became therefore the subject of increasing interest in recent years. A sensitive method for the determination of arsenic species has been developed. The proposed procedure involves the use of high-performance liquid chromatography and inductively coupled plasma mass spectrometry (HPLC-ICP-MS). Six arsenic compounds were separated by anion-exchange chromatography with isocratic elution using tartaric acid as mobile phase with an elution order: arsenocholine, arsenobetaine, dimethylarsinic acid, methylarsonic acid, arsenous acid and arsenic acid. The chromatographic parameters affecting the separation of the arsenic species were optimized. Analytical characterization of the method has been realized with standard solutions. The detection limits for six arsenic compounds were from 0.04 to 0.6 g/L as As element. The repeatability (expressed by R.S.D) was better than 7% for all investigated compounds. The HPLC-ICP-MS system was successfully applied to the determination of arsenic compounds in environmental and biological samples in g/L level.  相似文献

In this study, selenium species from Se containing proteins in mushrooms (Agaricus bisporus and Lentinula edodes) were investigated with size-exclusion liquid chromatography coupled to UV and inductively coupled plasma mass spectrometry (ICP-MS). Different protein extraction protocols were investigated. Variability of the fractionation patterns with three extraction media (0.1M NaOH, 30 mM Tris-HCl, and enzymatic digestions) was evaluated for both mushroom types. A 24 h Tris-HCl extraction followed by acetone addition was found to be optimal for protein precipitation. Presumably protein bound selenoamino acids were released using enzymes (proteinase K, protease XIV and trypsin). The selenium speciation of the proteolytic extract of the water soluble proteins fraction was carried out by using reversed-phase ion-pairing high performance liquid chromatography (RP-HPIPC) coupled on-line to ICP-MS for selenium specific detection. Selenocystine, selenomethionine, methylselenocysteine and inorganic selenium were established in both samples utilizing retention time standards and standard additions to the sample.  相似文献

建立了用反相离子对色谱和电感耦合等离子体质谱的联用技术同时测定As(Ⅲ)、 As(Ⅴ)、 MMA、 DMA、 Se(Ⅳ)、 Se(Ⅵ)、 SeMet和SeCys的砷、硒化学形态分析方法. 分别从流动相pH值、离子对试剂的浓度、甲醇量和流速4个方面进行了分离测定条件的优化. 利用碰撞池技术(CCT)较好地解决了^40Ar^35Cl^+复合离子对^75As的干扰, 并使^80Se的测定成为可能, 有效地提高了灵敏度. 将该方法应用于上海市苏州河底泥样品的微波辅助萃取液的形态分析中, 砷和硒的检出限分别达到0.4~1.3 和0.5~1.9 μg/L.  相似文献

Nanoliter high‐performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high‐performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high‐pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high‐performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high‐performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9–1.8 μg/L were obtained with precisions variable in the range of 1.6–4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87–102%).  相似文献

Inductively coupled plasma mass spectrometry (ICP/MS) is utilized as a detector for several organotin species separated by high-performance liquid chromatography. Detection limits obtained by ICP/MS are 3 orders of magnitude lower than those obtained with inductively coupled plasma atomic emission spectrometry (ICP/AES) detection under the same chromatographic conditions. Chromatographic detection limits are higher than conventional solution nebulization for the same compound by a factor of 20. Ion-exchange chromatography yields linear response over 3 orders of magnitude, while ion pair chromatography gives a linear response of only 2 orders of magnitude as a result of poor resolution. The relative standard deviation for the injection of 20 ng of tin compounds is less than 10%.  相似文献

Sulfur mustard (HD), bis(2-chloroethyl)sulfide, is one of a class of mustard agents which are chemical warfare agents. The main chemical warfare hydrolysis degradation products of sulfur mustards are: thiodiglycol, bis(2-hydroxyethylthio)methane, 1,2-bis(2-hydroxyethylthio)ethane, 1,3-bis(2-hydroxyethylthio)propane, and 1,4-bis(2-hydroxyethylthio)butane. The aim of this study is to identify these five hydrolysis degradation products utilizing reversed-phase high-performance liquid chromatography coupled with inductively coupled plasma mass spectrometry (ICP-MS) for element-specific sulfur detection using a collision/reaction cell and electrospray ionization mass spectrometry to confirm the identification. To date, this is the first study utilizing ICP-MS with 32S element-specific detection for the analysis of vesicant chemical warfare agent degradation products.  相似文献

A novel interface design for coupling gas chromatography and inductively coupled plasma mass spectrometry (GC/ICP-MS) was used to perform mercury speciation in biological tissues. Three derivatization approaches were optimized and compared for this purpose: anhydrous butylation using a Grignard reagent, aqueous ethylation by means of NaEt(4)B and aqueous propylation with NaPr(4)B. The last reagent was synthesized in the laboratory as it is not commercially available. Detection limits obtained by GC/ICP-MS ranged between 100 and 200 fg (as absolute mass) for methylmercury and between 500 and 600 fg for inorganic mercury using a 1 microl injection. Quantification of methyl- and inorganic mercury was carried out by resorting to aqueous calibration, using ethylmercury as internal standard for both propylation and butylation derivatization techniques. For ethylation procedures, a methylpropylmercury solution was used as internal standard. The absence of transmethylation during sample preparation was checked using a 97% enriched (202)Hg inorganic standard. The accuracy of the three derivatization approaches was evaluated by the analysis of the certified reference material DOLT-2 (dogfish liver) from the National Research Council of Canada and certified for methylmercury, with satisfactory results.  相似文献

The use of high-performance liquid chromatography/inductively coupled plasma mass spectrometry (HPLC/ICPMS) with sulphur-specific detection was investigated as a method for obtaining metabolite profiles for the drug omeprazole administered as a 1:1 mixture of (32)S- and (34)S-labelled material. Analysis based on the monitoring of the chromatographic eluent at either m/z 32 or 34 was not successful due to insufficient sensitivity caused by interferences from polyatomic ions. However, reaction of sulphur with oxygen in the hexapole collision cell, combined with monitoring at m/z 48 (for (32)S) or m/z 50 (for (34)S), provided a facile method for metabolite profiling. Detection of m/z 48 was superior in sensitivity to detection of m/z 50.  相似文献

Iodine determination in food samples was performed by inductively coupled plasma mass spectrometry (ICP-MS) after digestion by microwave-induced combustion (MIC). Sample masses up to 500 mg of bovine liver, corn starch, milk powder, or wheat flour were completely combusted using the MIC system. Ammonium nitrate (6 mol l−1 solution, 50 μl) was used as an aid for ignition and vessels were charged with 15 bar of O2. The use of H2O, 0.9 mmol l−1 H2O2, 10 to 50 mmol l−1 (NH4)2CO3 and 56 mmol l−1 tetramethylammonium hydroxide was investigated as absorbing solutions, as well as the suitability of performing a reflux step after the combustion process. Digestion of food samples by pressurized microwave-assisted acid digestion, microwave-assisted extraction and conventional extraction of iodine in alkaline solution were also evaluated. Iodine recoveries higher than 99% were obtained using MIC and 50 mmol l−1 (NH4)2CO3 or 56 mmol l−1 tetramethylammonium hydroxide as absorbing solution and with 5 min for the reflux step. Accuracy was evaluated using certified reference materials (bovine muscle, corn bran, and milk powder) and agreement better than 97% was obtained. The limit of quantification by MIC and further ICP-MS determination was 0.002 μg g−1. Blanks were always low and no memory effects were observed. Digestion by MIC allowed the processing of up to eight samples by each run in 25 min with high efficiency of digestion (residual carbon content lower than 1%) providing a suitable medium for further iodine determination by ICP-MS.  相似文献

A reliable and sensitive method for determination simultaneously of monomethylmercury (MeHg) and monoethylmercury (EtHg) in various types of foods by gas chromatography inductively coupled plasma mass spectrometry (GC-ICP/MS) was developed and validated. Samples were digested with pancreatin and then hydrochloric acid. MeHg and EtHg in the extract were derivatized in an aqueous buffer with sodium tetraphenylborate. After phase separation, the extract was directly transferred to analysis. The analyses were conducted by GC-ICP/MS with monopropylmercury chloride (PrHgCl) as surrogate standard. Concentrations of 254±5.1, 13.7±0.69 and 162±6.2 μg Hg kg(-1) (one standard deviation, n=3) were obtained for MeHg in NIST SRM 1947 (Superior Lake fish), SRM 1566b (oyster tissue) and NRC Tort-2 (lobster Hepatopancreas), respectively. These are in good agreement with the certified values of 233±10, 13.2±0.7 and 152±13 μg Hg kg(-1) (as 95% confidence interval), respectively. The method detection limits (3σ) for MeHg and EtHg are 0.3 μg Hg kg(-1). The method detection limit was estimated by using a 0.5 g of subsample, sufficiently low for the risk assessment of MeHg and EtHg in foods. The spiked recoveries of MeHg and EtHg in different food matrices were between 87 and 117% and the RSDs were less than 15%. When isotopic dilution mass spectrometry (IDMS) analysis was performed with a commercial available (201)Hg-enriched monomethylmercury (Me(201)Hg) solution as internal standard, concentrations of 244±13.4, 13.9±0.25 and 161±1.3 μg Hg kg(-1) were obtained for MeHg in NIST SRM 1947, SRM 1566b and NRC Tort-2, respectively. It shown clearly that IDMS analysis got improvement in precision and accuracy, however, EtHg cannot be analyze simultaneously and the cost of analysis is higher.  相似文献

A procedure for overcoming polyatomic interferences in liquid chromatography–quadrupole inductively coupled plasma mass spectrometry based on the mathematical correction point by point along the chromatogram is described. Correction factors, that are part of the elemental equations used for mathematical correction, were calculated from the height or the area of the peaks obtained after measurement of interference stock solutions by flow injection. Due to the sequential nature of quadrupole mass spectrometers, the selection of the acquisition parameters with respect to the peak width and the number of isotopes monitorised is critical. A sampling frequency which allows getting a minimum of 20 points per peak is recommended to avoid artifacts in the corrected profiles of the peaks. The effect of polyatomic interferences from carbon and chlorine on the determination and speciation of chromium by high performance size exclusion chromatography coupled to quadrupole inductively coupled plasma mass spectrometry in compost lixiviates is studied. Biased results of + 50% are successfully corrected by the proposed procedure.  相似文献

Of all the inorganic mass spectrometric techniques, laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) plays a key role as a powerful and sensitive microanalytical technique enabling multi- element trace analysis and isotope ratio measurements at trace and ultratrace level. LA-ICP-MS was used to produce images of detailed regionally-specific element distribution in 20 microm thin sections of different parts of the human brain. The quantitative determination of copper, zinc, lead and uranium distribution in thin slices of human brain samples was performed using matrix-matched laboratory standards via external calibration procedures. Imaging mass spectrometry provides new information on the spatially inhomogeneous element distribution in thin sections of human tissues, for example, of different brain regions (the insular region) or brain tumor tissues. The detection limits obtained for Cu, Zn, Pb and U were in the ng g(-1) range. Possible strategies of LA-ICP-MS in brain research and life sciences include the elemental imaging of thin slices of brain tissue or applications in proteome analysis by combination with matrix-assisted laser desorption/ionization MS to study phospho- and metal- containing proteins will be discussed.  相似文献

Several approaches to the determination of selenomethionine, selenocystine, selenite and selenate by high-performance liquid chromatography with online detection by atomic absorption spectrometry are described. The N?2,4-dinitrophenyl derivatives of selenomethionine, selenoethionine, selenocystine and phenylmercury(II) cystineselenoate were recovered from aqueous solution, separated on a Nucleosil 5-NO2 reversed-phase HPLC column with a methanolic mobile phase containing acetic acid and triethylamine, and detected with a quartz thermochemical hydride-generating interface–atomic absorption spectrometry (AA) system. The restriction of having to perform chromatography with an organic mobile phase (to support the combusion process) was overcome with a new interface design capable of operation with either organic or aqueous HPLC mobile phases. Using aqueous acetic acid (0.015% v/v) containing 0.1% (w/v) ammonium acetate delivered at 0.5cm3 min?1, selenate, selenite, selenomethionine, selenocystine and selenoethionine were separated virtually to baseline on a cyanopropyl-bonded phase HPLC column. Other selenium compounds which were investigated included methane seleninic and methane selenonic acids as well as the crude oxidation product mixtures resulting from the treatment of selenomethionine and selenocystine with hydrogen peroxide. A procedure for extracting selenate, selenite, selenomethionine, selenocystine and selenoethionine from spiked water or ground feed supplement into liquefied phenol resulted in acceptable recoveries for the latter four analytes but was unacceptably low for selenate.  相似文献

Selenium exists in several oxidation states and a variety of inorganic and organic compounds, and the chemistry of selenium is complex in both the environment and living systems. Selenium is an essential element at trace levels and toxic at greater levels. Interest in speciation analysis for selenium has grown rapidly in this last decade, especially in the use of chromatographic separation coupled with inductively coupled plasma-mass spectrometry (ICP-MS). Complete characterization of selenium compounds is necessary to understand selenium's significance in metabolic processes, clinical chemistry, biology, toxicology, nutrition and the environment. This review describes some of the essential background of selenium, and more importantly, some of the currently used separation methodologies, both chromatographic and electrophoretic, with emphasis on applications of selenium speciation analysis using ICP-MS detection.  相似文献

This work investigates for the first time the potential of mixed-mode (anion-exchange with reversed-phase) high performance liquid chromatography coupled to inductively coupled plasma mass spectrometry (ICP-MS) for the simultaneous retention and selective separation of a range of inorganic and organically-bound selenium (Se) species. Baseline separation and detection of selenocystine (SeCys2), Se-methyl-selenocysteine (SeMC), selenomethionine (SeMet), methylseleninic acid (MSA), selenite, γ-glutamyl-methyl-selenocysteine (γ-glutamyl-SeMC), and selenate in a Se standard mixture by mixed-mode HPLC-ICP-MS was achieved by switching between two citrate mobile phases of different pH and ionic strength within a single chromatographic run of 20 min. Limits of detection obtained for these Se species ranged from 80 ng kg?1 (for SeMC) to 123 ng kg?1 (for selenate). Using this approach as developed for selenium speciation, an adequate separation of inorganic and organic As compounds was also achieved. These include arsenite, arsenate, arsenobetaine (AsB) and dimethylarsenic acid (DMA), which may coexist with Se species in biological samples. Application of the newly proposed methodology to the investigation of the elemental species distribution in watercress (used as the model sample) after enzymatic hydrolysis or leaching in water by accelerated solvent extraction (ASE) was addressed. Only SeMet, SeMC and selenate could be tentatively identified in watercress extracts by mixed-mode HPLC-ICP-MS and retention time matching with standards. Recoveries (n = 3) of these Se species from samples spiked with standards averaged 102% (for SeMC), 94.9% (for SeMet) and 98.3% (for selenate). Verification of the presence of SeMet and SeMC in an enzymatic watercress extract was achieved by on-line HPLC-ESI MS/MS in selected reaction monitoring (SRM) mode.  相似文献

This work describes an arsenic speciation analysis in aqueous effluent from a shale industrial plant using liquid chromatography coupled to inductively coupled plasma mass spectrometry (LC–ICP–MS). Arsenic species have been separated through an anion-exchange column and several parameters investigated, such as retention time, pH, flow rate and concentration of the mobile phase (ammonium carbonate), chloride interference and column conditioning time. The best conditions have been found by fixing the pH of the mobile phase at 8.7. Keeping the mobile phase flow rate at 1.5 ml min− 1, arsenic species were separated by varying the concentration of the mobile phase and the time of elution, as follow: 1.5 mmol l− 1 for 10 min, 12 mmol l− 1 for 10 min and 20 mmol l− 1 for 10 min, respectively. Up to 13 As species present in the samples were separated under these conditions and the following species could be identified and quantified: arsenite [As(III)], dimethylarsinic acid (DMA), monomethylarsonic acid (MMA) and arsenate [As(V)]. The limits of detection of the LC–ICP–MS method were 0.02, 0.06, 0.04 and 0.10 μg l− 1 of As(III), DMA, MMA, and As(V), respectively. The concentration of these species in the samples were from 3.7 to 6.4 μg l− 1, 6.9 to 13.2 μg l− 1, 100 to 142 μg l− 1 and 808 to 1363 μg l− 1 for As(III), DMA, MMA and As(V), respectively. The accuracy, evaluated by recovery tests, varied from 94 to 105% and the precision, evaluated by the relative standard deviation was typically lower than 10%.  相似文献

Achievement of optimum selectivity, sensitivity and robustness in speciation analysis using high performance liquid chromatography (HPLC) with inductively coupled mass spectrometry (ICP-MS) detection requires that each instrumental component is selected and optimized with a view to the ideal operating characteristics of the entire hyphenated system. An isocratic HPLC system, which employs an aqueous mobile phase with organic buffer constituents, is well suited for introduction into the ICP-MS because of the stability of the detector response and high degree of analyte sensitivity attained. Anion and cation exchange HPLC systems, which meet these requirements, were used for the seperation of selenium and arsenic species in crude extracts of biological samples. Furthermore, the signal-to-noise ratios obtained for these incompletely ionized elements in the argon ICP were further enhanced by a factor of four by continously introducing carbon as methanol via the mobile phase into the ICP. Sources of error in the HPLC system (column overload), in the sample introduction system (memory by organic solvents) and in the ICP-MS (spectroscopic interferences) and their prevention are also discussed. The optimized anion and cation exchange HPLC-ICP-MS systems were used for arsenic speciation in contaminated ground water and in an in-house shrimp reference sample. For the purpose of verification, HPLC coupled with tandem mass spectrometry with electrospray ionization was additionally used for arsenic speciation in the shrimp sample. With this analytical technique the HPLC retention time in combination with mass analysis of the molecular ions and their collision-induced fragments provide almost conclusive evidence of the identity of the analyte species. The speciation methods are validated by establishing a mass balance of the analytes in each fraction of the extraction procedure, by recovery of spikes and by employing and comparing independent techniques. The urgent need for reference materials certified for elemental species is stressed.  相似文献
