Self‐adjuvanting tricomponent vaccines were prepared and assessed for their self‐assembly and immunological activity in mouse models. The vaccines each consisted of a peptide or glycopeptide antigen that corresponds to a complete copy of the variable‐number tandem repeat (VNTR) of the tumor‐associated mucin 1 (MUC1) glycoprotein, the universal T‐cell helper peptide epitope PADRE, and the immunoadjuvant Pam3CysSer. The vaccines were shown to spontaneously self‐assemble in water to form isotropic particles varying in size from 17 to 25 nm and elicited robust humoral responses in murine models without the addition of an external adjuvant. The serum antibodies could recognize tumor‐associated MUC1 epitopes on the surface of MCF7 breast‐cancer cells and B16 melanoma cells, which overexpress this tumor‐associated glycoprotein.  相似文献

Hospital‐acquired infections are an increasingly serious health concern. Infections caused by carpabenem‐resistant Klebsiella pneumoniae (CR‐Kp ) are especially problematic, with a 50 % average survival rate. CR‐Kp are isolated from patients with ever greater frequency, 7 % within the EU but 62 % in Greece. At a time when antibiotics are becoming less effective, no vaccines to protect from this severe bacterial infection exist. Herein, we describe the convergent [3+3] synthesis of the hexasaccharide repeating unit from its capsular polysaccharide and related sequences. Immunization with the synthetic hexasaccharide 1 glycoconjugate resulted in high titers of cross‐reactive antibodies against CR‐Kp CPS in mice and rabbits. Whole‐cell ELISA was used to establish the surface staining of CR‐Kp strains. The antibodies raised were found to promote phagocytosis. Thus, this semi‐synthetic glycoconjugate is a lead for the development of a vaccine against a rapidly progressing, deadly bacterium.  相似文献

A safe and effective vaccine against human immunodeficiency virus type 1 (HIV‐1) is urgently needed to combat the worldwide AIDS pandemic, but still remains elusive. The fact that uncontrolled replication of an attenuated vaccine can lead to regaining of its virulence creates safety concerns precluding many vaccines from clinical application. We introduce a novel approach to control HIV‐1 replication, which entails the manipulation of essential HIV‐1 protein biosynthesis through unnatural amino acid (UAA*)‐mediated suppression of genome‐encoded blank codon. We successfully demonstrate that HIV‐1 replication can be precisely turned on and off in vitro.  相似文献

With advances in polymerization techniques as well as selective chemical modification of carbohydrates, glycopolymers and glyco‐nanoparticles are emerging as an important class of materials with tailored properties or novel nanotechnology‐based platforms for a number of applications. The field of the so‐called glyco‐nanotechnology is starting to show some promises for future clinical applications. Glyco‐nanoparticles, due to their versatile nature, could offer a platform for the design of carbohydrate‐based vaccines and possibly allow the development of new single‐dose vaccines in disease areas of unmet need. This paper surveys the emerging roles of carbohydrate‐based polymeric and nanomaterials for biomolecular recognition processes and vaccine development.

Co‐assembling vaccines composed of a lipidated HER2‐derived antigenic CH401 peptide and either a lipophilic adjuvant, Pam3CSK4, α‐GalCer, or lipid A 506, were evaluated as breast cancer vaccine candidates. This vaccine design was aimed to inherit both antigen multivalency and antigen‐specific immunostimulation properties, observed in reported self‐adjuvanting vaccine candidates, by using self‐assembly and adjuvant‐conjugated antigens. Under vaccination concentrations, respective lipophilic adjuvants underwent co‐assembly with lipidated CH401, which boosted the anti‐CH401 IgG and IgM production. In particular, α‐GalCer was responsible for the most significant immune activation. Therefore, the newly developed vaccine design enabled the optimization of adjuvants against the antigenic CH401 peptide in a simple preparatory manner. Overall, the co‐assembling vaccine design opens the door for efficient and practical self‐adjuvanting vaccine development.  相似文献

Sialyl‐Tn (STn) is a tumor‐associated carbohydrate antigen (TACA) rarely observed on healthy tissues. We synthesized two fully synthetic N‐acetyl and N‐propionyl STn trimer (triSTn) vaccines possessing a T‐helper epitope and a TLR2 agonist, since the clustered STn antigens are highly expressed on many cancer cells. Immunization of both vaccines in mice induced the anti‐triSTn IgG antibodies, which recognized triSTn‐expressing cell lines PANC‐1 and HepG2. The N‐propionyl triSTn vaccine induced the triSTn‐specific IgGs, while IgGs induced by the N‐acetyl triSTn vaccine were less specific. These results illustrated that N‐propionyl triSTn is a valuable unnatural TACA for anticancer vaccines.  相似文献

Many materials as immune adjuvant are researched to help raise immnogenicity of subunit vaccines. Among them, peptide‐based hydrogels are gradually coming into notice because of their application in drugs delivery, cancer cell inhibition, vaccine adjuvants and detection of important analytes. High‐aliphatic‐content linear nylons were produced with an 18‐carbon diacid with diamines containing both odd and even methylene segments. The resulting polymers were characterized with viscosimetric, thermal, and spectroscopic techniques. We have developed and validated a high‐performance liquid chromatography method that uses monolithic silica disk‐packed spin columns and a monolithic silica column for the simultaneous determination of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, and NG,NG′‐dimethyl‐l ‐arginine in human plasma. For solid‐phase extraction, our method employs a centrifugal spin column packed with monolithic silica bonded to propyl benzenesulfonic acid as a cation exchanger. After pretreatment, the methylated arginines are converted to fluorescent derivatives with 4‐fluoro‐7‐nitro‐2,1,3‐benzoxadiazole, and then the derivatives are separated on a monolithic silica column. l ‐Arginine concentration was also determined in diluted samples. Standard calibration curves revealed that the assay was linear in the concentration range 0.2–1.0 μM for methylated arginines and 40–200 μM for l ‐arginine. Linear regression of the calibration curve yielded equations with correlation coefficients of 0.999 (r2). The sensitivity was satisfactory, with a limit of detection ranging from 3.75 to 9.0 fmol for all four compounds. The RSDs were 4.3–4.8% (intraday) and 3.0–6.8% (interday). When this method was applied to samples from six healthy donors, the detected concentrations of NG‐monomethyl‐l ‐arginine, NG,NG‐dimethyl‐l ‐arginine, NG,NG′‐dimethyl‐l ‐arginine and l ‐arginine were 0.05 ± 0.01, 0.41 ± 0.07, 0.59 ± 0.11, and 83.8 ± 30.43 μM (n = 6), respectively.  相似文献

Tumor‐associated carbohydrate antigens (TACAs) are key components of cancer vaccines. A variety of vaccines based on native TACAs such as α‐Tn have shown immunogenicity and protection in preclinical animal studies, however, their weak immunogenicity, low in vivo instability, and poor bioavailability, have discouraged their further evaluations in clinical studies. A new improved vaccine prototype is reported. It is composed of four clustered Tn‐antigen mimetics and a immunogenic peptide epitope that are conjugated to a cyclopeptide carrier. The immunization of mice with this vaccine 1) was safe, 2) induced a strong and long‐lasting Tn‐specific response with IgM/IgG antibodies able to recognize native carbohydrate antigens; 3) produced high titers of IgG1, IgG2a, and IgG3 antibodies; and 4) produced a significant antibody‐dependent regression of tumors and conferred protection. Altogether, these findings pave the way for the clinical development of safe and effective therapeutic vaccines against Tn‐expressing cancers.  相似文献

A series of well‐defined amphiphilic triblock copolymers [polyethylene glycol monomethyl ether]‐block‐poly(ε‐caprolactone)‐block‐poly[2‐(dimethylamino)ethyl methacrylate] (mPEG‐b‐PCL‐b‐PDMAEMA or abbreviated as mPEG‐b‐PCL‐b‐PDMA) were prepared by a combination of ring‐opening polymerization and atom transfer radical polymerization. The chemical structures and compositions of these copolymers have been characterized by Fourier transform infrared spectroscopy, 1H NMR, and thermogravimetric analysis. The molecular weights of the triblock copolymers were obtained by calculating from 1H NMR spectra and gel permeation chromatography measurements. Subsequently, the self‐assembly behavior of these copolymers was investigated by fluorescence probe method and transmission electron microscopy, which indicated that these amphiphilic triblock copolymers possess distinct pH‐dependent critical aggregation concentrations and can self‐assemble into micelles or vesicles in PBS buffer solution, depending on the length of PDMA in the copolymer. Agarose gel retardation assays demonstrated that these cationic nanoparticles can effectively condense plasmid DNA. Cell toxicity tests indicated that these triblock copolymers displayed lower cytotoxicity than that of branched polyethylenimine with molecular weight of 25 kDa. In addition, in vitro release of Naproxen from these nanoparticles in pH buffer solutions was conducted, demonstrating that higher PCL content would result in the higher drug loading content and lower release rate. These biodegradable and biocompatible cationic copolymers have potential applications in drug and gene delivery. © 2010 Wiley Periodicals, Inc. A series of stable organosuperbases, N‐alkyl‐ and N‐aryl‐1,3‐dialkyl‐4,5‐dimethylimidazol‐2‐ylidene amines, were efficiently synthesized from N,N′‐dialkylthioureas and 3‐hydroxy‐2‐butanone and their basicities were measured in acetonitrile. The derivatives with tert‐alkyl groups on the imino nitrogen were found to be more basic than the tBu P1 (pyrr) phosphazene base in acetonitrile. The origin of the high basicity of these compounds is discussed. In acetonitrile and in the gas phase, the basicity of the alkylimino derivatives depends on the size of the substituent at the imino group, which influences the degree of aromatization of the imidazole ring, as measured by 13C NMR chemical shifts or by the calculated ΔNICS(1) aromaticity parameters, as well as on solvation effects. If a wider range of imino‐substituents, including electron‐acceptor substituents, is treated in the analysis then the influence of aromatization is less predominant and the gas‐phase basicity becomes more dependent on the field‐inductive effect, polarizability, and resonance effects of the substituent.  相似文献

The synthesis and molecular characterization of a new amphiphilic conetwork (APCN) designed for silicone hydrogel use is described. The synthesis strategy, outlined in Scheme 1 , calls for the preparation, by the RAFT technique, of a new methacrylate‐telechelic amphiphilic pentablock, MA‐PHEA‐b‐PDMAAM‐b‐PDMS‐b‐PDMAAm‐b‐PHEA‐MA, and its crosslinking to the target APCN. The sketch shows the architecture of the APCN (dotted lines, PDMAAm; solid lines, PDMS; clusters, MA‐based crosslinking sites; see Fig. 3 ). All six synthesis steps proceed smoothly and efficiently, and the products are optically clear, colorless membranes exhibiting properties appropriate for ophthalmic use. © 2007 Wiley Periodicals, Inc. An alkylated semiconducting polymer comprising alternating bithiophene‐[all]‐S,S‐dioxide and aromatic monothiophene units in the polymer backbone was synthesized with the intent of modifying the energy gap and lowest unoccupied molecular orbital for use as a stable n‐type semiconductor. Films spun from this semiconducting polymer were characterized utilizing X‐ray scattering, near edge X‐ray absorption fine structure spectroscopy, ultraviolet photoelectron spectroscopy, and thin‐film field effect transistors to determine how oxidation of the thiophene ring systems impacts the structural and electronic properties of the polymer. The thiophene‐S,S‐dioxide polymers have lower optical and electrical band gaps than corresponding thiophene polymers. X‐ray scattering results indicate that the polymers are well ordered with the π–π stacking distances increased by 0.4 Å relative to analogous thiophene polymers. The electrical stability of these polymers is poor in transistors with a drop in the field effect mobility by approximately one order of magnitude upon addition of just 5% of the thiophene‐S,S‐dioxide unit in a copolymer with thiophene. © 2012 Wiley Periodicals, Inc. The new N‐heterocyclic carbene (NHC) precursors 4, ‐dicyano‐1, ‐dimesityl‐ ( 9 ) and 4, 5‐dicyano‐1, 3‐dineopentyl‐2‐(pentafluorophenyl)imidazoline ( 14 ) were synthesized. The structure of 9 could be determined by X‐ray crystallography. With the 2‐pentafluorophenyl‐substituted imidazolines 9 and 14 , the [AgCl(NHC)], [RhCl(COD)(NHC)], and [RhCl(CO)2(NHC)] complexes [NHC = 4, 5‐dicyano‐1, 3‐dimesitylimidazol‐2‐ylidene ( 3 ) and 4, 5‐dicyano‐1, 3‐dineopentylimidazol‐2‐ylidene ( 4 )] were obtained. Crystal structures of [AgCl( 3 )] ( 15 ), [RhCl(COD)( 3 )] ( 17 ), [RhCl(COD)( 4 )] ( 18 ), and [RhCl(CO)2( 3 )] ( 19 ) were solved and with the crystal data of 19 , the percent buried volume ( %Vbur) of 31.8(±0.1) % was determined for NHC 3 . Infrared spectra of the imidazolines 9 and 14 and of the complexes 15 – 20 were recorded and the CO stretching frequencies of complexes 19 and 20 were used to determine the Tolman electronic parameters of the newly obtained NHCs 3 (TEP: 2060 cm–1) and 4 (TEP: 2061 cm–1), thus proving that 1, 3‐substitution of maleonitrile‐NHCs does not have a significant effect for the high π‐acceptor strength of these carbenes.  相似文献

Anions of enolized heteroaromatic 1,3‐dicarbonyl systems, such as the title compounds 1, 9,14 , and 19 , react in dimethylformamide in the presence of potassium carbonate with diaryl disulfides 2 to yield arylsulfenyl derivatives ( 3, 10, 15, 20 ). The arylthiolate anions 4 formed in this reaction can be oxidized by air to yield the starting disulfides 2 again. Tetraalkylthiuram disulfides 7 react in the same manner to yield dialkylaminothiocarbonylthio derivatives ( 8, 13, 18 ) of the title compounds. Oxidation of the arylsulfenyl derivatives with hydrogen peroxide in sodium hydroxide solution usually leads to sulfoxides ( 5, 11, 16 ), whereas oxidation with peracetic acid affords sulfones ( 6, 12, 17 ).  相似文献
