The mandibular gland in Melipona bicolor workers and queens was studied by scanning and transmission electron microscopy. There is no difference in the gland anatomy between the castes, but the transmission electron microscopy showed variation of the cellular ultrastructure according to the secretory phase of the gland in both castes. Smooth endoplasmic reticulum was abundant in the secretory cells of physogastric queens, indicating that these cells produce lipid secretion that is stored in granules with multi-lamellar bodies. Mitochondrial variations during the cell secretory cycle indicates their participation in the lipid synthesis. After secretion, release in the reservoir lumen through the collecting canals, the secretory cells contain many myelinic bodies, indicative of cellular regression.  相似文献

The mandibular glands of Hymenoptera are structures associated with the mandibles and constitute part of the salivary glands system. Histological studies in workers of Atta sexdens rubropilosa revealed that this gland contains two portions: a secretory and a storage portion or reservoir. Both portions are connected by means of canaliculi. The object of the present work was the study of the ultratructure of the mandibular glands of minima, media and soldier ant of A. s. rubropilosa by TEM techniques. The glands, in the three castes studied, possess a reservoir, constituted by a simple pavementous epithelium surrounded by the cuticular intima and the secretory portion is constituted by cells of rounded shape. The secretory cells, mainly of minima and soldier, were rich in smooth endoplasmic reticulum. The media worker and soldier presented a large number of mitochondria, of varying shape. Well-developed Golgi complexes were also present in the soldiers. The secretory cells in minima, media and soldier were provided with collecting intracellular canaliculi, which were linked to the reservoir through the extracellular portion. The cytoplasm of the canaliculi-forming cell was poor in organelles. In the individuals of the three castes of A. s. rubropilosa, the presence of lipid secretion granules suggested, beyond the other functions, also a possible pheromonal action. The different roles executed by the different insect castes are directly dependent on the glandular products and, consequently, on the secretory cellular characteristics.  相似文献

The eusociality developed in Hymenoptera and Isoptera is driven by an efficient interaction between exocrine glands and jointed appendages, both in close interaction with the environment. In this context, the mandible of ants plays an important role, since, in addition to being the main jointed appendage, it possess glandular functions. As an example we might name the two glands associated with the mandible: the mandibular and the intramandibular glands. The intramandibular gland is found inside the mandible and consists of a hypertrophied secretory epithelium and secretory cells in the mandible's lumen. The secretion of the secretory epithelium is liberated through intracuticular ducts that open at the base of hairs at the mandible's surface. The secretion of the intramandibular gland (epithelium and secretory cells) reacted positively to tests for the detection of polysaccharides and proteins, thus suggesting that it consists of glycoproteins. The ultrastructure of the secretory epithelium presents variations related to the developmental stage of the individual, showing a large number of ribosomes and microvilli close to the cuticle in young individuals, while in the older specimens it was possible to note the formation of an intracellular reservoir. These variations of secretory epithelium, as also the interaction between the cellular groups inside the mandible, are important information about this gland in leaf-cutting ants.  相似文献

Spittlebugs are insects that suck sap from plants and regurgitate saliva containing toxic enzymes into the leaves. As a consequence, the conductive channels are blocked resulting in dry leaves, thus giving a burned aspect to the plantation. This work performed ultrastructural analyses of the salivary glands of the sugarcane spittlebug Mahanarva fimbriolata, since these organs produce the enzymes that are injected into the plants, thus being responsible for the economic losses in the production of sugarcane. Three kinds of secretory cells are found in principal gland, forming the lobules I-IV. The main differences among these cells relate to size, morphology and electron density of the secretory vesicles. The accessory glands contain different secretory vesicles to those in the principal gland. Muscular cells are found around the entire gland. The different secretory vesicles found in both principal and accessory glands indicate that the gland produces different substances or that the secretion in the interior of cells passes through a maturation process.  相似文献

This paper describes the flight muscles changes in relation to the age/function of the adult members of the colonies of two advanced species of eusocial bees: Apis mellifera (Apini) and Scaptotrigona postica (Meliponini). Here, are reported the results obtained through transmission electron microscopy studies, first describing a general overview of the flight muscle ultrastructure and second reporting on the ultrastructural changes that occur along the life stages/functions of workers, queens and males. The workers emerge with immature flight muscles, and the maturation takes about 20 days. In contrast, queens and males emerged with more advanced muscle differentiation, similar to workers after the 20 days of maturation. In both forager workers and laying queens, flight muscles showed signs of senescence, but not in sexually mature males. The differences among life phases, individual classes and species are discussed in relation to their functions in the colony.  相似文献

Apis mellifera is an interesting model to neurobiological studies. It has a relatively small brain that commands the complex learning and memory tasks demanded by the social organization. An A. mellifera colony is made up of a queen, thousands of workers and a varying number of drones. The latter are males, whereas the former are the two female castes. These three phenotypes differ in morphology, physiology and behavior, correlated with their respective functions in the society. Such differences include the morphology and architecture of their brains. To understand the processes generating such polymorphic brains we characterized the cell division and cell death dynamics which underlie the morphogenesis of the mushroom bodies, through several methods suitable for evidence the time and place of occurrence. Cell death was detected in mushroom bodies of last larval instar and mainly in black-eyed pupae. Cell division was observed in mushroom bodies, primarily at the start of metamorphosis, exhibiting temporal differences among workers, queens and males.  相似文献

The Triatominae subfamily has medical sanitary importance, since these insects are vectors of Trypanosoma cruzi, etiologic agent of Chagas Disease, and Trypanosoma rangeli, which develops in the salivary glands and it is frequently found in mixed infections with T. cruzi. Triatomines of Triatoma and Panstrongylus genera possess a salivary gland complex composed of a pair with three well differentiated units: the anterior (D1), median (D2) and posterior (D3). Saliva is secreted during blood meal and antagonizes hemostatic, inflammatory and immunological systems imposed by the host, which facilitate the hematophagy. In order to identify nuclear structures, we studied interphase nuclei of salivary gland cells of adult insects, males and females, of Triatoma infestans and Panstrongylus megistus. The glands were removed from insects, fixed in acetic acid (45%) and lactic acid (50%), squashed and submitted to different cytochemical methods: lacto-acetic orcein, silver ion impregnation, Feulgen reaction, Toluidine Blue, Variant method of critical electrolyte concentration and C-banding. The preparations were examined with a Zeiss Jenaval photomicroscope and photographed. The results evidenced predominance of binucleated cells in D1 and D2 glands and mononucleated ones in D3. In all salivary glands were observed bulky and polyploid nucleus, a clear association between nucleolar and heterochromatic corpuscles, cytoplasmatic metachromasy and many pre-secretion vesicles in cytoplasm. Such characteristics were associated with intense synthesis activity to produce the saliva. Species were mainly differentiated by a larger heterochromatic corpuscle observed only in T. infestans (called as chromocenter), while P. megistus showed a predominance of euchromatin, with some heterochromatic corpuscles just in males. Females of both species showed a smaller quantity of heterochromatin, which could be related to the high metabolism because of the oviposition.  相似文献

The mushroom bodies are structures present in the insect brain described as centers for the neural basis of learning, memory, and other higher functions. Honeybees (Apis mellifera) are insects with a sophisticated system of spatial orientation and possess well-developed learning and memory capabilities, which are associated with neural and brain structures. Thus, the present study aimed to compare the mushroom bodies during post-embryonic development and in newly emerged males, workers, and queens using light and transmission electron microscopy to examine how differential morphological characteristics are established during development. Measurements of structures were also taken in several post-embryonic developmental phases in order to evaluate size differences during the process and in the adult organs. The results show that workers, queens, and males exhibit temporal and size differences during the post-embryonic development of mushroom bodies, probably as adaptations to differences in behavior complexity. The mushroom bodies of workers are precociously formed and are larger than those of queens and drones. Thus, workers have the largest mushroom bodies resulting from differential development during metamorphosis.  相似文献

贻贝对各种海洋环境具有广泛适应性,范围从浅海跨越到深海热液、冷泉等极端环境。贻贝主要通过其足组织分泌的蛋白形成足丝附着于岩石等固体表面,这种蛋白是一种可再生、不受水环境影响的性能良好的天然生物胶黏剂,得益于分泌蛋白的性质,足丝在水下具有黏附性强、韧性高、耐水性优良等特性,在生物材料学、医学等方面具有很好的开发潜力和应用前景,已经是国内外研究热点之一。拉曼光谱是一种非接触的、无损的可以提供分子生物化学信息的检测技术。足丝是贻贝足腺体的外在表达形式,结合扫描电镜和共聚焦显微拉曼光谱技术,从贻贝足丝的表观差异到贻贝足腺体的分泌蛋白组分和分布特征,基于深海和浅海贻贝足丝的扫描电镜表征的表观形态差异,对两种贻贝足组织分别进行共聚焦显微拉曼光谱检测,得到两种贻贝的3个腺体的拉曼光谱和腺体局部区域的2D拉曼彩色分布图,从外在表现形式足丝到内部足腺体分布,通过对比两种贻贝的3个腺体的成分以及相对分布,分析造成两种贻贝足丝差异的内在腺体分布情况,此外结合两种贻贝生存环境的差异,认为贻贝的足丝外观差异以及其内部腺体分布是贻贝应对浅海和深海冷泉完全不同理化环境的一种环境适应机制。基于实验结果得到如下结论,拉曼光谱表明两种贻贝足腺体组成:表征核心腺体的amideⅢ信号位于1 242和1 269 cm^-1位置的2个峰的峰强度相对其他两个峰(1 318和1 337 cm^-1)较高,表现为有序高级的蛋白构象,外皮和粘附盘腺体含有丰富的酪氨酸(643, 830, 850和1 615 cm^-1)和3, 4-二羟基苯丙氨酸(多巴, DOPA,785 cm^-1);浅海贻贝在1 043 cm^-1位置有高强度的胶原蛋白信号。拉曼成像呈现两种贻贝腺体分布特征:深海贻贝表现为较为集中的腺体分布,浅海贻贝腺体分布较为分散,表明贻贝为适应不同环境形成不同的腺体分布机制。由此可见,拉曼光谱可以用于研究不同环境下生存的贻贝的足腺体分布特性,并在生物样品微观分析中更多的应用。  相似文献

The post-pharyngeal gland (PPG) occurs in ants and some Sphecidae wasps. Among its several roles is the storage of lipids from food. In order to investigate the effect of lipids on the cell, especially on mitochondria and peroxisomes, the present study was aimed at examining the peroxisomal and mitochondrial population in the PPG of Atta sexdens rubropilosa after lipid supplementation by confocal laser scanning microscopy and transmission electron microscopy. Soybean oil provided as lipid supplement was not toxic for A. sexdens rubropilosa workers for the first 48 h and 120 h. However, the ultrastructural cytochemical analysis revealed an accumulation of lipid droplets in the PPGs of ants after lipid supplementation at 48 h and 120 h, and smaller lipid droplets in the basal membrane of the PPG epithelium, showing lipid mobilization from the PPG to the hemolymph. The lipid supplementation reduces the life expectancy of medium workers, probably due to the high lipid metabolism. Most importantly, the PPGs of medium workers of leaf-cutting A. sexdens rubropilosa is probably a specialized gland in the lipid metabolism, due to the increased mitochondrial and peroxisomal population inside cells after lipid supplementation; participation of peroxisomal population in the β-oxidation of long chain fatty acids into smaller chains and participation of mitochondrial population in the β-oxidation of fatty acids for energy, or mobilization of lipid derivatives from the PPG to hemolymph, a process that requires energy. However, the hypothesis that the PPGs convert lipids from food in aldehydes and/or hydrocarbons must be better investigated.  相似文献

Ice-cell interactions especially the question whether and how ice crystals invade tissues and organs during freezing were examined in small transparent organs (salivary glands) in which many structural details remained visible despite of freeze-induced cell darkening. In most glands no invasion of ice into the lumen was observed since ice dendrites stopped growing after touching the gland. Here I report that in rare cases a so far unknown type of ice crystals developed which aggressively pushed against cell membranes before invading the gland via paracellular pathways (septate junctions). Aggressive ice crystals were also observed within a salivary gland cell which deformed and finally invaded the nucleus. In cell strands it was observed that intracellular freezing is indeed a two-step event in which ice developed in cytoplasm several seconds before invading the nucleus.  相似文献

Histological and histochemical analyses were carried out in order to evaluate the influence of the topical application of a synthetic juvenile hormone on the secretory cycle and degeneration of the venom gland of Apis mellifera. Newly emerged workers received the topical application of synthetic hormone and the results were compared to the normal development of the secretory cycle in virgin and mated queens. The first worker group received the juvenile hormone diluted in hexane (2 microg/microL), the second received only 1 microL of hexane, and the third did not receive any kind of application. After the application the workers were returned to the colony and collected at the ages of 14 and 25 days of adult life. The groups with virgin queens and the other with mated queens, did not receive the treatment. The results show that the individuals treated with juvenile hormone and with pure hexane presented differences in the histological and cytochemical aspects of the secretory cells of the venom gland. The data indicate that both the juvenile hormone and hexane accelerate the activity of the secretory cycle and the degeneration of the venom gland; however, the juvenile hormone proved to be more effective than hexane.  相似文献

Leaf salt glands of Limoniastrum guyonianum were examined by scanning and transmission electron microscopes and energy dispersive X-ray analysis (EDAX) system, after growing for three months on sandy soil with or without 300 mM NaCl. Results showed that salt glands were irregularly scattered on both leaf sides and sunk under the epidermal level. Salt excretion occurred in both conditions and is mainly composed of calcium and magnesium in control plants, and essentially sodium and chloride in plants subjected to salt treatment. A salt gland is comprised of collecting, accumulating, and central compartments, and is made up of total thirty-two cells. The collecting cells were characterized by large central vacuoles. Accumulating cells contain numerous, large, and unshaped vacuoles and rudimentary chloroplasts. The central compartment was comprised of four basal cells and each one is surmounted by an apical cell. The basal cells are granulated, containing large nucleus, numerous mitochondria, endoplasmic reticulum, ribosomes, polyribosomes, and small vacuoles or vesicles. Equally, the apical cells are rich in organelles. Application of 300 mM NaCl to the culture medium increased vacuoles number and size, and organelles density especially the mitochondria which suggests energy requirement for ions transport. The reduction in size and number of vacuoles toward the interior of salt glands of treated plants and the fusion of the smallest ones with the plasma membrane substantiate the implication of such vacuoles in salt excretion process. The current study which is the first report on L. guyonianum salt gland has provided an in-depth understanding on structure–function relationship in the multicellular salt glands.  相似文献

Different histochemical techniques were applied to examine the morphological features of the secretory cells of hypopharyngeal glands in the wasp Polistes versicolor. The results showed that most analyzed individuals present active glands with secretion stored in the cytoplasm. In some glands, morphological analyses revealed the presence of degenerative characteristics. Analyses of cellular integrity, however, did not detect dead cells. The results showed that, in P. versicolor, the development and regression of the hypopharyngeal glands were not age related, unlike glands of social bees.  相似文献

The venom gland of queens of Apis mellifera was examined through light and transmission electron microscopy and subjected to electrophoretic analyses. Virgin queens exhibited prismatic secretory cells containing large amounts of rough endoplasmic reticulum with dilated cisternae, open secretory spaces, numerous vacuoles and granules scattered in the cytoplasm, and spherical nuclei with numerous nucleoli. The secretion produced was non-refringent under polarized light and the electrophoretic analysis of glandular extracts revealed five main protein bands. In mated queens, the venom gland exhibited a high degree of degeneration. Its secretion was refringent under polarized light and one of the main bands was absent in the electrophoretic pattern obtained. The morphological aspects observed are in agreement with the function of this gland in queens, given that virgin queens use venom in battles for the dominance of the colony, a situation that occurs as soon as they emerge, while fertilized queens rarely use venom.  相似文献

Mouth dryness is one of the complications of diabetes mellitus but there has been little work on morphological changes of the salivary glands. In the present study, the effects of diabetes mellitus on the serous and mucous acini of submandibular gland of male and female rats, 4 and 12 weeks after diabetes induction were studied. Male and female rats were divided into experimental and control subgroups. Diabetes was induced to experimental rats by streptozotocin. At the end of 4 and 12 weeks, the submandibular glands were removed, random sections obtained and volume-weighted mean acini volume was estimated by point-sampled intercepts method. The data revealed that volume reduction occurred only in serous acini in both male and female rats 12 weeks after diabetes induction and the others remained unchanged. The present research using stereological methods demonstrates that diabetes make some morphological changes in serous acini, the main exocrine part of the rat submandibular gland.  相似文献

The vector leafhopper Psammotettix striatus (L.) (Hemiptera: Cicadellidae) is an important economic pest that is a serious threat to wheat in Northwest China, causing great losses to agricultural production by transmitting wheat blue dwarf (WBD) in a persistent circulative manner. Studies on morphology and ultrastructure of the digestive system were made using light, scanning and transmission electron microscopy. The gross morphology of the alimentary canal consists of esophagus (foregut), midgut, hindgut, and special filter chamber. The esophagus, a narrow and slender tube, runs through the whole thorax extending into the filter chamber and connecting with the anterior midgut. The midgut consists of three major regions, anterior, middle and posterior midgut. The anterior midgut is composed of exceptionally thick epithelial cells surrounding a large lumen. Numerous well-developed microvilli occur at the basal region of the epithelium. A large number of electron-lucent and lipid-like vesicles are observed under the microvilli. The posterior midgut is smaller than the anterior midgut in diameter. Numerous different concentric circular bodies are characteristic of the posterior midgut. The hindgut arises from the posterior midgut at the boundary of the filter chamber. It extends downward toward the anus where the hindgut enlarges to form the rectum. The rectum is formed by various cells typical of epithelium, whose nuclei are relatively smaller than those of the midgut. Relatively large muscle cells are present at the well-defined and thick basement membrane. There are two pairs of Malpighian tubules. Numerous mitochondria and lamellar rough endoplasmic reticulum in the cells of the anterior segment of the Malpighian tubules; the sub-anterior segment of the tubules resemble the distal segment which are wave-like, amount vesicles in the cells; numerous tightly packed large vesicles contain two types of brochosomes in the inflated segment of the Malpighian tubules. The salivary system of P. striatus contains one pair of salivary glands, which are made up of the principal and accessory gland. The principal glands are divided into anterior, mid- and posterior lobes. About nineteen acini are observed in the principal glands of P. striatus, and vary in structure and colour. Five cellular types are found in the principal glands by transmission electron microscopy, which are rich in secretory granules, modified in size, electron-dense and shape. Rough endoplasmic reticulum in the shape of vesicular and lamellar, mitochondria and Golgi complexes are observed. The accessory gland has only an oval or a rod-shaped acinus.  相似文献

Morphological studies in male reproductive system of insect, and accessory glands functions, can contribute to the knowledge of reproductive aspects in Triatominae. The present study aims to describe the structure of male reproductive system under scanning electron microscopy, and the ultrastructure of accessory glands during the process of maturation, in T. brasiliensis and T. melanica of different ages.These system consist of two testis, two vas deferens, two seminal vesicles, four pair of accessory glands and one ejaculatory duct. The four mesodermal male accessory glands of T. melanica and T. brasiliensis, present secretion storage in the firsts days of adult life in starved males. During the maturation process of male accessory glands of T. brasiliensis and T. melanica, granules with different sizes, shapes and electron density were seen. In T. melanica small vacuoles are released into the gland lumen in one-day-old males, however, after three and five days few granules are found in the cell cytoplasm. The secretory granules in T. brasiliensis increase in size and amount, in the five days old adult. The secretory pathway in the male accessory glands of the two species seems to be merocrine and apocrine suggesting that different substances are being produced at different times. The cell microvilli in T. brasiliensis are longer and narrower, these can be absent in some regions of the cell surface of T. melanica. Maturation of male accessory glands in the triatomine species studied occurred without adult blood meal. Its function can led to new insights for the comprehension of reproductive aspects in Triatominae.  相似文献
