This paper describes a novel, rapid, and sensitive analytical method for monitoring four triazolone herbicides in cereals (wheat, rice, corn, and soybean), using a quick, easy, cheap, effective, rugged, and safe sample extraction procedure followed by ultrahigh performance liquid chromatography coupled with tandem mass spectrometry. The four triazolone herbicides (amicarbazone, carfentrazone‐ethyl, sulfentrazone, and thiencarbazone‐methyl) were extracted using acidified acetonitrile (containing 1% v/v formic acid) and subsequently purified with octadecylsilane (C18) prior to sample analysis. Ultrahigh performance liquid chromatography coupled with tandem mass spectrometry was operated in positive and negative ionization switching mode. Amicarbazone and carfentrazone‐ethyl were detected in the positive mode (ESI+), while sulfentrazone and thiencarbazone‐methyl were detected in the negative mode (ESI?). All compounds were successfully separated in less than 3.0 min. Further optimization achieved desired recoveries ranging from 74.5 to 102.1% for all analytes with relative standard deviation values ≤17.2% in all tested matrices at three levels (10, 100, and 500 μg/kg). The limits of detection for all compounds were ≤2.3 μg/kg, and the limits of quantitation did not exceed 7.1 μg/kg. The developed method showed excellent linearity (R2 ≥ 0.994) and was proven to be highly efficient and reliable for the routine monitoring of triazolone herbicides in cereals.  相似文献

The present study reports the in vivo and in vitro identification and characterization of metabolites of fluvastatin, the 3‐hydroxy‐3‐methyl‐glutaryl‐coenzyme A reductase inhibitor, using liquid chromatography–mass spectrometry (LC–MS). In vitro studies were conducted by incubating the drug with human liver microsomes and rat liver microsomes. In vivo studies were carried out by administration of the drug in the form of suspension to the Sprague–Dawley rats followed by collection of urine, faeces and blood at different time points up to 24 h. Further, samples were prepared by optimized sample preparation method, which includes freeze liquid extraction, protein precipitation and solid phase extraction. The extracted and concentrated samples were analysed using ultrahigh‐performance liquid chromatography–quadruple time‐of‐flight tandem mass spectrometry. A total of 15 metabolites were observed in urine, which includes hydroxyl, sulphated, desisopropyl, dehydrogenated, dehydroxylated and glucuronide metabolites. A few of the metabolites were also present in faeces and plasma samples. In in vitro studies, a few metabolites were observed that were also present in in vivo samples. All the metabolites were characterized using ultrahigh‐performance liquid chromatography–quadruple time‐of‐flight tandem mass spectrometry in combination with accurate mass measurement. Finally, in silico toxicity studies indicated that some of the metabolites show or possess carcinogenicity and skin sensitization. Several metabolites that were identified in rats are proposed to have toxicological significance on the basis of in silico evaluation. However, these metabolites are of no human relevance. Using spiramycin as a dummy template, a molecularly imprinted polymer monolithic micro‐column with high selection to azithromycin was prepared in a micropipette tip. The imprinting factor of the monolithic micro‐column prepared was approximately 2.67 and the morphological structure of the polymers was characterized by scanning electron microscopy. A simple, sensitive, and reproducible method based on the imprinted monolithic micro‐column coupled to liquid chromatography with tandem mass spectrometry was developed for determining the residues of azithromycin in pork. Pork samples were extracted with acetonitrile, cleaned up under the optimal monolithic micro‐column conditions, and analyzed using liquid chromatography with tandem mass spectrometry in the multiple reaction monitoring mode. The assay exhibited a linear dynamic range of 0.50–50 μg/L with the correlation coefficient (r2) above 0.99. In the three spiking levels of 0.50, 1.0, and 10 μg/kg, the average recoveries of azithromycin from pork samples were between 85.8 and 96.5% with a relative standard deviation below 10%. The limit of detection and limit of quantitation were 0.03 and 0.1 μg/kg, respectively.  相似文献

In this work, gas chromatography tandem with electron ionization and full‐scan high‐resolution mass spectrometry with a time‐of‐flight mass analyzer was evaluated for analyzing pesticide residues in teas. The relevant aspects for mass spectrometry analysis, including the resolution and mass accuracy, acquisition rate, temperature of ion source, were investigated. Under acquisition condition in 2‐GHz extended dynamic range mode, accurate mass spectral library including 184 gas chromatography detectable pesticides was established and retrieval parameters were optimized. The mass spectra were consistent over a wide concentration range (three orders) with good match values to those of NIST (EI‐quadrupole). The methodology was verified by the validation of 184 pesticides in four tea matrices. A wide linear range (1–1000 μg/kg) was obtained for most compounds in four matrices. Limit of detection, limit of quantification, and limit of identification values acquired in this study could satisfy the requirements for maximum residue levels prescribed by the European Community. Recovery studies were performed at three concentrations (10, 50, and 100 μg/kg). Most of the analytes were recovered at an acceptable range of 70–120% with relative standard deviations ≤ 20% in four matrices. The potential extension of qualitative screening scope makes gas chromatography tandem with electron ionization and mass spectrometry with a time‐of‐flight mass analyzer a more powerful tool compared with gas chromatography with tandem mass spectrometry.  相似文献

The present work describes the development of a sensitive and reliable analytical method based on solid‐phase extraction followed by analysis using liquid chromatography with tandem mass spectrometry for the simultaneous determination of pharmaceuticals from antibiotics (fluoroquinolones, sulfonamides, and their N4‐acetyl metabolites, and trimethoprim as sulfonamides synergist) and anthelmintics groups. SPE was optimized using different cartridges (Strata‐X, Oasis HLB, Strata C18‐E, Isolute C18, SampliQ C8/Si‐SCX). The highest recovery was achieved using Strata X cartridge (>80%) with good reproducibility (RSDs < 5%) despite various physicochemical properties of the compounds. Investigated analytes were identified and quantitatively determined by liquid chromatography with tandem mass spectrometry using multiple reaction monitoring. The method was shown to be linear over the concentration range of 0.05–30 μg/L for febantel and albendazole, and 0.10–60 μg/L for all other pharmaceuticals. Correlation coefficients were >0.99 for all compounds except for sulfamethazine (0.98). In order to demonstrate the applicability of the developed method, wastewater from the veterinary industry was analyzed. Results evidenced the presence of febantel, praziquantel, albendazole, enrofloxacin, sulfamethazine, and sulfadiazine.  相似文献

A liquid chromatography with tandem mass spectrometry method was developed and validated to simultaneously determine chlorpyrifos, imidacloprid, and imidacloprid metabolites in soil, wheat grain, and wheat straw matrices. Satisfactory linearity (R2 ≥ 0.9965) of the method was obtained for all analytes. The ranges of limits of detection and limits of quantification for seven analytes in three matrices were 0.17–66.7 and 0.5–200 μg/kg, respectively. Average recoveries were 72.85–81.25% for chlorpyrifos, 78.54–84.70% for imidacloprid, 73.83–81.03% for imidacloprid olefin, 71.47–80.61% for 5‐hydroxy imidacloprid, 71.79–81.32% for imidacloprid urea, 70.42–82.20% for imidacloprid nitroguanidine, and 70.91–82.46% for imidacloprid 6‐chloronicotinic acid in soil, wheat grain, and wheat straw. The intra‐ and interday relative standard deviations were less than 8%. The established method was successfully applied for the residual analysis of chlorpyrifos, imidacloprid, and imidacloprid metabolites in actual soil, wheat grain, and wheat straw samples. The results indicated that the established method could be used to detect trace amounts of chlorpyrifos, imidacloprid, and imidacloprid metabolites in wheat and that the method might be able to provide some data on the detection of these seven compounds in other crops.  相似文献

A simple and efficient method for the determination of 28 carbamates in high‐fat cheeses is proposed. The methodology is based on a modified quick, easy, cheap, effective, rugged, and safe procedure as sample treatment using a new sorbent (Z‐Sep+) followed by ultra‐high performance liquid chromatography with tandem mass spectrometry determination. The method has been validated in different kinds of cheese (Gorgonzola, Roquefort, and Camembert), achieving recoveries of 70–115%, relative standard deviations lower than 13% and limits of quantification lower than 5.4 μg/kg, below the maximum residue levels tolerated for these compounds by the European legislation. The matrix effect was lower than ±30% for all the studied pesticides. The combination of ultra‐high performance liquid chromatography and tandem mass spectrometry with this modified quick, easy, cheap, effective, rugged, and safe procedure using Z‐Sep+ allowed a high sample throughput and an efficient cleaning of extracts for the control of these residues in cheeses with a high fat content.  相似文献

A procedure for multiresidue analysis was developed for the extraction and determination of 17 pesticides, including herbicides, fungicides, and insecticides, as well as certain degradation products, in vineyard soils from La Rioja region (Spain). Different solvents and mixtures were tested in spiked pesticide‐free soils, and pesticides were comparatively evaluated by gas chromatography with mass spectrometry and liquid chromatography with mass spectrometry. Recoveries >70%, with relative standard deviations <9%, were obtained when a mixture of methanol/acetone or a mixture of methanol/CaCl2 0.01 M for the most polar compounds was selected as the extraction solvent. Method validation was accomplished with acceptable linearity (r2 ≥ 0.987) within the concentration range of 0.005–1 μg/mL corresponding to 1.667–333.4 μg/kg and 0.835–167.1 μg/kg for liquid chromatography with mass spectrometry and gas chromatography with mass spectrometry, respectively, and detection limits <0.4 μg/kg for the compounds were studied. The extraction method was applied to 17 real vineyard soil samples, and terbuthylazine and its metabolite desethylterbuthylazine were the most ubiquitous compounds, as they were detected in the 100% of the soils analyzed. The presence of fungicides was also high, and the presence of insecticides was lower than other pesticides. The results confirm the usefulness of the optimized procedure for monitoring residues in vineyard soils.  相似文献

The separation of basic macrolide antibiotics suffers from peak tailing and poor efficiency on traditional silica‐based reversed‐phase liquid chromatography columns. In this work, a C18HCE column with positively charged surface was applied to the separation of macrolides. Compared with an Acquity BEH C18 column, the C18HCE column exhibited superior performance in the aspect of peak shape and separation efficiency. The screening of mobile phase additives including formic acid, acetic acid and ammonium formate indicated that formic acid was preferable for providing symmetrical peak shapes. Moreover, the influence of formic acid content was investigated. Analysis speed and mass spectrometry compatibility were also taken into account when optimizing the separation conditions for liquid chromatography coupled with tandem mass spectrometry. The developed method was successfully utilized for the determination of macrolide residues in a honey sample. Azithromycin was chosen as the internal standard for the quantitation of spiramycin and tilmicosin, while roxithromycin was used for erythromycin, tylosin, clarithromycin, josamycin and acetylisovaleryltylosin. Good correlation coefficients (r2 > 0.9938) for all macrolides were obtained. The intra‐day and inter‐day recoveries were 73.7–134.7% and 80.7–119.7% with relative standard deviations of 2.5–8.0% and 3.9–16.1%, respectively. Outstanding sensitivity with limits of quantitation (S/N ≥ 10) of 0.02–1 μg/kg and limits of detection (S/N ≥ 3) of 0.01–0.5 μg/kg were achieved.  相似文献

建立了液相色谱–三重四极杆串联质谱测定水果及其制品中氯吡脲的方法。样品经乙腈提取,氨基固相萃取小柱净化后,用ZORBAX Extend-C18柱(150 mm×2.1 mm,5μm)分离,以甲醇–水为流动相等度洗脱,采用多反应监测正离子模式检测,外标法定量。氯吡脲的质量浓度在4.0~200.0 ng/m L范围内线性良好,相关系数大于0.999,在5.0,10.0,20.0μg/kg 3个添加水平下,氯吡脲的平均加标回收率为86%~92%,测定结果的相对标准偏差为5.3%~7.6%(n=5),方法定量下限为2.0μg/kg。方法灵敏度高,操作简便,定量准确,可满足梨、柑桔、黄桃等水果及其罐头制品中氯吡脲残留的检测与确证需要。  相似文献

A modified Quick, Easy, Cheap, Effective, Rugged and Safe (QuEChERS) method for the simultaneous determination of spirotetramat and its four metabolite residues in citrus, peel, pulp and soil was developed and validated by liquid chromatography with tandem mass spectrometry (LC‐MS/MS). The samples were extracted with acetonitrile (1%, glacial acetic acid, v/v) and purified using primary secondary amine and octadecylsilane. The limit of detection was 0.01–0.13 mg/kg, whereas that of quantification was 0.02–0.40 mg/kg for spirotetramat and its metabolites. The average recoveries of spirotetramat, spirotetramat‐enol, spirotetramat‐mono‐hydroxy, spirotetramat‐enol‐glucoside and spirotetramat‐ketohydroxy in all matrices were 73.33–107.91%, 75.93–114.85%, 76.44–100.78%, 71.46–103.19% and 73.08–105.27%, respectively, with relative standard deviations < 12.32%. The dissipation dynamics of spirotetramat in citrus and soil followed first‐order kinetics, with half‐lives of 2.3–8.5 days in the three sampling locations. The terminal residues of spirotetramat in four matrices at the three locations were measured below the 1.0 mg/kg maximum residue limit set by China, and residues were found to be concentrated on the peel. The risk assessment of citrus was evaluated using risk quotients. The risk quotient values were found to be significantly <1, suggesting that the risk to human health was negligible when using the recommended doses of spirotetramat in citrus. These results could provide guidance for the safe and proper application of spirotetramat in citrus in China.  相似文献

Lentinula edodes, one of the most cultivated edible fungi in the world, are usually neglected for mycotoxins contamination due to the initial thinking of its resistance to mycotoxingenic molds. In the present study, a sensitive and reliable liquid chromatography with tandem mass spectrometry method was developed for the simultaneous quantification of 33 mycotoxins in L. edodes. Targeted mycotoxins were extracted using a quick, easy, cheap, effective, rugged, and safe procedure without any further clean‐up step, and analyzed by liquid chromatography with tandem mass spectrometry on an Agilent Poroshell 120 EC‐C18 column (100 × 3 mm, 2.7 μm) with a linear gradient elution program using water containing 5 mM ammonium acetate and methanol as the mobile phase. After validation by determining linearity (R2 > 0.99), sensitivity (LOQ ≤ 20 ng/kg), recovery (73.6–117.9%), and precision (0.8–19.5%), the established method has been successfully applied to reveal the contamination states of various mycotoxins in L. edodes. Among the 30 tested samples, 22 were contaminated by various mycotoxins with the concentration levels ranging from 3.3–28 850.7 μg/kg, predicting that the edible fungus could be infected by the mycotoxins‐producing fungi. To the best of our knowledge, this is the first report about real mycotoxins contamination in L. edodes.  相似文献

An ultrasensitive method for the simultaneous analysis of pesticides residues in tobacco was developed with online size exclusion chromatography with gas chromatography and tandem mass spectrometry. Tobacco samples were extracted with the solvent mixture of cyclohexane and acetone (7:3, v/v) and centrifuged. Then, the supernatant liquors were injected directly into the online size exclusion chromatography with gas chromatography and tandem mass spectrometry without any other purification procedures after being filtered with a 0.22 μm organic phase filter. The matrix interferences were effectively removed and recoveries of most pesticides were in the range of 72–121%. Especially, for chlorothalonil, the analysis efficiency of this method was much more favorable than that of the general method, in which dispersive solid‐phase extraction was used as an additional purified procedure. In addition, the limits of quantitation of this method were from 1 to 50 μg/kg. Therefore, a rapid, cost‐effective, labor‐saving method was proposed in the present work, which was suitable for the analysis of 41 pesticide residues in tobacco.  相似文献

Analytical methods based on multiplug filtration cleanup coupled with pulse glow discharge‐ion mobility spectrometry and liquid chromatography tandem mass spectrometry were developed for the analysis of tricaine mesylate residue in fish and fish‐raising water samples. A silica fiber holder and an appropriate new interface were designed to make the direct introduction of the fiber into the pulse glow discharge‐ion mobility spectrometry introduction mechanism. The multiplug filtration cleanup method with adsorption mixtures was optimized for the determination of tricaine mesylate in fish samples. Good linear relationships were obtained by the two methods. For fish samples, limits of detection were 6 and 0.6 μg/kg by ion mobility spectrometry and liquid chromatography with tandem mass spectrometry, respectively. The matrix effect of the established liquid chromatography tandem mass spectrometry method was negligible for fish samples but that of the ion mobility spectrometry method was not. The two methods were compared. The ion mobility spectrometry system could be used a rapid screening tool on site with the advantage of rapidity, simplicity, and portability, and the liquid chromatography tandem mass spectrometry system could be used for validation in laboratory conditions with the advantage of lower limit of detection, stability, and precision.  相似文献

A one‐step ultrasound/microwave‐assisted solid–liquid–solid dispersive extraction procedure was used for the simultaneous determination of eight neonicotinoids (dinotefuran, nitenpyram, thiamethoxam, clothianidin, imidacloprid, acetamiprid, thiacloprid, imidaclothiz) in dried Dendrobium officinale by liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry in multiple reaction monitoring mode. The samples were quickly extracted by acetonitrile and cleaned up by the mixed dispersing sorbents including primary secondary amine, C18, and carbon‐GCB. Parameters that could influence the ultrasound/microwave‐assisted extraction efficiency such as microwave irradiation power, ultrasound irradiation power, temperature, and solvent were investigated. Recovery studies were performing well (70.4–113.7%) at three examined spiking levels (10, 50, and 100 μg/kg). Meanwhile, the limits of quantification for the neonicotinoids ranged from 0.87 to 1.92 μg/kg. The method showed good linearity in the concentration range of 1–100 μg/L with correlation coefficients >0.99. This quick and useful analytical method could provide a basis for monitoring neonicotinoid insecticide residues in herbs.  相似文献

A simple and reliable method of ultra high performance liquid chromatography coupled with photo‐diode array detection has been proposed for the simultaneous determination of deoxynivalenol and its acetylated derivatives in wheat flour and rice, especially focusing on the optimization of sample extraction, cleanup, and chromatographic separation conditions. Sample pretreatment consisted of a first step using a quick, easy, cheap, effective, rugged, and safe based extraction procedure and a subsequent cleanup step based on solid‐phase extraction. The method was extensively validated in wheat flour and rice, obtaining satisfactory analytical performance with good linearity (R2 ≥ 0.999), acceptable recoveries (80.0–104.4%), and repeatability (RSDs 1.3–10.7%). The limits of detection (21.7–57.4 μg/kg) and quantitation (72.3–191.4 μg/kg) for deoxynivalenols were lower than those usually permitted by various countries’ legislation in these food matrices. The method was applied to 34 wheat and rice samples. The results were further compared with results of ultra high performance liquid chromatography with electrospray ionization tandem mass spectrometry.  相似文献

A modified quick, easy, cheap, effective, rugged and safe method was established for simultaneous determination of atropine, anisodamine, and scopolamine in goji berries by using ultra‐high‐performance liquid chromatography with tandem mass spectrometry. The graphene/hexagonal boron nitride hybrids were prepared and first applied as a cleanup adsorbent. Compared to classical cleanup adsorbent (C18), the graphene/hexagonal boron nitride hybrids as adsorbent had better extraction efficiency for the detection of analytes. Under the optimal conditions, the proposed analytical method achieved satisfactory linearity (R> 0.995), and obtained desirable recoveries ranged from 77.4 to 94.0% with the relative standard deviation of 1.2–6.1% at the concentration levels of 3.2–13.4 µg/kg. The limits of quantitation of atropine, anisodamine, and scopolamine were, respectively, 3.2, 4.6, and 4.5 µg/kg with linearity ranged from 3.2 to 25.4 µg/kg. The modified quick, easy, cheap, effective, rugged, and safe sample preparation with ultra‐high‐performance liquid chromatography and tandem mass spectrometry method was successfully applied to evaluate the safety of goji berries collected from 30 plant areas in China, suggesting its applicability and suitability for the routine analysis of three tropane alkaloids in goji berries.  相似文献

A novel, rapid and sensitive liquid chromatography tandem–mass spectrometry method for quantification of vemurafenib in human plasma, that also for the first time allows for metabolite semi‐quantification, was developed and validated to support clinical trials and therapeutic drug monitoring. Vemurafenib was analysed by precipitation with methanol followed by a 1.9 min isocratic liquid chromatography tandem masspectrometry analysis using an Acquity BEH C18 column with methanol and formic acid using isotope labelled internal standards. Analytes were detected in multireaction monitoring mode on a Xevo TQ. Semi‐quantification of vemurafenib metabolites was performed using the same analytical system and sample preparation with gradient elution. The vemurafenib method was successfully validated in the range 0.5–100 μg/mL according to international guidelines. The metabolite method was partially validated owing to the lack of commercially available reference materials. For the first time concentration levels at steady state for melanoma patients treated with vemurafenib is presented. The low abundance of vemurafenib metabolites suggests that they lack clinical significance. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

We developed an analytical method using liquid–liquid extraction (LLE) and liquid chromatography–tandem mass spectrometry (LC‐MS/MS) to detect and quantify tebufenozide (TEB) and indoxacarb (IND) residues in animal and aquatic products (chicken muscle, milk, egg, eel, flatfish, and shrimp). The target compounds were extracted using 1% acetic acid (0.1% acetic acid for egg only) in acetonitrile and purified using n‐hexane. The analytes were separated on a Gemini‐NX C18 column using (a) distilled water with 0.1% formic acid and 5 mm ammonium acetate and (b) methanol with 0.1% formic acid as the mobile phase. All six‐point matrix‐matched calibration curves showed good linearity with coefficients of determination (R2) ≥0.9864 over a concentration range of 5–50 μg/kg. Intra‐ and inter‐day accuracy was expressed as the recovery rate at three spiking levels and ranged between 73.22 and 114.93% in all matrices, with a relative standard deviation (RSD, corresponding to precision) ≤13.87%. The limits of quantification (LOQ) of all target analytes ranged from 2 to 20 μg/kg, which were substantially lower than the maximum residue limits (MRLs) specified by the regulatory agencies of different countries. All samples were collected from different markets in Seoul, Republic of Korea, and tested negative for tebufenozide and indoxacarb residues. These results show that the method developed is robust and may be a promising tool to detect trace levels of the target analytes in animal products.  相似文献

An ultra high performance liquid chromatography with tandem mass spectrometry method was established for the rapid and simultaneous analysis of seven antiviral drugs, amantadine, rimantadine, memantine, moroxydine, imiquimod, oseltamivir, and acyclovir, in chicken liver, muscle, and egg. Homogenized samples were extracted with trichloroacetic acid and acetonitrile solutions and then purified by cation‐exchange solid‐phase extraction. The target drugs were analyzed by liquid chromatography with a UPLC BEH Amide column (2.1 mm × 100 mm, 1.7 μm) coupled with a tandem mass spectrometer operating in the positive multiple‐reaction mode. A perfectly linear relationship was obtained within the concentration ranges of 0.5–20 μg/L for acyclovir and 0.1–10 μg/L for the other six antiviral drugs. The average recoveries of the seven antiviral drugs using four addition levels in chicken liver, muscle, and eggs were 82.67–90.10, 82.30–92.27, and 81.98–93.77%, respectively, and the acceptable coefficients of variation were 5.18–9.88, 4.84–11.2, and 42.8–9.95%, respectively. The detection limits and detection capabilities of the analysis method for the seven antiviral drugs were in the ranges of 0.04–0.64 and 0.11–0.78 μg/kg, respectively. Additionally, an inter‐laboratory study among five laboratories further validated the method.  相似文献
