In this paper, two immunoassay methods based on SERS are developed for multiplex analysis, both of which stemmed from the concept of forming a sandwich structure ‘capture antibody substrate/antigen/Raman‐reporter‐labeled immuno‐nanoparticles’. They are two‐molecule labeled one‐nanoparticle and one‐molecule labeled two‐nanoparticle methods. In both the methods, two different antibodies covalently bound to a solid substrate can specifically capture two different antigens from a sample. The captured antigens in turn bind selectively to their corresponding antibodies immobilized on Raman‐reporter‐labeled nanoparticles. Multianalyte immunoassay is successfully demonstrated by the detection of characteristic Raman bands of the probe molecules only when the antigen and antibody are matched. Here, we report the nature of new di‐α‐amino (L1–L3) and α‐amino‐α‐hydroxyphosphinic (L4–L6) acids, which are considered potential inhibitors of the aminopeptidase N, adsorbed on a colloidal silver surface by means of surface‐enhanced Raman scattering (SERS) spectroscopy. In order to reveal the adsorption mechanism of these species from their SERS spectra, Fourier‐transform Raman (FT‐RS) spectra of these nonadsorbed molecules were measured. By examining the enhancement, shift in wavenumbers, and changes in breadth of the SERS bands due to the adsorption process, we revealed that the tilted compounds interact with the colloidal silver substrate mainly through the benzene ring, amino group, and phosphinic moiety in the following way. The benzene ring of L2 and L3 is ‘standing up’ on the colloidal silver surface, and the C N bond is almost vertical to it, while the tilt angle between the O PO bond and this surface is greater than 45°. On the other hand, for L1, L4, and L5, the aromatic ring and C N bond are arranged more or less tilted, and the tilt angle between the O PO bond and the silver substrate is smaller than 45°. The elongation of the bond to the benzene ring, the L6 case, produces an almost horizontal orientation of the benzene ring and the O PO bond on the silver nanoparticles. For these ligands, the complement inhibition IC50 tested in vitro using porcine kidney leucine aminopeptidase was correlated mainly with the behavior of the O PO and C CH N fragments on the silver surface. Here we report the synthesis of 2–5 nm size gold nanoparticle labels for surface‐enhanced Raman Spectroscopy (SERS) based immunoassay to detect protein molecules. The Au nanoparticles were conjugated with fluorescein isothiocyanate (FITC) and goat anti‐h‐IgG (immunoglobin) and the resultant particles were used for the detection of h‐IgG. Commercially available nitrocellulose strip and silver enhancement method were used for SERS‐based immunoassays. The FITC acts as a Raman probe, and vibrational fingerprint of this molecule was used for the detection of h‐IgG in concentration ranging from 1 to 100 ng/µl. Our Raman probe is robust and small in size and has high water solubility with minimum steric effect during antigen–antibody binding. Copyright © 2007 John Wiley & Sons, Ltd.  相似文献

Surface‐enhanced Raman scattering (SERS) is an extremely powerful tool for the analysis of the composition of bimetallic nanoparticle (BNP) surfaces because of the different adsorption schemes adopted by several molecules on different metals, such as Au and Ag. The preparation of BNPs normally implies a change in the plasmonic properties of the core metal. However, for technological applications it could be interesting to synthesize core–shell structures preserving these original plasmonic properties. In this work, we present a facile method for coating colloidal gold nanoparticles (NPs) in solution with a very thin shell of silver. The resulting bimetallic Au@Ag system maintains the optical properties of gold but shows the chemical surface affinity of silver. The effectiveness of the coating method, as well as the progressive silver enrichment of the outermost part of the Au NPs, has been monitored through the SERS spectra of several species (chloride, luteolin, thiophenol and lucigenin), which show different behaviors on gold and silver surfaces. A growth mechanism of the Ag shell is proposed on the basis of the spectroscopic and microscopic data consisting in the formation and deposit of Ag clusters on the Au NP surface. Electromagnetic coupling between localised plasmons on metal nanoparticles and the strong localised fields on a micro‐structured surface is demonstrated as a means to increase the enhancement factor in surface‐enhanced Raman scattering (SERS) spectroscopy. Au nanoparticles of diameter 20 nm were deposited on a micro‐structured Au surface consisting of a periodic array of square‐based pyramidal pits (Klarite). The spectra of 4‐aminothiophenol (4‐ATP) were compared before and after deposition of Au nanoparticles on the micro‐structured surface. The addition of Au nanoparticles is shown to provide significantly higher signal intensities, with improvements of the order of ∼103 per molecule compared with spectra obtained from the micro‐structured substrate alone. This hybrid approach offers promise for combining nanoparticles with micro‐ and nano‐structured surfaces in order to design SERS substrates with higher sensitivities. Zinc oxide nanowires with two distinct morphologies were synthesized on silicon substrates using a simple thermal evaporation and vapor transport method in an oxidizing environment. The as‐synthesized nanowires were coated with gold to allow excitation of surface plasmons over a broad frequency range. SERS studies with near‐IR excitation at 785 nm showed significant enhancement (average enhancement > 106) with excellent reproducibility to detect monolayer concentrations of 4‐methylbenzenethiol (4‐MBT) and 1,2‐benzendithiol (1,2‐BDT) probe molecules. The Raman enhancement showed a strong dependence on the gold film thickness, and the peak enhancement was observed for a ∼40‐nm‐thick film. The Raman enhancement was stronger for randomly oriented nanowires compared to aligned ones suggesting the importance of contributions from the junctions of nanowires. In surface‐enhanced Raman scattering (SERS), the scattered intensity is drastically increased due to a resonant interaction with surface plasmons of coin metals. SERS is a nondestructive spectroscopic method applied also to biomedical samples. It inherits the advantages of normal Raman spectroscopy and at the same time overcomes the inherent low sensitivity problem. These properties endow SERS with exciting opportunities to be a successful analytical tool for cell analysis. SERS can be used to detect only molecules located on or close to the metallic nanostructures which can support surface plasmon resonances for the enhancement of the Raman signals. Therefore, these metallic nanostructures play a key role in the application of SERS in cell analysis. By incorporating the SERS substrates into the biosamples, molecular structural probing and cellular imaging become possible. In the past decade, analysts worldwide have developed many schemes to study the chemical changes and component distribution in cells by using SERS. We show in this paper how zinc oxide (ZnO)/silver (Ag) composite microspheres can be prepared by the reduction of Ag(NH3)2+ with the reducing agent formaldehyde in aqueous solution on the surface of ZnO microspheres. During the preparation, Sn2+ was absorbed on the surface of ZnO microspheres for sensitization and activation, and then Ag(NH3)2+ was reduced to Ag nanoparticles by the reducing agent to obtain ZnO/Ag composite microspheres. SEM and TEM images revealed silver nanoparticles with a diameter ranging from tens to 100 nm. X‐Ray photoelectron spectra (XPS), X‐ray diffraction (XRD) patterns and UV‐vis spectra were used to characterize the structure of the ZnO/Ag composite microspheres. The origin of the surface‐enhanced Raman scattering properties was traced to the surface of the ZnO/Ag composite microspheres. Ag film over nanosphere (AgFON) substrates for surface‐enhanced Raman spectroscopy (SERS) are shown to be ineffective for the detection of proteins in phosphate buffer solution (PBS) because of the decomposition of the substrate resulting in a total loss of SERS activity. However, modification of these substrates with SiO2 overlayers overcomes this problem. The SiO2 overlayers are produced by filtered arc deposition (FAD) and are characterised by atomic force microscopy (AFM). Their porosity is examined using Raman spectroscopy and the detection of cytochrome c and bovine serum albumin in PBS is successfully demonstrated. Compacted powders of commercially available nano‐ and microparticles of silver were used to successfully induce the surface‐enhanced Raman scattering (SERS) effect in spruce milled‐wood lignin (MWL). For the two silver particle sizes used in this investigation, the spectra were mostly similar. Some general characteristics of the lignin SERS spectrum are described. The SERS technique was found to be sensitive for detecting lignin. Significant spectral changes were present between the SERS and normal Raman spectra of MWL. The SERS spectrum was assigned on the basis of literature‐reported vibrational assignments of lignin and its models. Based on significant changes in Raman features, we propose that the lignin is strongly adsorbed on silver. To determine whether SERS of lignin can be obtained directly from wood without its isolation, Wiley‐milled spruce wood (WMW) adsorbed on silver was studied. The results indicated that not only the surface‐enhancement effect was successfully induced in the WMW, but that its spectrum was similar to MWL SERS. Moreover, for WMW, no signals from the carbohydrate components were observed, and therefore, lignin was detected selectively. This nano‐ and microparticle‐based molecularly specific method is expected to make a significant contribution in identifying and investigating lignin in various lignin‐containing materials. Surface‐enhanced Raman scattering (SERS) spectroscopy was first employed to detect oxyhemoglobin (OxyHb, the common type of hemoglobin) variation in type II diabetic development without using exogenous reagents. Using silver nanoparticles as SERS‐active substrate, high‐quality SERS spectra are obtained from blood OxyHb samples of 49 diabetic patients and 40 healthy volunteers. Tentative assignment of the observed SERS bands indicates specific structural changes of OxyHb molecule in diabetes, including heme transformation and globin variation. Furthermore, partial least squares and principal component analysis combined with linear discriminate analysis diagnostic algorithms are employed to analyze and classify the SERS spectra acquired from diabetic and healthy OxyHb, yielding the diagnostic accuracies of 90.0% and 95.5%, respectively. This exploratory work suggests that the silver nanoparticles‐based OxyHb SERS method in combination with multivariate statistical analysis has great potential for the label‐free detection of type II diabetes. We investigated the interfacial structures of various aromatic (each compound contains one or two phenyls) di‐α‐amino ( L1 – L3 ) and α‐amino‐α‐hydroxyphosphinic ( L4 – L6 ) acids immobilized onto an electrochemically roughened silver electrode surface in an aqueous solution using surface‐enhanced Raman scattering (SERS). These structures were compared to those on a colloidal silver surface to determine the relationship between adsorption strength and geometry. The presence of an enhanced ν19a ring band in the SERS spectra of L6 , L2 , and L3 on the electrode indicated that the benzene rings of those molecules interact with the electrode surface through localized CC bond(s). We observed significant band broadening of the benzene ring modes for all α‐hydroxyphosphinic acids on both substrates, except for L1 deposited onto the electrode surface. This suggests the possibility of direct interaction between the ring and Ag, although the benzene ring–surface π overlap is weaker for the colloidal silver than for the Ag electrode. The downward shift in wavenumber and alternations in the enhancement of a ν12 ring band indicate a general increase of tilt angle on both silver substrates in the order L3 < L4 < L5 < L6 . The altered enhancement of the bands due to the vibrations of the  NH2 and O PO fragments, a finding observed on both silver substrates, strongly suggests that the groups interact with different strength and geometry with these substrates. A simple method is demonstrated to detect DNA at low concentrations on the basis of surface‐enhanced Raman scattering (SERS) via polyvinyl alcohol‐protected silver grasslike patterns (PVA‐Ag GPs) grown on the surface of the common Al substrate. By the SERS measurements of sodium citrate and thymine, the PVA‐Ag GPs are shown to be an excellent SERS substrate with good activity, stability and reproducibility. With the use of the tested molecule of thymine, the enhancement factor of the PVA‐Ag GPs is up to ~1.4 × 108. The PVA‐Ag GPs are also shown to be an excellent SERS substrate with good biocompatibility for DNA detection, and the detection limit is down to ~10−5 mg/g. Meanwhile, the assignations of the Raman bands and the adsorption behaviors of the DNA molecules are also analyzed. In this work, the geometry optimization and the wavenumber analysis of adenine–Ag and guanine–Ag complexes for the ground states are performed using density functional theory, B3LYP functional and the LanL2DZ basis set. The transition energies and the oscillator strengths of adenine–Ag and guanine–Ag for the lowest six singlet excited states were calculated by using the time‐dependent density functional theory method with the same functional and basis set. The results show that the charge transfer in the adenine–Ag and guanine–Ag complexes should be the chemical factor for the SERS of the DNA molecules. Lastly, this method may be employed in large‐scale preparation of substrates that have been widely applied in the Raman analysis of DNA because the fabrication process is simple and inexpensive. Large area (3 × 3 cm2) substrates for surface‐enhanced Raman scattering were fabricated by combining femtosecond laser microstructuring and soft lithography techniques. The fabrication procedure is as follows: (i) femtosecond laser machining is used to create a silicon master copy, (ii) replicates from polydimethylsiloxane are made, and (iii) a 50‐nm‐thick gold film is deposited on the surface of the replicates. The resulting substrates exhibit strongly enhanced absorption in the spectral region of 350 ∼ 1000 nm and generate enhanced Raman signal with enhancement factor of the order of 107 for 10‐ 6 M rhodamine 6G. In this work, the surface‐enhanced Raman scattering (SERS) spectra of seven genomic DNAs from leaves of chrysanthemum (Dendranthema grandiflora Ramat.), common sundew (Drosera rotundifolia L.), edelweiss (Leontopodium alpinum Cass), Epilobium hirsutum L., Hypericum richeri ssp. transsilvanicum (Čelak) Ciocârlan, rose (Rosa x hybrida L.) and redwood (Sequoia sempervirens D. Don. Endl.), respectively, have been analyzed in the wavenumber range 200–1800 cm−1. The surface‐enhanced Raman vibrational modes for each of these cases, spectroscopic band assignments and structural interpretations of genomic DNAs are reported. A high molecular structural information content can be found in the SERS spectra of these DNAs from leaf tissues. Based on this work, specific plant DNA–ligand interactions or accurate local structure of DNA might be further investigated using surface‐enhanced Raman spectroscopy. We report here, for the first time, a simple method to prepare size‐controllable Au nanoparticles (NPs) in aqueous solutions from bulk Au substrates. First, chitosan (Ch)‐capped Au‐containing complexes were prepared by electrochemical oxidation–reduction cycles in 0.1 N NaCl and 1 g/l Ch solutions. Then the solutions were heated from room temperature to boiling at different heating rates to synthesize size‐controllable Au NPs. The particle sizes of the prepared Au(111) NPs could be controlled from 5 to 30 nm with an increase of the heating rate during preparation. Experimental results indicate that the prepared Au(111) NPs with diameters ranging from 10 to 30 nm can serve as surface‐enhanced Raman scattering active probes for molecules of rhodamine 6G. Carbendazim (MBC) is a fungicide widely used in agriculture, and there are serious concerns regarding the health risks that could be caused by this fungicide. Here, we explore its ultrasensitive detection by surface‐enhanced Raman scattering (SERS). First, to obtain maximum SERS signal, the adsorption of the target molecule onto metallic surface is essential. Therefore, we study the adsorption of the MBC onto the nanoparticle surface by SERS under different experimental conditions, such as different synthesis methods of nanoparticle, variable excitation wavelength, and fungicide concentration with the aim to detect MBC at low concentrations. Experiments are carried out with three kinds of colloidal nanoparticles: Ag and Au reduced by citrate and Ag reduced by hydroxylamine. However, mainly Ag colloids are highly efficient in the SERS detection of MBC. In addition, theoretical calculations of MBC Raman spectrum and that of the surface complex are used to help with the understanding the mechanisms responsible for the interaction between MBC and Ag. In this work, we use electrochemical oxidation–reduction cycles (ORC) methods to prepare surface‐enhanced Raman scattering (SERS)‐active gold substrates modified with SiO2 nanoparticles to improve the corresponding SERS performances. Based on the modified substrates, the SERS of Rhodamine 6G (R6G) exhibits a higher intensity by 3‐fold of magnitude, as compared with that of R6G adsorbed on a SERS‐active Au substrate without the modification of SiO2 nanoparticles. Moreover, the SERS enhancement capabilities of the modified and the unmodified Au substrates are seriously destroyed at temperatures higher than 250 and 200 °C, respectively. These results indicate that the modification of SiO2 nanoparticles can improve the thermal stability of SERS‐active substrates. The aging in SERS intensity is also depressed on this modified Au substrate due to the contribution of SiO2 nanoparticles to SERS effects. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

We created and studied a novel nanoprobe for spectroscopic molecular imaging of the epidermal growth factor (EGF) receptor, whose over‐expression is a hallmark of a wide range of cancers. Silver nanoparticles (AgNPs) of 45 nm diameter were synthesized and coupled to EGF by α‐lipoic acid, a short ligand that exhibits excellent silver binding affinity. Time‐of‐flight mass spectroscopy demonstrates formation of the protein complex. Enzyme‐linked immunosorbent assay verifies the protein complex is 100% active for the EGF receptor, alone and, following conjugation to silver nanoparticles. Compared with its monosulfide analog, 6‐mercaptohexanoic acid, α‐lipoic acid is stabilized by binding to silver with a total energy that is lower by 1.38 eV, as found from Density Functional Theory (DFT)/natural bond analysis calculations. A Highest Occupied Molecular Orbital (HOMO)‐Lowest Unoccupied Molecular Orbital (LUMO) gap energy of 5.25 (spin‐up electrons) and 5.74 eV (spin‐down electrons) was obtained for the silver‐α‐lipoic acid complex. This is the first report of silver nanoparticles being attached to EGF, and the first theoretical and experimental report on the surface enhanced Raman spectroscopy spectral interpretation of α‐lipoic acid bound to silver. These nanoprobes exhibit surface enhanced Raman spectroscopy, when aggregated in solution, at picomolar concentrations and have the necessary properties – specificity, sensitivity and stability – to serve as molecular imaging agents. Fourier‐transform infrared (FT‐IR), Raman (RS), and surface‐enhanced Raman scattering (SERS) spectra of β‐hydroxy‐β‐methylobutanoic acid (HMB), L ‐carnitine, and N‐methylglycocyamine (creatine) have been measured. The SERS spectra have been taken from species adsorbed on a colloidal silver surface. The respective FT‐IR and RS band assignments (solid‐state samples) based on the literature data have been proposed. The strongest absorptions in the FT‐IR spectrum of creatine are observed at 1398, 1615, and 1699 cm−1, which are due to νs(COOH) + ν(CN) + δ(CN), ρs(NH2), and ν(C O) modes, respectively, whereas those of L ‐carnitine (at 1396/1586 cm−1 and 1480 cm−1) and HMB (at 1405/1555/1585 cm−1 and 1437–1473 cm−1) are associated with carboxyl and methyl/methylene group vibrations, respectively. On the other hand, the strongest bands in the RS spectrum of HMB observed at 748/1442/1462 cm−1 and 1408 cm−1 are due to methyl/methylene deformations and carboxyl group vibrations, respectively. The strongest Raman band of creatine at 831 cm−1w(R NH2)) is accompanied by two weaker bands at 1054 and 1397 cm−1 due to ν(CN) + ν(R NH2) and νs(COOH) + ν(CN) + δ(CN) modes, respectively. In the case of L ‐carnitine, its RS spectrum is dominated by bands at 772 and 1461 cm−1 assigned to ρr(CH2) and δ(CH3), respectively. The analysis of the SERS spectra shows that HMB interacts with the silver surface mainly through the  COO, hydroxyl, and  CH2 groups, whereas L ‐carnitine binds to the surface via  COO and  N+(CH3)3 which is rarely enhanced at pH = 8.3. On the other hand, it seems that creatine binds weakly to the silver surface mainly by  NH2, and C O from the  COO group. Copyright © 2006 John Wiley & Sons, Ltd.  相似文献
